Industrial potential of domestic Zanthoxylum piperitum and Zanthoxylum schinifolium: Protective effect of both extracts on high glucose-induced neurotoxicity (국내산 초피와 산초의 산업적 활용 가능성: 고당으로 유도된 뇌신경세포 독성에 대한 추출물의 보호 효과)

  • This study focused on the in vitro investigation of antioxidant and anti-diabetic activities, along with neuroprotection against high glucose-induced cytotoxicity, in order to evaluate the physiological effects of Zanthoxylum piperitum and Zanthoxylum schinifolium. The highest total phenolic content was measured in the 40% ethanolic extracts of Zanthoxylum piperitum (EZP) and Zanthoxylum schinifolium (EZS). The in vitro EZP antioxidant activity showed a relatively higher ABTS/DPPH radical scavenging activity and malondialdehyde inhibitory effect than that of EZS. The EZP inhibited carbohydrate hydrolysis (α-glucosidase and α-amylase) more efficiently than EZS in anti-diabetic tests. However, EZS showed a more efficient inhibition of advanced glycation end-products formation than EZP. In addition, both EZP and EZS effectively protected human-derived neuronal cells from high glucose-induced cytotoxicity. Finally, the physiological compounds were analyzed using UPLC IMS-QTOF/MSE, and the main EZP (quercetin-3-O-glucoside and 3-caffeoylquinic acid) and EZS (5-caffeoylquinic acid) compounds were identified as phenolic compounds.

Removal Efficiency of the Deodorization Equipment and Characteristics of Malodor during the Process in Co-treatment of Sewage and Food Waste of Su-young Wastewater Treatment Plant in Busan (부산수영하수처리장 하수와 음식물쓰레기 병합처리 시 공정별 악취특성 및 후처리시설 효율평가)

  • Environmental issues are being paid more attention due to income growth, urban overcrowding, and population growth in Korea. Among the various environmental problems, odor damage is the one of the serious factors. To take example for food waste combination treatment in Su-young wastewater treatment plant in Busan, many complaints occurred because this plant locate around residential areas. The purpose of this work is not only to analyze odorous elements and their contributions but also to evaluate odor quotient (OQ), sum of odor quotient (SOQ), and treatment efficiency of bio-filter. The results of dilution sensory test of complex odor, grinder, leachate, hopper indicated higher order complex odors happen in July and August. The main odorous elements consisted of hydrogen sulfide, ammonia, methly mercaptan and acetaldehyde, which were analyzed by instrumental detection method, and methyl mercaptan was exceeded over 3,571 times of threshold. In addition, result of contribution of odor was methyl mercaptan (49.95 to 59.08%), hydrogen sulfide (20.43 to 29.27%), trimethylamine (8.82 to 13.42%) and acetaldehyde (9.17 to 11.35%). Other facilities were compared with the contribution of the odor using OQ and SOQ during the process. Sulfur compounds, acetaldehyde, and trimethylamine are high contribution of odor using OQ as well as odor intensity of grinding process is highest. As a result, sulfur compounds (e.g., methyl mercaptan and hydrogen sulfide) are highest for OQ and SOQ of grinding process is highest as 7,067. The removal efficiency of deodorization equipment was more than 90.00% in ammonia and amines, but the average efficiency of sulfur compounds was 53.51%. Thus, this facility is more higher contribution of acetaldehyde and trimethylamine than other treatment facilities. And food waste treatment in environmental area needs to consider appropriate capacity and refers to other bio-filter operating conditions.

Study on the Qualitative Discrimination of White, Red, and Black Ginseng Extract (백삼,홍삼과 흑삼 추출물의 정성적 구별법에 관한 연구)

  • This study analyzed the maltol quality, composition ratio of fatty acids, and contents of phenolic compounds in white ginseng extracts(four types), red ginseng extracts(five types), black ginseng extracts(two types), and Chinese ginseng extracts(nine types). By examining patterns in these measurements, we determined the characteristic factors of the extracts and measured the possibility of qualitative analysis. In the analysis of maltol using TLC, white ginseng extracts were not detected while red and black ginseng extracts were detected, so the possibility of detection was considered as a characteristic factor for qualitative analysis. Regarding the composition of fatty acids, palmitic and linoleic acids were the main fatty acids in the ginseng extracts palmitic acid was high in white ginseng extracts while linoleic was low in red ginseng extracts. Regarding the ratio(Pal/Lin) of the two fatty acids, there was a large difference between white ginseng extracts(56.7~64.3%) and red ginseng extracts(32.0~38.5%), and these figures seemed to be characteristic factors for the analysis. For the phenolic compounds, extracts contained maltol, caffeic acid, syringic acid, p-coumaric acid, ferulic acid, and cinnamic acid. White ginseng extracts contained similar percentages of phenolic compounds while red ginseng extracts had high maltol content. According to the measurement results of the percentages of maltol and cinnamic acid, white ginseng extracts showed values below five, whereas red and black ginseng extracts showed 53~289, which was also a characteristic factor for qualitative analysis. Consequently, we found that we can differentiate between ginseng extracts using characteristic factors that we analyzed in an experiment on white ginseng extracts from China.

Free Radical Scavenging Effect and Oxidative Stress Protective Activity of Domestic Processed Polygoni Multiflori Radix (국내산 법제 하수오의 라디칼 소거능 및 산화적 스트레스 개선 효과)

  • In this study, we confirmed biological compounds from methanol (MeOH) extract of processed Polygoni Multiflori Radix (PPMR), and the radical scavenging effect and oxidative stress protective activity of MeOH extract of PPMR were investigated under in vitro conditions using LLC-$PK_1$ renal epithelial cells. In HPLC analysis, MeOH extract of PPMR contained four species of biological compounds named 2,3,5,4'-tetrahydroxystilbene 2-O-${\beta}$-D-glucoside, emodin, chrysophanol, and rhein. 2,3,5,4'-Tetrahydroxystilbene 2-O-${\beta}$-D-glucoside was detected as the main compound in PPMR as 115.02 mg/kg. MeOH extract of PPMR showed 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) diammonium salt (ABTS), and hydroxyl radical scavenging activities in a concentration- dependent manner. In particular, upon $50{\mu}g/mL$ of PPMR extract treatment, DPPH, ABTS, and hydroxyl radical scavenging activities were approximately 48.4%, 57.9%, and 81.2%, respectively. LLC-$PK_1$ cell viability declined in response to oxidative stress induced by pyrogallol, sodium nitroprusside (SNP), and morpholinosydnonimine (SIN-1) generators of NO, $O_2{^-}$, and $ONOO^-$, respectively. However, MeOH extract of PPMR significantly and dose-dependently inhibited oxidative-stressed LLC-$PK_1$ cell cytotoxicity. In fact, upon $50{\mu}g/mL$ of PPMR extract treatment, LLC-$PK_1$ cell viabilities were approximately 82.1%, 89.1%, and 77.6% compared to stress levels induced by pyrogallol, SNP, and SIN-1, respectively.

Antifungal activity of extracts from Chamaecyparis obtusa and Pseudotsuga menziesii against Trichoderma spp. (국내산 침엽수 추출물의 Trichoderma spp.에 대한 항균활성)

  • The aim of this study was development of natural antifungal compounds from softwood. We investigated antifungal activities of extracts from Pseudotsuga menziesii and Chamaecyparis obtusa against Tricholderma genus which is virus causing green mold disease and analyzed antifungal compounds by Gas chromatography -Mass Spetrometer. Extracts from P. menziesii had inhibition activities against Tricholderma genus on 1,000 ppm and had high antifungal activities against T. viride by 70.1%, T. harzianum by 67.3% and T. aggressivum by 64.7% on 4,000 ppm. And extracts from C. obtusa had antifungal activities against Tricholderma genus on 1,000 ppm and had high antifungal activities against T. viride by 63.2%, T. harzianum by 59.3% and T. aggressivum by 59.1% on 4,000 ppm. But mixing compounds which are made from P. menziesii and C. obtusa extracts by variety ratio had lower antifungal activities than original extracts. Main antifungal active components of P. menziesii extracts against Tricholderma genus were 2-Isopropoxy-ethylamine 46.5%, epifluorohydrin 8.6%, trans-2,3-Di-methyloxirane 7.6%, (IR)-(-)-Myrtenal 6.0%, 2-Methoxy-4-Vinylphenol 3.9% and benzaldehyde 2.8%. In case of C. obtusa extracts, they were ${\alpha}$-Terpinenyl acetate 14.9%, Sabinene 10.9%, dl-Limonene 9.6%, ${\alpha}$-Terpinolene 7.5% and ${\alpha}$-Pinene 7.1%. As mentioned above, these results revealed extracts from P. menziesii and C. obtusa of softwood could be used as potential agents to inhibit Trichoderma genus.

Effects of Compounds Isolated from an Ethanol Extract of the Sclerotium of Wolfiporia hoelen on Osteoblast Differentiation and Osteoclast Formation (복령 균핵의 에탄올 추출물에서 분리한 화합물의 조골세포 분화 촉진 및 파골세포 생성 억제 효과)

  • Wolfiporia hoelen (Fr.) Y.C.Dai & V. Papp, commonly known as Poria cocos, is a significant traditional herb used for medicinal and culinary purposes Asian and European countries. Many studies have confirmed that the main components of W. hoelen have pharmacological activities and thatits extract has been shown to affect bone metabolism. This study aimed to the potential of a 50% ethanol extract of the sclerotium of W. hoelen for preventing and treating bone diseases. The ethanol extract was systematically fractionated using n-hexane, dichloromethane, and ethyl acetate. The dichloromethane fraction caused an approximately 29% increase in alkaline phosphatase (ALP) differentiation activity in C2C12 cells compared to the control. Four compounds isolated from this active dichloromethane fraction were identified through instrumental analysis and literature references as 3α-dehydrotrametenolic acid, ergosterol, pachymic acid, and dehydrotumulosic acid. All four compounds were evaluated at increasing concentrations (1, 3, 10, 30, and 100 μM) to determine their effects on ALP differentiation activity in C2C12 cells and RANKL-induced inhibition activity in bone marrow macrophages (BMMs), with a concurrent assessment of cytotoxicity at these concentrations. At a concentration of 3 μM, dehydrotumulosic acid caused a 160% increase in ALP activity, 24% higher than in the BMP-2 control. BMMs treated with dehydrotumulosic acid at concentrations between 10 and 100 μM showed a substantial 15-86% decrease in RANKL-induced inhibition activity compared to the control, with distinct patterns of RANKL inhibition and cytotoxicity observed at 10 μM. These findings suggest that the ethanol extract from the sclerotium of W. hoelen has potential to modulate bone-cell differentiation, while highlighting the possible benefits of dehydrotumulosic acid isolated from the dichloromethane fraction of W. hoelen for preventing and treating osteoporosis.

Change of Fragrant Components by Flowering Stages in Lilium Oriental Hybrid 'Casa Blanca' (오리엔탈 나리 '카사블랑카'의 개화단계별 향기성분 변화)

  • This experiment was conducted to find out suitable extraction time of available fragrant component in Lilium Oriental Hybrida 'Casa Blanca' based on qualitative and quantitative variation of its fragrant component in its flowering stages. The content of essential oil in its flowering stages increased with the progress of flowering stages except flower bud stage and it had the most oil content in its stage after full bloom. The analysis of the essential oil in Lilium Oriental Hybrida 'Casa Blanca' in its flowering stages shows that its main constituents were farnesol (18.96%), benzyl salicylate (13.81%), butyl-hydroxy toluene (12.87%), geranyl linalool isomer (7.46%), isoeugenol (7.17%) in its each stage. Benzenoids had much content at half bloom stage and full bloom stage while fatty acid derivatives had much content at initial flowering stage and after flowering stage. Most benzenoids such as butyl-hydroxy toluene and isoeugenol, which are some of main constituents of Lilium Oriental Hybrida 'Casa Blanca' had strong antioxidant effect. So they were expected to be used as antioxidant agents for food, feed, vegetable oil. The content of monoterpene compounds like geranyl linalool isomer increased at its later stage. Sesquiterpene such as farnesol, which is main component of lily existed only at full bloom stage. Therefore farnesol and geranyl linalool isomer of Lilium Oriental Hybrida 'Casa Blanca' are expected to be used for important spices with good fragrance. The suitable extraction time for the usable main constituents of Lilium Oriental Hybrida 'Casa Blanca' is as follows; farnesol and benzyl salicylate: at full bloom stage, butyl-hydroxy toluene: at half bloom stage, geranyl linalool isomer and isoeugenol: at stage after full bloom. Finally there was variation in essential oil components and contents of Lilium Oriental Hybrida 'Casa Blanca' in its flowering stages and they are expected to be used usefully for flavor industry, food industry, aromatherapy, when they are extracted at their suitable extraction time.

Nutritional Effects of Paper Board Sludge on the Soybean(Glycine max. L.) (대두(大豆)에 대(對)한 제지(製紙) Sludge의 영양학적(營養學的) 연구(昭究))

  • The paper board sludge(PBS) itself and compost sludge manure(CSM) mixed with sawdust, fowl droppings and urea to the PBS were treated to soybean plants to find the effects of growing characters, yield components, and nutritional compounds in the plant tissues. 1. Percentages of missing plants were 5-9% and 3.6-4.1% in the treatments of PBS and CSM, respectively. After that, the plants were restored to normal conditions. Anyway it is not desirable to use the paper board sludge and immature compost sludge manure in seeding time or to young seedlings. 2. Growth of the plant height was retarded in early growing phase, but it was normal in later stages. And the width and length of the largest leaf, numbers of main stem nodes and pods were not significant among the treatments. 3. The yield intends to increase through the treatments of 1,200, 1,600, and 2,000Kg PBS per 10a. In the CSM treatments with high rate of sawdust, fowl droppings and urea, the numbers of pods and grains were higher than the treatment of high content of PBS. 100-grain and one liter weights were opposite intention. It was suggested that the excess nitrogen amounts from the compost sludge manure than conventional fertilization affected to the yield components. 4. The contents of the main chemical compounds such as N, $P_2O_5$, $K_2O$, Ca, Mg were determined. The concentrations of nitrogen were higher in the treatment of PBS and CSM than none and control. 5. In conclusion, the nutritional effects of PBS were in evidence. To use the sludges, it should be fermented with adequate additives to improve the aeration, C/N ratio, activity of microbial, and other conditions. The sludges could be used to crops as a fertilizers effectively.

Pollution Characteristics and Loading Flux of Polycyclic Aromatic Hydrocarbons(PAHs) in Riverine Waters of Jinhae Bay, Korea (진해만 하천에서 다환방향족탄화수소 유입부하량과 오염특성)

  • PAHs(Polycyclic Aromatic Hydrocarbons) compounds may enter into the marine environment in a number of ways, and PAHs in coastal area and estuary adjacent to urban area and industrial activities region are mainly introduced into marine environment via atmosphere or river. The field surveys to evaluate the pollution of PAHs in rivers of Jinhae bay were carried out in August, 2008. The water samples collected from seven main rivers and Deokdong WWTP(Waste water Treatment Plant) which lead to Jinhae bay. 16 PAHs which had been selected as priority pollutants by EPA were analyzed. The range and mean of dissolved Total PAHs concentrations in the rivers and Duckdong WWTP that lead to Jinhae bay were shown to be 9.79~128.25(mean 36.94)ng/L, while the range and mean of Total PAHs concentrations in SPM(Suspended Particulate Matter) were shown to be 1,81434~8,893.37(mean 4,657.73)${\mu}g$/kg dry wt. The dissolved Total PAHs and Total PAHs in SPM were shown to be high concentrations in the Semhocheon which leads to Masan bay that is the most polluted area of Jinhae bay, while those were shown to be low concentrations at rivers which lead to the west area of the bay. The ranges and means of the loading fluxes from rivers and Duckdong WWTP were calculated at 0.06~12.05(mean 1.86)g/day for dissolved Total PAHs and at 0.12~16.00(mean 2.41)g/day tor Total PAH in SPM. The loading flux of Total PAHs from Duckdong WWTP was shown to be the greatest, occupying more than 80% of Total loading flux from all rivers and WWTP to Jinhae bay. The composition patterns of PAHs compound by the number of benzene ring between dissolved PAHs and PAHs of SPM were different each other. Low molecuar weight PAHs of dissoloved compounds were in high concentration, while high molecular weight PAHs of SPM were in high concentration. These results were due to physical and chemical characteristics of PAHs and were similar to those of other studies. The total PAHs concentrations of dissolved and SPM in this study are lower than those of other studies. The extent of PAHs pollution appeared to be not serious in reverine waters of Jinhae bay.

Inhibitory Effect of Hot-Water Extract of Paeonia japonica on Oxidative Stress and Identification of Its Active Components (백작약 열수추출물의 산화적 스트레스 억제효과 및 유효성분 동정)

  • This study was carried out to investigate the antioxidative activity and to identify the active components of hot-water extract of Paeoniajaponica (PJ), which was a main ingredient of a herb mixture preparation recently established as a potent candidate of radioprotector in our laboratory. The water extract was fractionated with CHCl$_3$, EtOAc and n-BuOH. The extract and its fractions showed very low activity in hydroxyl radical scavenging test. In lipid peroxidation test, the extract, EtOAc and water fractions showed moderate inhibition with the ratio above 50%. In DPPH radical scavenging test, the extract, EtOAc and water fraction showed high activity with the ratio above 80%, especially. EtOAc fraction scavenged the radicals as much as synthetic antioxidant (BHA), even at low concentration. It is suggested that mai or partition for antioxidative activity of Paeonia japonica was EtOAc fraction. Subsequently, two active compounds (PJE021-1 and JE024-1) from EtOAc fraction were isolated by using MCI gel and silica gel column chromatography The two compounds inhibited remarkedly the $H_2O$$_2$-induced DNA damage in human peripheral blood lymphocytes, measured by single-cell gel electrophoresis (SCGE). PJE021-1 protected the cells to almost negative control level, dose-dependently. PJE024-1 exhibited a potent inhibition with the ratio of 71% at even low concentration (0.5 $\mu\textrm{g}$/$m\ell$). Finally, their chemical structures were identified as gallic acid (PJE021-1) and (+)-catechin (PJE024-1), respectively, on the basis of the speculation of spectral and physical data.
