Selection of Human-Originated Lactobacillus acidophilus For Production of Probiotics

  • Lactobacillus acidophilus KFRI 233, a strain isolated from human, was selected as a candidate for probiotics due to its excellent growth in MRS broth where no special anaerobic condition is required. Both simultaneous and deferred agar diffusion assays exhibited Lb. acidophilus KFRI 233 to possess an antagonistic effect against Clostridium perfringens. Its antagonistic effect was pH dependent Associative culture of KFRI 233 and Cl. perfringens in broth resulted in maximum 94.04% inhibition of Cl. perfringens. $\beta$-Galactosidase activity of KFRI 233 was higher than other tested strains that are sold as commercial probiotics. Survival of KFRI 233 in pasteurized skim milk (4$^{\circ}C$) and Sherbet mix (-15$^{\circ}C$) after 7 days of storage were 71.9 and 105.5%, respectively.

10-Hydroxyoctadecanoic Acid Produced by Lactococcus lactis subsp. lactis as a Part of Flocculent Aggregate

  • A flocculent aggregate produced by Lactococcus lactis subsp. lactis in broths containing Tween 80, including MRS broth, had a microscopic structure of intertwined thread-like filaments. The filamentous structure was not elongated bacterial cells, but consisted of an organic solvent-soluble portion and an insoluble solid. L. lactis subsp. lactis grown at $25^{\circ}C$ for 15 days in tryptic soy broth with 0.1% Tween 80 and 1.0% malt extract produced 13 mg/l of flocculent aggregate, which contained 0.84 g/g of organic solvent-soluble component. The organic solvent-soluble part was identified as 10-hydroxyoctadecanoic acid.

  • The purpose of this study was to evaluate the rate of adherence of bacteria on various denture relining materials and to find out the effects of chitosan, when it was added to denture relining materials. Denture relining materials such as Tokuso rebase normal $set^{(R)}$, Mild $rebaron^{(R)}$, $Kooliner^{TM}$, and New $truliner^{TM}$ were used in this study. The adherence of Streptococcus mutans was studied on the surfaces in the denture relining materials with chitosan and in those without chitosan. When chitosan was added to M17 broth and MRS broth, the viable cell count of Streptococcus mutans was reduced. The viable cell count of Streptococcus mutans on the specimens decreased in the following order : Mild rebaron, Tokuso rebase normal set, and Newtruliner and Kooliner. The deture relining materials with chitosan showed a lower rate of adherence of Streptococcus mutans than those without chitosan.

Leuconostoc mesenteroides CJNU 0147 and Lactobacillus casei CJNU 0588 Improve Growth of a Bifidobacterium lactis Strain in Co-cultures

  • Previous studies have confirmed that fermented whey produced by Leuconostoc mesenteroides CJNU 0147 or Lactobacillus casei CJNU 0588 display bifidogenic growth stimulator (BGS) activity. The present study sought to determine if the strain itself can improve the growth of bifidobacteria in co-cultures. In reinforced clostridial medium (RCM), both strains stimulated the growth of a Bifidobacterium lactis strain during the exponential phase and also stimulated the growth during almost all growth phases in whey broth. Fermented whey containing viable Leu. mesenteroides CJNU 0147 and L. casei CJNU 0588 cells maintained viability of the B. lactis strain stored at $10^{\circ}C$ in MRS broth. Viable cell count of the B. lactis strain without the fermented whey was decreased to 5.6 log cfu/mL after 15 days, whereas that of the strain with the fermented whey was slightly increased to 7.1 log cfu/mL as compared with initial viable cell count of 6.9 log cfu/mL.

  • The increase in the serum cholesterol level of the rabbit fed a high cholesterol diet was prevented by the administration of a Lactobacillus sporogenes preparation, proportionally to the amount of the preparation. About 90% of the increase in the serum cholesterol level was inhibited in the rabbits fed the preparation (2%) as feed additives. In vitro, L. sporogenes cultured in MRS broth containing PPLO serum fraction as a cholesterol source decreased the cholesterol concentration in the spent broth and increased the intracellular cholesterol concentration. The ability of L. sporogenes to accumulate cholesterol was enhanced by the presence of oxgall in the medium. These results suggest that L. sporogenes may directly assimilate the cholesterol in the gastrointestinal tracts and may prevent the increase of the serum cholesterol level.

Optimization of Bacteriocin ST311LD Production by Enterococcus faecium ST311LD, Isolated from Spoiled Black Olives

  • Bacteriocin ST311LD is approximately 2.3 kDa in size. Low levels of bacteriocin activity were recorded in BHI and M17 broth (800 AU/ml) and in $10\%$ (w/v) soy milk (3,200 AU/ml). No bacteriocin pro-duction was recorded in $10\%$ (w/v) molasses, despite good growth. Optimal levels (12,800 AU/ml) were detected in MRS broth which had been supplemented with tryptone (20.0 g/l), saccharose (5.0 or 10.0 g/l) or vitamin C (1 ppm). Increased potassium levels did not result in higher levels of activity, and glycerol (1.0 g/l) inhibited the production of bacteriocin ST311LD.

Occurrence of Glutathione Sulphydryl (GSH) and Antioxidant Activities in Probiotic Lactobacillus spp.

  • The antioxidative ability on the basis of reduced glutathione sulphydryl level, the inhibition activities of linoleic acid peroxidation of cell free extract of Lactobacillus spp. and the effects of types of media and growth phase of the cells on the cellular GSH level have been determined. Correlation between reduced glutathione sulphydryl level and antioxidative ability of Lactobacillus spp. was analyzed: Lactobacillus casei HY 2782 contained 25.15 $\mu$mole/g of GSH, the cellular GSH level of L. casei HY 2782 reached maximum after 24 h of cultivation and tended to decrease on further cultivation up to 72 h. There was a significantly higher level of cellular GSH when grown in de Man Rogosa and Sharpe (MRS) broth than in tryptone phytone yeast extract (TPY) broth or bromcresol pruple dextrose (BCP) broth (p<0.05). The antioxidant activity of cell free extract of Lactobacillus spp. have been shown to be significantly different among strains in the inhibition of linoleic acid peroxidation by thiobarbituric acid (TBA) test (p<0.01). L. casei HY 2782 and L. acidophilus ATCC 4356 revealed a high degree of antioxidative effect in linoleic acid oxidation system. Spearmans' rank correlation coefficient between inhibitory activity on linoleic acid peroxidation and cellular GSH levels of Lactobacillus spp. was 0.65, which means a significant positive correlation.

Production and antifungal effect of 3-phenyllactic acid (PLA) by lactic acid bacteria

  • Phenyllactic acid (PLA), which is a known antimicrobial compound, can be synthesized through the reduction of phenylpyruvic acid (PPA) by lactate dehydrogenase of lactic acid bacteria (LAB). PLA-producing LAB was isolated from coffee beans, and the isolated LAB was identified as Lactobacillus zeae Y44 by 16S rRNA gene sequence analysis. Cell-free supernatant (CFS) from L. zeae Y44 was assessed for both its capability to produce the antimicrobial compound PLA and its antifungal activity against three fungal pathogens (Rhizoctonia solani, Botrytis cinerea, and Colletotrichum aculatum). PLA concentration was found to be 4.21 mM in CFS when L. zeae Y44 was grown in MRS broth containing 5 mM PPA for 12 h. PLA production could be promoted by the supplementation with PPA and phenylalanine (Phe) in the MRS broth, but not affected by 4-hydroxy-phenylpyruvic acid, and inhibited by tyrosine as precursors. Antifungal activity assessment demonstrated that all fungal pathogens were sensitive to 5 % CFS (v/v) of L. zeae Y44 with average growth inhibitions ranging from 27.8 to 50.0 % (p<0.005), in which R. solani was the most sensitive with an inhibition of 50.0 %, followed by B. cinerea and C. aculatum. However, pH modification of CFS to pH 6.5 caused an extreme reduction in their antifungal activity. These results may indicate that the antifungal activity of CFS was caused by acidic compounds like PLA or organic acids rather than proteins or peptides molecules.

도축 폐혈액 단백질을 이용한 Probiotics생산에서의 동결건조 조건 (Optimization of Freeze-drying Conditions for Probiotics Production with Animal Blood Proteins Added Medium.)

  • 본 연구는 도축 폐기물인 가축혈액의 이용을 위해 사료첨가제로서 혈분 성분이 함유된 유산균 probiotics의 생산을 목적으로 하여 수행되었다. 혈액으로부터 얻은 혈장을 질소원으로 이용한 BBPB 배지에, 시판 중인 미생물 사료첨가제로부터 분리된 probiotics용 Lactobacillus sp.를 배양하여 대조구인 MRS배지의 74%에 이르는 높은 생균수를 얻을 수 있었다. 얻어진 배양액의 동결 건조는 배양시간, 배양액의 pH보정, 배양액의 농축, 급속동결 등의 조건에 의해 균체의 생존율이 영향을 받았으며, 24시간 배양한 배양액을 pH 6.4로 보정한 뒤 10배로 농축하여 -5$0^{\circ}C$에서 3시간 동안 동결한 후 건조하는 조건으로 최적화되었다. BBPB 배지에 잔여하는 혈장 단백질 성분은 유산균의 동결건조 안정화 효과도 가지고 있는 것으로 나타났고 그 수준은 MRS와 유사하였다. 제제화를 위한 동결건조 안정제로는 Sucrose가 높은 효과를 나타내었고 10%첨가시 48.3%의 생존율로 $3.0{\times}^{10}$ CFU/g의 유산균 제제를 얻을 수 있었다. 본 연구에서의 동결건조조건을 이용하면 도축 폐혈액을 이용한 problotics의 산업적 생산이 가능할 것으로 사료된다.

황 흡착 알긴산의 Lactobacillus acidophillus 성장에 미치는 영향 및 항 돌연변이 작용 (The Antimutagenic Activity of Sulfated Alginate and Its Effect on the Growth of Lactobacillus acidophillus)

  • 톳으로부터 산ㆍ알칼리 (AASA) 방법으로 추출한 알긴산에 인위적으로 sulfate를 흡착시킨 후 ICP-AES를 이용해서 확인한 결과 3% alginate용액에서 1843 ppm 농도의 sulfate가 검출되었다. 배지 자체적으로 황을 함유하고 있는 MRS broth와 황이 없는 peptone water배지 에 S-alginate와 alginate를 0.25, 1, 2% 농도별로 첨가하여 Lactobacillus acidophilus를 접종한 후 배양하면서 유산균의 성장에 대한 영향을 살펴 본 결과 모든 농도에서 이들 배지에 S-alginate를 첨가한 것이 유산균의 성장을 증가시키는 것을 알 수 있었다. S-alginate와 alginate를 0.25% 첨가한 경우 유산균의 성장은 비슷한 값을 나타내었으나 농도가 증가할수록 S-alginate를 첨가한 군이 alginate만 첨가한 군보다는 유산균의 성장을 늘리는 것으로 나타났다. Ames test를 통해서 S-alginate는 돌연변이성을 전혀 나타내지 않았으며 항 돌연변이성는 직접변이원에 대하여 TA98균주는 14∼19%, TA100 균주에 대해서는 12∼17%까지의 억제율을 보였으며 2%의 S-alginate가 가장 높은 억제율을 나타내었다.
