Identification of Sex-specific Expression Markers in the Giant Tiger Shrimp (Penaeus monodon)

  • Bulked segregant analysis (BSA) and AFLP were used for isolation of genomic sex determination markers in Penaeus monodon. A total of 256 primer combinations were tested against 6-10 bulked genomic DNA of P. monodon. Five and one candidate female- and male-specific AFLP fragments were identified. Female-specific fragments were cloned and further characterized. SCAR markers derived from FE10M9520, FE10M10725.1, FE10M10725.2 and FE14M16340 provided the positive amplification product in both male and female P. monodon. Further analysis of these markers using SSCP and genome walk analysis indicated that they were not sex-linked. In addition, sex-specific (or differential) expression markers in ovaries and testes of P. monodon were analyzed by RAP-PCR (150 primer combinations). Twenty-one and fourteen RAP-PCR fragments specifically/differentially expressed in ovaries and testes of P. monodon were successfully cloned and sequenced. Expression patterns of 25 transcripts were tested against the first stranded cDNA of ovaries and testes of 3-month-old and broodstock-sized P. monodon (N = 5 and N = 7 - 10 for females and N = 4 and N = 5 - 7 for males, respectively). Five (FI-4, FI-44, FIII-4, FIII-39 and FIII-58) and two (M457-A01 and MII-51) derived RAP-PCR markers revealed female- and male-specific expression patterns in P. monodon. Surprisingly, MII-5 originally found in testes showed a higher expression level in ovaries than did testes of juvenile shrimps but a temporal female-specific pattern in P. monodon adults.

  • Eye growth and lens diameter of brown croaker Miichthys miiuy were positively allometric between hatching and 180 days post-hatch (d.p.h.). Eye growth in relation to head length and head height was nearly isometric. Eyes were formed completely at 14 d.p.h. At this age, the eye has a crystalline lens, an optic nerve fiber layer, a ganglion cell layer, an inner plexiform layer, an inner nuclear layer, an outer plexiform layer, an outer nuclear layer, an outer limiting membrane, and a pigment epithelium. The essential demands that must be met by the retina in this species pertain to light sensitivity and spatial resolution.

  • Biomass and community composition of microphytobentos in tidal flats were studied by HPLC analysis and also investigated to examine the relationship between microphytobenthic pigments and Adenosine-5' triphosphate (ATP) as an index of total microbial biomass in intertidal environments (muddy and sandy sediment) of Gyeonggi Bay, west coast of Korea. Microphytobenthic pigments and ATP concentration in muddy sediment were the highest at the surface while the biomass of microphytobenthos in sandy sediment was the highest at the sub-surface (0.75 cm sediment depth). The detected pigments of microphytobenthos were chlorophyll a, b (euglenophytes), $c_3$, peridinin (dinoflagellates), fucoxanthin (diatom or chrysophytes), diadinoxanthin, alloxanthin (cryptophytes), diatoxanthin, zeaxanthin (cyanobacteria), ${\beta}$-carotein, and pheophytin a (the degraded product of chlorophyll a). Among the pigments which were detected, the concentration of fucoxanthin was the highest, indicating that diatoms dominated in the microphytobenthic community of the tidal flats. There was little significant correlation between OC (Organic Carbon) and ATP in both sediments. However, a positive correlation between chlorophyll a concentration and ATP concentration was found in sandy sediment, suggesting that microbial biomass could be affected by labile OC derived from microphytobenthos. These results provide information that may help us understand the relationship between microphytobenthos and microbial biomass in different intertidal sediment environments.

동해 연안에 출현하는 도루묵(Arctoscopus japonicus)의 위내용물 조성 (Diet composition of the sandfish, Arctoscopus japonicus in the coastal waters of East Sea, Korea)

  • The diet composition of Arctoscopus japonicus was studied using 448 specimens collected in the coastal waters of East Sea, Korea. The size of the specimens ranged from 6.2-25.3 cm in total length (TL). A. japonicus were fed mainly on amphipods that constituted 76.1% in IRI. Euphausiids were the second largest prey component. The diets also include small quantities of crabs, copepods, mysids, cephalopods, monogeneans, and fishes. A. japonicus showed ontogenetic changes in feeding habits. Smaller individuals (6-10 cm, TL) fed mainly on amphipods. The proportion of amphipods decreased as body size increased, whereas the consumption of euphausiids gradually increased (10-14 cm, TL). As the body size more increased, the proportion of amphipods increased whereas euphausiids decreased (14-26 cm, TL).

ON ANALYTICAL SOLUTION OF NON LINEAR ROLL EQUATION OF SHIPS

  • Out of all types of motions the critical motions leading to capsize is roll. The dynamic amplification in case of roll motion may be large for ships as roll natural frequency generally falls within the frequency range of wave energy spectrum typical used for estimation of motion spectrum. Roll motion is highly non-linear in nature. Den are various representations of non-linear damping and restoring available in literature. In this paper an uncoupled non-linear roll equations with three representation of damping and cubic restoring term is solved using a perturbation technique. Damping moment representations are linear plus quadratic velocity damping, angle dependant damping and linear plus cubic velocity dependant damping. Numerical value of linear damping coefficient is almost same for all types but non-linear damping is different. Linear and non-linear damping coefficients are obtained form free roll decay tests. External rolling moment is assumed as deterministic with sinusoidal form. Maximum roll amplitude of non-linear roll equation with various representations of damping is calculated using analytical procedure and compared with experimental results, which are obtained form forced tests in regular waves by varying frequency with three wave heights. Experiments indicate influence of non-linearity at resonance frequency. Both experiment and analytical results indicates increase in maximum roll amplitude with wave slope at resonance. Analytical results are compared with experiment results which indicate maximum roll amplitude analytically obtained with angle dependent and cubic velocity damping are equal and difference from experiments with these damping are less compared to non-linear equation with quadratic velocity damping.

First Record of the Giant Grouper Epinephelus lanceolatus (Perciformes: Serranidae: Epinephelinae) from Jeju Island, South Korea

  • A single specimen of Epinephelus lanceolatus, belonging to Epinephelinae of the family Serranidae, was newly collected from Jeju Island, South Korea, in December 2009. Epinephelus lanceolatus is characterized by anterior lateral-line scales with branched tubules, cycloid scales, irregular white spots, and fins with numerous small black spots. We propose a Korean name, "Dae-wang-ba-ri' for this species.

  • Potassium N-heterobiaryltrifluoroborates were successfully prepared via a selective Cu-catalyzed C-N coupling reaction. The $BF_3K$ moiety was well tolerated under the reaction conditions involving CuI and dimethyl-ethylenediamine (DMEDA) in the presence of DMSO. The Pd-catalyzed Suzuki-Miyaura cross couplings of potassium N-heterobiaryltrifluoroborates with bromoarenes were studied to prepare the N-heterotriaryl compounds. Moreover, homocoupling, iodination, and hydroxylation of potassium N-heterobiaryltrifluoroborates provided the corresponding products in high yields.

Simulation of a two-stroke diesel engine for propulsion in waves

  • Propulsion in waves is a complex physical process that involves interactions between a hull, a propeller, a shaft and a prime mover which is often a diesel engine. Among the relevant components, the diesel engine plays an important role in the overall system dynamics. Therefore, using a proper model for the diesel engine is essential to achieve the reasonable accuracy of the transient simulation of the entire system. In this paper, a simulation model of a propulsion system in waves is presented with emphasis on modeling a two-stroke marine diesel engine: the framework for building such a model and its mathematical descriptions. The models are validated against available measurement data, and a sensitivity analysis for the transient performance of the diesel engine is carried out. Finally, the results of the system simulations under various wave conditions are analyzed to understand the physical processes and compare the efficiency for different cases.
