The native connective tissue attachment of the periodontium is known to be a complex consisting of gingival fibroblasts, periodontal ligament cells, gingival epithelial cells, cementum, alveolar bone and extensive extracellular matrix (collagen, glycoprotein and proteoglycans). The purpose of this study was to evaluate the effects of natural extracts on DNA, collagen and protein synthesis and inhibition of cytokine production in the gingival and periodontal ligament fibroblasts and gingival epithelial cells. Healthy gingival tissue was obtained from orthodontic treatment patients, and gingival epithelial cells, gingival fibroblasts and periodontal ligament cells were isolated and cultured from the samples. After treated with Ginseng protein, Pluronic F-68, Scutellariae Radix, centella asiatica, PDGF, IGF, DNA synthesis, total protein and collagen synthesis, and cytokine production of gingival epithelial cell, gingival fibroblast and periodontal ligamentcells were measured. MTT method for DNA synthesis, Peterkofsky and Dingerman method for total protein and collagen synthesis, and IL-1 ELISA kit for cytokine production were used. The proliferation of epithelial cells was enhanced in Centella asiatica, Ginseng protein, Pluronic F-68 and Scutellariae Radix. The activities of PDL cells were increased in PDGF, IGF, and Pluronic F-68. Higher collagen synthesis was observed in Scutellariae Radix and total protein synthesis was increased in Scutellariae Radix and PDGF. The inhibitory effects on IL-1, IL-6, $TNF-{\alpha}$ were observed in all exrracts.

In order to find potential anticancer agents, we extracted pheophytin from silkworm feces according to various dry and storage methods such as sun dry, shade dry, fresh freezing dry and freezing dry after freezing storage (for 1∼3 years). The pheophytin extracts, mainly 10-hydroxypheophytin a, little b, of various storage silkworm feces were analyzed by reversed-phase high-performance liquid chromatography with photodiode array and fluorescence detection. The content of those pheophytih in old silkworm for 3 years (freezing storage and freezing dried in use, or freezing dried and cold storage) was better than others. The cytotoxicity of the pheophytin extracts and ethanol extracts of various storage silkworm feces were measured using Jurkat cells originated from human leukemia, using dye uptake assay (MTT) in order to find effective photodynamic therapeutic agents. The anticancer activity of those pheophytin extracts in various storage methods showed little difference among them. But ethanol extracts of fresh freezing dried silkworm in the current year was good cytotoxic activity than those of any other silkworm feces. With regards to these results, fresh ethanol extracts of silkworm feces were better than old ones. On the other hands, the pheophytin extracts of old silkworm feces contained the highest percentage of pheophytin content and showed good cytotoxicity against cancer cells by changing the pheophytin into pheophobide in the degradative process.

Background: Development of a nanosized polymeric delivery system for erlotinib was the main objective of this research. Materials and Methods: Poly caprolactone-polyethylene glycol-polycaprolactone (PCEC) copolymers with different compositions were synthesized via ring opening polymerization. Formation of triblock copolymers was confirmed by HNMR as well as FT-IR. Erlotinib loaded nanoparticles were prepared by means of synthesized copolymers with solvent displacement method. Results: Physicochemical properties of obtained polymeric nanoparticles were dependent on composition of used copolymers. Size of particles was decreased with decreasing the PCL/PEG molar ratio in used copolymers. Encapsulation efficiency of prepared formulations was declined by decreasing their particle size. Drug release behavior from the prepared nanoparticles exhibited a sustained pattern without a burst release. From the release profiles, it can be found that erlotinib release rate from polymeric nanoparticles is decreased by increase of CL/PEG molar ratio of prepared block copolymers. Based on MTT assay results, cell growth inhibition of erlotinib has a dose and time dependent pattern. After 72 hours of exposure, the 50% inhibitory concentration (IC50) of erlotinib hydrochloride was appeared to be $14.8{\mu}M$. Conclusions: From the obtained results, it can be concluded that the prepared PCEC nanoparticles in this study might have the potential to be considered as delivery system for erlotinib.
Objective : This experimental study was performed to investigate the effects of Houttuyniae Herba extract on anti-inflammation and anti-oxidation. Methods : The cytotoxicity of Houttuyniae Herba water extract and ethanol extract about viability of Raw 264.7 cell were tested using a colormetric tetrazolium assay(MTT assay). The purpose of this study was to evaluate the effects of DFPR on differentiation of mouse calvarial cell in vitro, to examine the possibility for periodontal regeneration. $10{\mu}g/ml$ of DFPR was used as experimental concentration. osteogenic medium only was assigned as control, Experimental 1 was supplemented with 10nM dexamethasone, Experimental 2 with $10{\mu}g/ml$ DFPR and Experimental 3 with l0nM dexamethasone + $10{\mu}g/ml$ DFPR. cellular activity was evaluated by MTT method at 8, 12, 16 days, expression of mRNA of ALP, osteopontin, osteocalcin, collagen type-l was detected by RT-PCR method at 4, 8, 12, 16 days of culture. extent of mineralization was observed by Von Kossa staining at 16 day of culture. The results are as follows 1)Any acceleration of differentiation was not observed at expression of differentiation marker, 2) Decrease in expression of extracelluar matrix and in bone nodule formation was observed The results suggested that DFPR have negative effect on the rate of differentiation on rat calvarial cell, decrease extracelluar matrix formation ,decrease bone nodule formation. Ongoing studies are necessary in order to determine effect of DFPR on periodontal regeneration.
Purpose: Oncostatin M(OSM) is a multifunctional cytokine that belongs to the interleukin(IL)-6 family. Although there have been a number of studies that focused on the role and mechanism of OSM in various organs and tissues, there are few reports on its effect on wound healing. The final purpose of this project is to evaluate the effect of OSM on wound healing. This pilot study was designed to investigate the effect of OSM on proliferation and matrix synthesis of human dermal fibroblasts, which are the major components of the wound healing. Methods: Excess skin that was obtained from patients who underwent skin grafts, was used for this study. From this material, fibroblasts were isolated and cultured. The cultured fibroblasts were treated with one of four concentrations of OSM. The OSM concentrations used were 0, 50, 100, and 200 ng/ml, respectively. After the OSM treatment, cell proliferation was determined by the MTT assay, collagen synthesis by the C1CP method, GAG levels by the Blyscan Dye method. The parameter levels of each group were compared. Results: OSM treatment increased all the components tested in the study. In particular, cell proliferation, GAG synthesis demonstrated statistically significant increases(p<0.05 in the Mann-Whitney U-test). The highest increase in all the components was obtained at a 100 ng/ml concentration of OSM.Conclusion: The results of the present study indicate that OSM stimulates proliferation and matrix synthesis of human dermal fibroblast and the optimal concentration for wound healing is 100 ng/mL.
The antioxidant, antimicrobial, and cytotoxic activities of ovotransferrin were investigated in vitro. The antioxidant capacity of ovotransferrin was evaluated using the 2,2-Diphenyl-1-picryl hydrazyl (DPPH) radical scavenging method, antimicrobial effects using the agar well diffusion method, and cytotoxicity using the 3-(4,5-dimethylthizol-2-yl)-2,5-diphenylatetetrazolium bromide (MTT) assay. The DPPH radical-scavenging capacity of ovotransferrin at 1 mg/mL level reached approximately 60% after 48 h of reaction. The antimicrobial effects of ovotransferrin against common food-borne pathogens, Staphylococcus aureus KCCM 32395, Bacillus cereus KCCM 40935, Listeria monocytogenes ATCC 15313, Escherichia coli O157:H7 ATCC 43895, and Helicobacter pylori HpKCTC 26695 were dose dependant. Gram-positive bacteria was more sensitive to ovotransferrin than gram-negative bacteria. Ovotransferrin showed stronger antimicrobial effect against L. monocytogenes than other gram-positive bacteria tested. The cytotoxicity of ovotransferrin was evaluated in human cancer cell lines, various tissue origins, including the larynx (Hep-2), stomach (AGS), lung (SK-MES-1), liver (HepG2), breast (MCF-7), cervix (HeLa), and colon (HT-29). Ovotransferrin displayed relatively high cytotoxicity (${\leq}60%$ inhibition effects) at 40 mg/mL. At lower concentrations (${\leq}10mg/mL$), however, ovotransferrin cytotoxic effects were not significant in all cancer cell lines tested. These results indicated that ovotransferrin has potential to be used as an antioxidant or antimicrobial agent in foods or a pharmaceutical agent against cancers.
Under the active search for biologically active novel agents for cancer prevention and treatment, some agents have been found from plants which are easily available. Our previous research on them revealed that C. annuum L. var. angulosum Mill have high antiproliferating effect on cancer cells. However, it has not been known whether the anticancer efficacy is different according to each part of C. annuum L. var. angulosum Mill or whether it can be changed by timing of harvest or solvent for extraction. Thus we compared the efficacy of each part of C. annuum L. var. angulosum Mill and assessed how much difference in the efficacy can be made according to the time of harvest or solvents for extraction. We observed the morphologic change and apoptosis 48 hr after treatment with the extract of each part of C. annuum L. var. angulosum Mill in MCF-7 mammary gland adenocarcinoma cells and human hepatoma cells. We also counted cancer cells by trypan blue method and MTT method to check the cytotoxicity. The leaf extract showed the highest anticancer effect among all the parts of C. annuum L. var. angulosum Mill; 50% and 70% reduction in the number of cancer cells was observed at 25 $\mu\textrm{g}$/mι and 50 $\mu\textrm{g}$/mι, respectively. It was more than 2 times as potent as 5-fluorouracil (5-FU). We found chromosomal fragmentation, clumping, and destuction by PI staining, and DNA fragmentation by electrophoresis. In conclusion, this study suggests that leaf extraction using water as solvent has the highest antiproliferative and apoptotic activity in cancer cells compared with other parts of extraction.

Objective : The aim of our study is to investigate the cytotoxic, antioxidant, and antimicrobial effects of newly synthesized boron compounds in U87MG glioblastoma cell treatment. Methods : We synthesized boron glycine monoester (BGM) and boron glycine diester (BGD) structures containing boron atoms and determined their cytotoxic activities on glioblastoma by the MTT method. The inhibitory concentration 50 (IC50) value was calculated with GraphPad Prism 5.0 program. The IC50 values were administered 48 hours on U87MG glioblastoma cell. Catalase (CAT), acid phosphatase (ACP) and alkaline phosphatase (ALP) enzyme activity, malondialdehyde (MDA), total glutathione (GSH), and total protein levels were detected using spectrophotometric methods. We determined the antimicrobial activities of BGM and BGD with the disc diffusion method. Results : After 48 hours of BGM and BGD application to U87MG glioblastoma cells, we found the IC50 value as 6.6 mM and 26 mM, respectively. CAT and ACP enzyme activities were decreased in BGM and BGD groups. MDA which is a metabolite of lipid peroxidation was increased in both boron compounds groups. GSH level was reduced especially in BGD group. BGM and BGD have been found to be antimicrobial effects. Conclusion : Boron compounds, especially the BGM, can provide a new therapeutic approach for the treatment of glioblastoma with their anticancer, antioxidant, and antimicrobial effects.
Objective : Phellinus spp. mushroom is an object of interest because it has excellent anticancer effect. Owing to the similalarities in the morphology, Phellinus linteus and Phellinus baumii are often used as same Sang Hwang Mushroom in the Korean market.. The quality control for mushrooms is needed because there are many differences in the efficacy according to cultivation method and cultivation area. Therefore, a reliable authentication method of these herbal medicine is necessary to compare and measure the amount of beta-glucan which is known to have a hypoglycemic effect, from the mushrooms collected in various regions Methods : 7 samples of medicinal mushrooms supplying phellinus spp. were collected in Korea, China and Cambodia. We investigated the hardness, colors, extract ratio, ${\alpha}-amylase$ and ${\alpha}-glucosidase$ inhibitory activities, glucose transporter 4 (GLUT-4) expressions of water extracts from Phellinus spp and also MTT assay were examined for cell toxicity. Results : The results revealed that Phellinus spp.water Ext.inhibited ${\alpha}-glucosidase$ activity. glucose transporter 4 (GLUT-4), the key insulin signaling pathway transcription factor, was remarkably increased by the Phellinus baumii water extract Conclusions : These results suggest that The more yellowish the mushroom is, the lower the hardness, the more the content of ${\beta}-glucan$ is proportional. Because the more ${\beta}-glucan$, the greater the effect of hypoglycemia. compared to the hypoglycemic effect, Phellinus Baumii grown at hanging on selves for 7 month in the green house is the best.
