Recapitulation of previously reported associations for type 2 diabetes and metabolic traits in the 126K East Asians

  • Over the last decade, genome-wide association studies (GWASs) have provided an unprecedented amount of genetic variations that are associated with various phenotypes. However, previous GWAS were mostly conducted in European populations, and these biased results for non-Europeans may result in a significant reduction in risk prediction for non-Europeans. An issue with the early GWAS was the winner's curse problem, which led to misleading results when constructing the polygenic risk scores (PRS). Therefore, more non-European population-based studies are needed to validate reported variants and improve genetic risk assessment across diverse populations. In this study, we validated 422 variants independently associated with glycemic indexes, liver enzymes, and type 2 diabetes in 125,872 samples from a Korean population, and further validated the results by assessing publicly available summary statistics from European GWAS (n = 898,130). Among the 422 independently associated variants, 284, 320, and 361 variants were replicated in Koreans, Europeans, and either one of the two populations. In addition, the effect sizes for Koreans and Europeans were moderately correlated (r = 0.33-0.68). However, 61 variants were not replicated in both Koreans and Europeans. Our findings provide valuable information on effect sizes and statistical significance, which is essential to improve the assessment of disease risk using PRS analysis.

MicroRNA-Gene Association Prediction Method using Deep Learning Models

  • Micro ribonucleic acids (miRNAs) can regulate the protein expression levels of genes in the human body and have recently been reported to be closely related to the cause of disease. Determining the genes related to miRNAs will aid in understanding the mechanisms underlying complex miRNAs. However, the identification of miRNA-related genes through wet experiments (in vivo, traditional methods are time- and cost-consuming). To overcome these problems, recent studies have investigated the prediction of miRNA relevance using deep learning models. This study presents a method for predicting the relationships between miRNAs and genes. First, we reconstruct a negative dataset using the proposed method. We then extracted the feature using an autoencoder, after which the feature vector was concatenated with the original data. Thereafter, the concatenated data were used to train a long short-term memory model. Our model exhibited an area under the curve of 0.9609, outperforming previously reported models trained using the same dataset.

  • As environmental Problems are increasing, many attempts to solve problems and preserve environment are appearing around the world. Not only in construction, but also dwelling life, residents attend many activities to improve environmental problems. Ecological development example of Freiburg in Germany was focused in this study to show the direction of development of environmental friendly housing complex in Korea. The target of study is Vauban Housing Complex of Freiburg in Germany. The study methods are literature study and field trip. Contents of the study include 1) formation process of the complex 2) physical environment of the complex 3) environmental friendly factors of the complex 4) environmental friendly life style of the residents, etc. Result of this study is as follows. Vauban Housing Complex was remodeled from the existed army buildings by residents themselves through education and negotiations. Residents' life satisfaction and housing adaptation level were high on the whole. The principle of the complex formation was use of natural feature itself in maximum against developing in minimum. For instance, construction materials, energy saving, resources saving were planned and used carefully. The residents led community successfully according to environmental friendly life principles. In conclusion, the persistent study on environmental components is necessary that is suitable to Korean situation.

  • Octadecyltrimethylammonium-TCNQ(1:1) complex was synthesized by the reaction of LiTCNQ with octadecyltrimethylammonium bromide and the reaction product was identified with FT-IR and UV/Vis spectroscopies. In order to prepare the LB films, ${\pi}-A$ isotherm characteristics of octadecyltrimethylammonium-TCNQ(1:1) complex were investigated. The LB films of octadecyltrimethylammonium-TCNQ(1:1) complex were formed on various substrates and the formation of ultrathin films of octadecyltrimethylammonium-TCNQ(1:1) complex was confirmed by using FT-IR and UV-Vis spectroscopies.

Human Health Risk Assessment of Trichloroethylene from Industrial Complex A

  • This study investigated the human health risks of trichloroethylene from Industrial Complex A. The excessive carcinogenic risks for central tendency exposure were $1.40{\times}10^{-5}$ for male and female residents in the vicinity of Industrial Complex A. The excessive cancers risk for reasonable maximum exposure were $2.88{\times}10^{-5}$ and $1.97{\times}10^{-5}$ for males and females, respectively. These values indicate that there are potential cancer risks for exposure to these concentrations. The hazard index for central tendency exposure to trichloroethylene was 1.71 for male and female residents. The hazard indexes for reasonable maximum exposure were 3.27 and 2.41 for males and females, respectively. These values were over one, which is equivalent to the threshold value. This result showed that adverse cancer and non-cancer health effects may occur and that some risk management of trichloroethylene from Industrial Complex A was needed.

  • The significance of complexation facilities in Japan is to recognize the problems resulting from social/economic changes such as rapid informatization, globalization, low rates of birth, and aging population, and to strengthen the national competitiveness and urban advantages. Likewise, there have been increasing demands for community facilities that can meet the new requirements in terms of efficient administration and resident-centered administrative service system in line with changes in the society and administrative conditions. Thus, analyzed are cases concerning complex community facilities in Japan that have coped with changes in consciousness in the community earlier than in Korea. Specifically, the concept of modernday complex community facility is investigated, and the complexation of Korea Japan complex community facilities is divided to physical complexation and functional complexation to grasp the types. The complex community facilities are compared, and the characteristics in terms of spatial composition are also examined. The result of analyzing complex community facility cases is comprehensively analyzed to find out new possibilities in this regard.

  • This study is a research for revitalized characteristics of compounded community center which is a complex aid facility with a key position of regional culture and welfare. Recent complex conditions of community center and public facility is studied on this research. The methods are searches in the Internet, telephone interviews, and documents of present local conditions. The results are as follow : The community center in Seoul City support 2 million people per a center in average and self-supporting financial rate came out low. The community center formed physical shape in one building with community facilities. A district complex community center type, which is a village office united to public facilities, expected to be increased. The community center compound 1 or 2 facilities, divided 15 types. Most of community center is complex type of community center with village office and the other types are complex type of community center with a hall for the aged, nursery, and library. Accordingly, the community center keeps up the complex type of community center with village office type as a physical shape in one building and the district complex community center type complexes with hall for the aged, nursery and library.

Modulation of autophagy by miRNAs

  • MicroRNAs (miRNAs) can regulate the expression of genes that are involved in multiple cellular pathways. However, their targets and mechanism of action associated with the autophagy pathway are not fully investigated yet. EWSR1 (EWS RNA-Binding Protein 1/Ewing Sarcoma Break Point Region 1) gene encodes a RNA/DNA binding protein that is ubiquitously expressed and plays roles in numerous cellular processes. Recently, our group has shown that EWSR1 deficiency leads to developmental failure and accelerated senescence via processing of miRNAs, but its role in the regulation of autophagy remains elusive. In this context, we further investigated and found that EWSR1 deficiency triggers the activation of the DROSHA-mediated microprocessor complex and increases the levels of miR125a and miR351, which directly target Uvrag. Interestingly, the miR125a- and miR351-targeted reduction of Uvrag led to the inhibition of autophagy in both ewsr1 knockout (KO) MEFs and ewsr1 KO mice. In summary, our study demonstrates that EWSR1 is associated with the posttranscriptional regulation of Uvrag via miRNA processing. The regulation of autophagy pathway in miRNAs-Uvrag-dependent manner provides a novel mechanism of EWSR1 deficiency-related cellular dysfunction. [BMB Reports 2015; 48(7): 371-372]
