DNA methylation is involved in epigenetic processes such as X-chromosome inactivation, imprinting and silencing of transposons. DNA methylation is a highly plastic and critical component of mammalian development The DNA methyltransferases (Dnmts) are responsible for the generation of genomic methylation patterns, which lead to transcriptional silencing. The maintenance DNA methyltransferase enzyme, Dnmt 1, and the de novo methyltransferase, Dnmt3a and Dnmt3b, are indispensable for development because mice homozygous for the targeted disruption of any of these genes are not viable. The occurrence of DNA methylation is not random, and it can result in gene silencing The mechanisms underlying these processes are poorly understood. It is well established that DNA methylation and histone deacetylation operate along a common mechanistic pathway to repress transcription through the action of methyl-binding domain proteins (MBDs), which are components of, or recruit, histone deacetylase (HDAC) complexes to methylated DNA. As a basis for future studies on the role of the DNA-methyl-transferase in porcine development, we have isolated and characterized a partial cDNA coding for the porcine Dnmt1. Total RNA of testis, lung and ovary was isolated with TRlzol according to the manufacture's specifications. 5 ug of total RNA was reverse transcribed with Super Script II in the presence of porcine Dnmt 1 specific primers. Standard PCRs were performed in a total volume of 50 ul with cDNA as template. Two DNA fragmenets in different position were produced about 700bp, 1500bp and were cloned into pCR II-TOPO according to the manufacture's specification. Assembly of all sequences resulted in a cDNA from 158bp of 5'to 4861bp of 3'compare with the known human maintenance methyltransferase. Now, we are cloning the unknown Dnmt 1 region by 5'-RACE method and expression of Dnmt 1 in tissues from adult porcine animals.

This study identified the materials and construction methods of 'Old Wall' in 13 villages which were designated as National Registered Cultural Heritage at the time of designation and examined the their structural changes based on field survey. The results are as follows: First, the 'Old Wall' consisted of 10 Soil-Stone Wall and 5 Stone Wall. At the time of designation, Stone Wall, which was built irregularly by dry-construction of natural stones, is similar in shape, but Soil-Stone Wall showed difference by the construction method of making used stones, joints, and faces. Second, the study extracted the changes of 'Old Wall' by repair and examined the changes of construction methods as well as the substitution and addition of materials of structure. The wall-roof was built with cement roof-tile and asbestos slate which have the advantage improve durability and cost-effectiveness. In addition, tile-mouth soil was added to korean traditional roof-tile to prevent rainwater from flowing in. Besides, to improve constructional convenience, the natural stone of the wall-body was replaced with blast stone, float stone and cut stone. Cement block, cement brick and cement mortar were frequently used to repair as well. As Soil-Stone Wall was transformed from irregular pattern-construction to comb pattern-construction and wet-construction was changed to dry-construction, it caused landscape and structural problems. Also, the layer of cement mortar applied to wall-foundation blocked the flow of rainwater that was induced by dry-construction of natural stones. Third, the study regarded that the problem with the repair of 'Old Wall' may occur as it is located in living space, because the owner of the wall could repair for the minor damages without technical knowledge. In addition, it is difficult for repair companies in charge of maintenance of Cultural Heritage to supply local materials, and it is differential construction specifications are not applied.

The majority of high-speed railway bridges along the domestic Gyeongbu and Honam lines feature a PSC-box type structure with a span length ranging from 35 to 40m, which typically exhibits a first bending natural frequency of approximately 4 to 5Hz. When KTX high-speed trains transverse these bridges at speeds ranging from 290 to 310km/h, the vibration induced by the trains approaches the first bending natural frequency of the bridge. Furthermore, with the upcoming operation of a EMU-320 high-speed train and the anticipated increase in the speeds of these high-speed trains, there is a need to analyze the dynamic response of high-speed railway bridges. For this, based on measured responses from actual railway bridges, a numerical model was constructed using a numerical model updating technique. The dynamic response of the updated numerical model exhibited a strong agreement with the measured response from the actual railway bridges. Subsequently, this updated model was utilized to analyze the dynamic response characteristics of the bridges when KTX and EMU-320 trains operate at increased speeds. The maximum vertical displacement and acceleration at the mid-span of the bridges were also compared to those specified in the railway design standard with the increasing speed of KTX and EMU-320.

Fracture toughness is a material property for crack initiation and propagation in fracture mechanics. For mode I fracture toughness measurement in concrete, RILEM committees 89-FMT proposed three-point bend tests based on the two-parameter fracture model. But, there is no proposed test method as a standard for mixed mode test for now. And RILEM three-point bend test procedure is complicate. Therefore, in this study, brazilian disks of various size were designed as the concrete with a similar specified concrete strength and maximum size of coarse aggregate($G_{max}$) were respectively 20mm and 40mm. And mode I fracture toughness of brazilian disks was compared with that of RILEM three-point bend test. As a result, it was suggested appropriate size(thickness, diameter) and notch length ratio of brazilan disk on the $G_{max}$. And it was verified that stress intensity factors for mixed mode can be easily calculated with the disk specimen. Stress intensity factors of a concrete brazilian disk were evaluated with finite element analysis and five terms approximation for comparison.

Number of long-span bridge construction has been increased recently so that seismic consideration of design has become significant. To adapt such significance to design, seismic design in the newly revised 'Cable Steel Bridge Design Handbook' specifies some of wave travelling effect and local site effect. In this study, a cable-stayed bridge with main span of 500m is analysed having variables of uniform excitation, wave travelling effect, and wave travelling effect plus local site effect. Result shows that wave travelling effect in cable-stayed bridge affects considerably to its seismic response under weak soil condition even though the span length is relatively short. What's more, regardless of soil type, the seismic response has become higher for analysis with wave travelling effect and local site effect than with wave travelling effect only. Consequently, in seismic response analysis of long-span bridge, consideration should be given to application of wave travelling effect and local site effect.

In this paper, large scale experimental model tests were performed to investigate the distribution of earth pressure acting on embedded rigid pipes having different bedding conditions. For these tests, very light weighted EPS blocks were installed at top and bottom of the rigid pipe and Jumunjin Standard Sand was used as a ground material. As results of model tests, for the case of no bedding on the pipe, the measured pressure at the bottom of the pipe was $4.96_{tf/m^2}$ whereas they were in the range of $1.87{\sim}4.96_{tf/m^2}$ in the case of EPS beddings being installed at the top and the bottom of the pipe. Therefore, for the case of EPS bedding being installed, the ratio of reduced pressures acting on the pipe, compared with the case of no EPS beddings, were in the rage of 16~62%. As a result of parametric test with changing the locations of EPS bedding, the trend of reducing the stress acting on the pipe was in the order of bottom bedding, top bedding, and top and bottom bedding. Effect of bedding positions on the reduced magnitude of acting pressure on the pipe was more significant in the case of top bedding than in the case of the bottom bedding.

Sounding data is the essential source for the safety of ships navigation system, and fundamental to the reasonable usage and maintenance of the ocean as well. As IT tech, positioning equipment such as GPS and INS, echo sounder are developed, recently, the precise submarine topography database bas been built by Multi-Beam Echo Sounder. However, MBES data includes some inevitable error caused by several factor, and some data have errors where the terrain is wobble. The error, which causes the $moir\acute{e}$ pattern error is the main factor hindering the accuracy of MBES data results, and therefore it is necessary to figure out the main cause of the error for the improvement of the accuracy by removing error data. On this research, the main cause of the error data is studied by analyzing motion sensor value of data including the $moir\acute{e}$ pattern error. Thus, as the result of examination, it turns out that the $moir\acute{e}$ pattern error is related to the standard deviation of Roll, and error data values are results of the non-correspondence between Swath data and Roll values caused by the drastic change of Roll values. Accordingly, the error data is removed by comparing between the gradient of Swath data and Roll values. Finally, as the result of removing error data, it is expected to be able to estimate the quality of MBES using the standard deviation of Motion sensor's Roll value, and calculate the additive error factor, which minimize non-corresponding data, and also this research must be contributed to improve the accuracy of sounding for small vessels with lots of motion in the bad circumstance for navigation.

Recently, many accidents occur frequently because sprinklers aren't installed or don't work right when there are outbreak of fire in houses and aggregate buildings. Therefore, they can result in damage for humans and loss of property. Sprinklers are the most appropriate which can extinguish buildings' fire in the initial stages. Through lack of legal system, in domestic cases, sprinklers cannot operate their inherent performance. Domestic standard simply classifies installation objects according to types of business and forms of buildings, also divides into uses and floors of buildings. Especially it only regulates that sprinklers must be installed every floors in particular fire buildings that have more than eleven floors. While it doesn't need to install sprinklers below ten floors, so we are threatened the safety. In this study, we derived causes and implications by analyzing concepts of sprinklers facilities, installation standards in domestic and foreign legal system, and recent cases that expanded damages in fire accidents because of weak point of installation and control standards. In domestic cases, as a result, government has to provide an institutional strategy and law that regulate duty to install sprinklers to all aggregate buildings regardless of floors in terms of new buildings. Also, if someone who has existing buildings wanted to install them, government would guarantee subsidy to encourage installation. In addition, government supervises fire-fighting activities when there are fire by compensating standards about regular inspection by a qualified technician, operation and maintenance of sprinklers as well as reinforcement of administrative criteria.

Many studies have been implemented to manage solar plant being supplied widely in recent years. This study analyzed heat emission of solar cell using unmanned aerial vehicle(UAV)-based thermal infrared sensor, and major conclusions are as belows. Firstly, orthomosaic image and digital surface model(DSM) data were acquired using UAV-based RGB sensor, and solar light module layer necessary to analyze the heat emission of solar cell was constructed by these data. Also as a result of horizontal error into validation points using virtual reference service(VRS) survey for evaluating the location accuracy of solar light module layer, higher location accuracy could be acquired like standard error of $dx={\pm}2.4cm$ and $dy={\pm}3.2cm$. And this study installed rubber patch to test the heat emission of solar cell and could analyzed efficiently the location of rubber patch being emitted heat using UAV-based thermal infrared sensor. Also standard error showd as ${\pm}3.5%$ in analysis between calculated cell ratio by rubber patch and analyzed cell ratio by UAV-based thermal infrared sensor. Therefore, it could be efficiently analyzed to heat emission of solar cell using UAV-based thermal infrared sensor. Also efficient maintenance of solar plant could be possible through extracting the code of solar light module being emitted of heat automatically.
The use of water by cities is increasing owing to industrialization, the concentration of population, and the enhancement of the standard of living. Accordingly, the amount of waste water is also increasing, and the degree of pollution of the water system is rising. In order to solve this problem, it is necessary to remove organisms and suspended particles as well as the products of eutrophication such as nitrates and phosphates. This study developed a high-end treatment engineering solution with maximum efficiency and lower costs by researching and developing a advanced treatment engineering solution with the use of Biosorption. As a result, the study conducted a test with a $50m^3/day$ Pilot Scale Plant by developing treatment engineering so that only the secondary treatment satisfies the standard of water quality and which provided optimal treatment efficiency along with convenient maintenance and management. The removal of organisms, which has to be pursued first for realizing nitrification during the test period, was made in such a way that there would be no oxidation by microorganisms in the reactor while preparing oxygen as an inhibitor for the growth of microorganism in the course of moving toward the primary settling pond. The study introduced microorganisms in the endogeneous respiration stage to perform adhesion, absorption, and filtering by bringing them into contact with the inflowing water with the use of a sludge returning from the secondary settling pond. Also a test was conducted to determine how effective the microorganisms are as an inner source of carbon. The HRT(Hydraulic Retention Time) in the nitrification tank (aerobic tank) could be reduced to two hours or below, and the stable treatment efficiency of the process using the organisms absorbed in the NAR reactor as a source of carbon could be proven. Also, given that the anaerobic condition of the pre-treatment tank becomes basic in the area of phosphate discharge, it was found that there was excellent efficiency for the removal of phosphate when the pre-treatment tank induced the discharge of phosphate and the polishing reactor induced the uptake of phosphate. The removal efficiency was shown to be about 94.4% for $BOD_5$. 90.7% for $COD_{Cr}$ 84.3% for $COD_{Mn}$, 96.0% for SS, 77.3% for TN, and 96.0% for TP.
