This study describes an alternative mixed culture fermentation technology to anaerobically convert lignocellulosic biomass into butyric acid, a valuable product with wide application, without supplementary cellulolytic enzymes. Rice straw was soaked in 1% NaOH solution to increase digestibility. Among the tested pretreatment conditions, soaking rice straw at $50^{\circ}C$ for 72 h removed ~66% of the lignin, but retained ~84% of the cellulose and ~71% of the hemicellulose. By using an undefined cellulose-degrading butyrate-producing microbial community as butyric acid producer in batch fermentation, about 6 g/l of butyric acid was produced from the pretreated rice straw, which accounted for ~76% of the total volatile fatty acids. In the repeated-batch operation, the butyric acid production declined batch by batch, which was most possibly caused by the shift of microbial community structure monitored by denaturing gradient gel electrophoresis. In this study, batch operation was observed to be more suitable for butyric acid production.
Fire can affect microbial community structure of soil through altered environmental conditions, nutrient availability, and biotic source for microbial re-colonization. We examined the influence of fire on chemical properties and soil enzyme activities of soil for 10 months. We also characterized the soil microbial community structure through ester-linked fatty acid analysis(EL-FAME). For this study, we established five burned plots(1*1 m) and 5 unburned plots outside the margin of fire. Soil was sampled three soil cores in a each plots and composited for analysis at 1, 3, 5, 8, and 10 month after fire. The fire caused an increase in soil pH, exchangeable Ca, and Mg, organic matter, available $P_2O_5$ compared to unburned sites. The content of $NH_4-N$ in burned site was significantly higher than that of unburned site and this effect continued for 8 months after fire. There was no difference of $NO_3-N$ content in soil between burned and unburned site. Fire caused no change in acid phosphatase and arylsulfatase activities but $\beta$-glucosidase and alkaline phosphatase activities in burned site were increased compared to unburned site. Microbial biomass as estimated by total concentration of EL-FAMEs in burned sites was significantly higher than that of unburned sites at one month after fire. Burned site decreased the EL-FAMEs indicative of gram-positive bacteria and tended to increase the fatty acid associated with gram-negative bacteria at one and three months after fire. The sum of EL-FAME compound $18:2{\omega}6,9c$ and $18:1{\omega}9c$ as served fungal biomarkers was decreased in burned site compared to unburned site.
Response surface methodology (RSM) was successfully applied to optimize for the production of Ganoderma lucidum in batch fermentations using the whey (40,000 mg latose/L) as substrate. This study was performed according to the central composite design (CCD) with respect to pH and temperature, where the designed intervals were 3.3$22.9^{\circ}C$$37.1^{\circ}C$, respectively. A second-order factorial design of the experiments was used to build empirical models providing a quantitative interpretation of the relationships between the two variables. The optimum conditions to maximize the production of G. lucidum were pH 4.2 and $28.3^{\circ}C$. At optimum conditions, the mycelial dry weight (MDW) and residual soluble COD (SCOD) were simultaneously used to evaluate the biokinetic coefficients assocoated with substrate inhibition model by nonlinear least squares method with 95% confidence interval. The. maximum microbial growth rates (${\mu}m$), half saturation coefficient ($K_s$), and the inhibition substrate concentration ($K_{is}$) were determined to be 0.095 l/hr, 128,000 mg SCOD/L and 49,000 mg SCOD/L, respectively. And the microbial yield coefficient (Y), biomass decay rate coefficient ($K_d$), and the maintenance energy coefficient ($m_s$) were determined to be 0.37 mg MDW/mg SCOD, 0.001 1/hr, and 0.0015 1/hr, respectively.
Bacillus subtilis ATCC 6633 was grown in BHIB medium supplemented with $Mn^{2+}$ for 96 h at $37^{\circ}C$ in a shaker incubator. After removing the microbial biomass, a lipopeptide biosurfactant was extracted from the supernatant. Its structure was established by chemical and spectroscopy methods. The structure was confirmed by physical properties, such as Hydrophile-Lipophile Balance (HLB), surface activity and erythrocyte hemolytic capacity. The critical micelle concentration (cmc) and erythrocyte hemolytic capacity of the biosurfactant were compared to those of surfactants such as SDS, BC (benzalkonium chloride), TTAB (tetradecyltrimethylammonium bromide) and HTAB (hexadecyltrimethylammonium bromide). The maximum hemolytic effect for all surfactants mentioned was observed at concentrations above cmc. The maximum hemolytic effect of synthetic surfactants was more than that of the biosurfactant produced by B. subtilis ATCC 6633. Therefore, biosurfactant would be considered a suitable surface-active agent due to low toxicity to the membrane.
Three typical biological solid wastes, namely, animal manure, garbage, and sewage sludge, were compared with regard to the composting process and the changes in microbial community structure. The effects of different bulking agents such as rice straw, vermiculite, sawdust, and waste paper were compared in manure compost. The differences in the microbial community were characterized by the quinone profile method. The highest mass reduction was found in garbage composting (56.8%), compared with manure and sludge (25% and 20.2%, respectively). A quinone content of $305.2\;{\mu}mol/kg$ was observed in the late stage of garbage composting, although the diversity index of the quinone profile was 9.7, lower than that in manure composting. The predominant quinone species was found to be MK-7, which corresponds to Gram-positive bacteria with a low G+C content, such as Bacillus. The predominance of MK-7 was especially found in the garbage and sludge composting process, and the increase in quinones with partially saturated long side-chains was shown in the late composting process of manure, which corresponded to the proliferation of Actinobacteria. The effects of different bulking agents on the composting process was much smaller than the effects of different raw materials. High organic matter content in the raw materials resulted in a higher microbial biomass and activity, which was connected to the high mass reduction rate.
본 연구에서는 회분여과방식을 이용하여 부하율에 따라 생성되는 생물대사성분의 특성 및 분포를 관찰하였다. 실험에 사용된 기질은 단일 탄소인 phenol을 사용하였으며, 분자량 분포실험을 위하여 분자량이 각각 30K, 100K Dalton 및 $0.45{\mu}$ membrane filter를 이용하여 구하였다. 페놀농도가 120, 230 및 440 mg/L 일 때 비기질이용율(q)은 각각 0.639, 1.281, 1.744 (mgTOC/mg MLSS/day)로 나타났으며 Run C일 때 가장 높은 이용율을 나타냈다 . 내생단계에서 미생물의 사멸율($K_d$)는 각각 0.0536, 0.0661, 0.0749($day^1$)이며 생성계수 ($SMP_e$) 는 각각 0.006, 0.0058, 0.0057($day^1$)로 나타났다. 초기 유입된 기질이 기질분해에 의해 생성된 $SMP_s$로 분해되어지며, 시간경과에 따라 $SMP_{nd}$ 로 진행됨을 알수 있었다. 기질분해 완료 후 미생물의 내생단계에 접어들면서 $SMP_e$성분으로 전환되었다. 유입부하율에 따른 분자량 분포 측정결과는 운전시간이 경고함에 따라 점차 저분자 물질이 고분자의 난분해성 물질로 전환되었다.
양파는 2모작 작물로써 소득이 큰 작물이면서 건강에 좋은 기능성분이 많이 함유하고 있어 생산자와 소비자에게 인기가 높은 식품이다. 최근 건강과 환경을 중시하는 생활패턴 때문에 소비자는 유기농 양파를 더 선호할 것으로 예상되나 자연생태계를 고려하면서 안정적이고 건강한 먹거리를 생산하기는 매우 어렵다. 유기농법의 한계를 극복하고자 양파 잔재물에서 증류법으로 양파 추출액을 채취하여 유기농양파 재배기간 중에 500배로 희석하여 7번 되돌려 주고 생산성과 관련된 생육, 수량, 양분 및 토양미생물을 비교한 결과는 다음과 같았다. 1. 양파의 생육은 대체로 대조구<순환구<관행구 순으로 좋았으며, 초기에는 순환구가 관행구보다 현저히 불량하였으나 후기에 갈수록 처리간 차이가 줄어져 비슷하였다. 2. 양파의 수량은 순환구가 대조구보다 9.3% 증가되었고 관행구보다 11.0% 감소되었으며, 300 g 이상 구중 분포비율이 높을수록 수량도 많아지는 경향이었다. 3. 양파의 무기성분 함량은 순환구가 대조구와 관행구보다 많았으며, 특히 인산, 철, 아연 함량 등은 관행구에 비하여 47%, 78%, 172% 현저히 증가되었다. 4. 양파의 유기성분 함량은 순환구가 대조구와 관행구보다 많았으며, 퀘세틴 함량은 순환구가 대조구와 관행구에 비하여 각각 33.9%, 44.0% 증가되었다. 5. 토양의 내생 균근균은 순환구가 대조구와 관행구에 비하여 각각 40%, 31.7% 더 많았고 미생물 스트레스 지표는 순환구가 대조구와 관행구에 비하여 15%, 20% 감소되었다.
유류 및 중금속으로 오염된 토양에서 Pb 및 Cd에 내성을 갖는 미생물을 분리하여 미생물 내 중금속 흡착 특성을 조사하였다. 분리한 토착미생물의 Pb 및 Cd의 흡착특성과 흡착에 영향을 미치는 요인 중에 생장단계, 중금속 농도, 생물량, pH, 온도에 따른 영향을 비교하였다. 또한 흡착등온식을 적용하여 중금속의 흡착용량과 흡착강도를 알아보았다. 낮은 중금속 초기 농도와 높은 생물량에서 높은 중금속 제거 효율을 가지며 중금속 마다 다른 흡착 효율을 보여 주었다. 흡착 효율은 미생물 생장 말기, pH 5~9 조건에서 최적의 효율을 나타내었으나, 25~$35^{\circ}C$에서 온도 변화에 따른 영향은 미미하였다. 생물흡착 과정을 Langmuir 등온 흡착식에 적용하면, 이론적 최대 흡착량은 Pb와 Cd에 대해서 각각 62.11과 192.31 mg/g로 나타났고, $R^2$가 0.71과 0.98로 계산되었다. Cd는 세포 표면의 단일 층에 단분자 흡착에 의한 생물흡착이 진행되었으나, Pb는 미생물 대사 작용을 통한 세포 내로의 축적 작용과 미생물 내 음이온과의 반응에 의한 침전물 형성작용 둥을 통하여 생물흡착이 진행된 것으로 판단된다.
Understanding N mineralization dynamics in soil is essential for efficient nutrient management. An anaerobic incubation experiment was conducted to examine N mineralization potential and N mineralization rate of the organic amendments with different C:N ratio in paddy soil. Inorganic N in the soil sample was measured periodically under three temperature conditions ($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$) for 90 days. N mineralization was accelerated as the temperature rises by approximately $10%^{\circ}C^{-1}$ in average. Negative correlation ($R^2=0.707$) was observed between soil inorganic N and C:N ratio, while total organic carbon extract ($R^2=0.947$) and microbial biomass C ($R^2=0.824$) in the soil were positively related to C:N ratio. Single exponential model was applied for quantitative evaluation of N mineralization process. Model parameter for N mineralization rate, k, increased in proportion to temperature. N mineralization potential, $N_p$, was very different depending on C:N ratio of organic input. $N_p$ value decreased as C:N ratio increased, ranged from $74.3mg\;kg^{-1}$ in a low C:N ratio (12.0 in hairy vetch) to $15.1mg\;kg^{-1}$ in a high C:N ratio (78.2 in rice straw). This result indicated that the amount of inorganic N available for crop uptake can be predicted by temperature and C:N ratio of organic amendment. Consequently, it is suggested that the amount of organic fertilizer application in paddy soil would be determined based on temperature observations and C:N ratio, which represent the decomposition characteristics of organic amendments.
