고본양정방 투여가 TCDD 유도 랫드의 생식독성에 미치는 영향 (The Effects of Gobonyangjeonbang Administration on Reproductive Toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induced Rats.)

  • TCDD로 유발할 수 있는 남성 난임 원인 중 가장 큰 비율을 차지하는 것은 정자형성 장애와 정자의 질적 저하가 있다. 이를 해결하기 위해 GYB의 한의학적 처방을 이용하여 TCDD-유도 생식독성을 유발한 랫드에서 내분비기능 및 생식독성 관련 지표에 미치는 영향을 조사하고자 본 연구를 시행하였다. 수컷 SD 랫드를 5개의 그룹으로 7마리씩 나누어 실험하였다. 정상 대조군은 vehicle 및 saline을 투여하였으며, TCDD 단독투여그룹은 TCDD(2 ㎍/kg, weeks) 및 생리식염수를 복강 내 투여하였으며, 시험 군은 6주 동안 GYB (75, 150, 300 mg/kg)를 3가지 농도로 나누어 경구로 투여하였다. TCDD를 투여한 모든 군에서 체중 감소가 나타났으며, 호르몬 변화에서 GYB 300 mg/kg 투여군에서 free testosterone의 유의한(p<0.01) 감소가 나타났다. 또한 GYB 투여군의 고환조직에서 생식세포 일부 파괴, 정세관 위축 그리고 정자 수의 감소가 농도 의존적으로 개선되는 것이 관찰되었다. 또한 Johnsen's score, serotoli cell index(SCI)에서도 농도 의존적으로 개선되었다(p<0.05). 이러한 발견은 향후 남성 난임의 해결방안으로 의학적 시술이 아닌 약물요법을 효과적으로 사용할 수 있을 것으로 기대할 수 있다.

Effects of nandrolone decanoate on expression of steroidogenic enzymes in the rat testis

  • Objective: Nandrolone decanoate (ND) is an anabolic-androgenic steroid frequently used for clinical treatment. However, the inappropriate use of ND results in the reduction of serum testosterone level and sperm production. The suppressive effect of ND on testosterone production has not been investigated in detail. The present study was designed to examine the effect of ND on the expression of steroidogenic enzymes in the rat testis. Methods: Male Sprague Dawley rats at 50 days of age were subcutaneously administrated with either 2 or 10 mg of ND/kg body weight/week for 2 or 12 weeks. The changes of transcript and protein levels of steroidogenic enzymes in the testis were determined by real-time polymerase chain reaction and western blotting analyses, respectively. Moreover, immunohistochemical analysis was employed to determine the changes of immunostaining intensity of these enzymes. The steroidogenic enzymes investigated were steroidogenic acute regulatory protein, cytochrome P450 side chain cleavage enzyme, $17{\alpha}-hydroxylase$, $3{\beta}-hydroxysteroid$ dehydrogenase, and cytochrome P450 aromatase. Results: The treatment of ND resulted in depletion of Leydig cells and sloughing of germ cells in the testis. The ND treatment caused significant expressional decreases of steroidogenic enzymes at transcript and protein levels, and the destructive effects of ND on the testis were more apparent with a higher dose and a longer period of the treatment. Evident reduction of immunostaining intensity present in Leydig cells was clearly detected by the ND treatment. Conclusion: The exposure to ND in young male results not only in histological changes of the testis but also in aberrant gene expression of testicular steroidogenic enzymes, consequently leading into the reduction of testosterone production in the testis and thus likely disruption of spermatogenesis.

Annual Reproductive Cycle of Acheilognathus rhombeus, in Korea

  • In this study, an experiment was conducted to investigate the annual reproductive cycle of a Korean flat bittering, Acheilognathus rhombeus, from Ogok-myeon located in Seomjin River. The reproductive cycle was examined histologically regarding water temperature and day length of the habitat, the gonadosomatic index (GSI), and developmental characteristics of female and male gonads. The maximum GSI was found to be 3.50±0.53 and 1.36±0.14 for females and males, respectively, when the water temperature and day light was 16.9℃ and 11.3 hours, respectively in October 2018. On the other hand, the minimum GSI was found to be 0.16±0.09 and 0.69±0.15 for males and females in December 2018 and February 2019, respectively. The ovipositor of females appeared from August to November 2018. We compared and calculated the stages of germ cell developmental characteristics in the testis and ovaries to determine the reproductive cycle. According to the result, we classified the female A. rhombeus reproductive cycle into four phases, which are ripe and spawning phase (October), degenerative phase (November to December), growing phase (January to March) and mature phase (April to September). The annual reproductive cycle of male A. rhombeus was categorized into four phases: mature phase (June to October), degenerative phase (November to March), resting phase (April) and growing phase (May). The Korean flat bittering is an autumn-spawner as the main spawning season in October. In male, testicular spermatogonia appeared all year-round, and the ripe and releasing phase, which is characteristics of the spawning season in other fish, did not appear.

Radioprotective Effect of Panax ginseng against Giant Cell Formation in The Testis of Irradiated Mice

  • Panax ginseng (family- Araliaceae) is a native plant of Korea and has been used for past several years among oriental people. To evaluate the radioprotective potential of P. ginseng on the formation of giant cells in the testis of Swiss albino mice, the animals were divided into four groups: -(I)-Only vehicle was administered. (II)P. ginseng treated group: -The animals received 10 mg/kg body weight P. ginseng root extract (in DDW) i.p. continuously for 30 days. (III) Irradiated group: -The animals were exposed to 8 Gy gamma radiation at the dose rate of 1.69 Gy/min at the distance of 80 ems. (IV) Combined treatment group: -Animals were given P. ginseng extract for four days and on fourth day they were irradiated to 8 Gy gamma radiation after 30 minute of extract administration. The animals of these three groups were autopsied on day 1,3, 7, 14 and 30 days. In ginseng treated group, active spermatogenesis was observed without any toxic effect. Histopathological studies of irradiated group (II) revealed reduction in germ cell count, loss of sperms and formation of multinucleated giant cells on day 7th. These giant cells were formed by round nuclei of early or late spermatids. In combination group (III), although germinal epithelium was still disorganized with loss of cells in few tubules, but no giant cell formation was observed. In order to know the mechanism of radioprotection of ginseng, LPO and GSH were estimated. It was observed that pretreated irradiated animals showed inhibition of LPO and increase in GSH. Thus the present study suggests ginseng protects male gonads. This may be attributed to the inhibition of LPO and increase synthesis of GSH byginseng.

Epidemiology of Primary Brain and Central Nervous System Tumors in Korea

  • Objective : The aim of this report is to provide accurate nationwide epidemiologic data on primary central nervous system (CNS) tumors in Korea. Despite its importance, there are no accurate statistics on primary CNS tumors in Korea. We analyzed primary CNS tumors diagnosed in 2005 from the nationwide registry. Methods : Data on primary CNS tumors diagnosed in 2005 were collected from the Korean Central Cancer Registry and the Korean Brain Tumor Society. Crude and age-standardized rates were calculated in terms of gender, age, and histological type. Tumors of uncertain histology were investigated individually at the corresponding hospitals and had their diagnoses confirmed. Results : A total of 5,692 patients diagnosed with primary CNS tumors in 2005 were included in this study. CNS tumors occurred in females more often than in males (female to male, 1.43 : 1). The most common tumor was meningioma (31.2%). Glioblastoma accounted for 30.7% of all gliomas, and 19.3% of all malignant primary CNS tumors. In children under 19 years of age, both germ cell tumor and embryonal/primitive/medulloblastoma were the most common tumors. Conclusion : This article is the first nationwide primary CNS tumor epidemiology report in Korea. Data from this study should provide valuable information regarding the understanding of primary CNS tumors epidemiology in Korea.

Effect of 11-ketotestosterone (11-KT) on Gonadal Sex Reversal and Spermatogenesis of Honeycomb Grouper Epinephelus merra

  • We investigated the androgenic effects of 11-ketotestosterone (11-KT) on gonadal sex reversal and spermatogenesis in honeycomb grouper Epinephelus merra by method of gonadal biopsy. 11-KT was injected intramuscularly at a concentration of 1 and $10{\mu}g$ body weight. The proportion of cross sectional area of the gonad occupied by each germ cell type was measured and compared pre- and post-injection group. During the sex change phase, the distribution ratio of oocytes was decreased in all fish of 11-KT treatment group while the distribution ratio of spermatocytes was increased than pre-injection group. In male phase, all fish of 11-KT treatment group shown the increased distribution ratio of spermatocytes, but the distribution ratio of spermatozoa was decreased than pre-injection group. The present results suggest that 11-KT can stimulate degeneration of oocytes, proliferation of spermatocytes and spermiation in honeycomb grouper.

  • N-ethyl-N-nitrosourea (ENU), which is a toxin and a carcinogen, as well as a mutagen, has a variety of effects on mice. ENU induces point mutation in male germ cell. Number of mutant animals are developed with ENU treatment. However, potentiality ot ENU as a carcinogen is not fully understood, even though, mutagenicity of ENU is broadly studied, In the present study, the gene expression profiling and histopathological investigation of ENU treated mouse's liver and brain were investigated. Also, the expression patterns of cancer related genes in ENU-treated mouse were analyzed.

  • Homeobox genes play essential roles in embryonic development and reproduction. Recently, a large cluster of homeobox genes, reproductive homeobox genes on the X chromosome (Rhox) genes, was discovered as three gene clusters, ${\alpha}$, ${\beta}$, and ${\gamma}$ in mice. It was found that Rhox genes were selectively expressed in reproduction-associated tissues, such as those of the testes, epididymis, ovaries, and placenta. Hence, it was proposed that Rhox genes are important for regulating various reproductive features, especially gametogenesis in male as well as in female mammals. It was first determined that 12 Rhox genes are clustered into ${\alpha}$ (Rhox1-4), ${\beta}$ (Rhox5-9), and ${\gamma}$ (Rhox10-12) subclusters, and recently Rhox13 has also been found. At present, 33 Rhox genes have been identified in the mouse genome, 11 in the rat, and three in the human. Rhox genes are also responsible for embryonic development, with considerable amounts of Rhox expression in trophoblasts, placenta tissue, embryonic stem cells, and primordial germ cells. In this article we summarized the current understanding of Rhox family genes involved in reproduction and embryonic development and elucidated a previously unreported cell-specific expression in ovarian cells.

종격동 내배엽동 종양의 세침흡인 세포학적 소견 (Fine Needle Aspiration Cytology of Endodermal Sinus Tumor of the Mediastinum)

  • Endodermal sinus tumor of the mediastinum is a very rare malignant tumor, usually affecting young adolescents, and its histologic findings are characteristic as that of gonadal germ cell origin. We describe the cytologic finding of fine needle aspiration of mediastinal endodermal sinus tumor in a 19-year-old male patient, comparing with tissue section. The tumor cells in smears were usually in tight clusters with large overlapping cells, which were arranged in a papillary or microacinar pattern, suggesting carcinoma. The tumor cells were large, round or oval with a small amount of cytoplasm which was occasionally vacuolated and had indistinct border. The nuclei were large, pleomorphic, and vesicular with large prominent nucleoli. The tissue sections showed typical findings of endodermal sinus tumor. Careful correlation of cytological findings and the serum alpha -fetoprotein level would be helpful to confirm the diagnosis.

Spermatid Differentiations During Spermiogenesis and Mature Sperm Ultrastructure in Male Crassostrea nipponica (Seki, 1934, Pteroirmorphia: Ostreidae)

  • Spermatid differentiations during spermiogenesis and mature sperm ultrastructure in male Crassostrea nipponica were investigated by transmission electron microscope observations. The morphology of the spermatozoon of this species has a primitive type and is similar to those of other bivalves. Mature spermatozoa consist of broad, cap-shaped acrosomal vesicle and an axial rod in subacrosomal materials on an oval nucleus showing deeply invaginated anteriorly, two triplet substructure centrioles surrounded by four spherical mitochondria, and satelite fibres, which appear near the distal centriole. The acrosomal vesicle of spermatozoa of C. nipponica resemble to those of other investigated ostreids. Especially, two transverse bands (stripes) appear at the anterior region of the acrosomal vesicle, unlikely 2-3 transverse bands (stripes) in C. gigas. It is assumed that differences in this acrosomal substructure are associated with the inability of fertilization between the genus Crassostrea and other genus species in Ostreidae. Therefore, we can use sperm morphology in the resolution of taxonomic relationships within the Ostreidea. The sperm is approximately $48-50{\mu}m$ in length including an oval sperm nucleus (about $1.0{\mu}m$ in length and $1.41{\mu}m$ in width), an acrosome (about $0.48{\mu}m$ in length and 0.30 in width) and tail flagellum ($46-48{\mu}m$). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9 + 2 structure. These morphological charateristics of acrosomal vesicle belong to the family Ostreidae in the subclass Pteriomorphia.
