Objectives : This research was observed by examining the external and internal parts and the quantity of the surface components of the wild type and cultivar of Angelica tenuissima in korea. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by the microscope and examined the quantity of the surface components. Results : 1. The cultivar has comparatively many thick roots while the wild type has many radicles, with its root and stem constituting the major part, and the size of its root is small. 2. In case of ferulic acid, the wild type contains 0.066% of it, while the cultivar, 0.031%, The wild type contains 4.258% of z-ligustilide and the cultivar, 1.956%. Conclusions : It was thought that there might be also some differences in their genetic characteristics, etc. On this, however, further in-depth study is needed.

Results : In the shape of the roots, the wild type had a very thin main root and 2 long roots, while the cultivar had a long root branched out from the short and thick main root making a square pyramid shape. Isoimperatorin was detected at Rt $17{\sim}18$ minutes. The wild type contained 0.56% while the cultivar contained it of 0.355%, thus the component of the wild type was about 1.5 times of that of the cultivar. Conclusions : It was thought that there might be also some differences in their genetic characteristics, etc.
Objctive:To clarify the criterion, the characteristic of varaious age of ginseng radix cultivated in Korea and China were studied. Method:The surface of the transverse section of the specimen was made into a slid by the Paraffin Section method, and then dyed by Safranine Malachite Green method. The samples were observed at the power of 400 by an optic microscope(Olympus, Japan). The component and flavor of ginseng radix were analyzed by TLC(Thinlayer Chromatography) and electronic nose(FOX3000, France). Result:Ginseng radix according to the growing district and various age were comparative analyzed by optic microscope, TLC and electronic nose. The results were as followings. 1. The external form of Korean ginseng is longer and brightness then Chinese ginseng. 2. The internal form of Korean and Chinese ginseng are similar to each other. 3. The component of Korean and Chinese ginseng in TLC are similar to each other. 4. The fragrance of Korean and Chinese ginseng are clearly different. Conclusion:The results in this study demonstrate that morphology and component of Korean ginseng are similar to Chinese, on the other hand, fragrance of Korean and Chinese ginseng are different.
In ancient times, many kinds of different inorganic pigments were used as colorants for making objects. These pigments are still evidentin well known objects such as Danchung, mural painting and Buddhist painting. This study discusses the results obtained from an analysis of the pigments used on the Buddha pigments. The results can be briefly summarized as below; Firstly, the microcrystalline structures revealed on the cross section of analyzed pigments, samples of which were taken from various parts of Buddha pigments show that different sizes and shapes of pigment particle were used for different purposes such as coloring, toning of the pigments. The arrangement of pigments and their usages are varied according to the owner temples and the place where it was created. Secondly, a result of the analysis on the composition and structure of the pigments shows that the main components in their composition are: Red pigments - Red lead($Pb_3O_4$) and Cinnabar(HgS)Green pigments - Malachite[$CuCO_3$.$Cu(OH)_2$] and Prussian Blue[$Fe4(Fe(CN)_6)_2$]Gold pigments - pure gold(Au)Yellow pigments - Orpiment($A_s2S_3)White pigments - Lead Cyanamide[$Pb_3(CO_3)_2(OH)_2]Ultramarine pigments - Azulite[$Cu_3(CO_3)_2(OH)_2]Especially, we knew that pigments used on the Ssanggye temple not repaired to the artificial synthetic pigment
The Wooden Seated Amitabha Buddha Triad and Altarpiece have not been the subjects of definitive species identification and scientific analysis. In this study, visual investigation, portable X-ray fluorescence (p-XRF), species identification, and lacquer layer observations were carried out to determine the original materials and conservation status. Cracks, exfoliation and discoloration were detected during the visual investigation. The p-XRF data indicated that chrome oxide green, malachite, gold, cadmium red, cinnabar, minium, hematite, lead white, ink stick (Carbon), and copper were used for pigments and the coronet accessory. Tilia spp., Abies spp., and Pinus spp. were identified from both the Buddha Triad and Altarpiece. Finally, lacquer layer analyses of the base layer, lacquer layer, orange pigment layer, and gold leaf layer observed modern synthetic pigments likely used in previous conservation. As the Korean Cultural Heritage Charter and the International Charter for the Conservation and Restoration of Monuments and Sites clearly state that objects of cultural heritage must be conserved using their original materials, future conservation of these objects should utilize the data obtained in this study to employ traditional materials. Furthermore, a deterioration map diagnosis can be applied together with the obtained analysis data to understand the conservation status of and inform an appropriate and authentic conservation treatment for the Buddha Triad and Altarpiece.

Several microorganisms from rat and human feces and lumen fluid of cows were screened for their ability to decolorize the synthetic dyes. Consequently, a novel dye-degrading strain AB&J was isolated. Taxonomic identification including 165 rDNA sequencing and phylogenetic analysis indicated that the isolate had 99.9% homology in its 165 rDNA base sequence with Clostridium perfringens. After 27 h Incubation with the strain, brilliant blue R, bromophenol blue, crystal violet, malachite green, methyl green, and methyl orange were decolorized by about 69.3%, 97.7%, 96.3%, 97.9%, 75.1%, and 97.2%, respectively. The triphenlmethane dye, bromophenol blue, was decolorized extensively by growing Clostridium perfringens AB&J cells in liquid cultures under anaerobic condition, although their growth was strongly inhibited in the initial stage of incubation. This group of dyes is toxic, depending on the concentration used. The dye was significantly decolorized at a relatively lower concentration of below 50 $\mu g \;ml^{-1}$, however, the growth of the cells was mostly suppressed at a dye concentration of 100 $\mu g \;ml^{-1}$. The decolorization activity in cell-free extracts was much higher in cytoplasm than in periplasm and cytoplasmic membrane. Therefore, the enzyme related uptake of bromophenol blue seemed to be localized in cytoplasm. The optimal pH and temperature of bromophenol blue uptake fur decolorization activities were 7.0 and 4$0^{\circ}C$, respectively.
Marasmius scorodonius secretes an extracellular laccase in potato dextrose broth, and this enzyme was purified up to 206-fold using $(NH_4)_2SO_4$ precipitation and a Hi-trap Q Sepharose column. The molecular mass of the purified laccase was estimated to be ~67 kDa by SDS-PAGE. The UV/vis spectrum of the enzyme was nontypical for laccases, and metal content analysis revealed that the enzyme contains 1 mole of Fe and Zn and 2 moles of Cu per mole of protein. The optimal pH for the enzymatic activity was 3.4, 4.0, and 4.6 with 2,2'-azino-bis(3-ethylbenzothazoline-6-sulfonate) (ABTS), guaiacol, and 2,6-dimethoxy phenol as the substrate, respectively. The optimal temperature of the enzyme was $75^{\circ}C$ with ABTS as the substrate. The enzyme was stable in the presence of some metal ions such as $Ca^{2+}$, $Cu^{2+}$, $Ni^{2+}$, $Mg^{2+}$, $Mn^{2+}$, $Ba^{2+}$, $Co^{2+}$, and $Zn^{2+}$ at a low concentration (1 mM), whereas $Fe^{2+}$ completely inhibited the enzymatic activity. The enzymatic reaction was strongly inhibited by metal chelators and thiol compounds except for EDTA. This enzyme directly decolorized Congo red, Malachite green, Crystal violet, and Methylene green dyes at various decolorization rates of 63-90%. In the presence of 1-hydroxybenzotriazole as a redox mediator, the decolorization of Reactive orange 16 and Remazol brilliant blue R was also achieved.
순환여과장치에 의한 어류사육시 어병방제의 목적으로 약품을 사용하는 일이 많다. 이 때, 이들 약품이 여과 조중의 산화분해세균을 사멸시켜 여과기능을 마비시킬 우려가 있어 여과를 중단시키고 약품을 처리하므로, 큰 불편을 주고 있다. 이번에 뱀장어 양식시에 흔히 사용되는 formalin 33ppm+malachite green 0.15ppm 혼합처리, dipterex 0.25ppm 처리 및 furanace 0.1ppm 처리가 여과기능에 미치는 영향을 주로 DO 소비량 및 ammonia 처리능력에서 시험검토하여 본 결과 별다를 영향을 미치지 않는다는 것을 알았다. 따라서 이상의 약물을 사용할 경우에 여과를 중단시키지 않아도 상관이 없다고 인정된다.
문화재 복원을 비롯한 회화용으로 많이 사용되는 녹색 안료 중 천연 무기안료인 석록과 뇌록의 물리적 특성 규명을 위하여 본 연구를 수행하였다. 석록은 Malachite가 주 구성광물이고, 뇌록은 Celadonite가 주 구성광물이다. 본 연구를 위하여 K사(社)의 A등급 석록, B등급 석록, 소성석록의 물리적 특성을 평가하였으며, 이와 비교하기 위하여 J사(社)의 송엽녹청, 소녹청을 평가하였다. 뇌록은 K사(社)의 뇌록을 평가하였다. 석록 및 송엽녹청의 소성 여부는 색도를 제외한 다른 물리적 특성에 큰 영향을 미치지 않는 것으로 판단되며, K사(社)의 석록과 소성석록과의 색차가 J사(社)의 송엽녹청과 소녹청의 색차보다 훨씬 높았다. K사(社)는 녹색도 $+a^*$가 J사(社)보다 가로로 넓게 분포하여 녹색의 다양한 색표현이 가능하다. 뇌록은 색도, 비중, 흡유량 등의 물리적 특성이 석록과는 많은 차이를 보였다. 뇌록의 색도는 $a^*$가 0에 가까이 분포하여 석록에 비하여 무채색에 더 가까웠고, 비중은 석록에 비해 작고, 흡유량은 2배 정도 컸다. 또한 뇌록과 암녹은 비록 구성 물질에 차이가 있어도 물리적 특성이 비슷하여 대체하여 사용 가능 할 것으로 보인다. 본 연구결과를 통하여 천연안료의 물리적 특성을 규명하여 품질기준을 확립하고, 문화재 보존, 복원의 선택적 사용에 대한 기초적인 자료로 활용되길 기대한다.
고대 대가야시대(6세기) 고분벽화에 사용된 제작기술을 연구하기 위해 벽화 구조와 재질에 대한 정밀분석을 실시하였다. 벽화는 석실을 축조 후 벽면과 천정에 모르타르를 바르고 그 위에 바탕칠층을 조성하여 채색하였다. 천장을 제외한 사면의 벽면에는 모르타르 위로 대부분 진흙이 덧발려져 있다. 모르타르는 모래는 사용되지 않았고, 순수한 칼슘성분의 물질들로 이루어져 있다. 또한 소성(燒成)이 완전히 진행되지 않은 불규칙한 크기의 패각들이 혼합되어 있으며, 모르타르의 석회질은 굴 껍질을 소성하여 얻어진 것으로 추정되었다. 바탕칠층은 백토(kaolinite($Al_2Si_2O_5(OH)_4$) 또는 halloysite($Al_2Si_2O_5(OH)_4{\cdot}2H2_O$))를 사용하였으며, 채색에 사용된 적색안료는 진사(cinnabar(HgS) 또는 vermilion), 녹색은 공작석(malachite($Cu_2CO_3(OH)_2$)) 그리고 백색은 연백(lead white($PbCO_3{\cdot}Pb(OH)_2$))이 쓰인 것으로 나타났다. 모르타르 위로 조성된 토양층은 주로 중립사 이하 크기의 입도가 낮은 점토질을 사용하여 얇고 치밀하게 조성하였으며, 모르타르층이 박락되고 상당기간이 경과 된 후 보수목적으로 마감한 것으로 추정된다. 굴 껍질을 소성하여 많든 석회를 모르타르로 사용한 것은 한국 고대 고분벽화 제작에 있어 유래 없는 사례이며, 내구성이 매우 떨어지는 특성을 지니고 있어 가야인들의 모르타르 제작기술이 동일시기의 고구려인들에 비해 상대적으로 떨어졌던 것으로 추측된다.
