IgG Humoral Immune Response to Extract Proteins of Malassezia Pachydermatis Isolated from a Dog with Atopic Dermatitis (Ad) (아토피 견에서 분리한 M. pachydermatis 추출 단백질에 대한 IgG 체액성 면역 반응의 연구)

  • Malassezia pachydermatis (M. pachydermatis) is a component of the normal cutaneous flora of the dog and atopic dermatitis (AD) is one of the most common diseases associated with Malassezia overgrowth in dogs. The purpose of this study was to investigate the humoral response (IgG) to extracts of M. pachydermatis of in a dog with AD. We used Western immunoblotting to identify allergens of M. pachydermatis. Gel electrophoresis of extracts proteins and immunoblotting of sera samples in both an atopic dog and a non-atopic dog were compared. Proteins of 18, 21, 26, 32, 34, 38, 40, 42, 46, 58, 64, 75, 85, and 120 kDa were observed in a serum of atopic dog. However, when serum of a non-atopic dog was used, protein bands were not identified except binding in 50 kDa protein. The results of this study indicate that atopic dogs with M. pachydermatis dermatitis may induce IgG response and also suggest that humoral response to M. pachydermatis could be important in pathogenesis of AD in dogs. However, further studies are required to identify roles of humoral response to M. pachydermatis in canine AD.

Production of Intracelluar Tyrosinase Inhibitor from Malassezia pachydermatis (Malassezia pachydermatis에 의한 세포 내 Tyrosinase 저해제의 생산)

  • A yeast strain SL-27 found to produce active intracellular tyrosinase inhibitor was screened from 972 kinds of yeasts. It was identified as Malassezia pachydermatis based on microbiological characteristics. The optimum pH and temperature for the growth of Malassezia pachydermatis SL-27 were pH 7.0 and $37^{\circ}C$, respectively. The optimal culture conditions for the production of tyrosinase inhibitor by Malassezia pachydermatis SL-27 were investigated. The optimal medium cimposition for tyrosinase inhibitor production was determined to be 1.0% casamino acid, 2.0% glucose, 0.1% $KH_2PO_4$, 0.05% $MgSo_{4-}7H20$ and each 0.01 of $CaCl_2$ and NaCl. Optimal initial pH and temperature for the production of tyrosinase inhibitor were pH 5.0 and $30^{\circ}C$, respectively. The maximum tyrosinase inhibitory activity of 84%/mL of cell-free extract was showed after 12 h of cultivation under the optimal culturing conditions.

A case of dermatitis associated with Malassezia pachydermatis in a dog (Malassezia pachydermatis에 의한 개의 피부염 치료 증례)

  • A 4-year-old female American cocker spaniel was hospitalized with 6 months prolonged dermal problems in The Veterinary Medical Teaching Hospital of Seoul National University. Initial noticed signs by owner of this dog were rashes and papules on ear part and they were spreaded whole body. On initial physical examinations, papules, pustules, crusts, and erythema were presented on whole body. Diameters of focal scaly and erythematous alopetic lesions were varied ranging 1 to 4cm. Skin scraping for detecting ectoparasites and wood's light examinations for dermatophyte were negative. Any bacteria were not detected but Malassezia pachydermatis was detected on cytological examination and fungal culture. The dog was treated with Itraconazole (5mg/kg, q12h) and Cephalexin (20mg/kg, q12h) orally and then bathed with 2% chlorhexidine shampoo twice per week for 5 weeks. From 12 weeks after starting therapy, all dermal problems were disappeared although some mild pyoderma were once relapsed. Regular chlorhexidine bathing treatment so far has been continued for preventing recurrence.

Risk Factors for Malassezia pachydermatis-associated Dermatitis in dogs: a Case-control Study (개에서 Malassezia pachydermatis에 의한 피부염의 위험요인에 관한 Case-control 연구)

  • Malassezia(Pityrosoprum) pachydermatis에 의한 피부염의 위험요인을 파악하고자 1997년 1월에서 1998년 12월까지 서울대학교 수의과대학 부속동물병원에 내원하여 본 질환으로 진단된 29두와 동일한 기간 동안 피부과 검사에서 임상적으로 건강한 개로 확인된 97 두의 대조군을 대상으로 분석하였다. 각 설명변수에 대한 교차비(odds ratio, OR)와 신뢰구간(confidence interval, CI)은 변수보정법을 사용하여 계산하였다. 다른 공변수를 보정한 상태에서 다중 로지스틱 회귀분석(multiple logistic regression analysis)을 시행한 결과 피부병기왕력(p=0.0059; OR, 4.44; CI, 1.5∼12.8)과 비만(p=0.0013; OR, 5.38; CI, 1.9∼14.9)은 유의한 위험요인으로 나타났다. 단변량 분석(univariate analysis)에서 주 사료로 고기를 섭취하는 경우 피부염의 위험이 높은 변수로 작용하였지만(p=0.0398; OR, 6.38; CI, 1.1∼37.3) 다른 공변수를 보정한 분석에서는 유의하지 않은 것으로 나타났다. 성별, 품종, 및 예방접종은 위험요인으로 작용하지 않았다.

Canine Dermatitix Associated with Malassezia pachydermatis (개에 있어서 Malassezia pachydermatis에 의한 피부염 1예)

  • 지방친화성 진균인 M. pachydermatis가 두 마리의 수캐에서 피부병의 원인균으로 증명되었다. 두 마리의 환축에서 흥반, 인설, 탈모 등이 여러 부위에서 발생하였으며, 광역항생물질과 코르티코스테로이드의 45일간 투여에 대해 아무런 임상적 반응을 나타내지 않았다. 발아를 나타내는 난원형 내지 원주형의 효모균이 KOH 및 PHOL 방법에 의해 환축의 피부병변으로부터 다수 증명되었고, 이들은 형태학적으로 M. pachydermatis와 일치하였다. 두 환축의 피부 병변부위 시료를 chloramphenicol을 첨가한 %37{\diamond}C$의 Saubouraud dextrose agar에 배양하면 이 진균이 반복하여 배양되었다. ketoconazole의 경구투여와 sulphur lime의 국소적용에는 치료반응을 잘 나타내었다.

Synergistic Growth Inhibition of Herbal Plant Extract Combinations against Candida albicans

  • Many skin diseases are caused by microbial infections. Representative pathogenic fungus and bacterium that cause skin diseases are Candida albicans and Staphylococcus aureus, respectively. Malassezia pachydermatis is a fungus that causes animal skin diseases. In this study, we propose a method for removing pathogenic microorganisms from the skin using relatively safe edible herbal extracts. Herbal extracts were screened for skin health through the removal of pathogenic microorganisms, and combinations for effective utilization of the screened extracts were identified. In this study, among methanol extracts of 240 edible plants, C. albicans, S. aureus, and M. pachydermatis were killed by extracts of 10 plants: Acori Gramineri Rhizoma, Angelicae Tenuissimae Radix, Cinnamomi Cortex, Cinnamomi Ramulus, Impatientis Semen, Magnoliae Cortex, Moutan Cortex Radicis, Phellodendri Cortex, Scutellariae Radix, and Syzygii Flos. By evaluating the synergistic antifungal activities against C. albicans using all 45 possible combinations of these 10 extracts, five new synergistic antifungal combinations, Acori Gramineri Rhizoma with Magnoliae Cortex extracts, Acori Gramineri Rhizoma with Phellodendri Cortex extracts, Angelicae Tenuissimae Radix with Magnoliae Cortex extracts, Magnoliae Cortex with Phellodendri Cortex extracts, and Phellodendri Cortex with Syzygii Flos extracts, were identified. By utilizing the selected extracts and five combinations with synergistic antifungal effects, this work provides materials and methods to develop new and safe methods for treating candidiasis using natural products.

Immune Reaction to Infection by Malassezia pachydercatis in Canine Ezternal Ear Canals (개 외이도내 Malassezia pachydercatis 감염에 대한 면역 반응)

  • Malassezia pachydercatis균은 개 외이염의 주된 원인균으로 알려져 있으나, 그 감염률은 지역에 따라 많은 차이가 있다. 그러나 그 기병론적인 역할에 대해서는 아직 명확히 밝혀지지 않았다. 이에 본문에서는 국내에서 사육되는 개의 외이염에서 Malassezia pachydercatis 균의 분리율을 조사하고, Malassezia pachydercatis에 대한 혈중 항체와 외이염과의 연관성을 밝혀 Malassezia pachydercatis에 의한 외이염 진단을 위한 도구로서 간접 ELISA 법의 유용성을 평가해 보고자 본 연구를 수행하였다. 총 112두 중, 외이염에 감염된 개 44두와 정상견 68두를 대상으로, 심부외이도에서 취한 가검물을 배양검사하였고, Malassezia pachydercatis 항원에 대한 혈청내 IgG와 IgM 역가를 간접 ELISA법에 의해 측정하였다. 살아있는 Malassezia pachydercatis 균을 3두의 정상견 외이도에 접종하여 Malassezia pachydercatis 균의 실험적 감염에 대한 조직병리학적 소견, 조직내 침투 정도, 그리고 접종 후 시간경과에 따른 IgG와 IgM의 혈중 역가변화를 측정하였다. Malassezia pachydercatis 균은 외이염이 있는 개 귀에서 가장 높게 분리(50.0%)되었고, 정상귀에서는 낮게 분리(6.3%)되었다. Malassezia pachydercatis 항원을 이용한 간접 ELISA 법 진단에서, Malassezia pachydercatis 양성배양인 외이염 개체에서 IgG와 IgM에 대한 민감도는 각각 41.7%, 20.8%이었고 특이도는 88.2%, 85.2% 이었다. 실험적으로 Malassezia pachydercatis 를 접종한 3두중 1두에서만 조직병리학적 변화와 모낭에 Malassezia pachydercatis 균체가 확인되었으며, IgG와 IgM 혈중역가도 유의성 있게 증가하였다. 국내에서 사육되는 개 외이염의 주된 원인균은 Malassezia pachydercatis 균이었고 이 균의 혈중항체는 유의성 있게 증가하였으며(p<0.005), 간접 ELISA법은 Malassezia pachydercatis 에 의한 개외이염의 유용한 진단도구임을 증명하였다.

High Prevalence of Lipid-Dependent Malassezia Infections in Dogs (개에서 조사된 높은 지방 친화성 Malassezia 감염율)

  • Malassezia (M.) is a member of normal mycobiota in warm-blooded vertebrates. Increased humidity is likely to be crucial in this infection. We studied the proportion of the species infected in dog during summer of Korea. Fifty samples were analyzed by PCR-RFLP and direct sequencing from June 2006 to October 2006. The study showed that lipid-dependent species was main pathogen (M. furfur; 86%, M. obtusa; 10%) while M. pachydermatis (4%) has only small portion. This result suggests that Malassezia infection has endemic characters that can be affected by the climate (temperature and humidity) in dogs.

Microfloras of otitis externa and normal external ear canals in dogs (개에서 외이도염 및 정상외이도에서 분리한 세균총에 대한 연구)

  • This study was conducted to examine the outbreak rate and the causative agents of otitis externa in 26 dogs (49 ears ; 23 dogs = bilateral, 3 dogs = unilateral), and the normal microfloras of external ear canal in 68 dogs(133 ears ; 65 dogs = bilateral, 3 dogs = unilateral ) in Taegu, 1997. The breed, living environment, sex, age and season distribution of otitic dogs were as follows : Dogs with erect and hairy ears(42.3%), pendulous and hairy ears(38.5%), indoor(92.3%), female(65.4%) and below one year old(38.5%) were more prevalent. According to season, otitis externa was mainly occurred between July and October. The major causative agents of canine otitis externa were Malassezia pachydermatis (32.7%), Staphylococcus aureus (26.5%) and S intermedius (16.3%). In the microorganism isolated 39 otitic ear canals, single infection was 53.8% and mixed infection was 46.2%. The normal microfloras of canine external ear canal were fungi including M pachydermatis, Aspergillus spp, Microsporum canis, Alternaria spp, Verticillium spp and Yeast, and bacteria including Staphylococcus spp(10 species including S xylosus), Bacillus spp, Corynebacterium spp, Listeria spp, Actinomyces pyogenes and Escherichia coli. No growth was 34.6%.
