Effects of GHX02 on Chronic Obstructive Pulmonary Disease Mouse Model

  • Objectives: Chronic obstructive pulmonary disease (COPD) is characterized by chronic inflammation and irreversible airflow. This study aimed to evaluate the effects of GHX02 in a COPD-induced mouse model. Methods: The COPD mouse model was established by exposure to cigarette smoke extract and lipopolysaccharide which were administered by intratracheal injection three times with a 7 day interval. GHX02 (100, 200, 400 mg/kg) and all other drugs were orally administrated for 14 days from Day 7 to Day 21. Results: GHX02 significantly decreased the neutrophil counts in bronchoalveolar lavage fluid (BALF) and the number of $CD4^+$, $CD8^+$, $CD69^+$, and $CD11b^+/GR1^+$ cells in BALF and lung cells. GHX02 also suppressed the secretion of tumor necrosis factor-alpha ($TNF-{\alpha}$), interleukin-17A, macrophage inflammatory protein 2 (MIP2), and chemokine (C-X-C motif) ligand 1 (CXCL-1) in BALF and ameliorated the lung pathological changes. Conclusions: Thus, GHX02 effectively inhibited airway inflammation by inhibiting migration of inflammatory cells and expression of pro-inflammatory cytokines. Therefore, GHX02 may be a promising therapeutic agent for COPD.

Therapeutic Effects of Sesamum Indicum Extracts on Atopic Dermatitis-Like Skin Lesions of NC/Nga Mouce (호마자 추출물이 NC/Nga 생쥐의 아토피 피부염에 미치는 영향)

  • Objectives The purpose of this study is to investigate the effects of Sesamum indicum extracted (SEI) on atopic dermatitis in an in-vitro and in-vivo experiment using a MC/9 murine mast cells and a NC/Nga mouse. Methods In-vitro experiment, IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ and GM-CSF mRNA expression were evaluated by Real-time PCR, IL-13, MIP-$1{\alpha}$ production by ELISA and manifestations of NFAT-1, NFAT-2, c-jun, c-fos, NF-${\kappa}B$ p65 transcription factors by western blotting. In-vivo experiment, we measured WBC, Eosinophil, Neutrophil, and serum IL-5, IL-13 in NC/Nga atopic dermatitis mouse, IL-5, IL-13, IFN-${\gamma}$, IL-4 in the spleenocyte culture supernatant by ELISA, the absolute cell numbers of CD4+, CD8+, +Gr-1+CD11b, B220+CD23+ in the axillary lymph node (ALN), peripheral blood mononuclear cells (PBMCs) and dorsal skin tissue, IL-5, IL-13 by Real-time PCR, the distribution of tissue inflammation and cellular infiltration by H&E and toluidine blue. Results SEI decreased IL-4, IL-5, IL-6, IL-13, GM-CSF, TNF-${\alpha}$ mRNA expression, IL-13, MIP-$1{\alpha}$ production and the expression of transcription factors including NFAT-1, c-jun, NF-${\kappa}B$ p65 in MC/9 murine mast cells. SEI orally administration decreased cell number of WBC, Eosinophil, the level of serum IgE, total cell number of ALN and dorsal skin tissue, absolute cell number of CD4+, CD8+, B220+CD23+ in the ALN. SEI orally administration also increased absolute cell number of CD8+/CD3+ and decreased Gr-1+/CD11b+ in PBMCs, decreased CD4+ in dorsal skin tissue, inhibited IL-5, IL-13 mRNA expression. Infiltration levels of inflammatory immune cells, mast cells and thickness of epidermis decreased in dorsal skin tissue. Conclusions SEI can regulate allergic inflammatory response suppressed the gene expression and production of cytokines that mediate allergic reactions, and will be able to be effectively utilized in the treatment of atopic dermatitis future.

The Effects of Dictamni Radicis Cortex Extracts on DNCB Induced Atopic Dermatitis-like Skin Lesions of NC/Nga Mouse (백선피(白鮮皮) 추출물이 DNCB로 유발된 NC/Nga 생쥐의 아토피 피부염에 미치는 영향)

  • Objectives Dictamni Radicis Cortex extracts (DRC) has been known to suppress allergic reaction, however the cellular target of DRC and its mode of action remain unclear. The purpose of this study is to investigate the effects of Dictamni Radicis Cortex extracts on DNCB induced atopic dermatitis-like skin lesions of NC/Nga mouse. Methods This study was designed to investigate the effects of DRC extract in the DNP-IgE-induced activation of MC/9 murine mast cell lines in vitro and in the DNCB-induced activation of NC/Nga mouse in vivo. For this investigation, We examined IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ and GM-CSF mRNA expression by Real-time PCR, IL-13, MIP-$1{\alpha}$ production by ELISA analysis and manifestations of NFAT1, NFAT2, AP-1 and NF-${\kappa}B$ p65 transcription factors by western blotting in vitro. Then, we examined WBC, eosinophil and neutrophil in NC/Nga mouse, IL-5, IL-13 in serum, IFN-${\gamma}$, IL-4 in the spleenocyte culture supernatant, the absolute cell numbers of $CD4^+$, $CD8^+$, $^+Gr-1^+CD11b$, $B220^+CD23^+$ in the ALN, PBMCs and dorsal skin, IL-5, IL-13 in the dorsal skin by Real-time PCR and the distribution of mast cells by H&E and toluidine blue. Results In vitro the mRNA expression of IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$, GM-CSF and IL-13, MIP-$1{\alpha}$ production by ELISA analysis were completely abolished by DRC and the western blot analysis decreased the expression of mast cell-specific transcription factors including NFAT-1, NF-${\kappa}B$ p65. In vivo DRC oral adminstration also decreased the counts of WBC, eosinophils and inflammatory cytokines such as IL-13 and IgE in the serum. DRC oral adminstration elevated IL-4 level in the spleenocyte culture supernatant. DRC oral adminstration decreased total ALN cells, total skin cells, cell numbers of $CD4^+$, $B220^+CD23^+$ in the ALN, $^+Gr-1^+CD11b$ in the PBMCs and $CD4^+$, $CD8^+$ in the dorsal skin. The mRNA expression of IL-5, IL-13, thickness of epidermis, inflammation immune cells and mast cells were abolished by DRC in the dorsal skin. Conclusions Histological examination showed that infiltration levels of immune cells in the skin of AD-induced NC/Nga mouse were much improved by DRC oral adminstration. These results, therefore, suggest that DRC can regulate molecular mediators and immune cells that are functionally associated with atopic dermatitis induced in NC/Nga mouse, and may play an important role in recovering AD symptoms.

  • In this thesis we observe microvascular structure in mice by using micro-computed tomography (CT), which is high-resolution X-ray imaging equipment that can acquire Real-time dynamic image, and it aims to investigate the usefulness of micro-CT developed by Institute for Radiological Imaging Science Wonkwang University School of Medicine. After acquiring the systemic images of rats, contrast-enhanced 3D images of vascular structures could be acquired by using Maximum Intensity Projection (MIP) and Volume Rending Technique (VRT), This was divided into each vascular system of head, abdomen and heart and systemic vascular system.

Real-Time Implementation of Acoustic Echo Canceller for Mobile Handset Using TeakLite DSP Core (Teaklite DSP Core 를 이용한 이동통신 단말기용 음향반향제거기의 실시간 구현)

  • In this paper, we developed an acoustic echo canceller in real-time using TeakLite DSP Core, which will be placed in the vocoder chip of a mobile handset. Considering the limited computational capacity given to the acoustic echo canceller in a vocoder chip, we employed a FIR-type adaptive filter using a conventional NLMS algorithm. To begin with, we designed and implemented an acoustic echo canceller with floating-point format C-source code, and then converted it into fixed-point format through integer simulation. Then we programmed and optimized it in the assembler level to make it run ill real-time. After optimization procedure, the implemented echo canceller has approximately 624 words of program memory and 811 words of data memory. With 8 KHz sampling rate and 256 filter taps in the echo canceller that corresponds to 32 msec of echo delay, it requires 14.12 MIPS of computational capacity. For coverage of 16 msec echo delay, i.e., 128 filter taps, 9 MIPS is requited.

Analysis of Intrafractional Mass Variabilities Using Deformable Image Registration Program (영상변조 프로그램을 이용한 호흡 위상 간 종양의 움직임 특성 분석)

  • The aim of this study is to compare the geometric characteristics of the lung tumor, such as tumor centroid, HU change relative to breath phase, depending on tumor location and adhesion using 4DCT and deformable image registration program (MIMVista). The Y axis change was most significant and the mean Y axis centroid fluctuation was $7.32{\pm}6.88mm$ in lower lung tumor. The mean HU variation in lower lung mass has changed more than other locations, and its mean HU variation was $7.7{\pm}4.97%$ and non-adhered mass was more changed. Correlation for the mass volume between 3DCT and MIP was very high and its coefficient was 0.998. The effect of tumor location, adhesion and diaphragm excursion to geometric uncertainties was analyzed by linear regression model, it was influenced to mass deformation and geometrical variation so much except diaphragm excursion. but intra-fractional and inter-patient's uncertainties were great, so it couldn't find any exact deformation trend.

Analysis of Inflammatory Cytokines from the Cecum and Proximal Colon of Mice Infected with Enterotoxigenic Bacteroides fragilis

  • Enterotoxigenic Bacteroides fragilis (ETBF) causes inflammatory diarrhea in humans and animals and is also implicated in colorectal cancer. ETBF-infected mice exhibit a prominent large intestinal inflammation characterized by neutrophil infiltration and induction of the Th17 response. In this study, we examined differences in the secreted cytokine profile of the cecum and proximal colon of ETBF-infected mice using an antibody array. Of the cytokines examined, we found that the cecal tissues from ETBF-infected mice secreted elevated levels of G-CSF, IL-6, IL-17 and LIX compared to non-toxigenic Bacteroides fragilis (NTBF) and Mock infected mice. The proximal colon tissues from ETBF-infected mice secreted higher levels of G-CSF, IL-6, KC, LIX, MIP-1g and MCP-1. This study demonstrates that the cecum and colon should be considered separately when assays are used to determine immune responsiveness to enteric infections.

Classification of Alkali Activated GGBS Mortar According to the Most Suitable Usage at the Construction Site

  • The usage of OPC-free alkali activated ground granulated blast furnace slag(GGBS) mortar has been widely studied on the previous years, due to its advantages on sustainability, durability and workability. This paper brings a new view, aiming to classify the best application in situ for each mortar, according to the type and activator content. By this practical implication, more efficiency is achieved on the construction site and consequently less waste of materials. In order to compare the different activators, the following experiments were performed: analysis of compressive strength at 28 days, setting time measured by needles penetration resistance, analysis of total pore volume performed by MIP and permeability assessment by RCPT test. In general, activated GGBS had acceptable performance in all cases compared to OPC, and remarkable improved durability. Following the experimental results, it was confirmed that each activator and different concentrations impose distinct outcome performance to the mortar which allows the classification. It was observed that the activator Ca(OH)2 is the most versatile among the others, even though it has limited compressive strength, being suitable for laying mortar, coating/plaster, adhesive and grouting mortar. Samples activated with NaOH, in turn, presented in general the most similar results compared to OPC.

  • Internet Engineering Task Force(IETF) proposed the Hierarchical Mobile IPv6(HMIPv6) to support mobility efficiently. The HMIPv6 was developed to reduce the signaling overhead and delay concerned with Binding Update in Mobile IPv6. However, the HMIPv6 still need a further enhancement for supporting the real-time application because HMIPv6 only concerns with the latency resulted within a MAP For providing seemless handoff we propose a scheme that can reduce latency when Mobile Node changes a MAP. Also we compare the HMIPv6 with the proposed scheme through a analysis model.

High-yield Purification and Characterization of Recombinant Human Leukotactin-1 in Pichia pastoris

  • The human chemokine, the short version of leukotactin-1(shLkn-1;molecular weight=7.2 kD and 66 amino acids), was expressed and secreted into a culture medium using the me-thylotrophic yeast, Pichia pastoris. The recombinant shLkn-1 was purified from the culture supernatant using a simple two-step procedure consisting of cation exchange and reverse phase chromatography(RPC), in which shLkn-1 was highly purified (99.5%) with a high recovery yield of 82.7%. The C-terminal truncated derivative of shLkn-1 was found in the supernatant and was separated by RPC. The physicochemical properties of the purified shLkn-1 were verified to be the same as expected. The biological activity of the purified recombinant shLkn-1 was also quantified using a chemotaxis assay. It was observed that the recombinant shLkn-1 had the maximum migration activity at a concentration of 10nM, as potent as MIP-1${\alpha}$.
