An Acetophenone Derivative, Clavatol, and a Benzodiazepine Alkaloid, Circumdatin A, from the Marine-Derived Fungus Cladosporium

  • The crude extract of the mycelium of Cladosporium was found to exhibit antimicrobial activity against the Staphylococcus aureus, methicillin-resistant S. aureus, and multidrug-resistant S. aureus. Bioassayguided fractionation of an organic extract led to the isolation of an acetophenone derivative, clavatol (2',4'-dihydroxy-3',5'-dimethylacetophenone) (1), and a benzodiazepine alkaloid, circumdatin A (2). Compound 1 showed moderate antibacterial activity against S. aureus, methicillin-resistant S. aureus, and multidrug-resistant S. aureus with minimum inhibitory concentration (MIC) values of 62.5, 62.5, 31.0 $\mu$g/mL, respectively, but compound 2 was inactive. Compounds 1 and 2 exhibited UV-A protection activity with ED$_{50}$ values of 227.0 and 82.0 $\mu$M, respectively, indicating that they were more potent than the positive control, oxybenzone (ED$_{50}$ 350 $\mu$M), a common sunscreen agent.

  • Ascophyllum nodosum (L.) Le Jolis has a systemic infection with the ascomycete Mycophycias ascophylli (Cotton) Kohlmeyer and Volkmann-Kohlmeyer with which it establishese a mutualistic symbiosis. In addition, A. nodosum is the host for the obligate red algal epiphyte, Vertebrata lanosa (L.) Christensen. Using light and electron microscopy we describe morphological and cytochemical changes occurring as a consequence of rhizoid penetration of V. lanosa into cortical host tissue. Rhizoids induce localized cell necrosis based on physical damage during rhizoid penetration. Host cells adjacent to the rhizoid selectively undergo a hypersensitive reaction in which they become darkly pigmented and become foci for hyphal development. Light and electron microscopy show that M. ascophylli forms dense hyphal aggregations on the surface of the V. lanosa rhizoid and extensive endophytic hyphal growths in the rhizoid wall. This is the first morphological evidence of an interaction between M. ascophylli and V. lanosa. We speculate that M. ascophylli may be interacting with V. lanosa to limit tissue damage to their shared host. In addition, the fungus provides a potential pathway for the transfer of materials (e.g., nutrients and photosynthate) between the two phototrophs.

Ralstonia pickettii Enhance the DDT Biodegradation by Pleurotus eryngii

  • DDT is a hydrophobic organic pollutant, which can be bio-accumulated in nature and have adverse consequences on the physical condition of humans and animals. This study investigated the relationship between the white-rot fungus Pleurotus eryngii and biosurfactant-producing bacterium Ralstonia pickettii associated with the degradation of DDT. The effects of R. pickettii on fungal development were examined using in vitro confrontation assay on a potato dextrose agar (PDA) medium. R. pickettii culture was added to the P. eryngii culture at 1, 3, 5, 7, and 10 ml ($1ml{\approx}1.44{\times}10^{13}CFU$). After 7 d incubation, about 43% of the initial DDT ($12.5{\mu}M$) was degraded by the P. eryngii culture only. The augmentation of 7 ml of R. pickettii culture revealed a more highly optimized synergism with DDT degradation being approximately 78% and the ratio of optimization 1.06. According to the confrontational assay, R. pickettii promoted the growth of P. eryngii towards the bacterial colony, with no direct contact between the bacterial cells and mycelium (0.71 cm/day). DDD (1,1-dichloro-2,2-bis(4-chlorophenyl) ethane), DDE (1,1-dichloro-2,2-bis(4-chlorophenyl) ethylene), and DDMU (1-chloro-2,2-bis(4-chlorophenyl) ethylene) were identified as metabolic products, indicating that the R. pickettii could enhance the DDT biodegradation by P. eryngii.

Aspergillus fumigatus-derived demethoxyfumitremorgin C inhibits proliferation of PC3 human prostate cancer cells through p53/p21-dependent G1 arrest and apoptosis induction

  • Human prostate cancer is the second most frequently diagnosed cancer worldwide, and its incidence rate continues to increase. Advanced prostate cancer is more difficult to treat than early forms due to its chemotherapy resistance. There is need for more effective agents that can inhibit the progression of advanced prostate cancer. Demethoxyfumitremorgin C (DMFTC) was isolated from the fermentation extract of the marine fungus Aspergillus fumigatus. Antiproliferative activity of DMFTC against human prostate cancer PC3 cells was examined through cell cycle analysis by flow cytometry, the fluorescent nuclear imaging analysis with propidium iodide (PI), and proteins expression related to cell cycle arrest and apoptosis were investigated via Western blotting. DMFTC inhibited PC3 cells growth through G1 phase cell cycle arrest and apoptosis induction. It activated the tumor suppressor p53 and the Cdk inhibitor p21, which regulate the cell progression into the G1 phase. Additionally, PI-positive late apoptotic non-viable cells were increased and the expression levels of the G1-positive downstream regulators cyclin D, cyclin E, Cdk2, and Cdk4 were decreased by DMFTC treatment. These results suggest that DMFTC induces G1 arrest and apoptosis induction through regulation of p53/p21-dependent cyclin-Cdk complexes, and it may be a useful therapeutic agent for the treatment of human advanced prostate cancer.

Effect of Rice Fermented using Poria cocos (a Wood-decay Fungus) Mycelium on Fermentation of Doenjang (Soybean Paste) (복령(Poria cocos) 균사체 발효쌀의 첨가가 된장의 품질에 미치는 영향)

  • We investigated changes in doenjang quality upon addition of rice fermented with Poria cocos mycelium. Sensory evaluation showed that rice addition to 15% (w/w) was optimal. The content of free amino acids was 1,899.17 mg% in doenjang fermented for 90 days with P. cocos mycelium. The rice contained seven essential amino acids: leucine, valine, phenylalanine, isoleucine, lysine, threonine, and methionine. The levels of essential amino acids increased after fermentation with P. cocos mycelium. Vitamin A (7.47 RE/100 g), Vitamin D (0.45 mg/100 g), and Vitamin E (5.73 mg/100 g) were detected in the experimental preparation but not in the control. In terms of electron-donating ability, the highest scores were 73.8% (in a water extract) and 76% (in an ethanolic extract) of doenjang fermented with P. cocos mycelium. Nitrite-scavenging ability was higher in the ethanolic extract than in the water extract of rice fermented with P. cocos mycelium.

Changes in Bioluminescence of Omphalotus japonicus Mycelia under Environmental Stress Conditions (환경 스트레스 조건에 따른 화경버섯 균사의 생물발광 변화)

  • Bioluminescence refers to the production and emission of light in living organisms. This phenomenon arises from luciferase-catalyzed oxidation reaction of luciferin. Bioluminescence is widely observed in marine vertebrates and invertebrates, as well as in some microorganisms and fungi. To date, approximately 80 species of fungi have been reported to be luminous. One such example is Omphalotus japonicus, which is a luminous fungus found in Korea. In this study, we examined the bioluminescence of Omphalotus japonicus mycelia. Light emission was detected at the edges of mycelia grown on solid agar medium. Notably, the intensity of bioluminescence was found to be significantly enhanced following wound induction. The increase in light intensity peaked at 3 h after mechanical damage. We also investigated the effects of extreme temperatures on bioluminescence. Unlike mechanical damage, high and low temperatures repressed the light emission from mycelia. Further investigations are required to reveal the physiological and ecological properties of fungal bioluminescent responses to environmental stresses.

Effects of combination treatment with sulfur dioxide generating pad and modified atmosphere packaging (MAP) on the quality of 'Campbell Early'grape under simulated export conditions (모의수출조건에서 유황패드와 MAP 복합처리가 '캠벨얼리' 포도의 품질에 미치는 영향)

  • The effects of combination treatment with sulfur dioxide generating pad ($SO_2$ pad) and modified atmosphere packaging (MAP) on grape quality were examined under simulated exporting condition and actual export to Los Angeles, USA. The 'Campbell Early' grape harvested in Hwaseong, Gyeonggi was precooled at $0^{\circ}C$, selected and packaged at $10^{\circ}C$, and stored at $0^{\circ}C$ for 30 days. The treatment was as follows: general export packaging (control), only $SO_2$ pad, combination of $SO_2$ pad and MAP (perforated polyethylene film, $SO_2$ pad+MA). In case of control and only $SO_2$ pad under simulated exporting condition, the grape quality changes with storage time were decrease in hardness and brush length, increase in stem browning, and increased in shattering rate. The treatment of $SO_2$ pad+MA was the most effective in preserving the grape quality since $SO_2$ concentration inside the package remained around 2.9 ppm by MA film. Grape exports from harvest to local distribution of the USA took 30 days, and only $SO_2$ pad+MA package showed no fungus, maintained brush length and rachis color, and was lowest at 4.0% of shattering rate since the concentration of $SO_2$ in the package was about 4.0 ppm. As a result, it was considered that the combination of $SO_2$ pad and MA was the most effective way to maintain freshness of grape during long-term marine transportation and extend the shelf-life in exporting countries.
