소맥(Triticum aestivum L. em Thell)의 출수기 유전에 관한 연구 (Studies on the Inheritance of Heading Date in Wheat(Triticum aestivum L. em Thell))

  • 소맥의 조숙품종육성을 위하여는 춘파성품종의 조숙인자를 추파성품종에 도입하는 것이 중요한 과제이므로 춘추파성 소맥품종의 일장과 온도조건에 따른 출수반응 및 출수기를 지배하는 생리적 요인의 구명과 출수기의 유전에 관한 지견을 얻고저 본실험을 실시하였다. 수원 작물시험장 전작포장 및 온실에서 1970년부터 조숙품종 Yecora F70, 중숙품종 수계 169호, 육성 003 만숙품종 장광, Bezostaia, Sturdy, Blueboy 등 8개품종을 상호교배하여 1972년부터 1973년에 온실 및 초자실을 이용하여 양친과 Diallel cross F$_1$을 고온장일, 고온단일, 저온장일, 저온단일의 4처리조건 및 포장조건에서 3조합의 F$_1$, F$_2$, BC$_1$, BC$_2$ 및 양친들은 고온단일조건에서 F$_1$, F$_2$ 9조합 및 그 양친은 포장에서 각각 재배 실험하였다. 그 결과를 요약하면 다음과 같다. 1. 춘화 처리후의 주요엽수는 일장 및 온도조건에 관계없이 일정하였으나 주간의 출엽속도에만 영향을 주었으며 춘파성품종은 추파성품종보다 주간엽수가 적었다. 2. 지엽전개일수의 일장 및 온도반응은 출수속도에 의하여 결정되며 일장과 온도에 따른 출엽속도의 차이는 하위엽보다 상위엽에서 컸다. 3. 춘화처리후의 출수기에 대한 생리적 요인은 일장반응과 순수조만생이며, 출수일수의 품종간 변이는 고온단일에서 가장 크고 포장조건에서 가장 적었으며 고온장일, 저온장일, 저온단일을 비슷하였다. 4. 일장차이에 의한 출수일수의 품종간 반응은 장광, Parker가 크고, Yecora F70, 수계 169 호 등 기타 품종들은 비교적 적었으며, 장광, Parker는 단일이 의하여 출수일수가 크게 지연되는 감광성 품종이었다. 5. 일장반응은 온도의 영향을 크게 받고 출수일수의 품종간차이는 저온일 빼보다 고온일 때 현저히 나타났다. 6. 온도차이에 의한 출수일수의 품종간 반응은 비슷하였으며, 어떠한 품종이라도 고온에 의하여 출수가 촉진되고 저온에 의해서 지연되었다.7. 출수일수에 대한 유전은 일장 및 온도조건이 변하여도 조숙은 만숙에 대하여 부분우성이었으며, 우성의 정도는 단일에서 크고 장일에서 적었다. 8. 고온장일조건에서 출수일수의 품종간 차이는 수수조만성의 차이라고 할 수 있으며, 춘파성품종인 Yecora F70은 추파성품종에 대하여 우성정도가 컸고 추파성품종간에는 우성정도가 없거나 적었다. 이때의 유효 우성 유전자수는 1쌍의 대립유전자에 의해서 지배되나 다소의 미동유전자가 관여되었다. 9. 출수일수에 관여하는 비감광성은 감광성에 대하여 우성단인자차에 의하여 지배되었고 저온조건에서는 단일감응성의 발현정도가 둔화 또는 억제되었다. 10. 순수 조만성과 단일감응성은 모두 조숙이 만숙에 대하여 부분우성으로 표현되었다. 11. 출수기에 관여하는 감온성의 품종간반응은 대립유전자 및 비대립유전자의 지배를 받으며 같은 대립유전자를 가진 품종군에서는 비감온성이 감온성에 대하여 우성으로 표현되나 일장 및 순수 조만성보다는 그 정도가 미미하다. 12. 장광과 수계 169호 사이에는 감광성에 관여하는 1쌍의 대립인자 ee와 EE에 의하여 조만이 결정되었고, 장광과 Yecora F70사이에는 ee, enen과 EE, EnEn의 2쌍의 대립인자에 의하여 지배된다고 추정되었다. 이때 EE와 EnEn는 조숙방향에 상가적으로 작용하여 단일조건하에서는 EE의 효과가 EnEn의 효과보다 크다. 장일 조건하에서는 EE의 효과는 나타나지 않고 EnEn효과만 발현된다. En과 en 사이에 우열관계는 E와 e 사이보다 적고 상가적 작용을 하고 있다고 본다. 13. 본 실험의 포장조건에서는 2개정도의 우성주동유전자를 가정할 수 있으나 생육초기의 저온단일과 생육후기의 고온장일의 영향으로 2개의 우성유전자 표현이 불확실하며 F$_2$분리에 있어서는 일장 및 온도의 영향과 미동유전자의 영향으로 연속변이를 하였다. 14. 출수일수의 유전력은 0.51-0.72로서 어느 조건에서나 높은 경향으로 초기세대의 선발효과가 큰 것으로 인정되었으며, 환경에 의하여 다소의 차이가 있어 장일보다는 단일에서 유전력이 컸고, 저온보다는 고온에서 큰 경향이었다. 또한 감광성의 유전력은 0.86-0.76으로 컸으나 온도효과의 유전력은 적었다. 15. 출수기가 빨라지므로서 1수입수, 천입중, 수량이 낮아지는 상관관계를 보여 조숙다수성 품종선발이 어렵다는 결과를 보였으나 수수와는 부의 상관(유의성은 없음)을 보여 조숙품종으로서 수수형 품종을 선발하면 수량을 올릴수 있는 약간의 가능성도 보였다.

세포 외 $\textrm{K}^{+}$의한 혈관 수축신 조절 기전: 혈관평활근 수축성과 내피세포 의존성 이완에 미치는 영향 (Regulatory Mechanism of Vascular Contractility by Extracellular $\textrm{K}^{+}$: Effect on Endothelium-Dependent Relaxation and Vascular Smooth Muscle Contractility)

  • 외부 자극에 의하여 세포 내 $Ca^2$$^{+}$이 증가하면 세포 내 $K^{+}$이 유출되어 세포 외 $K^{+}$ 농도는 수 mM 범위에서 증가할 수 있다. 이러한 세포 외 $K^{+}$의 증가가 혈관 수축성에 미치는 영향을 규명하고자, 세포 외 $K^{+}$가 혈관평활근 수축성, 내피세포 의존성 이완과 혈관내피세포 $Ca^2$$^{+}$ 농도에 미치는 영향을 알아보고자 하였다. 토끼에서 분리한 경동맥, 상장간막동맥 분지, 기저동맥과 쥐의 대동맥에서 등장성 수축을 기록하였으며 배양한 쥐의 대동맥 혈관내피세포와 인간 제대정맥 내피세포에서 세포 내 $Ca^2$$^{+}$ 변화를 측정하였다. 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하는 경우 도관동맥인 토끼 경동맥은 수축성에 변화가 없는 반면 저항혈관인 기저동맥과 상장간막동맥분지는 이완하였다. 이러한 $K^{+}$ 유발 이완은 혈관 종류에 따라 차이가 있었는데 기저동맥에서는 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하였을 때보다 세포 외 $K^{+}$ 농도를 1에서 3 mM로 증가하였을 때 더 크게 이완하였으며 상장간막동맥의 분지에서는 반대로 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하였을 때 더 크게 이완하였다. 그리고 세포 외 $K^{+}$ 농도를 6에서 12 mM로 증가하였을 때의 이완은 $Ba^2$$^{+}$에 의하여 억제되는 반면 1에서 3 mM로 증가에 의한 이완은 억제되지 않았다. 쥐 대동맥에서도 토끼 경동맥과 동일한 효과가 관찰되었는데 세포 외 $K^{+}$ 농도를 6 mM에서 12 mM로 변화시켜도 norepinephrine혹은 prostaglandin $F_2$$_{\alpha}$에 의한 수축력은 유의한 변화가 없었다. 또한 세포 외 $K^{+}$ 농도를 점차 증가시키는 경우 12 mM 이상 증가가 되면 혈관평활근이 수축하기 시작하였지만 12 mM 이하의 증가에 의해서는 혈관평활근의 수축력은 증가하지 않았다. 한편 쥐 대동맥에서 acetylcholine에 의하여 유발된 내피세포 의존성 이완은 세포 외 $K^{+}$ 농도를 정상 6 mM에서 12 mM로 증가시키면 억제되었다. 한편 배양한 쥐 대동맥 내피세포에서는 acetylcholine 혹은 ATP에 의하여 세포 내 $Ca^2$$^{+}$이 증가하였다. 증가한 세포 내 $Ca^2$$^{+}$은 세포 외 $K^{+}$농도를 6 mM에서 12 mM로 증가시키면 가역적 및 농도 의존적으로 감소하였다. 세포 외 $K^{+}$ 증가에 의한 세포 내 $Ca^2$$^{+}$ 억제 효과는 인간 제대정맥 내피세포에서도 관찰되었다. 그리고 세포 외 $K^{+}$ 증가에 의한 내피세포 의존성 이완의 억제효과는 $Na^{+}$- $K^{+}$ pump 억제제인 ouabain과 $Na^{+}$-C $a^2$$^{+}$exchanger 억제제인 N $i^2$$^{+}$에 의하여 억제되었다. 이러한 실험 결과로 미루어 세포 외 $K^+$의 증가는 저항혈관 평활근을 이완시키는데 그 기전은 혈관 종류에 따라 차이가 있었다. 그리고 세포 외 $K^{+}$의 증가는 혈관내피세포 $Ca^2$$^{+}$을 감소시켜 내피세포 의존성 이완을 억제하는데 이는 $Na^2$$^{+}$- $K^2$$^{+}$pump를 활성화시켜 일어나는 것으로 생각된다.

붕소-중성자 포획치료를 위한 미세 속중성자 선량 특성 연구 (Dosimetry of the Low Fluence Fast Neutron Beams for Boron Neutron Capture Therapy)

  • 목적 : 붕소-중성자 포획치료법(Boron Neutron Capture Therapy, BNCT)을 위해 원자력병원 싸이클로트론에서 발생되는 최대에너지 34.4 MeV의 속중성자(Fast neutron)를 70 cm 파라핀으로 감속시킨 후 선량 특성을 조사하였다. 그 결과를 토대로 열외중성자(Epithermal neutron) 선량 측정법에 대한 프로토콜을 확립하여 원자로에서 방출되는 열외 중성자 선량 특성 평가의 기초를 삼고, 가속기를 이용한 BNCT 연구에 대한 타당성 여부를 조사하고자 한다. 대상 및 방법 : 공기 중 선량 및 물질 내 선량 분포 측정을 위해 Unidos 10005 (PTW, Germany) 전기계와 조직 등가 물질인 A-150 플라스틱으로 제작된 IC-17 (Far West, USA) 및 IC-18, ElC-1 이온함을 사용하였고, 감마선의 측정을 위해서는 마그네슘으로 제작된 IC-l7M 이온함을 이용하였으며 조직등가 기체와 아르곤 기체를 분당 5cc 씩 주입하며 측정하였다. 중성자, 광자, 전자가 혼합된 장의 모의 수송 해석을 위해 이용되는 Monte Carlo N-Particle (MCNP) transport code를 사용하여 2차원적 선량 분포 및 에너지 분포를 계산하였으며 이 결과를 측정값과 비교하였다. 결과 : BNCT에서의 유효 치료 깊이인 물 팬텀 4 cm에서의 선량은 치료기 1 MU 당 $6.47\times10^{-3}\;cGy$로 미세하였으며, 이때 감마 오염도(contamination)는 $65.2{\pm}0.9\%$로 중성자보다는 감마선에 의한 선량 기여분이 우세하였다. 깊이에 따른 선량 분포 특성에서는 중성자 선량은 선형적으로 감쇠 되었고, 감마선량은 지수적으로 보다 급격히 감쇠되는 경향을 보였으며 전체 선량의 $D_{20}/D_{10}$은 0.718 이었다. MCNP에 의한 에너지 분포 전산 계산의 결과 2.87 MeV 이하에서 중성자 피크가 나타났으며, 저에너지 영역에서는 감마선이 연속적으로 분포되는 양상을 보였다. 결론 : 벽 물질이 서로 다른 두 개의 이온함을 사용한 직접 선량 측정과 MCNP 전산 시뮬레이션을 이용한 공간 선량분포 계산으로 미세 속중성자 빔에 대한 선량 특성을 파악할 수 있었으며, 원자로 열외중성자 주(Epithermal neutron column)에 대한 선량 평가 자료로 확보하였다. 아울러 가속기에 대한 연구가 진행되어 고전압, 고전류를 발생시키는 전원 공급장치와 표적핵(Target) 물질이 개발되고 비스무스나 납 등에 의해 감마 오염도를 줄일 경우, 싸이크로트론에 의한 보론-중성자 포획치료도 가능해질 것으로 판단된다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. To use as the structural matrix of the composite, collagen was purified from human umbilical cord. The obtained collagen was treated by pepsin to remove telopeptides, and finally, the immune-free atelocollagen was produced: The cross linked atelocollagen was highly resistant to the collagenase induced collagenolysis.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer.
