Single dose toxicity study of Jengjengamiyjintang in rats (정전가미이진탕(正傳加味二陳湯)의 랫드에서의 단회투여독성시험(單回投與毒性試驗))

  • The single dose toxicity of Jengjengamiyjintang, a herbal drug for duodenal ulcer was evaluated in male and female Sprague-Dawley (SD) rats. Jengjengamiyjintang was once administered to both sexes of rats at the dose levels of 2000, 1000, 500, 250 and 125mg/kg for oral route according to KFDA guidelines for single dose toxicity test (1999-61). In addition, vehicle control group was added in order to compare clinical signs, body weight changes and abnormal necropsy findings. After single administration, clinical signs were observed every twice a day for 14 days and body weights were measured 5 times including initial measurement on day 0. When observation period was over, the animals were sacrificed and macroscopic examination of major organs was conducted. Neither significant clinical signs nor death after administration was observed during the observation periods except for soft feces or diarrhea that were restricted to Day 1 of 2000 and 1000mg/kg-dosing groups. Although some accidental findings such as gross and histopathological changes of lung that were demonstrated in some animals of all experimental groups including vehicle control group, no abnormal necropsy findings and changes of body weight were observed at terminal necropsy in both sexes. From these results, it is considered that $LD_{50}$ of Jengjengamiyjintang is over 2000mg/kg in oral administration in both sexes of rats and approximated lethal dose was considered over 2000mg/kg.

EFFECTS OF PROPLAST I AND II IMPLANTATION ON THE SURROUNDING TISSUE RESPONSE AND BONE FORMATION IN RABBIT MANDIBLE (가토 하악골에 Proplast I과 II 이식후 주위 조직반응 및 골형성)

  • The purpose of this study was to compare the response of adjacent tissue and new bone formation after implantation by different methods of subperiosteal using using Proplast I and II in rabbit mandible. Microstructure of Proplast I and II was observed by scanning electron microscope. And the implantation procedure was carried out by dividing into tow groups, A and B. a group consisted of subperiosteal graft on the cortex, and the other B group was made up onlay graft following artificial decortication in the madibular body of rabbit. The experimental animals were sacrificed on the 1st, 2nd, 4th and 8th week after grafting for macroscopic and histopathologic examination. The samples extracted at the 6th postgrafting week were also used for biometric test. The result ere as follows : 1. By scanning electron microscopic observation, pore size was $50{\sim}180{\mu}m$ in the Proplast I and $100{\sim}220{\mu}m$ in Proplast II. 2. Macroscopically, infection of the graft site, deformation and displacement of the implanted materials were not observed in all experimental groups. 3. In the tissue response, infiltration of inflammatory cells and multinucleated giant cells were observed from the 2nd to the 8th week in Proplast I. Inflammatory cells decreased in number from the 2nd week in Proplast II suggesting that Proplast II is better than Proplast I. 4. Bone formation was not observed until the 8th week in the group A, but new bone formation from the surrounding graft bed and the periostium was appeared from the 4th week in the group B. 5. The maximum mean values of shear stress mere serially $65.5gf/mm^2$ in Proplast II of group B, $32.9gf/mm^2$ in Proplast I of group B, $17.0gf/mm^2$ in Proplast II of group A, and $15.7gf/mm^2$ in Proplast I. of group A.

  • The present study was aimed to evaluate the effect of demineralized freeze-dried bone (DFDB) and resorbable hydroxyapatite (RHA) on bone formation in the extracted socket. The lower left and right 2nd and 3rd premolar were extracted in adult dogs. The one group was grafted with DFDB into the extracted socket, and the other group grafted with RHA. The extracted socket was sutured without any graft materials as control. The animals were killed on the 1st, 2nd, 4th, and 8th week after the graft for macroscopic and microscopic examination. Results obtained were as follows : 1. Macroscopically, nor infection of the graft site and dislodgement of the grafted material were noted in any animals used. 2. Young trabeculae of osteoid were formed in the socket wall in control group at 2 weeks after the graft. Osteoid tissue was formed in DFDB group at 1 week after graft, and a fine osteoid tissue was grown through the RHA particles in RHA group at 2 weeks graft. 3. The grafted groups showed more rapid bone formation than the control. Between the grafted groups, DFDB group showed more rapid formation than RHA group, DFDB group showed osteoinductive bone formation and RHA group showed osteoconductive bone formation. These results suggest that DFDB and RHA are useful to preserve the alveolar bone and to improve new bone formation by immediate grafting into the extraction sockets.

Injinhotang was once administered to both sexes of rats at the dose levels of 2000, 1000, 500, 250 and 125 mg/kg for oral route. After single administration, clinical signs were observed every day for 14 days and body weights were measured 5 times including initial measurement on day 0 (the days of administration). When observation period was over, the animals were sacrificed and macroscopic examination of major organs was conducted. In addition, the histopathological profiles of these major organs were also conducted. Neither significant clinical signs nor death after administration was observed during the observation periods except for soft feces or diarrhea. In addition, no abnormal necropsy findings, changes of body weight and histopathological profiles were observed at terminal necropsy in both sexes. From these results, it is considered that $LD_{50}$ of Injinhotang is over 2000 mg/kg in oral administration in both sexes of rats.

Surgically induced degenerative changes in the femorotibial joints by total medial meniscectomy in minipigs closely resemble late-stage osteoarthritis

  • Animal models of osteoarthritis (OA) have played a key role in understanding the etiology of OA and in the development of new therapeutic strategies. Although pigs have an advantage as an animal disease model due to their similarity to humans, there are few studies on the induction of OA in minipigs. Therefore, this study aimed to characterize disease progression of OA in total medial meniscectomy (TMM)-operated skeletally mature minipigs, up to day 180 postoperatively. There were no significant alterations in vital signs or hematological indices throughout the observation period. However, clinical manifestations of OA in the medial femoral condyles of TMM-operated minipigs were progressive, depending on postoperative duration, with respect to osteophytes formation and roughened surfaces on radiological observation, cartilage erosion under macroscopic examination, and severe cartilage defects including fibrillation, vertical fissures, and cartilage denuding on histopathological observation, with the highest score indicating late-stage OA on day 180 and without indicating apparent variation between subjects. In particular, the lateral femoral condyles were also degenerated, possibly due to localization of weight-bearing from both menisci to the lateral meniscus. Therefore, TMM in minipigs is suitable for reproducible induction of degenerative changes in the femorotibial joints that closely resemble late-stage OA, and is suitable for use in further research.

Experimental study trends on the prevention and treatment effects of herbal medicine for gastroesophageal reflux disease (GERD) - based on Pubmed (천연물의 위식도역류질환 예방, 치료 효과에 대한 실험연구 현황 – Pubmed를 중심으로)

  • Objectives : This study aimed to review the current trends in experimental studies on the use of natural products for treatment of gastroesophageal reflux disease (GERD). Methods : Experimental studies assessing the efficacy of natural products against GERD were searched on PubMed. Articles were selected based on predefined inclusion and exclusion criteria and then analyzed for experimental methods, interventions, and result analysis techniques. Results : A total 37 studies were included in this review. Predominantly, in vivo experiments were conducted to induce GERD through surgery, involving the ligation of the pylorus and the transitional junction between the corpus and the forestomach using 7-week-old male Sprague-Dawley rats. The acute induction model, sacrificing animals after a single administration following GERD induction, was mainly used.The utilization of cell experiments was relatively infrequent, with a focus on assessing antioxidant and anti-inflammatory effects via the treatment of the RAW 264.7 cell line with lipopolysaccharides treatment. Glycyrrhizae Radix et Rhizoma, Pinelliae Tuber, Ginseng Radix and Zingiberis Rhizoma were used as single ingredients, and herbal formula, STW-5 (iberogast), Rikkunshito (六君子湯), Banhasasim-tang (半夏瀉心湯), and Hewei Jiangni granule (和胃降逆湯) were used. Outcome analysis methods encompassed Macroscopic evaluation, esophageal function assessment, blood biomarker analysis, histological examination, protein analysis, gene expression analysis, and gastric juice analysis. Proton pump inhibitors were predominantly employed as positive controls. Conclusions : This study revealed the current trends in non-clinical research evaluating natural products for GERD. Based on the results of this study, we expect that non-clinical research on clinically effective natural products will be revitalized.

A Study on the Tissue Response and Bone Formation after Augmentation Using Proplast and Porous Polyethylene in Rabbit Mandible (Proplast와 Porous Polyethylene을 이용(利用)한 가토(家兎) 하악골(下顎骨) 증대술후(增大術後) 조직반응(組織反應) 및 골형성(骨形成)에 관(關)한 연구(硏究))

  • Proplast and Porous Polyethylene which have porous structures as low-modulus polymers have been recently used in maxillofacial plastic and reconstructive surgery. The purpose of this study was to compare the response of adajacent tissue, new bone formation and stability after augmentation by differen methods of subperiosteal graft using proplast and purous polythylene in rabbit mandible. The augmentation procedure was carried out by dividing into two groups, A and B. A group consisted of subperiosteal graft on the cortex, and the other B group was made up only graft following artificial decortication in the mandibular body of rabbit. The experimental animals were sacrificed on the 1st, 2nd, 4th and 8th week after grafting for macroscopic and light microscopic examination. The samples extracted at the 6th postgrafting week were also used for biometric testing and scanning electron microscopic examination. The results obtained from this study were as follows : 1. Macroscopically, infection of graft site, deformation and migration of graft material were not observed in all experimental groups. 2. B group showed more rapid and increased bone formation and the greater stability than A group, and tissue response was similar to each other. 3. In the tissue response, macrophages and cellular infiltrations were observed in Proplast group, but few in PHDPE group. 4. In bone formation of A group, Proplast group showed no bone formation until the 8th week, but PHDPE group showed small quantity of osteoid tissue from the 2nd week and appositional bone growth with new bone formation at the 8th week. 5. In bone formation of B group, both Proplast and PHDPE group showed bone formation, but PHDPE group showed more rapid and larger bone formation. 6. In pattern of bone formation, Proplast group mainly showed appositional bone growth pattern connected with graft site. On the other hand, PHDPE group showed mixed pattern of new bone formation in the pore connective tissue with appositional bone growth from graff site. 7. The maximum mean values of shear stress were serially $111.3gf/mm^{2}$ in PHDPE of B group, $84.8gf/mm^{2}$ in PHDPE of A group, $32.9gf/mm^{2}$ in Proplast B group, and $15.7gf/mm^{2}$ in Proplast of A group. From above results, It was suggested that the capacity of bone formation and stability between bone and graft material were dependent on the pore size and structure of graft material itself, the state of graft site and tissue response.

Evaluation of Mechanical Test Characteristics of Fillet Welding (필릿 용접의 기계적 시험 특성 평가)

  • FCAW is used mainly in the welding of carbon steel and alloy steel because it can be welded in all positions and can obtain excellent quality at sites with variable working conditions. Recently, many studies in Korea have estimated the fatigue strength, residual stress, and deformation, and to develop a fillet welding process. On the other hand, there have been few studies of the mechanical properties based on the strength, macro and magnetic particle test results for fillet welding. This study shows the following results through fillet welding, macro testing and strength testing using SM490A (solid-structure rolled steel) for thick plates using SS400 (rolled steel) for the upper plate and FCAW. The hardness test, macro test and magnetic particle test were then conducted. The hardness tests showed that all result values were smaller than the KS B 0893 standard values of 350Hv. The macro-test showed that each type of welded part was in a normal organic state and that there were no internal errors, bubbles, or impurities on the front of the welded part. Therefore, there were no concerns about lamination. The magnetic particle examination showed no problems.

The Future Direction of Learning Objectives and National Nurse's Licensing Examination in Adult Health Nursing (성인간호학 교육의 학습목표와 국가고시 문제 개선방안)

  • Purpose: The purpose of this study was to identify a model based on f the learning objectives in adult health nursing curriculums. The model can be eventually reflected in Korean National Nurse's Licensing Examination (KNNLE). Methods: The model was developed through a series of practical analysis by the task force team from June to October, 2011. The research team conducted the data synthesis and analysis from the learning objectives of adult health nursing drawn from selected national and international universities, from the adult health nursing areas in KNNLE, and from the current disease prevalence from clinical data in the representative university hospitals in Korea. Results: The current disease prevalence and mortality rates are on the increase especially for malignant neoplasm, heart diseases, suicide and diabetes. The items on the KNNLE of adult nursing are categorized by body systems into digestive system (15%), introduction (13.8%), respiratory system (11.3%), and cardiac system (11.3%). While the current system-based learning objectives covers extensive areas of adult health nursing, the core items are required to be selected based on core competences and core learning objectives to restructure the items of KNNLE. The first revised model is to consolidate and restructure the items microscopically in the subjects of adult health nursing by system. The second proposed model from macroscopic perspective is to build a comprehensive scheme of nursing curriculum by encompassing 8 subjects under current KNNLE and adjust the number of items accordingly. Conclusion: The items of KNNLE need to be gradually redesigned by considering the intervention need at clinical practice and the modified learning objectives of adult health nursing. Based on the job analysis on core competences for newly employed nurses performed by the Korean Accreditation Board of Nursing, it is necessary to establish the core learning objectives of adult health nursing and set up standards for core items in KNNLE.

A Study of the Effect of a Mixture of Hyaluronic Acid and Sodium Carboxymethyl Cellulose ($Guardix-sol^{(R)}$) on the Prevention of Pericardial Adhesion (Hyaluronic Acid와 Sodium Carboxymethyl Cellulose 혼합용액($Guardix-sol^{(R)}$)의 섬유막유착 방지 효과에 관한 연구)

  • Background: This study was designed to evaluate the efficacy of a mixture of hyaluronic acid and sodium carboxymethyl cellulose ($Guardix-sol^{(R)}$) on experimental pericardial adhesion. Material and Method: Thirty rats were divided into 2 groups of 15 rats each and pericardial mesothelial injury was induced during surgery by abrasion. In the control group, blood and normal saline were administered into pericardium; in the test group, blood and HA-CMC solution were administered. Pericardial adhesions were evaluated at 2 weeks (n=5), 4 weeks (n=5), and 6 weeks (n=5) after surgery. The severity of adhesions was graded by macroscopic examination, and the adhesion tissue thickness was analyzed microscopically with Masson trichrome stain and an image processing program. Result: The test group had significantly lower macroscopic adhesion scores ($2.9{\pm}0.6$ : $3.9{\pm}0.4$, p<0.000) compared with the control group. For microscopic adhesion tissue thickness, the test group had lower scores compared with the control group, but this difference was not statistically significant ($91.73{\pm}49.91$ : $117.67{\pm}46.4$, p=0.106). Conclusion: We conclude that an HA-CMC solution ($Guardix-sol^{(R)}$) reduces the formation of pericardial adhesions in this animal model.
