Magnetic resonance imaging (MRI) has been extensively applied in the study of multiple sclerosis (MS), substantially contributing to diagnosis, differential diagnosis, and disease monitoring. MRI studies have significantly contributed to the understanding of MS through the characterization of typical radiological features and their clinical or prognostic implications using conventional MRI pulse sequences and further with the application of advanced imaging techniques sensitive to microstructural damage. Interpretation of results has often been validated by MRI-pathology studies. However, the application of MRI techniques in the study of neuromyelitis optica spectrum disorders (NMOSD) remains an emerging field, and MRI studies have focused on radiological correlates of NMOSD and its pathophysiology to aid in diagnosis, improve monitoring, and identify relevant prognostic factors. In this review, we discuss the main contributions of MRI to the understanding of MS and NMOSD, focusing on the most novel discoveries to clarify differences in the pathophysiology of focal inflammation initiation and perpetuation, involvement of normal-appearing tissue, potential entry routes of pathogenic elements into the CNS, and existence of primary or secondary mechanisms of neurodegeneration.
It is becoming more and more clear that each cell, even those of the same type, has a unique identity. This sophistication and the diversity of cell types in tissue are what are pushing the necessity for spatially distributed omics at the single-cell (SC) level. Single-cell chemical assessment, which also provides considerable insight into biological, clinical, pharmacodynamic, pathological, and toxicity studies, is crucial to the investigation of cellular omics (genomics, metabolomics, etc.). Mass spectrometry (MS) as a tool to image and profile single cells and subcellular organelles facilitates novel technical expertise for biochemical and biomedical research, such as assessing the intracellular distribution of drugs and the biochemical diversity of cellular populations. It has been illustrated that ambient mass spectrometry (AMS) is a valuable tool for the rapid, straightforward, and simple analysis of cellular and sub-cellular constituents and metabolites in their native state. This short review examines the advances in ambient mass spectrometry (AMS) and ambient mass spectrometry imaging (AMSI) on single-cell analysis that have been authored in recent years. The discussion also touches on typical single-cell AMS assessments and implementations.

The mechanical-collimation imaging is the most mature technology in prompt gamma (PG) imaging which is considered the most promising technology for beam range verification in proton therapy. The purpose of the present study is to compare the performances of two mechanical-collimation PG cameras, knife-edge (KE) camera and multi-slit (MS) camera. For this, the PG cameras were modeled by Geant4 Monte Carlo code, and the performances of the cameras were compared for imaginary point and line sources and for proton beams incident on a cylindrical PMMA phantom. From the simulation results, the KE camera was found to show higher counting efficiency than the MS camera, being able to estimate the beam range even for $10^7$ protons. Our results, however, confirmed that in order to estimate the beam range correctly, the KE camera should be aligned, at least approximately, to the location of the proton beam range. The MS camera was found to show lower efficiency, being able to estimate the beam range correctly only when the number of the protons is at least $10^8$. For enough number of protons, however, the MS camera estimated the beam range correctly, errors being less than 1.2 mm, regardless of the location of the camera.

Since the emergence of proteomics methods, many proteins specific for renal cell carcinoma (RCC) have been identified. Despite their usefulness for the specific diagnosis of RCC, such proteins do not provide spatial information on the diseased tissue. Therefore, the identification of cancer-specific proteins that include information on their specific location is needed. Recently, matrix-assisted laser desorption ionization (MALDI) mass spectrometry (MS) based imaging mass spectrometry (IMS) has emerged as a new tool for the analysis of spatial distribution as well as identification of either proteins or small molecules in tissues. In this report, surgical tissue sections of papillary RCC were analyzed using MALDI-IMS. Statistical analysis revealed several discriminative cancer-specific m/z-species between normal and diseased tissues. Among these m/z-species, two particular proteins, S100A11 and ferritin light chain, which are specific for papillary RCC cancer regions, were successfully identified using LC-MS/MS following protein extraction from independent RCC samples. The expressions of S100A11 and ferritin light chain were further validated by immunohistochemistry of human tissues and tissue microarrays (TMAs) of RCC. In conclusion, MALDI-IMS followed by LC-MS/MS analysis in human tissue identified that S100A11 and ferritin light chain are differentially expressed proteins in papillary RCC cancer regions.
Purpose: We developed the software for diffusion tensor imaging and evaluated its feasibility in norm brains. Method: Five normal volunteers, aged from 25 to 29 years, were examined on a 1.5 T MR system. the diffusion tensor pulse sequence used a SE-EPI with 6 diffusion gradie directions of (1, 1, 0), (-1, 1,0), (1, 0, 1), (-1, 0, 1), (0, 1, 1), (0, 1, -1) and also with no diffusion gradient. A b-factor of 500 sec/mm2 was used. Measurement parameter were as follows; TR/TE=10000 ms/99 ms, FOV=240 mm, matrix=128$\times$128, slice thickness/gap=6 mm/0 mm, bandwidth=91 kHz and the number of total slices=20. Four repeated axial diffusion images were averaged for diffusion tensor imaging. A total scan 11 of 4 min 30 sec was used. Six full diffusion tensor components of Dxx, Dyy, Dzz, Dxy, Dxz and Dyz were obtained using two-point linear regression model from 7 diffusion-weight images at each pixel and fractional anisotropy and lattice index images was estimated fr their eigenvectors and eigenvalues. Our program was written on a platform of IDL. W evaluated the qualities of fractional anisotropy and lattice index images of normal brains a knew whether our software for diffusion tensor imaging may be feasible.
목적: 고식적인 spoiled gradient echo(SPGR) 기법과 EPI기법간의 뇌의 운동피질에 대한 기능적 영상의 차이를 평가하고자 하였다. 대상 및 방법: 5명의 정상 성인을 대상으로 1.5T 자기공명영상기기에서 SPGR기법(TR/TE/flep angle=50ms / 40ms / $30^{\circ}$, FOV=300mm, matrix $size=256{\times}256$, 단면두께=5mm)과 기법(TR / TE / flip angle=3000ms / 40ms / $90^{\circ}$, FOV=300mm, matrix $size=128{\times}128$, 단면두께=5mm)을 사용하여 MR 에이터를 얻었다. EPI기법의 경우, 총 6개의 단면에 대해 총 960개의 영상을 얻었고 SPGR 기법 겨우, 위 6개의 단면 중 운동중추영역이 포함된 한 단면에 대해 총 160개의 영상을 얻었다. 사용된 paradigm은 각각 8번의 휴지 기간과 활성 기간으로 구성되었고 활성기간동안에 시행된 작업은 오른손 손가락의 쥠과 펼침 운동이었다. 뇌의 활성영역을 국소화 하기 위해 사용된 통게적인 처리방법은 cross-correlation 방법이었다. 두 기법의 활성화 영상에 대해 활성 영역에서의 휴지 기간과 활성 기간간의 신호 크기의 변화와 연속적인 MR 데이터의 신호 대 잡음비의 측정, 그리고 활성 영역의 위치와 그 범위를 비교 분석하였다. 결과: 두 기법 모두에서 운동중추영역인 전운동피질이 잘 관찰되었다. 휴지 기간과 활성 기간간의 신호 크기의 변화는 두 기법간의 유의한 차이가 없었다. 그러나, 연속적인 MR 데이터의 신호 대 잡음비에 있어서는 EPI 기법이 SPGR 기법보다 약 2배 높았다. 또한, 활성 영역에 있어서 EPI 기법이 SPGR기법보다 낮은 쪽으로의 p값의 분포를 보여 주었다. 결론: 본 연구는 운동중추영역에 대한 뇌의 기능적 영상을 얻는 데에 있어서 두 기법 모두 유용함을 보여주었다. 그러나, EPI 기법에서 얻을 수 있는 높은 신호 감도 특성에 의해 EPI 기법이 SPGR 기법보다 뇌의 기능적 영역에 대한 더 정확한 정보를 제공하는 장점을 지닌다.
최근에, 질량분석기술의 폴리머 분석에의 응용은 MALDI-TOF MS 개발 이후 급속도로 발전하였다. 이 리뷰 논문은 현재까지 연구된 MALDI-TOF MS의 폴리머 특성분석에의 응용에 관한 최신 논문을 정리하였다. MALDI-TOF MS는 바이오 폴리머와, 합성 폴리머의 평균분자량 분석, 폴리머의 시퀀스 분석을 통한 구조의 해석, 모노머의 조성분석에까지 이용되고 있다. 엔드그룹의 특성과 농도를 분석하는 연구도 많이 진행되었고, 복잡한 폴리머의 분자량의 분석에는 SEC와 MALDI-TOF MS를 연결한 분석법을 추천한다. MALDI에 tandem MS를 결합한 분석기술이나, 이온 모빌리티를 응용한 질량분석기, TOF-SIMS, MALDI-TOF-Imaging 기술도 급격히 발전하고 있으며, 이의 폴리머 특성분석에의 응용은 별도의 분리기술이 필요 없어 앞으로 더 많이 이용될 것으로 생각된다. 분자량, 시퀀스, 그리고 모노머의 조성을 정확하게 계산해주는 소프트웨어와 고분자량(> 100 kDa)의 분석을 가능하게 해주는 기술이 개발된다면, 폴리머를 연구하는 과학자들에게 MALDI-TOF MS의 이용은 문제점을 해결하고, 목적하는 폴리머를 합성하는 데 중요한 수단이 될 것이다.
Background: Sexual dimorphism is important for sex determination in the field of forensics. However, sexual dimorphism is commonly assessed using cone beam computed tomography (CBCT) rather than three-dimensional (3D) modeling software; therefore, studies using a more accurate measurement approach are necessary. This study assessed the sexual dimorphism of the MS using a 3D modeling program to obtain information that could contribute to the fields of surgery and forensics. Methods: The CBCT data of 60 patients (age, 20~29 y; 30 males and 30 females) admitted to the Department of Orthodontics at the Dankook University School of Dentistry were provided in Digital Imaging and Communications in Medicine (DICOM) format. The left MS and right MS were modeled based on the DICOM files using the Mimics (version 22; Materialise, Leuven, Belgium) 3D program and converted to stereolithography (STL) files used to measure the width, length, and height of the MS, infraorbital foramen (IOF), right MS, and left MS. The average of three repeated measurements was calculated, and a reliability test was performed to ensure data reliability (Cronbach's α=0.618). A canonical discriminant analysis was performed using a standard approach (left: Box's M=0.096; right: Box's M=0.115). Results: Males had greater values for all parameters (MS width, MS length, MS height, IOF, right MS, left MS) than females. The discriminant analysis identified six independent variables (MS width, MS height, MS length, IOF, right MS, left MS) that could identify sex. The left MS and right MS correctly identified the sex of 81.7% and 71.7% of the patients, respectively, with the left MS having higher accuracy. Conclusion: This study confirmed that, for Korean individuals, the left MS has a better ability to identify sex than the right MS. These results may contribute to sex identification in the fields of surgery and forensics.
In MR functional imaging, it is shown that the signal change during photic activation is composed of two terms, i.e. the inflow effect and the susceptibility effect. Relatively the inflow effect affects the data obtained by CGE on the condition of short $T_E$(15ms) and large $\alpha$(90degree). The susceptibility effect, however, mainly contributes to the data on the condition of large $T_E$(35ms) and small $\alpha$(30degree). In this apper, we will discriminate the susceptibility effect for the intermingled data affected both flow effect and susceptibility effect. Finally susceptibility only functional imaging is proposed by using TRFGE.
목적 본 연구는 gadoxetic acid 조영증강 간 자기공명영상에서 T1 이완시간이 만성간질환을 가진 환자에서 간섬유화의 발견과 병기설정에 유용한지 알아보고자 한다. 대상과 방법 국소간병변이 의심되는 103명 환자들이 간 자기공명영상과 Fibroscan을 시행하였다. Fibroscan은 간섬유화의 정도를 분류하는 참조표준검사로 사용되었다. T1 이완시간은 조영제 주입 전(preT1)과 주입 20분 후(postT1), 그리고 이들 간의 T1 이완시간 감소율(rrT1)을 3 테슬라 자기공명영상의 횡단 3D VIBE 시퀀스 하에 측정하였다. Receiver operating characteristic (이하 ROC) 분석을 통해 간섬유화 병기설정을 위한 최적의 cut-off 값이 결정되었다. 결과 METAVIR score (F0-F4)에 따른 간섬유화 병기가 증가함에 따라, preT1과 postT1은 증가하였고, rrT1은 감소하였다. PreT1의 F2와 F3 사이(F2, 836.0 ± 74.7 ms; F3, 888.6 ± 77.5 ms, p < 0.05), postT1의 F3와 F4 사이(F3, 309.0 ± 80.2 ms; F4, 406.6 ± 147.7 ms, p < 0.05), 그리고 rrT1의 F3와 F4사이(F3, 65.4 ± 7.7%; F4 57.3 ± 11.4%, p < 0.05)에서 통계적 유의미한 차이를 보였다. ROC 분석은 preT1과 postT1의 병용검사가 간섬유화 병기설정에 있어 가장 유용한 검사라는 것을 보여준다. 결론 preT1과 postT1은 간섬유화 병기가 증가함에 따라 증가하며, T1 mapping이 gadoxetic acid 조영증강 간 자기공명영상에서 간섬유화 병기설정에 있어 유용한 보조적 시퀀스로 사용될 수 있다.
