Chromatographic Fractionation of Protease Inhibitors from Fish Eggs (어류 알로부터 Protease Inhibitors의 크로마토그래피법에 의한 분획)

  • A protease inhibitor from fish eggs was fractionated using chromatographic methods. The fractionation efficiency was evaluated in terms of specific inhibitory activity (SIA, U/mg), purity (fold), total inhibitory activity (TIA, U), and recovery (%). The protease inhibitor (PI) from egg extracts of skipjack tuna (ST Katsuwonus pelamis), yellowfin tuna (YT Thunnus albacares) and Alaska pollock (AP Theragra chalcogramma) was fractionated using Sephadex G-50 gel filtration and DEAE-Sepharose CL-6B anion exchange chromatography based on protein size exclusion and net charge, respectively. Fractions exhibiting strong inhibitory activity were contained in the 30-50 kDa fraction on gel filtration and in the range of 0.4-0.7 M NaCl gradient fraction on anion exchange chromatography. The respective TIA and percent recovery of the fraction obtained with gel filtration toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 2,758.7 U and 29.6% for ST, 1,005.5 U and 25.6% for YT, and 1,267.5 U and 26.0% for AP. Gel filtration chromatography was more effective at fractionating PI than using ion exchange chromatography. These results suggest that fish eggs act as serine protease inhibitors and might be useful for protease inhibition in foodstuffs.

Debittering of Enzymatic Hydrolysate Using Exopeptidase Active Fractions from the Argentina Shortfin Squid Illex argentinus Hepatopancreas (원양산 오징어(Illex argentinus) 간췌장 유래 Exopeptidase 분획물의 쓴맛개선 효과)

  • Exopeptidase active fractions from the hepatopancreas of the Argentina shortfin squid Illex argentinus, were obtained with acetone (AC 30-40%), ammonium sulfate (AS 60-70% saturation), anion exchange chromatography (AE-II, 0.2 M NaCl) and gel filtration chromatography (GF-I, 30-50 kDa) fractionation methods. A bitter peptide solution that has a bitterness equivalent to that of 2% glycylphenylalanine and prepared by tryptic hydrolysis of milk casein, was treated with the exopeptidase active fractions. The GF-I fraction was the best based on aminopeptidase activity (35.3 U/mg), percentage of recovery (30.7%) and a sensory evaluation (1.7). The amount of released amino acids increased as incubation time increased, and the bitterness of the enzyme reaction mixtures decreased. Incubation with the GF-I fraction for 24 h resulted in the hydrolysis of several peptides as revealed by the reverse-phase high performance liguid chromatography profile, with three peaks (3, 5 and 6) decreasing in area (%) and three peaks (1, 2 and 4) increasing in area (%). Therefore, the GF-I fraction appeared to be ideally suited to reduce bitterness in protein hydrolysates by catalyzing the hydrolysis of bitter peptides.

  • In order to aid the researchers who analyze the biotoxins in marine molluscs, this paper introduces the official names and ecological characteristics of the domestically produced or distributed bivalve and gastropods that can be contaminated with marine biotoxins. Also, the paper intends to inform the location and scientific name of each organ of the representative group of species through anatomical maps. In the future, it is necessary to standardize and normalize the names of the species and the research institutions in food codes, scholarly papers and reports on the marine biotoxin analysis.

Evaluation of Life Time for Anti-Corrosive Methods for Marine Steel Sheet by Cyclic Corrosion Test (실내 가속부식시험을 통한 해양 강관합성 말뚝의 방식 기법 수명 평가)

  • When a steel sheet pipe applied to marine environment, an anti-corrosive coating should be treated to obtain long-term life-time for steels, especially, splash zone. Although anti-corrosive property of coatings is required to be tested in real marine environment, it is difficult because of long test time such as 20 years or more time. Therefore, we used cyclic corrosion tester in a laboratory, which has similar conditions with salt-dry-wet process such as real marine environment. Anti-corrosive properties of the coatings and two steels were tested their anti-corrosive properties under cyclic corrosion test conditions(KS D ISO 14993) and the results were compared with estimate life-time in real marine environment. According to cyclic corrosion test, accelerated corrosive factor of each anti-corrosive coating was investigated accelerated corrosive factor from impedance with EIS method. Accelerated corrosive factor of type SS400 carbon steel and A690 was also investigated their accelerated corrosive factor from the regression curves of weigh loss results. One of the anti-corrosive coatings showed about 50 years life-time compared with standard sample life-time. Carbon steel SS400 showed from 0.1 mm/yr to 0.06 mm/yr as its corrosion rate.

Economic Feasibility Analysis of Marine Debris Pollution Abatement Technology Program (해양쓰레기 오염대응 기술개발사업의 경제적 타당성 분석)

  • The Korean government is considering the implementation of the marine debris pollution abatement technology program (MDPATP) to mitigate the negative impacts of marine debris and systematically manage marine debris through scientific researches such as monitoring and environmental impact assessment of marine debris. In this regard, this study attempts to analyze the economic feasibility of the MDPATP in order to provide policy-maker with useful information. To this end, the indices for economic feasibility such as net present value (NPV), benefit/cost (B/C) ratio, and internal rate of return (IRR) are presented. The results show that NPV, B/C ratio, and IRR are computed to be 45.7 billion won, 2.72, and 17.12%, respectively, which are bigger than 0, 1.0, and 5.5%, and that the MDPATP passes the cost-benefit analysis. Thus, it is concluded that it is socially profitable to conduct the MDPATP.

Optimum Sterilization Conditions and Quality Characteristics of the Retort-Sterilized Crab Analog (레토르트 Crab Analog의 살균 최적화 및 품질 특성)

  • This study developed a normal temperature-circulatable retort-sterilized crab analog (RSCA) with a long shelf-life by retort sterilization. We examined the optimum sterilization conditions and quality characteristics of the RSCA. A central composite design for response surface methodology (RSM) was adopted to optimize of the retort sterilization. The central composite design consisted of 11 samples: four factorial points, four star points, and three central points. The sterilization temperature and $F_0$-value for retort sterilization were the independent variables, and the shearing force, whiteness, and sensory score for texture were the dependent variable. The RSM analysis of the multiple response optimization for the RSCA using the Minitap statistical programing gave an $F_0$-value of 3.3 min. at $117.5^{\circ}C$. The RSM analysis also indicated that the sterilization temperature during retort sterilization was the most influential factor, while the $F_0$-value had little effect on the quality of the RSCA. The moisture, pH and volatile basic nitrogen contents of the RSCA product were 73.1%, 7.3 and 17.2 mg/100 g, respectively, and viable cells count was >18 CFU/g. And sensory quality characteristics of this RSCA was similar to those of a commercial crab analog.

A Study on Mine Localization of Forward Looking Sonar Considering the Effect of Underwater Sound Refraction (수중 음파 굴절효과를 고려한 전방주시소나 기뢰 위치 추정기법 연구)

  • Mine detection has been mainly studied with images of the forward-looking sonar. Forward-looking sonar assumes the propagation path of the sound wave as a straight path, creating the surrounding images. This might lead to errors in the detection by ignoring the refraction of the sound wave. In this study, we propose a mine localization method that can robustly identify the location of mines in an underwater environment by considering the refraction of sound waves. We propose a method of estimating the elevation angle of arrival of the target echo signal in a single receiver, and estimate the mine location by applying the estimated elevation angle of arrival to ray tracing. As a result of simulation, the method proposed in this paper was more effective in estimating the mine localization than the existing method that assumed the propagation path as a straight line.

Characteristics of Films Based on Chitosans Isolated from Todarodes pacificus

  • Chitosans were obtained with varying deacetylation times using the ${\beta}$-chitin isolated from Todarodes pacificus, and their deacetylation degrees and average molecular weights were determined. Films prepared with the squid chitosans were characterized by estimating their tensile strengths, percent elongations, water vapor permeabilities, degree of swelling, and temperatures of glass transition and thermal decomposition. The results suggest that the squid chitosan films were comparable to common crustacean chitosan films in regard of mechanical, moisture barrier, and thermal properties, although further, multilateral investigations are necessary to make a more definitive conclusion.

  • This study was conducted to optimize the processing of high quality surimi gel from unmarketable cultured bastard halibut Paralichthys olivaceus. According to endogenous enzyme activity and processing optimization, high quality surimi gel from unmarketable cultured bastard halibut was prepared by mixing 3.0% (w/w) salt, 2.4% (w/w) starch, 5.0% (w/w) egg white and 4.8% (w/w) ice water in a Stephan mixer, set at $5^{\circ}C$ for 24 h, followed by boiling for 30 min, and finally cooling for 30 min. The strength of the surimi gel from unmarketable cultured bastard halibut prepared by the above processing method was $1,257\;g{\times}cm$, which was 33% higher than that of a commercial surimi gel from Alaska pollock, grade SA.
