GeneFishing PCR 기법을 이용한 한우 등심조직의 육질 등급 간 차등 발현 유전자의 발굴

  • 본 연구는 한우 근내 지방 축적 기작을 구명하고 고급육과 저급육에서 차등 발현되는 유전자를 발굴 동정하여 한우 육질 진단을 위한 분자 표지 마커로 활용하기 위해 GeneFishing PCR 기법을 이용하여 한우 육질등급에 따른 등심조직에서 차등적으로 발현되는 유전자를 분석하였다. 한우 육질 등급($1^+$ 등급 vs 3 등급)간에 총 10개의 차등 발현 유전자가 확인되었고 이 가운데 고급육 한우 등심에서 발현량이 높은 유전자가 4개 그리고 저급육 등심에서 발현량이 높은 유전자 6개가 각각 검출되었다. 발현량 차이 유전자를 cloning하여 염기서열을 분석하고 상동성 검색을 실시한 결과 고급육에서 발현량이 높은 DEG는 주로 EST(expressed sequence tag) 유전자들로 밝혀졌고 저급육에서 발현량이 높은 DEG는 malate dehydrogenase 2(MDH2), myosin heavy chain 2a, triosephosphate isomerase 1(TPI 1), actin, alpha 1, skeletal muscle(ACTA1 ) 유전자들로 동정 되었다. 본 연구를 통해 한우 육질간 차등 발현되는 유전자들은 한우 육질 및 등급판정을 위한 표지인자(marker)로 활용할 수 있어 유전자 마커를 이용한 고급육 생산 한우의 육질 조기진단이 가능할 것이다.

척추동물의 Isozyme에 관한 비교연구: III. 한국산 뱀목의 Lactate 및 Malate Dehydrogenase Isozyme

  • 한국에서 서식하는 뱀과, 살모사과, 및 북살모사과에 속하는 몇종의 파충류에서 각 장기별로 MDH 및 LDH isozyme을 조사하였던 바 MDH는 2개의 isozyme으로 구성되어 있는 점이 동일하였다. 그러나 대륙유혈목이의 MDH isozyme은 이동속도가 빠른 점이 다른 종과 다르다. 뱀과, 살모사과 및 북살모사과의 구성원 각 종에서 4개의 LDH isozyme을 확인할 수 있었다. 근과 간조직의 LDH isozyme은 근본적으로 같은 pattern이었고 $LDH_4$가 없는 것이 뱀과, 살모사과 및 북살모사과의 특징이었다. 한편 심장에서는 2개의 LDH isozyme이 발견되는데 근조직과는 반대되는 pattern 이었다. 위 homogenate에서 나타나는 LDH isozyme은 심장의 pattern과 동일하다고 볼수 있는데 살모사과와 북살모사과에서는 4개의 isozyme이 나타나는 점이 독특하였다. 따라서 실험 동물의 MDH 및 LDH isozyme pattern에서 의미있는 다양성을 발견할 수 없었다.

Characteristics of Sophorolipid as an Antimicrobial Agent

  • Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm, respectively. Also, 100 ppm of sophorolipid inhibited $50\%$ of cell growth of plant pathogenic fungus, Botrytis cineria. However, sophorolipid showed no effect on Escherichia coli, indicating that its selective antimicrobial activity depended on the cell wall structure. Treatment of B. subtilis with sophorolipid increased leakage of intracellular enzyme, malate dehydrogenase, indicating a possible interaction of sophorolipid with a cellular membrane. Comparing lactone-type and acid-type sophorolipids, the former showed a higher antimicrobial activity. Supplementing other surfactants showed no significant effects on the antimicrobial activity. Animal study showed that 5 g of sophorolipid per kg body weight by oral administration caused no toxicity, and sophorolipid induced no irritation on the skin. These results show potential use of sophorolipid as an active ingredient in healthcare products.

Effects of dietary leucine supplementation on the hepatic mitochondrial biogenesis and energy metabolism in normal birth weight and intrauterine growth-retarded weanling piglets

  • BACKGROUND/OBJECTIVES: The study was conducted to evaluate the effects of dietary leucine supplementation on mitochondrial biogenesis and energy metabolism in the liver of normal birth weight (NBW) and intrauterine growth-retarded (IUGR) weanling piglets. MATERIALS/METHODS: A total of sixteen pairs of NBW and IUGR piglets from sixteen sows were selected according to their birth weight. At postnatal day 14, all piglets were weaned and fed either a control diet or a leucine-supplemented diet for 21 d. Thereafter, a $2{\times}2$ factorial experimental design was used. Each treatment consisted of eight replications with one piglet per replication. RESULTS: Compared with NBW piglets, IUGR piglets had a decreased (P < 0.05) hepatic adenosine triphosphate (ATP) content. Also, IUGR piglets exhibited reductions (P < 0.05) in the activities of hepatic mitochondrial pyruvate dehydrogenase (PDH), citrate synthase (CS), ${\alpha}$-ketoglutarate dehydrogenase (${\alpha}$-KGDH), malate dehydrogenase (MDH), and complexes I and V, along with decreases (P < 0.05) in the concentration of mitochondrial DNA (mtDNA) and the protein expression of hepatic peroxisome proliferator-activated receptor-${\gamma}$ coactivator $1{\alpha}$ (PGC-$1{\alpha}$). Dietary leucine supplementation increased (P < 0.05) the content of ATP, and the activities of CS, ${\alpha}$-KGDH, MDH, and complex V in the liver of piglets. Furthermore, compared to those fed a control diet, piglets given a leucine-supplemented diet exhibited increases (P < 0.05) in the mtDNA content and in the mRNA expressions of sirtuin 1, PGC-$1{\alpha}$, nuclear respiratory factor 1, mitochondrial transcription factor A, and ATP synthase, $H^+$ transporting, mitochondrial F1 complex, ${\beta}$ polypeptide in liver. CONCLUSIONS: Dietary leucine supplementation may exert beneficial effects on mitochondrial biogenesis and energy metabolism in NBW and IUGR weanling piglets.

Carbonic Maceration처리에 의한 Campbell Early 발효액의 감산 효과: 사과산 대사 관련 효소활성의 영향 (Deacidification Effect of Campbell Early Must via Carbonic Maceration : Effect of Enzyme Activity Associated with Malic-Acid Metabolism)

  • Carbonic maceration처리 포도주에 있어 유기산 특히 사과산 함량을 감소시키는 주요 원인을 찾고자 포도를 2주 동안 온도별 carbonic maceration 처리하며 시기별로 산함량 및 사과산대사 관련 효소활성을 측정하였다. 온도별 carbonic maceration 처리 포도의 pH는 CM-$25^{\circ}C$와 CM-$35^{\circ}C$에서 처리 시간이 경과할수록 가장 높았으며, 총산은 초기에는 CM-$35^{\circ}C$에서 가장 낮은 함량을 나타내었지만 처리 6일 이후 서서히 증가하였고, CM-$25^{\circ}C$는 처리 완료일 까지 꾸준히 감소하는 것으로 나타났다. CM-$45^{\circ}C$는 초기와 비슷한 함량을 나타내어 다른 처리보다 높은 총산 함량을 나타내었다. 유기산 함량에 있어 사과산 함량은 CM-$35^{\circ}C$에서 가장 많이 감소하였고, 젖산 함량은 CM-$35^{\circ}C$에서 가장 높게 나타났다. 사과산 대사과련 효소활성을 살펴본 결과, malic enzyme과 malic dehydrogenase는 CM-$25^{\circ}C$와 CM-$35^{\circ}C$에서 가장 높은 효소활성을 나타내었지만 CM-$45^{\circ}C$에서는 초기부터 효소활성이 나타나지 않았다. oxalacetate decarboxylase 활성도 malic dehydrogenase 활성과 비슷한 경향을 나타내었다. pyruvate decarboxylase 활성은 다른 효소활성에 비해 활성이 낮았지만, CM-$45^{\circ}C$에서도 활성을 나타내었다. 반면 L-lactic dehydrogenase 활성은 어떤 처리구에서도 나타나지 않았다. 이와 같은 결과로부터 온도와 효소활성과의 관계에 있어 온도가 $40^{\circ}C$ 이상에서는 사과산 대사관련 효소가 활성을 나타내지 않는 것을 알 수 있었고, carbonic maceration 처리에서 사과산 감소가 효소의 작용에 크게 영향 받는 것을 확인할 수 있었지만, 젖산 생성에 대해서는 효소작용 외에 사과산 대사 미생물과 같은 다른 요인들에 대해 좀 더 깊이 있는 연구가 필요하다.

Differential Protein Expressions in Virus-Infected and Uninfected Trichomonas vaginalis

  • Protozoan viruses may influence the function and pathogenicity of the protozoa. Trichomonas vaginalis is a parasitic protozoan that could contain a double stranded RNA (dsRNA) virus, T. vaginalis virus (TVV). However, there are few reports on the properties of the virus. To further determine variations in protein expression of T. vaginalis, we detected 2 strains of T. vaginalis; the virus-infected ($V^+$) and uninfected ($V^-$) isolates to examine differentially expressed proteins upon TVV infection. Using a stable isotope N-terminal labeling strategy (iTRAQ) on soluble fractions to analyze proteomes, we identified 293 proteins, of which 50 were altered in $V^+$ compared with $V^-$ isolates. The results showed that the expression of 29 proteins was increased, and 21 proteins decreased in $V^+$ isolates. These differentially expressed proteins can be classified into 4 categories: ribosomal proteins, metabolic enzymes, heat shock proteins, and putative uncharacterized proteins. Quantitative PCR was used to detect 4 metabolic processes proteins: glycogen phosphorylase, malate dehydrogenase, triosephosphate isomerase, and glucose-6-phosphate isomerase, which were differentially expressed in $V^+$ and $V^-$ isolates. Our findings suggest that mRNA levels of these genes were consistent with protein expression levels. This study was the first which analyzed protein expression variations upon TVV infection. These observations will provide a basis for future studies concerning the possible roles of these proteins in host-parasite interactions.

Effect of ${\beta}$-Glucosidase as a Feed Supplementary on the Growth Performance, Digestive Enzymes and Physiology of Broilers

  • The effects of ${\beta}$-glucosidase on the overall growth performance and a set of physiological parameters of broilers were investigated. 240 male, one-day old Avine broiler chickswere randomly allocated to four treatment groups and fed with a corn-soybean meal supplemented with 0% (control), 0.2%, 0.4% and 0.6% ${\beta}$-glucosidase. The 0.2% ${\beta}$-glucosidase group, but not the 0.4% and 0.6% ${\beta}$-glucosidase groups, showed a significantly increased average daily weight gain (p<0.05) over that of the control. All three ${\beta}$-glucosidase feed groups showed significantly higher feed conversion ratios than the control group (p<0.05). Feed supplementation of 0.2% ${\beta}$-glucosidase significantly raised the contents of serum isoflavone aglycones as shown by decreases of genistin and daizin (p<0.01) and an increase of daidzein (p<0.01). The 0.2% ${\beta}$-glucosidase feeding significantly increased the intestinal amylase activity while it had little effect on lipase and trypsin activities (p>0.05). 0.2% ${\beta}$-glucosidase feeding also significant elevated the levels of highdensity lipoprotein cholesterol and malate dehydrogenase while lowering the level of low-density lipoprotein cholesterol (LDL-C). Finally, ${\beta}$-glucosidase improved the anti-oxidative activities of the animals; the 0.2% ${\beta}$-glucosidase feed group had higher activities of superoxide dismutase (p<0.05), glutathione peroxidase and glutathione reductase in the liver (p<0.05), and malondialdehyde level in the serum (p<0.05).

A Cytosolic Thioredoxin Acts as a Molecular Chaperone for Peroxisome Matrix Proteins as Well as Antioxidant in Peroxisome

  • Thioredoxin (TRX) is a disulfide reductase present ubiquitously in all taxa and plays an important role as a regulator of cellular redox state. Recently, a redox-independent, chaperone function has also been reported for some thioredoxins. We previously identified nodulin-35, the subunit of soybean uricase, as an interacting target of a cytosolic soybean thioredoxin, GmTRX. Here we report the further characterization of the interaction, which turns out to be independent of the disulfide reductase function and results in the co-localization of GmTRX and nodulin-35 in peroxisomes, suggesting a possible function of GmTRX in peroxisomes. In addition, the chaperone function of GmTRX was demonstrated in in vitro molecular chaperone activity assays including the thermal denaturation assay and malate dehydrogenase aggregation assay. Our results demonstrate that the target of GmTRX is not only confined to the nodulin-35, but many other peroxisomal proteins, including catalase (AtCAT), transthyretin-like protein 1 (AtTTL1), and acyl-coenzyme A oxidase 4 (AtACX4), also interact with the GmTRX. Together with an increased uricase activity of nodulin-35 and reduced ROS accumulation observed in the presence of GmTRX in our results, especially under heat shock and oxidative stress conditions, it appears that GmTRX represents a novel thioredoxin that is co-localized to the peroxisomes, possibly providing functional integrity to peroxisomal proteins.

Immunization with Brucella abortus recombinant proteins protects BALB/c mice from Brucella abortus 544 infection

  • This study evaluated the protective effects of a combination of eight B. abortus recombinant proteins that were cloned and expressed into a pMal vector system and $DH5{\alpha}$: nucleoside diphosphate kinase (rNdk), 50S ribosomal protein (rL7/L12), malate dehydrogenase (rMDH), DNA starvation/stationary phase protection protein (rDps), elongation factor (rTsf), arginase (rRocF), superoxide dismutase (rSodC), and riboflavin synthase subunit beta (rRibH). The proteins were induced, purified, and administered intraperitoneally into BALB/c mice. The mice were immunized three times at weeks 0, 2, and 5 and then infected intraperitoneally (IP) with $5{\times}10^4CFU$ of virulent B. abortus 544 one week after the last immunization. The spleens were collected and the bacterial burden was evaluated at four weeks post-infection. The results showed that this combination produced a significant reduction of the bacterial burden in the spleen with a log reduction of 1.01 compared to the PBS group. Cytokine analysis revealed induction of the cell-mediated immune response in that TNF (tumor necrosis factor) and proinflammatory cytokines IL-6 (Interleukin 6) and MCP-1 (macrophage chemoattractant protein-1) were elevated significantly. In summary, vaccination with a combination of eight different proteins induced a significant protective effect indicative of a cell mediated immune response.

Glutamic-oxaloacetic transaminase 1 regulates adipocyte differentiation by altering nicotinamide adenine dinucleotide phosphate content

  • Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p<0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism.
