사람 성장호르몬 유전자를 발현하는 형질전환생쥐의 불임성 (Infertility of Transgenic Mice Experssing Human Growth Hormone Gene)

  • Many transgenic mice expressing human growth hormone gene were infertile. To investigate the infertility of these transfenic mice, it was looked into the estrus cycle and sexual behaviour and also tested through in vitro fertilization whether the germ cells of these mice normal or not. The infertile female transgenic mice were mated to the fertile males of ICR strain, but in almost all of them the vaginal plugs were not detected and their estrus cycles by vaginal smear were almost irregular which kept up estrus or diestrus stage. Many male transgenic mice did not have the ability of sexual behaviour. Therefore the viability of germ cells in infertile male transgenic mice was investigated by in vitro fertilization, but the sperm were normally fertilized with the eggs and the transgene of parent was passed on to the progeny. These results consequently suggest that the infertility of transgenic mice experssing human growth hormone gene may be due to the physiological activity of human growth hormone, not germ cells.

Oral Single Dose Toxicity Study of Low Molecular Fucoidan in Mice

  • This study was conducted to obtain information of the oral dose toxicity of low molecular fucoidan (LMF) in male and female mice. In order to calculate 50% lethal dose ($LD_{50}$) and approximate lethal dose (LD), test material was once orally administered to male and female ICR mice at dose levels of 2000, 1000, 500, 250, 125 and 0 (vehicle control) mg/kg (body wt.). The mortality and the changes on body weight, clinical signs, gross observation and organ weight and histopathology of principle organs were monitored 14 days after LMF treatment. We could not find any mortalities, clinical signs, body weight changes and gross findings. In addition, significant changes in the organ weight and histopathology of principal organs were not observed except for some sporadic findings. The results obtained in this study suggest that LMF may not be toxic in mice and may be therefore safe for clinical use. The $LD_{50}$ and approximate LD in mice after single oral dose of LMF were considered over 2000 mg/kg in both female and male mice.

  • The object of this study was to evaluate the single oral dose toxicity of platycodin D, a saponin from the root of Platycodon grandiflorum in male and female mice. Platycodin D was administered to female and male mice as an oral dose of 2000, 1000, 500, 250 and 125 mg/kg (body wt.). Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after treatment, upon necropsy, organ weight and histopathology of 14 principle organs were examined. As the results, no platycodin D treatment related mortalities, clinical signs, changes on the body and organ weights, gross and histopathological observations against 14 principle organs were detected up to 2000 mg/kg in both female and male mice. Therefore, $LD_{50}$ (50% lethal dose) and approximate LD of playtcodin D after single oral treatment in female and male mice were considered over 2000 mg/kg - the limited dosages recommended by KFDA Guidelines [2009-116, 2009], respectively.

Effects of methanol extracts of Cuscuta reflexa Roxb. stem and Corchorus olitorius Linn. seed on male reproductive system of mice

  • The antifertility activity of methanol extract of Cuscuta reflexa Roxb. stem (MECR) and Corchorus olitorius Linn. seed (MECO) were studied on male Swiss albino mice. The extracts were found to decrease sperm count, percentage of motile sperm and testosterone level in treated mice when compared with vehicle control after 17 days of treatment. The weight of gonads, epididymis were decreased whereas no significant changes of the body weight of mice were observed after methanol extract treatments. The fertility test showed 100% negative result in MECR and MECO treated mice at medium and high dose level of treatment. MECR and MECO in low (25 mg/kg and 15 mg/kg, respectively), medium (50 mg/kg and 20 mg/kg, respectively) and high (75 mg/kg and 25 mg/kg, respectively) dose level caused a simultaneous fall in testicular ${\Delta}5$-$3{\beta}$-hydroxy steroid dehydrogenase and glucose-6-phosphate dehydrogenase activities which are involved in testicular steroidogenesis. Total cholesterol and ascorbic acid content in testis were increased significantly in gonads. The activities of lactate dehydrogenase, malic dehydrogenase and ascorbic acid oxidase were reduced whereas that of carbonic anhydrase was increased significantly in the testis of MECR and MECO treated mice. All these observations indicate that the methanol extract of C. reflexa stem and C. olitorius seed produced antifertility activity in sexually matured male mice, which may be due to inhibition of gonadal steroidogenesis. This activity may be attributed due to the presence of flavonoids and steroids, respectively.

  • Objective: The differences between type 1 and type 2 diabetes mellitus (T1DM and T2DM) in terms of their adverse effects on male reproductive parameters have never been elucidated. This study aimed to distinguish between the effects of the DM types in mice treated with multiple low doses of streptozotocin (STZ) to mimic human T1DM and coadministered a high-fat diet (HFD) to mimic human T2DM. Methods: The T1DM mice were intraperitoneally injected with STZ (40 mg/kg body weight) for 5 days. The T2DM mice received an HFD for 14 days prior to STZ injection (85 mg/kg body weight), followed by continuous feeding of an HFD. Male reproductive parameters were evaluated. Results: The reproductive organs of the DM mice weighed significantly less than those of controls, and the seminal vesicles plus prostates of the T1DM mice weighed less than those of the T2DM mice. Increased sperm abnormalities and incomplete DNA packaging were observed in the DM groups. Sperm concentration and the proportion of normal sperm were significantly lower in the T1DM group. The seminiferous histopathology of DM mice was classified into seven types. The penises of the DM mice were smaller than those of the controls; however, tunica albuginea thickness and the amount of penile collagen fibers were increased in these mice. Round germ cells were abundant in the epididymal lumens of the mice with DM. Conclusion: T1DM adversely affected reproductive parameters to a greater extent than T2DM.

흑진주벼로부터 색소추출물의 안전성 (Food Safety of Pigment in Black Rice cv. Heugjinjubyeo)

  • 흑진주벼 색소 분획의 급성독성 시험을 하기 위하여 ICR계 생쥐 각 암수 6마리를 1주일 이상 순화시킨 다음 경구투여를 실시하여 14일 동안 급성독성 평가를 한 결과는 다음과 같다. 1. 색소추출물의 급성독성 실험에서 생쥐의 암수 모두 9000mg/kg의 대량 경구 용량에서도 사용한 6마리가 모두 사망하지 않았다. 2 1000, 3000 및 9000mg/kg 경구 용량에서 14일간 측정한 체중변화는 대조군과 유의성 있는 변화가 없었고 증상의 이상도 보이지 않았다. 3. 경구 용량 시험의 부검결과 대조군과 유의성 있는 체중변화와 다른 증상의 이상이 관찰되지 않아서 안정성이 높은 물질로 확인되었다.

빈랑자(檳榔子) 추출물의 마우스 경구 단회 투여독성 평가 (Single Oral Dose Toxicity Test of Areca catechu Aqueous Extracts in Mice)

  • This study was to evaluate the single dose toxicity of Arecae Semen (AS) in male and female ICR mice. Aqueous extracts of AS (Yield = 13.15%) were administered as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 12 principle organs were examined. We could not find any mortality, clinical signs, and changes in the body and organ weight except for diarrhea. Diarrhea were observed in all three different dosage groups of male mice, and in 2000 mg/kg groups of female mice within 48hrs after administration. In addition, no AS extract related abnormal gross findings and changes in histopathology of principle organs were detected except for some sporadic accidental findings. Although the 50% lethal dose and approximate lethal dose of AS aqueous extracts in female and male mice were detected as over 2,000 mg/kg - the limited highest dosage recommended by KFDA guidelines. It should be carefully used in clinics because AS may be induced severe digestive tract disorders.

Mouse Single Oral Dose Toxicity Test of Bupleuri Radix Aqueous Extracts

  • The aim of this study was to evaluate the single oral dose toxicity of Bupleuri Radix (BR) aqueous extracts, it has been traditionally used as anti-inflammatory agent, in male and female mice. BR extracts (yield = 16.52%) was administered to female and male ICR mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 14 principal organs were examined. As the results, no BR extracts treatment related mortalities, clinical signs, changes on the body and organ weights, gross and histopathological observations against 14 principal organs were detected up to 2,000 mg/kg in both female and male mice, except for soft feces and related body weight decrease detected in male mice treated with 2,000 mg/kg. Therefore, $LD_{50}$ (50% lethal dose) and approximate LD of BR aqueous extracts after single oral treatment in female and male mice were considered over 2000 mg/kg, respectively. Although it was also observed that the possibilities of digestive disorders, like soft feces when administered over 2,000 mg/kg of BR extracts in the present study, these possibilities of digestive disorders can be disregard in clinical use because they are transient in the highest dosages male only.

Amisulpride의 지속 투여가 생쥐의 성별에 따른 체중 및 대사에 미치는 영향 (Effects of Amisulpride Administration on Body Weight and Metabolic Abnormalities in Mice)

  • 목 적 이 연구는 수컷과 암컷 생쥐에서 amisulpride를 투여하였을 때 먹이 섭취량, 체중 및 대사 지표에 대한 영향을 risperidone 및 정상 대조군과 비교하여 알아보고자 하였다. 방 법 수컷과 암컷 C57BL/6형 생쥐를 저용량 amisulpride 투여군(1.5mg/kg), 고용량 amisulpride투여군(15mg/kg), risperidone 투여군(0.1mg/kg)과 Vehicle군으로 나누었다. 약물은 21일간 매일 하루에 한 번 복강 주사를 통해 투여되었다. 체중은 일주일에 한 번씩 측정하였으며 먹이 섭취량은 매일 측정하였다. Triglyceride(TG), glucose, insulin 그리고 prolactin은 실험 종료 시에 측정하였다(실험 22일). 결 과 암컷 생쥐의 경우, 저용량과 고용량의 amisulpride 투여군에서는 유의한 체중 증가를 보였으며, risperidone 역시 유의한 체중 증가를 보였다. 그러나, 두 amisulpride군의 증가량은 risperidone군의 증가량에 비해 수치적으로 작았다. 수컷 생쥐의 경우, 고용량의 amisulpride만이 유의한 체중 증가를 가져왔다. 체중 증가를 보였던 집단 중에서, 고용량의 amisulpride를 투여하였던 수컷 생쥐만이 유의하게 증가된 먹이 섭취량과 연관성이 있었다. 암컷 생쥐에서 체중 증가를 보였던 군들은 먹이 섭취량과 유의한 상관성을 보이지 않았다. 수컷 생쥐에서, 두 개의 amisulpride 투여군 모두는 vehicle에 비해 유의하게 높은 혈장 insulin 수치를 보였다. 암컷과 수컷 생쥐에서, 저용량과 고용량의 amisulpride 투여군은 vehicle과 비교하여 유의하게 높은 혈장 prolactin수치를 보였다. Triglyceride는 모든 집단에서 유의하게 변화하지 않았으며 glucose는 수컷 risperidone 투여군에서만 유의하게 변화하였다. 결 론 Amisulpride는 암수 생쥐에서 체중 증가를 일으키지만, 대사 이상에 대한 결과는 성별에 따라 차이가 있었다. 본 연구의 결과는 amisulpride에 의한 체중 증가에 암수 생쥐에서 서로 다른 기전이 작용할 가능성을 시사한다.

Bisphenol a induces reproductive dysfunction in male mice

  • It has been suggested that bisphenol A (BPA), a known endocrine disruptor, interferes with the endocrine system, causing reproductive dysfunction. Recently, BPA has been found in waste water due to incomplete sewage purification, possibly threatening health through its ingestion via tap water. In this study, young male mice (6 - 7 weeks old) were administered water containing BPA (50 mg·kg-1) for four weeks, while control mice consumed water without BPA. Serum, epididymal spermatozoa and testicular sections were assessed after sacrificing the mice on day 28. No significant differences were obtained between the groups in the body, testis and seminal vesicle weights. However, the epididymal sperm motility and count levels were significantly reduced in BPA-fed mice. Significantly higher hepatotoxicity levels were also observed in mice ingesting BPA as compared to the control mice. The level of serum testosterone was reduced, and testicular sections revealed incomplete and irregular spermatogenesis in BPA-ingested mice. The sperm proteasomal-proteolytic activity level has been implicated in sperm function and is measured in motile spermatozoa using fluorometric substrates. High ubiquitin C-terminal hydrolase activity levels were observed in the control mice without BPA. During a mating trial, a low pregnancy rate (71.4%) was observed in females mated with males who had consumed BPA (100% in the control mice). Overall, BPA adversely affected spermatogenesis and quality, as indicated by decreased sperm motility, concentration and serum testosterone levels, resulting in reduced fertility competence.
