Effects of Green Tea Powder or Antioxidant Vitamin Supplementation on Lipid Peroxidation and Antioxidant Enzyme Activities in 9 Month- and 12 Month-old Rat Brain Regions (녹차 건분이나 항산화 비타민 보충이 9개월령과 12개월령 흰쥐의 부위별 뇌조직에 미치는 항산화 효과)

  • This study was performed to investigate the effects of green tea or antioxidant vitamins on lipid peroxidation and antioxidant enzyme system in various regions of rat brain aged 9 and 12 months. Male Sprague-Dawley rats were raised on the experimental diets; 3% green tea powder diet, antioxidant vitamins diet containing the $\beta$-carotene, vitamin C, vitamin E in the level same as in the 3% green tea powder diet, and control diet far 3 weeks. We measured concentrations of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD), catalase, and glutathione peroxide (GSH-Px) in various brain regions such as cortex, cerebellum, striatum, and hippocampus. Green tea powder or antioxidant vitamin supplementation decreased MDA concentrations in the striatum and the hippocampus, and increased SOB activities in the striatum, and GSH-Px activities in the cortex. There was no significant difference in the observed antioxidative effects between the green tea powder and antioxidant vitamin supplementation. A significant difference between 9 month- and 12 month-old rats was found in MDA concentrations and GSH-Px activities in all brain regions. These results suggest that green tea powder can have protective effects on various regions of rat brain and that these effects on lipid peroxidation and antioxidant enzymes are different by age. In inhibiting lipid peroxidation, there was no difference between green tea powder and antioxidant vitamins.

Effects of Biphenyldimethyl dicarboxylate(DDB) on the Lipid Peroxidation, Oxygen Free Radical Scavenging Enzymes Activities and Hepatic Functions in Ethanol-induced Hepatotoxic Rats (Biphenyldimethyl dicarboxylate(DDB)가 Ethanol 유발 간독성 흰쥐에서의 지질 과산화와 Oxygen Free Radical 제거 효소 활성도 및 간기능에 미치는 영향)

  • In an attempt to define the effects of Biphenyldimethyl dicarboxylate(DDB) on the lipid peroxidation, oxygen free radical scavenging enzymes activities and hepatic functions in ethanol-induced hepatotoxic rats, we studies malondialdehyde(MDA) level and the activities of catalse, superoxide dismutase(SOD), glutamic-oxaloacetic transaminase(GOT) and glutamic-pyruvic transaminase(GPT) in liver of the rats at 24, 48 and 72 hr after the injection of ethanol and DDB. Sprague-Dalwey albino rats weighing 250 to 280gm were injected intraperitoneally with ethanol(2.5 gm/kg ) only and ethanol plus DDB(300mg/kg ). The result obtained can be summarized as follows : 1) The group treated with ethanol showed significantly higher MDA level and lower catalase and SOD activities at 24, 48 and 72hr after the injection as compared with that of control group. 2) The group treated with ethanol showed significantly higher GOT and GPT activities at 24, 48 and 72hr after the injection as compared with that of control group. 3) The group treated with ethanol plus DDB showed significantly lower MDA level and higher catalase and SOD activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. 4) The group treated with ethanol plus DDB showed significantly lower GOT and GPT activities at 24, 48 and 72 hr after the injection as compared with that of ethanol group. These results suggest that the excessive oxygen free radicals resulting from the depression of the activities of catalase and superoxide dismutase is an important determinant in pathogenesis of ethanol-induced hepatotoxicity and DDB has antioxidant effects.

The Effects of Oxidative Stress Induced by Aluminum on Cellular Macromolecules in the Hippocampus and Cerebral Cortex of Rats (알루미늄을 투여한 흰쥐의 해마와 대뇌피질에서 Reactive Oxygen Species 생성으로 인한 생체거대분자의 산화적 손상)

  • This work aimed to study the effectiveness of cellular oxidative parameter (malondial-dehyde, protein carbonyl, and 8-hydroxy-2'deoxyguanosine). The experimental groups were aluminum treated rats and control rats. Aluminum treatd rats were given intraperitoneally aluminum nitrate nonahydrate ($Al^{3+}$, 0.2 mmol/kg) daily for 30 days except Sunday. Control rats were injected 1 ml of saline. After the dose, rats were decapitated and hippocampus and cerebral cortex were removed. The measured parameters were tissue malondialdehyde (MDA, index of lipid peroxidation), protein carbonyl (index of protein oxidation), 8-hydroxy-2'-deoxy-guanosine (8-OHdG, index of DNA oxidation), reduced glutathione (GSH) levels as well as glutathione reductase (GR) and catalase. AI concentrations in the tissues were also measured. All results were corrected by tissue protein levels. The results were as followed; 1. The concentrations of AI in the cortex and hippocampus were significantly higher in the AI-treated rats than in the control rats. 2. Antioxidative enzyme's activity, catalase and GR, were significantly higher in the AI-treated rats than the control rats. GSH levels were also higher in the AI-treated rats. 3. MDA, protein carbonyl, and 8-OHdG concentration of AI-treated rats were significantly higher than those of control rats. 4. The concentrations of antioxidants, and oxidative stress parameter were correlated with the concentrations of AI in hippocampus and cerebral cortex. Catalase and GR activity were also correlated with the concentration of AI. Based on these results, it can be suggested that intraperitoneally injected AI was accumulated in the brain and induced the increase of antioxidant levels and antioxidative enzyme activity. Also, the oxidative products of cellular macromolecules are significantly related to tissue AI concentration. Therefore MDA, protein carbonyl, and 8-OHdG are useful markers for oxidative stress on cellular macromolecules.

Salt Tolerance Assessment with NaCl of Stauntonia hexaphylla (Thunb.) Decene. and Raphiolepis indica var. umbellata (Thunb.) Ohashi (NaCl 처리에 따른 멀꿀과 다정큼나무의 내염성 평가)

  • Stauntonia hexaphylla and Raphiolepis indica, cold-tolerant broadleaved evergreens ranging through the southern region of South Korea, were assessed on salt tolerance with NaCl treatment using visual damage, chlorophyll florescence image, and malondialdehyde (MDA) analysis. As NaCl concentrations increased, the soil pH decreased and EC increased, and the soil of S. hexaphylla was affected more strongly by the treatment than that of R. indica. In visual damage, S. hexaphylla withered above 200 mM NaCl at 20 days after the treatment. All individuals of R. indica survived during the experiment though the leaves of R. indica showed visual damages up to 400 mM NaCl. The color changes in chlorophyll fluorescence showed a strong correlation with the degree of visual damage. As NaCl increased, the red color of the leaves of S. hexaphylla was distinctly changed to blue and chlorophyll fluorescence decreased starting from the margin to the middle of a leaf. R. indica showed subtle color changes and remained in red color during the experiment. At five days after the NaCl treatment, the MDA of S. hexaphylla was above $4.56nmol{\cdot}g^{-1}$ when plants showed the highest visual damage and EC. The MDA of R. indica in all treatments showed below $1.5nmol{\cdot}g^{-1}$ except 400 mM NaCl treatment during the experiment.

The Oxidative Stress in Cigarette Smokers and Antioxidant Vitamins

  • The purpose of this study was to find the extent of lipid peroxidation of erythrocytes in cigarette smokers, and to determine the relative effectiveness of $\beta$-carotene, canthaxanthin , and $\alpha$-tocopherol as antioxidants. Thirty smokers and 30 nonsmokers participated in this study . No significant differences according to age, sex, and height were shown. Cigarette smokers in this study had higher hemoglobin concentrations and more oxidation of hemoglobin than non-smokers. In addition, the erythrocytes of cigarette smokers had significantly higher MDA concentrations than crythrocytes of nonsmokers, which suggests that smokers may have tocopherol were studied in vitro by measuring the concentration of malondialdehyde(MDA) and precent hemolysis of erythrocytes. The addition of any antioxidant to erythrocytes significantly decreased MDA concentrations(p<0.05) while antioxidants showed nonsignificant inhibition of hemolysis. Among the antioxidant used in this study, canthaxanthin showed the greatest inhibition of both lipid peroxidationand hemolysis. Meanwhile, $\alpha$-tocopherol showed potent inhibition of lipid peroxidation, but not of hemolysis.

Effects of Onion Diet on Carbon Tetrachloride Toxicity of Rats (양파식이가 흰쥐에서 사염화탄소 독성에 미치는 영향)

  • This study designs to investigate effects of onion diet on carbon tetrachloride toxicity of rats. Experiments were performed with week's feeding, body weight, food intake, ratio of orgen weight/dody weight, serum lipid levels, superoxide dismutase and catalase activity and malondialdehyde(MDA) content in liver and kidney were determined. The content of serum total cholesterol in each group were lower than those of control group(p<0.05), especially at onion juice treated group. Serum HDL-cholesterol level of CBB and CBJ groups was significantly lower than that of control group (p<0.05). Superoxide dismutase activities of liver and kidney were significantly increased by carbon tetrachloride treatment and decreased by onion feeding. MDA contents in liver and kidney of CCl4 treated rats were significantly decreased by boiled and fresh onion fed group, compared with CCl4 treated control group. This result suggested that onion diet has a protective effect of CCl4 induced hepatotoxicity and nephrotoxicity of rat.

Modulation Effects of Antioxidant Vitamins on Ochratoxin A-induced Oxidative Toxicity in Mice (마우스에서 Ochratoxin A로 유발된 산화적 독성에 대한 항산화 비타민의 완화작용)

  • Ochratoxin A (OA), a naturally occurring mycotoxin, has been known to cause renal and hepatic lesion in human and animals. This study was carried out to investigate the modulation effects of antioxidant vitamins on OA-induced lipid peroxidation associated with oxidative damage. Vitamin C (10 mg/kg/day) and vitamin E (63.8 mg/kg/day) were administered by intraperitoneal (i.p.) injection to male ICR mice, and 1 hr later, OA which was dissolved in 0.1 M $NaHCO_3$, treated 4 mg/kg/day by i.p. injection. During 4 days repeated, and then measured superoxide dismutase (SOD) activity, catalase activity and malondialdehyde (MDA) formation in microsomes of liver and kidney. Additionally, the relationship between cell damage and modulation effects of antioxidant vitamins was evaluated by comet assay. Results were as followed; i) SOD, catalase activity and MDA level were significantly increased by OA treated, ii) SOD, catalase activity and MDA formation were significantly decreased by antioxidant vitamins combine treated, iii) blood cell damage associated with lipid peroxidation, induced by OA, also modulated by antioxidant vitamins. These results indicated that antioxidant vitamins might be used for prevention of renal and hepatic damage due to ochratoxicosis.

Antioxidant Characteristics and Phytoremediation Potential of 27 Taxa of Roadside Trees at Industrial Complex Area (공단지역에서 생육하는 가로수 27종의 항산화특성과 잠재적 환경정화능력)

  • In order to screen for the best species for mitigating air pollutants by plants at an industrial complex area, we investigated antioxidant capacity, lipid peroxidation and nitrogen content in the leaves of 27 taxa of woody plants that are mostly utilized as roadside trees. Among 27 taxa, the highest value of antioxidant capacity was given by Cedrus deodara (91.4%) and the lowest one was by Firmiana simplex (56.9%). At lipid peroxidation level, little malondialdehyde (MDA) was observed in Lagerstroemia indica and Ginkgo biloba, but Platanus occidentalis, Castanoposis cuspidata var, sieboldii, Machilus thunbergii and Juniperus chinensis showed high MDA content. Antioxidant capacity of the deciduous woody plants was not significantly different in comparison with that of the evergreen ones. But MDA content of the deciduous woody plants was lower than that of the evergreen ones. The 27 taxa of woody plants appeared to be classified into four types: those of high antioxidant capacity and low lipid peroxidation, those of high antioxidant capacity and high lipid peroxidation, those of low antioxidant capacity and low lipid peroxidation, and those of low antioxidant capacity and high lipid peroxidation. The taxa included in these types are 7 (first type), 6 (second one), 8 (third one) and 6 (fourth one) taxa. first or second type species which have a high antioxidant capacity represented low nitrogen content in their leaves. However, third or forth type species which have low antioxidant capacity showed high nitrogen content in their leaves. Metasequoia glyptostroboides, Platycarya strobilacra and P. occidentalis which belong to the first or second type had extraordinarily high antioxidant capacity and high nitrogen content. Thus, three species are considered to be good phytoremediators for an industrial complex area.

Effects of Benincasa Hispida Seed Supplementation on Glyeogen status and Lipid Peroxidatin in Streptozotecin-Induced Diabetic Rats (동과인의 보충이 당뇨 유발 흰쥐의 글리코겐과 지질과산화에 미치는 영향)

  • This study was designed to examine the effects of diets containing different levels of seeds of Benincasa hispida(wax gourd) on glycogen, protein levels and lipid profiles as well as malondialdehyde (MDA) in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley rats were induced diabetes mellitus by STZ injection (45 mg/kg) into the tail vein and were divided into four groups: normal, STZ-control, and two experimental groups. Normal and STZ-control groups were fed the AIN-93 diet and the two experimental groups were fed a modified diet containing 2.5% and 5.0% of wax gourd seed powder for four weeks. The liver, muscle, lung, kidney, and pancreas were excised after sacrifice, then the glycogen, protein, and lipid peroxidation products were measured. The rats fed 2.5% wax gourd seed group showed higher levels of liver glycogen compared with that of the STZ-control group. The levels of kidney protein were significantly increased in the 2.5% and 5.0% wax gourd seed groups. There were no significant difference cholesterol and liver triglyceride levels of the liver and MDA concentration in the liver, lung, and kidney among all four groups. These results show that wax gourd seed treatment of 2.5% and 5.0% doses did not exhibit profound anti-lipid peroxidation properities.

Preservative Effect of Leukocyte-Depleted Blood Cardioplegic Myocardium during Cardiac Surgery

  • The present study was prospectively designed to assess the clinical effect of leukocyte-depleted blood cardioplegic solution (BCS) on myocardium during cardiac surgery with cardiopulmonary bypass (CPB). 30 adult patients scheduled for elective cardiac surgery were divided into control group (n=15), which infused routine BCS, and leukocyte-depleted (LD) group (n=15), which infused leukocyte-depleted BCS. Total and differential leukocyte counts in BCS, malondialdehyde (MDA) and troponin-T (TnT) concentrations in coronary sinus blood, and cardiac index (CI) were measured at preoperative and postoperative period. The BCS in LD group had less total leukocyte counts with neutropenia than that in control group (P<0.01). MDA (3.70$\pm$0.35 vs 5.90$\pm$0.57 $\mu$mol/L, p<0.05) and TnT (0.42$\pm$0.03 vs 0.60$\pm$0.09 ng/mL, p<0.05) were significantly low in LD group compared with control group, while LD group had higher CI (3.28$\pm$0.16 L/min/$m^2$, p<0.05) than control group (2.69$\pm$0.18 L/min/$m^2$). These results suggest that leukocyte-depleted blood cardioplegic solution has a better myocardial protective effect with less generations of oxygen free radicals and ischemia/reperfusion injury.
