The Characteristics of Spatio-temporal Distribution on Environmental Factors After Construction of Artificial Structure in the Nakdong River Estuary (인공시설물 건설 이후 낙동강 하구 환경인자의 시·공간적분포특성)

  • Nakdong River Estuary is affected by the dam, barrage construction and dredge and reclaim worked artificially. So, we have studied the area input both freshwater and sea water to understand marine environment of Nakdong River estuary related to the effect of artificial work from 2013 to 2015. As a result, The discharge flow to the estuary remarkably decreased before barrage construction and the average of salinity at the estuary increased. So, the brackish water zone reduced under the influence of decreased discharge flow. The major sources of nitrate and silicate were freshwater, phosphate supplied from bottom and the open sea water. The concentration of phosphate and dissolved oxygen (DO) decreased remarkably in spring and summer. we investigated that phosphate in freshwater was removed under the influence of the estuary dam and phosphate in sea water was removed under the influence of phytoplankton. The low concentration of DO was due to decomposition of the organic compound by microorganism after phytoplankton blooms. Generally, the concentrations of chlorophyll-a in summer was higher than spring and fall. Therefore, the change of ecosystem in Nakdong river estuary was due to decrease of freshwater influx, the other change is facing because of the barrage.

Survey of the level of Microbial Contamination in Fish Farms on the Jeju-Island (제주도 어류양식장에 대한 미생물학적 오염도 조사)

  • The purpose of this study was to monitor and compare the contamination levels of heterotrophic bacteria and pathogenic bacteria (total coliforms, Escherichia coli O157:H7, Salmonella spp., Vibrio parachaemolyticus, Staphylococcus aureus and Bacillus cereus). The samples collected at fish farms located in the Jeju-island were investigated from March to August in 2006. The contamination levels of S. aureus, B. cereus and Salmonella spp. ranged $0-3.3{\times}10^2 CFU/ml,\;0-2.2{\times}10^2 CFU/ml\;and\;0-5.1{\times}10^1CFU/ml$, respectively, and 85% of moist pellet feeds were contaminated with S. aureus, B. cereus and total coliforms. But total coliforms and E. coli O157:H7 were not detected in oliver flounder (Paralichthys olivaceus). For the production of safe oliver flounder (Paralichthys olivaceus), education of sanitation for employees, control of fish feed, and continuous monitoring for microorganism will be required.

Studies on Proteolytic and Fibrinolytic Activity of Bacillus subtilis JM-3 Isolated from Anchovy Sauce (멸치액젓으로부터 분리한 Bacillus subtilis JM-3의 단백질 분해활성과 혈전 용해 활성에 관한 연구)

  • This study was performed to search for potential microorganism that has rapid fermenting and physiological function from anchovy sauce. We isolated three bacterial strains, JM-1, JM-2, and JM-3 with proteolytic and fibrinolytic activity from anchovy sauce. Among the 3 bacterial strains, JM-3 showed the strongest proteolytic and fibrinolytic activity. Bacterial strain JM-3 was gram-positive rod, motile and formed endospore. The 16S rRNA of bacterial strain JM-3 was amplified by PCR and then its sequence was determined by ABI 310 genetic analyzer. The 16S rRNA sequence of bacterial strain JM-3 was compared to BLAST DNA database and identified to Bacillus subtilis with 99% of homology. The optimum temperature, pH and NaCl concentration for growth of B. subtilis JM-3 were $40^{\circ}C$, 5.0 and 0%, respectively. The optimum temperature, pH and NaCl concentration for proteolytic and fibrinolytic enzyme production of B. subtilis JM-3 were same as optimum conditions for growth. At 20% of NaCl concentration which is common NaCl concentration of fish sauce, B. subtilis JM-3 showed about 60% of proteolytic and fibrinolytic activity of 0% NaCl concentration. From above results, we found that B. subtilis JM-3 will be able to used for starter of functional fish sauce.

A Study on Microorganism Dominant Species in Bench-scale Shipboard STP Using Combined SBR and MBR Process (SBR 및 MBR 복합공정을 적용한 Bench-scale Shipboard STP에서의 미생물 우점종에 관한 연구)

  • International Maritime Organization (IMO) is one of the most effective organizations in evolving international law for the protection and conservation of the marine environment. The IMO, MARPOL(Marine Pollution) 73/78 contains six Annexes that provide an overarching framework for the objectives of the international marine pollution. Annex IV was regulated by 64 th resolution in 2012 to control sea pollution from sewage. In 2014 large-scale wastewater treatment and nutrient removal device was developed with a grant from the Ministry of Oceans and Fisheries. A combined new process of Sequence Batch Reactor (SBR) and Membrane Bioreactor(MBR) was developed to overcome the pollution caused by shipboard sewage. In the present study, shipboard sewage wastewater was treated by mixing and aeration cycle in the newly developed SBR process. Furthermore, during analysis by NGS technique(Macrogen Co., Ltd.), dominant species of bacteria were found in the aeration tank of the Bench-scale wastewater treatment facility. Bacteroidetes and Gammaproteobacteria accounted for 27.1 % of the aerobicbacteria and 16.8 % of the anaerobicbacteria, respectively. Microorganisms play a vital role in shipboard wastewater treatment. A further detailed study is required to understand the precise role of the microorganisms in the wastewater treatment.

Characterization of Miniimonas sp. S16 isolated from activated sludge (활성슬러지로부터 분리된 Miniimons sp. S16 세균의 특성)

  • Biological factors (e.g. microorganism activity) in wastewater treatment plant (WWTP) play essential roles for degradation and/or removal of organic matters. In this study, to understand the microbial functional roles in WWTP, we tried to isolate and characterize a bacterial strain from activated sludge sample. Strain S16 was isolated from the activated sludge of a municipal WWTP in Daejeon metropolitan city, the Republic of Korea. The cells were a Gram-stain-positive, non-motile, facultative anaerobe, and rod-shaped. Strain S16 grew at a temperature of $15{\sim}40^{\circ}C$ (optimum, $30^{\circ}C$), with 0~9.0% (w/v) NaCl (optimum, 1.0~2.0%), and at pH 5.5~9.0 (optimum, pH 7.0~7.5). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain S16 was most closely related to the unique species Miniimonas arenae NBRC $106267^T$ (99.79%, 16S rRNA gene sequence similarity) of the genus Miniimonas. The cell wall contained alanine, glutamic acid, serine, and ornithine. Although the isolation source of the type strain NBRC $106267^T$ which considered as a marine microorganism is sea sand, that of strain S16 is terrestrial environment. It might raise an ecological question for habitat transition. Therefore, comparative genome analysis will be valuable investigation for shedding light on their potential metabolic traits and genomic streamlining.

Antibiotic Susceptibility of Vibrio spp. Isolated from West Sea (서해안에서 분리한 비브리오균의 항생제 감수성 특성)

  • Bacteria of genus Vibrio are Gram-negative, curved, halophilic, nonspore-forming bacteria, autochthonous inhabitans of the marine and estuarine environments. Some of the Vibrio species such as V. parahaemolyticus, V. vulnificus, and V. cholerae are associated with human disease. Each year many people have been suffering from food-borne disease caused by the ingestion of seafood. In this study, we have monitored antibiotic resistance of this microorganism in 6 coastal areas of West Sea by sampling shellfish monthly. Vibrio spp. were detected from 23.3% of 120 samples analyzed using TCBS agar plates as well as API 20E kit. Among 16 antibiotics tested, resistance to vancomycin and ampicillin was observed in 82.1% of the isolates, and Vibrio spp. resistant to rifampin (71.4%) and cephalothin (53.6%) were also high. Most of the isolates were sensitive to chloramphenicol (92.9%), sulfamethoxazole/trimethoprim (92.9%), and tetracycline (96.4%). About 71.4% of the isolates showed multiple drug resistance toward 3 antibiotics including vancomycin and ampicillin.

Effects of Pressure and Dissolved Oxygen Concentration on the Activated Sludge (압력 및 용존산소 농도가 활성슬러지에 미치는 영향)

  • This study was conducted to evaluate the effects of pressure and dissolved oxygen concentration on the activated slut비e and to determine the optimum depth of deep shaft process. Some results from this study were summarized as follows. 1. It is considered that low sludge product in the activated sludge system maintaining high dissolved oxygen concentration is attributed to the increase of endogeneous respiration rate caused by the increase of aerobic zone in the sludge floe. 2. The increase of dissolved oxygen concentration does not affect to the increase of organic removal efficiency greatly and therefore the limiting factor is the substrate transfer into the inner part of floe. 3. The yield coefficient, Y is decreased in proportion to the increase of oxygen concentration. In this study, Y values arre ranged from 0.70 to 0.41 according to the variation of dissolved oxygen concentration from 18.0mg/$\ell$ to 258 mg/$\ell$. 4. The optimum depth of deep shaft process should be determined within the limits of non-toxicity to the microorganism and it is about loom in this study.

A Novel pH-Stable, Bifunctional Xylanase Isolated from a Deep-Sea Microorganism, Demequina sp. JK4

  • A genomic library was constructed to clone a xylanase gene (Mxyn10) from Demequina sp. JK4 isolated from a deep sea. Mxyn10 encoded a 471 residue protein with a calculated molecular mass of 49 kDa. This protein showed the highest sequence identity (70%) with the xylanase from Streptomyces lividans. Mxyn10 contains a catalytic domain that belongs to the glycoside hydrolase family 10 (GH10) and a carbohydrate-binding module (CBM) belonging to family 2. The optimum pH and temperature for enzymatic activity were pH 5.5 and $55^{\circ}C$, respectively. Mxyn10 exhibited good pH stability, remaining stable after treatment with buffers ranging from pH 3.5 to 10.0. The protein was not significantly affected by a variety of chemical reagents, including some compounds that usually inhibit the activity of other related enzymes. In addition, Mxyn10 showed activity on cellulose. These properties mark Mxyn10 as a potential enzyme for industrial application and saccharification processes essential for bioethanol production.

Preparation of Calcium Peroxide Originated from Oyster Shell Powder and Oxygen Releasing Ability (패각 분말기반 과산화칼슘 제조와 산소 유리 특성)

  • Bioremediation in situ is heavily dependent on the oxygenic environment which would privide the dwelling microorganism with sufficient oxygen. The situation could be easily resolved with supply of an Oxygen Releasing Compound (ORC). In this paper we prepared that sort of material out of oyster shell powder (mostly calcium carbonate) that prevails every shore areas of the country. We used two different oxidizing methods in the first step of the whole manufacturing process-conventional heating in a furnace and an ultrasound generator to obtain calcium oxide. Then that calcium oxide was further oxidized into calcium peroxide which may release oxygen under a moisturized condition. The oxygen releasing experiments were run to test the performance of our products, and to determine the gas kinetics during the experiments. Interestingly, calcium peroxide derived from ultrasound treatment was much more energy-effective as ORC than that from furnace heating although the heat derived process was better than that of ultrasound in terms of oxygen content and its releasing rate. We also found that most of the data collected from the gas releasing experiments fairly supported an ordinary $1^{st}$ order kinetics to oxygen concentration, which shaped a sharp discharge of oxygen at the very early moment of each test.

Polymer coating for controlled release of biostimulants from Biostimulant balls (생물활성촉진제의 용출율 제어를 위한 폴리머 코팅)

  • We prepared biostimulant balls using sea sediment mixed with biostimulants viz acetate, nitrate and sulfate. The Biostimulant balls were coated with Cellulose Acetate (CA) and Polysulfone (PS) to control the release of the biostimulants. SEM images showed that CA coating was porous and irregular in the inside and very uniform and tight like beehive while PS coating was the same in the inside and outside and not porous. Biostimulants release was found to be high in sea water compared to distilled water. The release of nitrate was higher compared to sulfate. In turbulent environment the release of bionutrients was 50% higher than static environment.
