Human ChlR1 Stimulates Endonuclease Activity of hFen1 Independently of ATPase Activity

  • Human ChlR1 protein (hChlR1), a member of the cohesion establishment factor family, plays an important role in the segregation of sister chromatids for maintenance of genome integrity. We previously reported that hChlR1 interacts with hFen1 and stimulates its nuclease activity on the flap-structured DNA substrate covered with RPA. To elucidate the relationship between hChlR1 and Okazaki fragment processing, the effect of hChlR1 on in vitro nuclease activities of hFen1 and hDna2 was examined. Independent of ATPase activity, hChlR1 stimulated endonuclease activity of hFen1 but not that of hDna2. Our findings suggest that the acceleration of Okazaki fragment processing near cohesions may aid in reducing the size of the replication machinery, thereby facilitating its entry through the cohesin ring.

Activity Determination, Kinetic Analyses and Isoenzyme Identification of Gamma Glutamyltransferase in Human Neutrophils

  • Gamma-glutamyltransferase (GGT, EC 2.3.2.2) which hydrolyzes glutathione (GSH), is required for the maintenance of normal intracellular GSH concentration. GGT is a membrane enzyme present in leukocytes and platelets. Its activity has also been observed in human neutrophils. In this study, GGT was purified from Triton X-100 solubilized neutrophils and its kinetic parameters were determined. For kinetic analyses of transpeptidation reaction, $\gamma$-glutamyl p-nitroanilide was used as the substrate and glycylglycine as the acceptor. Apparent $K_m$ values were determined as 1.8 mM for $\gamma$-glutamyl p-nitroanilide and 16.9 mM for glycylglycine. The optimum pH of GGT activity was 8.2 and the optimum temperature was $37^{\circ}C$. It had thermal stability with 58% relative activity at $56^{\circ}C$ for 30 min incubation. L-serine, in the presence of borate, was detected as the competetive inhibitor. Bromcresol green inhibited neutrophil GGT activity as a noncompetetive inhibitor. The neutrophils seem to contain only the isoenzyme that is present in platelets. We characterized the kinetic properties and compared the type of the isoenzyme of neutrophil GGT with platelet GGT via polyacrylamide gel electrophoresis (PAGE) under a standart set of conditions.

Comparative Study for the Validation of TPNSim++ and its Applicability to Military Simulation (TPNSim++의 검증 및 군사 시뮬레이션 분야의 활용성)

  • TPNSim++ is the object-oriented framework for the discrete event simulation of military systems, developed by authors. The simulation world view of TPNSim++ is on the basis of activity scanning. TPNSim++ is implemented as C++ class library under Windows 95/NT. It uses the extended timed Petri nets which are called TPNSim nets for simulation modeling tool. The aim of this study is to conduct a comparative study of TPNSim++ and SLAM II in the simulation of military maintenance systems for the validation and the applicability of TPNSim++. From this study, TPNSim++ and SLAMII have given the same results under the equivalent assumption. Thus we can get the validation of TPNSim++ and its applicability to the simulation of maintenance systems.

  • Reliability analysis and prediction of next failure time is critical to sustain weapon systems, concerning scheduled maintenance, spare parts replacement and maintenance interventions, etc. Since 1981, many methodology derived from various probabilistic and statistical theories has been suggested to do that activity. Nowadays, many A.I. tools have been used to support these predictions. Support Vector Regression(SVR) is a nonlinear regression technique extended from support vector machine. SVR can fit data flexibly and it has a wide variety of applications. This paper utilizes SVM and SVR with combining time series to predict the next failure time based on historical failure data. A numerical case using failure data from the military equipment is presented to demonstrate the performance of the proposed approach. Finally, the proposed approach is proved meaningful to predict next failure point and to estimate instantaneous failure rate and MTBF.

  • Regression testing is a very costly process performed primarily as a software maintenance activity. It is the process of retesting the modified parts of the software and ensuring that no new errors have been introduced into previously tested source code due to these modifications. A regression test selection technique selects an appropriate number of test cases from a test suite that might expose a fault in the modified program. In this paper, we propose both a regression test selection and prioritization technique. We implemented our regression test selection technique and demonstrated in two case studies that our technique is effective regarding selecting and prioritizing test cases. The results show that our technique may significantly reduce the number of test cases and thus the cost and resources for performing regression testing on modified software.

  • This paper considers the differences in the software execution environments in the testing phase and the operational phase to determine the optimal release time and warranty period of software systems. We formulate equations for the total expected software cost until the end of the software life cycle based on the NHPP. In addition, we derive the optimal release time that minimizes the total expected software cost for an imperfect debugging software reliability model. Finally, we analyze the sensitivity of the optimal testing and maintenance design related to variation of the cost model parameters based on the fault data observed in the actual testing process, and discuss the quantitative properties of the proposed model.

  • Recently, the environment problems are risen to social problems. People have requested the convenience of a human activity and the enhancement of a human quality. The facility was, however, emphasized on the acquisition and treatment of water with the construction and maintenance of a irrigation and drainage structure. The concept of an environmental friendship has been introduced to change our society structure to an environment friendship at 21th century. But the concept lacks theoretical and systematic foundation. So, The maintenance technique, concrete products and construction methods will be made to be an environment friendship. The standards for environment friendship will be presented.

  • We have developed the global assessment system for medium voltage power cable systems. Our purpose of diagnostic activity is the economic discrimination and maintenance of bad cables which is likely to cause cable system failure. We have atapted the Voltage Decay Method to construct the power cable assessment system for our diagnostic purpose. From the field application test results, we have concluded that our system is successful and convenient for the discrimination and maintenance of the damaged cables economically.

  • The seasonal variation in the activity of sucrose synthase, ADP-glucose pyrophosphorylase and UDP-glucose pyrophosphorylase in roots of Panax ginseng C.A.Meyer have been studied. It was revealed that sucrose synthase and ADP-glucose pyrophosphorylase are adaptive enzymes and can serve as markers of sink strength, while UDP-glucose pyrophosphorylase is the maintenance enzyme. The average day temperature exceeded 24。C appeared to cause the disturbance in refilling process, affecting the starch synthesis. Study on the dependence of oxygen consumption in stele tissue with temperature revealed the sharp accelerating of this process after 24。C.

The Effect and Influencing Mechanism of TPM Factors to Performance

  • This study tries to analyze how TPM works in domestic manufacturing industry by estimating two-stage model. First stage tests the effects of five TPM-factor variables (TFV : (1) Small group activity & Autonomous maintenance, (2) Education & Training, (3) Planned maintenance, (4) improving effectiveness of each piece of facility (5) Safety & Environment) to two TPM-performance variables. Second stage tests how two TPVs affect the industry's productivity level. By combining these two stages, this study uses a model to explain how TPM, represented by TFVs, works to improve productivity via TPVs. Multivariate and univariate regression and correlation analyses were peformed. It is shown that five TFVs works in two different ways to improve the industry's productivity level. In the second stage, overall equipment effectiveness has relatively more significant effects to the productivity level.
