Estrogen Receptor Enhances the Antiproliferative Effects of Trichostatin A and HC-toxin in Human Breast Cancer Cells

  • Trichostatin A, an antifungal antibiotics, and HC-toxin are potent and specific inhibitors of histone deacetylase activity. Histone deacetylase inhibitors are new class of chemotherapeutic drugs able to induce tumor cell apoptosis and/or cell cycle arrest. In this study, the antiproliferative activities of trichostatin A and HC-toxin were compared between estrogen receptor positive human breast cancer cell MCF-7 and estrogen receptor negative human breast cancer cell MDA-MB-468. Trichostatin A and HC-toxin showed potent antiproliferative activity in both MCF-7 and MDA-MB-468 cells. In MCF-7 cells that contain high level estrogen receptor, trichostatin A and HC-toxin brought about three-times more potent cell growth inhibitory effect than estrogen receptor negative MDA-MB-468 cells. Both trichostatin A and HC-toxin showed cell cycle arrest at G$_2$/M phases of MCF-7 and MDA-MB-468 cells in a dose- and time- depen- dent manner. Trichostatin A and HC-toxin also induced apoptosis from MCF-7 and MDA-MB-468 cells in a dose- and time-dependent manner. Results of this study suggested that antipro-liferative effects of trichostatin A and HC-toxin might be involved in estrogen receptor signaling pathway, but cell cycle arrest and apoptosis of trichostatin A and HC-toxin might not be involved in estrogen receptor system of human breast cancer cells.

Genistein Inhibits the Proliferation of MDA-MB-231 Cells In Vivo

  • This study was conducted to evaluate the influence of diets containing genistein and soy extract on the growth of MDA-MB-231 cells implanted into female Balb/c mice. Four-week-old female athymic nude mice (Balb/c) were acclimated to an AIN-93G control diet for 1 week and then injected MDA-MB-231 cells ($1{\times}10^6$/site) and were continued on the on AIN-93G control diet. Five weeks after injecting the MDA-MB-231 cells ($1{\times}10^6$/site), two experimental groups were assigned to diets containing genistein (750 ${\mu}g/g$ AIN-93G diet) or 0.6% soy extract (containing genistein at 750 ${\mu}g/g$ AIN-93G diet) until they were sacrificed. Tumor growth was significantly reduced in the groups treated with genistein and soy extract compared to the control group. The results of the proliferating cell nuclear antigen (PCNA) assay also revealed that genistein and soy extract treatment reduced the proliferation of MDA-MB-231 cells in vivo. In the present study, dietary isoflavone was provided just before solid tumor formation, and thus the timing of dietary isoflavone administration may be critical to the suppression of tumor growth.

Performance Analysis for Spatial Multiplexing MIMO in MB-OFDM UWB Receivers (MB-OFDM UWB 시스템에서 공간 다중화 MIMO 수신기의 성능 분석)

  • This paper presents the spatial multiplexing MIMO system to increase data rate to double in MB-OFDM UWB system, which is ECMA standards, and compares BER performance of various receiver structures. The complexity and BER performance of various types of spatial multiplexing receivers are compared and analyzed using diagonal and horizontal encoding techniques for $2{\times}2$\;and\;2{\times}3$ antennas systems. Computer simulations exhibit that $2{\times}2$ MML and $2{\times}3$ ZF method show better BER performance than that of SISO system with simple complexity.

  • The retention and fatigue properties of ferroelectric LiNbO$_3$ thin films were studied. Metal-ferroelectric-semiconductor(MFS) devices by using rapid thermal annealed LiNbO$_3$/Si structures were successfully fabricated and demonstrated nonvolatile memory operations of the MFS devices. The I$_{D}$-V$_{G}$ characteristics of MFSFET\`s showed a hysteresis loop due to the ferroelectric nature of the LiNbO$_3$ thin film. The ferroelectric capacitors showed practically no polarization degradation up to about 10$^{10}$ switching cycles when subjected to symmetric bipolar voltage pulse (peak-to-peak 6V, 50% duty cycle) in the 500kHz. The retention properties of the LiNbO$_3$ thin films were quite good up to about 10$^{3}$ s . s .

Anticancer Effects of the Isoflavone Extract from Chungkukjang via Cell Cycle Arrest and Apoptosis in MDA-MB-453 Cells (청국장에서 얻은 Isoflavone의 MDA-MB-453세포에서 항암효과 및 관련 기전)

  • The objective of this study is to evaluate the anticancer effects of the isoflavone extract from Chungkukjang in human breast cancer, MDA-MB-453 cells. For this study, MDA-MB-453 cells were treated with 12.5, 25, and $50{\mu}g$ isoflavone extract for 24, 48, and 72 hr. Cell proliferations were decreased in a time- and dose-dependent manner. Reduced cell proliferation was suspected by apoptosis or cell cycle arrest. Therefore, after treatment of $50{\mu}g$ isoflavone extract, apoptotic cells were investigated by annexin V staining. The results indicated that isoflavone extract increased the number of early apoptotic cells compared with control. Cleaved PARP was also increased. Next, we investigated the cell cycle and related proteins. The isoflavone extract leads to cell cycle arrest at the G2/M phase. Moreover isoflavone extract had influenced cell cycle relate proteins such as cyclin B1, cyclin A, and p21. These results suggest that isoflavone extract from Chungkukjang induce apoptosis and cell cycle arrest at G2/M phase via regulation of cell cycle-related proteins in MDA-MB-453 cells.

Automatic Generation of MIB for Network Management (네트웍 관리를 위한 MIB의 자동생성)

  • Network management in TMN concerns to the operating system and communication equipments in network, and defines them as objects. GDMO(guidelines for the Definition of Managed Objects) is used to describe those objects. GDMO is not directly used for managing the network, but translated into a language with object-oriented paradigm. And GDMO refers to ASN.1(Abstract Syntax Notation One) for manage objects. This paper presents design and implementation techniques for the translator which automatically translates the specification of ASN.1 and GDMO to the object-oriented language for generating MIB(Managed object Instance Base). This system, unlike the existing source code generator, is designed to generate various object-oriented languages automatically, which are used to generate Managed object Instance Base(MIB). And the system includes various graphic user interface to enhance the development environment of ASn.1 and GDMO

Novel Suppressive Effects of Ketotifen on Migration and Invasion of MDA-MB-231 and HT-1080 Cancer Cells

  • The high mortality rates associated with cancer reflect the metastatic spread of tumor cells from the site of their origin. Metastasis, in fact, is the cause of 90% of cancer deaths. Therefore, considerable effort is being made to inhibit metastasis. In the present study, we screened ketotifen for anti-migratory and anti-invasive activities against MDA-MB-231 breast cancer and HT-1080 fibrosarcoma cancer cells. Cancer cell migration and invasion were measured using multi-well chambers. Additionally, western blots were used to examine the effects of ketotifen on the expressions of CDC42, Rho, Rac, and matrix metalloproteinase 9 (MMP-9). The results showed that ketotifen dose-dependently suppressed the migration and invasion of MDA-MB-231 and HT-1080 cells. Ketotifen also suppressed the expressions of CDC42, Rac, and Rho, which, significantly, are involved in MDA-MB-231 and HT-1080 cancer cell migration. Moreover, ketotifen suppressed the expression and activity of MMP-9, which is involved in degradation of the extracellular matrix leading to invasion. The overall data suggested that ketotifen suppresses the migration and invasion of MDA-MB-231 and HT-1080 cancer cells via inhibition of CDC42, Rac, Rho, and MMP-9 expression.

  • $CNT/TiO_2$ composites were prepared by using multiwall carbon nanotubes (MWCNT) and various titanium alkoxide precursors. The composites were comprehensively characterized by scanning electron microscopy (SEM), field-emission scanning electron microscopy (FE-SEM), X-ray diffraction (XRD), energy dispersive X-ray analysis (EDX) and UV-vis absorption spectroscopy. The photoactivity of the prepared materials under UV irradiation, was tested using the conversion of MB from model aqueous solution. Finally, according to the results of methylene blue (MB) removal experiment, we could see that sample CTOS have better MB removal effect than samples CTIP and CTPP.

Effect of Angelica keiskei Extract on Apoptosis of MDA-MB-231 Human Breast Cancer Cells (신선초 추출물이 인체 유방암 세포 MDA-MB-231의 세포 사멸에 미치는 영향)

  • We investigated the effect of Angelica keiskei ethanol (AKE) extract on cell death in MDA-MB-231 human breast cancer cells. MDA-MB-231 cells were cultured in the presence 125, 150 and 175 ${\mu}g$/mL concentrations of AKE for 24 hours. MTT assays demonstrated that mitochondrial dehydrogenase activities decreased in a dose-dependent manner in MDA-MB-231 cells (p<0.05). In contrast, the proportion of dual staining with Hoechst 33342/ethidium bromide(EtBr) for cell death increased in a dose-dependent manner in MDA-MB-231 cells (p<0.05). In particular, the levels of cell death caused by apoptotic program showed marked increases in the 150 and 175 ${\mu}g$/mL AKE groups, as revealed by flow cytometry. An apoptotic suppressor gene, Bcl-2, significantly decreased at the transcript level (p<0.05). The expression levels of proapoptotic genes, both Bax and caspase 3 significantly increased (p<0.05). Furthermore, the ratio of Bcl-2/Bax mRNA which is considered to be an important indicator of apoptosis, significantly decreased in a dose-dependent manner (p<0.05). These results taken together indicate that, the AKE extract used in this study induces cell death in MDA-MB-231 human breast cancer cells.

Rejection property of geosmin and 2-Methylisoborneol (MIB) with high concentration level at multi stage nanofiltration (NF) membrane system (다단 나노여과 공정에서 고농도 geosmin 및 2-Methylisoborneol (MIB)의 제거특성)

  • Algal problem in drinking water treatment is being gradually increased by causing deterioration of water supplies therefore, especially taste and odor compounds such as geosmin and 2-MIB occur mainly aesthetic problem by its unpleasant effects resulting in the subsequent onset of complaints from drinking water consumer. Recently, geosmin and 2-MIB are detected frequently at abnormally high concentration level. However, conventional water treatment without advanced water treatment processes such as adsorption and oxidation process, cannot remove these two compounds efficiently. Moreover, it is known that the advanced treatment processes i.e. adsorption and oxidation have also several limits to the removal of geosmin and 2-MIB. Therefore, the purpose of this study was not only to evaluate full scale nanofiltration membrane system with $300m^3/day$ of permeate capacity and 90% of recovery on the removal of geosmin and 2-MIB in spiked natural raw water sources at high feed concentration with a range of approximately 500 to 2,500 ng/L, but also to observe rejection property of the compounds within multi stage NF membrane system. Rejection rate of geosmin and 2-MIB by NF membrane process was 96% that is 4% of passage regardless of the feed water concentration which indicates NF membrane system with an operational values suggested in this research can be employed in drinking water treatment plant to control geosmin and 2-MIB of high concentration. But, according to results of regression analysis in this study it is recommended that feed water concentration of geosmin and 2-MIB would not exceed 220 and 300 ng/L respectively which is not to be perceived in drinking tap water. Also it suggests that the removal rate might be depended on an operating conditions such as feed water characteristics and membrane flux. When each stage of NF membrane system was evaluated relatively higher removal rate was observed at the conditions that is lower flux, higher DOC and TDS, i.e., $2^{nd}$ stage NF membrane systems, possibly due to an interaction mechanisms between compounds and cake layer on the membrane surfaces.
