Inhibition of the Expression of Matrix Metalloproteinases in Articular Chondrocytes by Resveratrol through Affecting Nuclear Factor-Kappa B Signaling Pathway

  • In the present study, we tried to examine whether resveratrol regulates the expression of matrix metalloproteinases (MMPs) through affecting nuclear factor-kappa B ($NF-{\kappa}B$) in articular chondrocytes. Rabbit articular chondrocytes were cultured in a monolayer, and reverse transcription-polymerase chain reaction (RT-PCR) was used to measure interleukin-${\beta}$ ($IL-1{\beta}$)-induced gene expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 and type II collagen. Effect of resveratrol on $IL-1{\beta}$-induced secretion of MMP-3 was investigated in rabbit articular chondrocytes using western blot analysis. To elucidate the action mechanism of resveratrol, effect of resveratrol on $IL-1{\beta}$-induced $NF-{\kappa}B$ signaling pathway was investigated in SW1353, a human chondrosarcoma cell line, by western blot analysis. The results were as follows: (1) resveratrol inhibited the gene expression of MMP-3, MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5, but increased the gene expression of type II collagen; (2) resveratrol reduced the secretion of MMP-3; (3) resveratrol inhibited $IL-1{\beta}$induced activation (phosphorylation) of inhibitory kappa B kinase (IKK), and thus phosphorylation and degradation of inhibitory kappa $B{\alpha}$ ($I{\kappa}B{\alpha}$); (4) resveratrol inhibited $IL-1{\beta}$-induced phosphorylation and nuclear translocation of $NF-{\kappa}B$ p65. This, in turn, led to the down-regulation of gene expression of MMPs in SW1353 cells. These results suggest that resveratrol can regulate the expression of MMPs through affecting $NF-{\kappa}B$ by directly acting on articular chondrocytes.

Effects of Hormones on the Expression of Matrix Metalloproteinases and Their Inhibitors in Bovine Spermatozoa

  • Proteases and protease inhibitors play key roles in most physiological processes, including cell migration, cell signaling, and cell surface and tissue remodeling. Among these, the matrix metalloproteinase (MMPs) pathway is one of the most efficient biosynthetic pathways for controlling the activation of enzymes responsible for protein degradation. This also indicates the association of MMPs with the maturation of spermatozoa. In an attempt to investigate the effect of MMP activation and inhibitors in cultures with various hormones during sperm capacitation, we examined and monitored the localization and expression of MMPs (MMP-2 and MMP-9), tissue inhibitors of metalloproteinases (TIMP-2 and TIMP-3), as well as their expression profiles. Matured spermatozoa were collected from cultures with follicle-stimulating hormone (FSH), luteinizing hormone (LH), and Lutalyse at 1 h, 6 h, 18 h, and 24 h. ELISA detected the expression of MMP-2, MMP-9, TIMP-2, and TIMP-3 in all culture media, regardless of medium type (FSH-supplemented fertilization Brackett-Oliphant medium (FFBO), LH-supplemented FBO (LFBO), or Lutalyse-supplemented FBO (LuFBO)). TIMP-2 and TIMP-3 expression patterns decreased in LFBO and LuFBO. MMP-2 and MMP-9 activity in FBO and FFBO progressively increased from 1 h to 24 h but was not detected in LFBO and LuFBO. The localization and expression of TIMP-2 and TIMP-3 in sperm heads was also measured by immunofluorescence analysis. However, MMPs were not detected in the sperm heads. MMP and TIMP expression patterns differed according to the effect of various hormones. These findings suggest that MMPs have a role in sperm viability during capacitation. In conjunction with hormones, MMPs play a role in maintaining capacitation and fertilization by controlling extracellular matrix inhibitors of sperm.

Comparative evaluation of Emblica officinalis as an etchant and an MMP inhibitor with orthophosphoric acid and chlorhexidine on the microshear bond strength of composite resin: an ex vivo study

  • Objectives: This study aimed to evaluate Emblica officinalis (Indian gooseberry or amla) as an acid etchant and matrix metalloproteinase (MMP) inhibitor, and to compare its effect on the microshear bond strength of composite resin with orthophosphoric acid (OPA) and 2% chlorhexidine (CHX) as an acid etchant and MMP inhibitor, respectively. Materials and Methods: The etching effect and MMP-inhibiting action of amla on dentin samples were confirmed by scanning electron microscopy (SEM) and gelatin zymography, respectively. Dentinal slabs (3 mm thick) from 80 extracted human molars were divided into 10 and 20 samples to form 2 control groups and 3 experimental groups. Groups 1, 2, and 4 were etched with OPA and groups 3 and 5 with amla juice. An MMP inhibitor was then applied: CHX for group 2 and amla extract for groups 4 and 5. Groups 1 and 3 received no MMP inhibitor. All specimens received a standardized bonding protocol and composite resin build-up, and were subjected to microshear bond strength testing. The force at which the fracture occurred was recorded and statistically analyzed. Results: Amla juice had a similar etching effect as a self-etch adhesive in SEM and 100% amla extract was found to inhibit MMP-9 by gelatin zymography. The microshear bond strength values of amla were lower than those obtained for OPA and CHX, but the difference was not statistically significant. Conclusions: Amla has a promising role as an acid etchant and MMP inhibitor, but further studies are necessary to substantiate its efficacy.

U-373-MG 세포의 이동 및 침윤에 미치는 simvastatin의 효과 (Effect of Simvastatin on the Migration and Invasion of U-373-MG Cells)

  • 본 연구에서는 simvastatin에 의한 U-373-MG 세포의 이동 및 침윤에 미치는 효과와 그 가능한 기작에 대해 조사하였다. U-373-MG 세포의 이동에 미치는 simvastatin의 효과를 확인한 결과, $0.01\;{\mu}M$의 simvastatin에서는 U-373-MG 세포의 이동이 약 43% 증가되었으나 $1\;{\mu}M$$20\;{\mu}M$에서는 세포의 이동이 각각 28%와 35% 억제되었다. 세포의 침윤 역시 $0.001\;{\mu}M$의 simvastatin에서는 23%, $0.01\;{\mu}M$에서는 26%의 침윤 증가가 관찰되었으나, $1\;{\mu}M$$10\;{\mu}M$에서는 세포의 침윤이 각각 45%와 54%가 억제되었다. $0.001\;{\mu}M$$0.01\;{\mu}M$의 simvastatin 처리에 의해 MMP-2의 분비량이 각각 30%와 59% 증가되었으나, $10\;{\mu}M$$20\;{\mu}M$의 simvastatin 처리에 의해 MMP-2의 분비량은 각각 31%와 60% 감소되었다. MMP-9 및 플라스민의 분비에 미치는 simvastatin의 효과도 MMP-2에 미치는 효과와 유사하였다. Simvastatin이 MMP-2와 MMP-9 단백질의 생성에 미치는 효과를 조사한 결과, $0.001{\sim}1\;{\mu}M$의 simvastatin에서는 MMP-2와 MMP-9 생성이 증가되었으나 $10{\sim}20\;{\mu}M$에서는 MMP-2와 MMP-9의 생성이 감소되었다. 이러한 결과는 낮은 농도의 simvastatin은 혈관신생을 유도하고 높은 농도의 simvastatin은 혈관신생을 억제한다는 것을 보여주는 것이다.

Brazilin Inhibits of TPA-induced MMP-9 Expression Via the Suppression of NF-${\kappa}B$ Activation in MCF-7 Human Breast Carcinoma Cells

  • Metastasis is the primary cause of from breast cancer mortality. Cell migration and invasion play important roles in neoplastic metastasis. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix (ECM), plays an important role in cancer cell invasion. NF-${\kappa}B$ is transcription factor important in the regulation of MMP-9, as the promoter of MMP-9 gene contains binding sites for NF-${\kappa}B$. Brazilin, an active component of sappan wood (Caesalpinia sappan), decreases TPA-induced MMP-9 expression and invasion in MCF-7 cells. Also, brazilin suppressed NF-${\kappa}B$ activation in TPA-treated MCF-7 cells. Taken together, we demonstrated that the inhibition of TPA-induced MMP-9 expression and cell invasion by brazilin is mediated by the suppression of the NF-${\kappa}B$ pathway in MCF-7 cells. This result suggest brazilin provide a potential therapeutic app roach for the treatment of breast cancer.

Characterization of Matrix Metalloproteinase Expression in Triglyceride Treated THP-1 Macrophages

  • Elevated blood triglyceride (TG) levels correlate with development of atherosclerosis suggesting that TG may promote the development of this disease. During atherosclerosis, TG is taken up by tissue macrophages which result in dramatic changes in various secreted factors. One such factor is the family of matrix metalloproteases (MMP) which are involved in tissue remodeling during both physiological and pathological processes. In this study, we examined the MMP expression profile in PMA-differentiated THP-1 macrophages treated with TG. We found that TG-treated THP-1 macrophages showed decreased expression of MMP-3, MMP-7, MMP-8 and MMP-9 in a time- and dose-dependent manner. In contrast, expression of MMP-1, MMP-2, and MMP-10 remained relatively unchanged after TG treatment. In addition, we found that expression of select MMPs was affected by various inhibitors of signaling pathways. In particular, expression of MMP-3 was slightly recovered by cRAF and PLC signaling pathway inhibitors. These data suggests a possible role of MMPs in macrophages during TG-induced atherosclerosis.

오미자(五味子)(Fructus Schisandrae)가 흰쥐 해마신경세포의 저산소증 모델에서 세포사에 미치는 영향 (Prevention of ROS Production and MMP Dissipation by Fructus Schisandrae(FS) in a Hypoxic Model of Cultured Rat Hippocampal Neurons)

  • Objectives : The purpose of this study was to investigate the effect of FS for the modulation of ROS and MMP in a hypoxic model of cultured rat cortical cells. Methods : For the effect of FS on the viability, FS was added to culture media (neurobasal supplemented with B27) and cell viability was measured by LDH assay. To investigate the effects of FS on ROS generation and MMP preservation, cells grown in FS-containing media were given a hypoxic shock(2% $O_2/5%$ $CO_2$, $37^{\circ}C$, 3 hrs) on DIV 10, stained with $H_2DCF-DA$(10 nM) and JC-1, respectively, and observed by fluorescent microscope. Results : 1. FS has a protective effect of cortical cells in both normoxia and hypoxia. 2. FS reduced the generation of ROS and this reduction was especially significant at 3 days after hypoxia. 3. FS was effective for the maintenance of MMP in hypoxia, and this efficacy was especially significant at 3 days after hypoxia. Conclusions : Taken together, these results indicate that FS attenuates ROS generation and MMP dissipation, which eventually protects from neuronal cell death in hypoxia.

근관 삼출액에서의 기질금속단백분해효소-8과 substance P의 수준에 관한 연구 (Matrix metalloproteinase-8 and substance P levels in root canal exudates of nonvital teeth)

  • 연구목적: 근관 치료를 전후로 근관 삼출액을 추출하여 MMP-8과 SP의 양을 측정하고 근관 치료에 따른 변화 및 이들간이 상관관계를 보는 것을 본 연구의 목적으로 하였다. 연구 재료 및 방법: 치수괴사, 혹은 이전 근관 치료 후 치근단 병소가 있어서 근관 치료가 예정된 35개의 치아를 대상으로 하였으며, 근관 치료 첫날, 2주 후, 4주 후 paper point를 이용하여 근관 삼출액을 채취하였다. MMP-8과 SP의 양은 enzyme-linked immunosorbent assay (ELISA)를 이용하여 정량 하였다. 삼출액의 채취 시점에 따른 MMP-8, SP 양의 변화 및 환자의 통증 정도, 임상증상에 따른 MMP-8, SP의 변화는 mixed model을 이용하여 통계 분석하였으며 MMP-8과 SP의 상관관계는 Pearson correlation coefficient를 구하여 분석하였다(p < 0.05). 결과: 근관치료가 진행됨에 따라 MMP-8, SP의 양은 유의성 있게 감소하였으며(p < 0.0001), 환자의 통증이나 타진검사 시 반응이 SP의 발현에 영향을 주는 것으로 나타났다. MMP-8의 경우 SP와 미약한 양의 상관관계가 있었지만 환자의 증상이나 다른 임상검사와 상관관계가 발견되지 않았다. 결론: 본 실험에서 사용된 paper point를 이용한 근관 삼출액의 채취를 통하여 치근단 염증과정에서 발현되는 MMP-8, SP를 검출할 수 있었으며 특히 SP의 경우 환자의 증상과 보다 관련이 깊은 것으로 나타났다.

Expression and Prognostic Value of Matrix Metalloproteinase-7 in Colorectal Cancer

  • The purpose of this study was to evaluate expression and prognostic value of matrix metalloproteinase-7 (MMP-7) in colorectal cancer (CRC) patients. CRC tissues and corresponding distal normal mucosa tissues of 118 CRC patients were assessed by immunohistochemistry. Correlations between MMP-7 expression, patients' clinic pathological features, and overall survival rate were analyzed. We found that positive expression of MMP-7 in CRC tissues was significantly higher than that in distal normal mucosa (61.0% vs. 39.8%, p =0.001). Poor histological differentiation, advanced clinical stage and lymph node metastasis were significantly correlated with MMP-7 expression in CRC. The overall survival rate was significantly higher in the MMP-7 negative group than the positive group (Log-rank test= 9.957, p= 0.002). MMP-7 appeared as a significant independent prognostic factor through multivariate survival analysis. Collectively, we found MMP-7 expression to be correlated with progression and metastasis of CRC and thus could be used as a predictive marker of prognosis in CRC patients.

발효 돈태반과 그 주요 다이펩타이드 섭취가 UVB 조사에 의한 무모 생쥐의 피부 주름생성에 미치는 효과 (The Effects of Orally Administered Fermented Porcine Placenta and Its Major Dipeptides on UVB-induced Wrinkle Formation in the Hairless Mice)

  • 본 연구에서는 발효 돈태반 (fermented porcine placenta, FPP)과 그의 주요 다이펩타이드인 L-Leucyl-Glycine (Leu-Gly), Glycyl-L-Leucine (Gly-Leu)의 섭취가 UVB 조사에 의한 무모 쥐의 피부 주름 생성에 미치는 효능을 알아보았다. Human Primary Dermal fibroblasts-Neonatal (HDF-N) 세포에서 세포 독성을 나타내지 않는 농도를 설정하여 평가하였을 때, FPP, Leu-Gly, Gly-Leu 처리 시 procollagen의 증가 및 MMP-1 (matrix metalloproteinase-1)의 감소를 확인하였다. 또한 무모 생쥐에 주 3회 UVB를 조사하여 광노화를 유도하였고, FPP 10, 100 mg/kg, Leu-Gly 10 mg/kg, Gly-Leu 10 mg/kg을 매일 총 8주간 경구투여 한 후, 주름생성, 홍반및 MMPs의 발현량을 측정하였다. 8주간 진행된 동물 실험 결과 UVB만 조사한 군과 비교하여 UVB를 조사하고 FPP, Leu-Gly, Gly-Leu을 섭취시킨 군에서 주름생성과 홍반이 감소하였고 피부 수분함량과 콜라겐 생합성이 증가하였다. 뿐만 아니라 FPP, Leu-Gly, Gly-Leu 섭취군에서 콜라겐 분해효소인 MMP-3, MMP-13의 mRNA 발현량이 감소하였고, MMP-2와 MMP-9의 활성이 감소하였다. 결과를 종합하였을 때, FPP의 주요 다이펩타이드인 Leu-Gly과 Gly-Leu은 자외선에 의한 주름 생성을 억제하고, 피부 손상을 회복시키는 효능을 갖는 피부미용식품 소재로서 활용 가능성이 클 것으로 사료된다.
