Recombinant Expression, Isotope Labeling and Purification of the Vitamin D Receptor Binding Peptide

  • The vitamin D receptor binding peptide, VDRBP, was overexpressed as a fused form with the ubiquitin molecule in Rosetta(DE3)pLysS, a protein production strain of Escherichia coli harboring an induction controller plasmid. The fusion protein was bound to the immobilized metal ions, and the denaturation and renaturation of the fusion protein were performed as a part of the purification procedure. After the elution of the fusion protein, the peptide hormone was released from its fusion partner by using yeast ubiquitin hydrolase (YUH), and subsequently purified by reverse phase chromatography. The purity of the resulting peptide fragment was checked by MALDI-TOF mass and NMR spectroscopy. The final yields of the target peptide were around 5 and 2 mg per liter of LB and minimal media, respectively. The recombinant expression and purification of this peptide will enable structural and functional studies using multidimensional NMR spectroscopy and X-ray crystallography.

  • Thd specimens were melited in high frequency induction furnace. The samples for measurements were prepared by machining cylinder of 9.5mm diameter and 15mm height. These samples were then hot-pressed according to strain rate ( .epsilon. ). These samples were decanned and cut out, and subsequently heat treated at 1000 .deg. C for 4hours. These were investigated for the change of microstructure, domain pattern, X-ray diffraction and magnetic properties. The stress distributions in the specimens during compressing process were calculated by a finite element method program(SPID). The calculated stresses were effective stress( .sigma. $_{eff}$), compressive direction stress( .sigma. $_{z}$), and shear stress( .tau. $_{rz}$ ). These stresses were compared with the experimental data.a.a.

  • Wireless power transfer technology is using the magnetic resonance recently drawing increased attention. It uses the resonance between transmitter and receiver coils to transfer power. Thus, it can improve the transfer distance and efficiency compared with the existing magnetic induction technique. The authors found from the previous study that the application of the superconductor coil to the magnetic resonance wireless power transfer system improved its efficiency. Its application to real life, however, requires the additional study on the effects of adjacent materials. In this study, the two resonance coils made by superconductor coils were used to aluminum and plastic shielding materials was placed between the coils. S-parameters were analyzed according to the position of the shielding material between the transmitter and receiver coils. As a result, the plastic of shielding material had no effect, but the aluminum of shielding material affected the wireless power transfer due to the shielding effectiveness.

  • A pulsed field magnetometer with maximum applied field of 130 kOe has been constructed. The pulsed field generated by a 18 kJ capacitor bank system combined with a pulse coil was damped oscillation with the period of 10.12 ms. Magnetic hysteresis loop was measured by induction method during 10.12 ms of a pulse duration from the first positive to the second positive peak. The difference from DC magnetic properties due to the eddy current effect was inferred below 3% for the NdFeB magnet with the diameter below 5 mm.

  • This paper is a study for accurate iron loss calculation of a cylindrical linear machine for free piston engine. This study presents that it is possible to accurately predict power loss in ferromagnetic laminations under magnetic flux by specially considering the dependence of hysteresis, classical, and excess loss components on the magnetic induction derivative. Significant iron loss in the armature core will not only compromise the machine efficiency, but may also result in excessive heating, which could lead to irreversible deterioration in the machine performance. Thus, correct prediction of power losses under a distorted flux waveform is therefore an important prerequisite to machine design, particularly when dealing with large apparatus where stringent efficiency standards are required. Finally, it will be discussed about the iron loss in various materials of cylindrical linear electric machine by geometric and electrical parameters. It will give elaborate information about the perfect design and design rules of cylindrical linear machine and in parallel tools for the calculation, simulation and design will be available.

  • In this thesis, we fabricate a zero-magnetostrictive amorphous ribbon measure the impeadance effect, and then Investigate possibility as a sensor material. $Co_{72.5}$F $e_{0.5}$M $o_{2}$ $B_{15}$ S $i_{5}$ is used as composition of specimen alloy. We first melt the specimen in high frequency induction furnace and then rapidly quench it by using single roll technique. As the result, we obtain a ribbon where thickness is 12${\mu}{\textrm}{m}$, width is 1mm and length is 93mm. Consequently, it is proved through this study that zero-magnetostrictive amorphous ribbon can be used as an excellent magnetic sensor material.rial.l.

  • The recent trains are almost being operated by the mechanical propulsion force to drive the gear and wheel with the traction motor. However Magnetic Levitation Vehicle is differently operated. Magnetic Levitation Vehicle is applied with Linear Induction Motor(LIM) that has many advantage like to high capability of going up to slope, low noise, easy to control of speed. So domestic and many advanced countries are interested in Magnetic Levitation Vehicle and they have been studying about it continuously. Thus this paper is studied the LIM test method of static states and guess the optimum driving point by characteristic of static states for LIM. The test items are measurement of thrust force by changed air gap, measurement of thrust force and normal force by changed slip frequency etc.

Comparative Measurement of Transverse Nuclear Magnetization of Polarized 129Xe and 131Xe by Spin-exchange Optical Pumping

  • We analyze the transverse nuclear magnetizations of 129Xe and 131Xe in a vapor cell containing natural Xe, 87Rb, and buffer gases. Th e Xe atoms are polarized th rough spin-exch ange optical pumping (SEOP) with Rb atoms under low-magnetic-field conditions. From the free-induction-decay (FID) signal, we measure the nuclear magnetization of the Xe atoms in the Xe-Rb vapor cell. Furthermore, we measure the dependence of the gyromagnetic ratio on the magnetization of 129Xe and 131Xe by examining the amplitude of the FID signal of each isotope, and we evaluate the relationship between the magnetic field gradient and transverse relaxation rate for both of the 129Xe and 131Xe isotopes.

  • In order to obtain specific magnetic properties, it is of paramount importance to increase the alloy density of components fabricated by powder metallurgy. An alternative to increase the density of alloys such as Fe-49Co-2V would be the use of elemental Fe and Co instead of the pre-alloyed powder. Trying to give some insight on the industrial application of this strategy, this paper investigates the replacement of more conventional pre-alloyed Fe-49Co-2V powders with elemental Fe and Co. A previous analysis shows that it is possible to achieve higher densities and leads to a noticeable improvement in some important magnetic properties.

  • Magnetic field is commonly used in low temperature processing plasmas in order to obtain high density. E $\times$ B magnetron or surface multipole configuration were most popular. However, the properties of capacitively coupled rf plasma confined by axially applied static magnetic fields have rarely been studied. In this paper, the effects of magnetic field on the characteristics of 13.56MHz/40KHz argon plasma will be reported. Ion saturation current, electron temperature and plasma potential were measured by Langmuir probe and omissive probe. At low pressure region ($\sim$10mTorr), ion current was increased by a factor of 3 - 4 due to reduction of diffusion loss of charged particles to the wall. It was observed that magnetic field induces large time variation of the plasma potential. The experimental result was compared with particle-in-cell simulation. It was also observed that electron temperature tend to decrease with increasing magnetic induction level for 40KHz discharge.
