A Study on the Effectiveness of the Mortar Jet Method in Increasing the Strength of the Soft Ground (시멘트 몰탈형 고압분사공법(MJM)에 의한 연약지반 보강효과에 관한 연구)

  • Although high pressure discharge method is widely used for improving soft ground, it has various problems including lack of strength increase and the possibility of water pollution and soil contamination. MJM(Morta Jet Method) uses sand in addition to cement as the injection material. MJM uses triple rods with a built-in nozzle that allows easier discharge of the slime, resulting in higher replacement area ratio and more uniform formation of pillar hydrates, and thus results in significant increase in strength. MJM is expected to perform especially well as piles in marine clays. This study investigates the field applicability of the MJM through extensive laboratory and field tests.

  • The high pressure jet grouting method is mainly used in the grouting. But, this method has problems that the scale and strength of improved body is not constant with ground condition. Considering these problems, triple rod MJM that results in the high-strength effect by the technology of the injected ${\phi}7mm$ cement mortar was developed. In this MJM, the unconfined strength is estimated with the various combination ratio and engineering characteristic, strength improvement effect of improved body, was checked through the field test. It is known that the application of MJM was verified with ground and construction condition.

Isolation and Structural Determination of Antifungal Antibiotic from Streptomyces hygroscopicus MJM1004 (Streptomyces hygroscopicus MJM1004가 생산하는 항진균성 항생 물질의 분리 및 구조 결정)

  • Several Streptomyces strains were tested for potent antifungal agents active against phytopathogenic fungi. Among the tested, S. hygroscopicus MJM1004 showed a potent antifungal activity when assayed using Candida albicans as indicator organism. With the strain of MJM1004, fermentation medium for the production of an antifungal agent was developed with varying carbon sources, nitrogen sources, and mineral elements, which resulted in the highest productivity in the medium containing 2% soybean meal, 1% glucose, 2% starch, 0.3% $CaCO_3$, 0.05% $MgSO_4{\cdot}7H_2O$, 0.05% $K_2HPO_4$. The active compound showed a broad spectrum of antifungal activity against several plant pathogenic fungi. The antifungal compound was purified and showed the physicochemical characteristics similar to azalomycin F complex in NMR and MS analysis.

Foliar Application of Extract from an Azalomycin-Producing Streptomyces malaysiensis Strain MJM1968 Suppresses Yam Anthracnose Caused by Colletotrichum gloeosporioides

  • Yam anthracnose caused by Colletotrichum gloeosporioides (C.g) is the most devastating disease of yam (Dioscorea sp.). In the present study, we evaluated the culture filtrate extract (CFE) of azalomycin-producing Streptomyces malaysiensis strain MJM1968 for the control of yam anthracnose. MJM1968 showed strong antagonistic activity against C.g in vitro. Furthermore, the MJM1968 CFE was tested for inhibition of spore germination in C.g, where it completely inhibited spore germination at a concentration of 50 μg/ml. To assess the in planta efficacy of the CFE and spores of MJM1968 against C.g, a detached leaf bioassay was conducted, which showed both the treatments suppressed anthracnose development on detached yam leaves. Furthermore, a greenhouse study was conducted to evaluate the CFE from MJM1968 as a fungicide for the control of yam anthracnose. The CFE non-treated plants showed a disease severity of >92% after 90 days of artificial inoculation with C.g, whereas the disease severity of CFE-treated and benomyl-treated yam plants was reduced to 26% and 15%, respectively, after 90 days. Analysis of the yam tubers from the CFE-treated and non-treated groups showed that tubers from the CFE-treated plants were larger than that of non-treated plants, which produced abnormal smaller tubers typical of anthracnose. This study demonstrated the utility of the CFE from S. malaysiensis strain MJM1968 as a biofungicide for the control of yam anthracnose.

Rice bran fermentation by lactic acid bacteria to enhance antioxidant activities and increase the ferulic acid, ρ-coumaric acid, and γ-oryzanol content

  • Rice bran is considered a natural source of antioxidants. In this study, rice bran was fermented with lactic acid bacteria to increase its antioxidant activity. Four strains isolated from fermented food, Lactobacillus plantarum MJM60383, Lactococcus lactis subsp. lactis MJM60392, Lactobacillus fermentum MJM60393, and Lactobacillus paracasei MJM60396, were confirmed as safe through stability tests such as safety assessment for biogenic amine production, hemolytic activity, and mucin degradation, and showed high reducing capacity. The antioxidant activity of rice bran fermentation altered by these strains was evaluated using several methods including measurement of $Fe^{2+}$ chelating activity and scavenging activity by 1,1-diphenyl-2-picryl-hydrazil (DPPH), 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and nitric oxide assays. In this study, the total phenolic content and ${\gamma}$-oryzanol were evaluated by high-performance liquid chromatography. Compared to non-fermented rice bran and a commercial product, rice bran fermented with Lactococcus lactis subsp. lactis MJM60392 showed the highest phenolic content (844.13 mg GAE/g). Moreover, the content of ferulic acids, ${\rho}$-coumaric acid, and ${\gamma}$-oryzanol in rice bran increased after fermentation with L. lactis subsp. lactis MJM60392 and L. fermentum MJM60393 compared to other samples. Indeed, the DPPH radical scavenging activity and NO scavenging activity were also found to be high in these fermented rice brans. These results indicated that fermentation with lactic acid bacteria increases the active compound levels and the potent antioxidant activities of rice bran.

Functional Characterization of an Exopolysaccharide Produced by Bacillus sonorensis MJM60135 Isolated from Ganjang

  • The present study focused on the production, characterization, and in vitro prebiotic evaluation of an exopolysaccharides (EPS) from Bacillus sonorensis MJM60135 isolated from ganjang (fermented soy sauce). Strain MJM60135 showed the highest production ($8.4{\pm}0.8g/l$) of EPSs compared with other isolates that were screened for EPS production based on ropy culture morphology. Furthermore, MJM60135 was cultured in 5 L of medium and the EPS was extracted by ethanol precipitation. The emulsification activity of the EPS was higher in toluene than in o-xylene. Fourier transform infrared spectroscopy analysis showed the presence of hydroxyl and carboxyl groups and glycosidic linkages. The isolated EPS contained mannose and glucose, as observed by thin-layer chromatography analysis of the EPS hydrolysate. Lactic acid bacteria (LAB) and pathogenic E. coli K99 and Salmonella enterica serovar Typhimurium were tested for their growth utilizing the EPS from B. sonorensis MJM60135 as the sole carbon source for its possible use as a prebiotic. All the tested LAB exhibited growth in the EPS-supplied medium compared with glucose as carbon source, whereas the pathogenic strains did not grow in the EPS-supplied medium. These findings indicate that the EPS from B. sonorensis MJM60135 has potential application in the bioremediation of hydrocarbons and could also be used as a prebiotic.

Kitasatospora sp. MJM383 Strain Producing Two Antitumor Agents, Streptonigrin and Oxopropaline G

  • MJM383, a rare actinomycete sp. strain originated from Chinese soils, was isolated through an antimicrobial screening system. The analysis of 16S rDNA sequences and biochemical characterization determined the strain to belong to genus Kitasatospora. Both NMR and ESI mass data of its purified bioactive compounds revealed Kitasatospora sp. MJM383 to produce two antitumor agents, streptonigrin and oxopropaline G, which have been known to be produced from Streptomyces species. This is the first report to demonstrate the presence of antitumor agents produced by genus Kitasatospora.

The Measurement of Seasonal Evapotranspiration above Corn Canopy Based on the Bowen ratio-Energy Balance Method (보우엔비(比)-에너지수지방법(收支方法)에 의한 옥수수군락(群落) 증발산(蒸發散)의 계절별(季節別) 관측(觀測))

  • Heat energy distribution forming net radiation above corn canopy was determined by means of the Bowen ratio-energy balance method. Total-global solar radiation above crop canopy during the growing season was $1,559MJm^{-2}$ and total latent heat flux density was $960MJm^{-2}$. The data showed that 61.6% of the global solar radiation was used for a heat source of evapotranspiration (ETa) above corn canopy. Mean daily ETa ranged from 2.7 to 5.6mm. Total ETa, total drymatter, and water use efficiency were 394mm, $2,214gm^{-2}$, and $5.6gm^{-2}mm^{-1}$, respectivively.

Comparison of prosthetic models produced by traditional and additive manufacturing methods

  • PURPOSE. The purpose of this study was to verify the clinical-feasibility of additive manufacturing by comparing the accuracy of four different manufacturing methods for metal coping: the conventional lost wax technique (CLWT); subtractive methods with wax blank milling (WBM); and two additive methods, multi jet modeling (MJM), and micro-stereolithography (Micro-SLA). MATERIALS AND METHODS. Thirty study models were created using an acrylic model with the maxillary upper right canine, first premolar, and first molar teeth. Based on the scan files from a non-contact blue light scanner (Identica; Medit Co. Ltd., Seoul, Korea), thirty cores were produced using the WBM, MJM, and Micro-SLA methods, respectively, and another thirty frameworks were produced using the CLWT method. To measure the marginal and internal gap, the silicone replica method was adopted, and the silicone images obtained were evaluated using a digital microscope (KH-7700; Hirox, Tokyo, Japan) at 140X magnification. Analyses were performed using two-way analysis of variance (ANOVA) and Tukey post hoc test (${\alpha}=.05$). RESULTS. The mean marginal gaps and internal gaps showed significant differences according to tooth type (P<.001 and P<.001, respectively) and manufacturing method (P<.037 and P<.001, respectively). Micro-SLA did not show any significant difference from CLWT regarding mean marginal gap compared to the WBM and MJM methods. CONCLUSION. The mean values of gaps resulting from the four different manufacturing methods were within a clinically allowable range, and, thus, the clinical use of additive manufacturing methods is acceptable as an alternative to the traditional lost wax-technique and subtractive manufacturing.

Isolation and Characterization of a New ${\gamma}$-Polyglutamic Acid Producer, Bacillus mesentericus MJM1, from Korean Domestic Chungkukjang Bean Paste

  • Poly-${\gamma}$-glutamic acid (PGA) is an unusual anionic polypeptide and has great potential as an environmentally and industrially significant biodegradable material. A new ${\gamma}$-PGA producer, Bacillus mesentericus MJM1, with high production capacity was isolated from Korean domestic Chungkuckjang bean paste. It produced ${\gamma}$-PGA at the level of 10 g/l in suitable media. The viscosities of 5% initially extracted mucin and purified ${\gamma}$-PGA solutions were 660 cps and 600 cps, respectively. The produced ${\gamma}$-PGA polymer consisted of 2,000 glutamic acid residues with even proportion of L and D types with molecular mass of about 200- 300 kDa. Bacillus mesentericus MJM1 displayed ${\gamma}$-glutamyltranspeptidase (${\gamma}$-GTP) activity that is known to play a key role in ${\gamma}$-PGA biosynthesis. The ${\gamma}$-GTP coding region was located on the plasmid of 5.8 kb. The plasmid, named pMMH1, is a rolling-circle replication (RCR) plasmid and additionally contained a replication origin and type I signal peptidase (sipP) coding region.
