Reaction Mechanism of Troleandomycin on the Activity of Human Liver Microsomal Cytochrome P450 3A4 (인체 간 조직의 Cytochrome P450 3A4의 활성에 대한 Troleandomycin의 작용기전)

  • Incubation of aflatoxin $B_1$ $(AFB_1)$ with microsomes isolated from human liver number 110 yielded two metabolite peaks which were aflatoxin $Q_1$ $(AFQ_1)$ and $(AFB_1)$-exo-8, 9-epoxide (exo-epoxide) in high performance liquid chromatography. Production ratio of $AFQ_1$ to exo-epoxide was 2.43$\pm $0.04. Metabolism of $(AFB_1)$ to $(AFQ_1)$ and exo-epoxide was inhibited by troleandomycin in a same degree although troleandomycin was not activated as a mechanism-based inhibitor. The inhibitory effect was dependent upon either the incubation time with $(AFB_1)$ or the preincubation time before the addition of $(AFB_1)$. Incubation of troleandomycin and NADPH by the microsomes resulted in the formation of a cytochrome P 450 (P450)-metabollc intermediate (MI) complex and the level was approximately 80% of total P450 3A4 in the microsomes. This figure was similar to that of the inhibitory effect of troleandomycin on $AFB_1$ metabolism. Glutathione which was reported that it prevented the formation of MI complex in rat liver microsomes did not inhibit the formation of MI complex in human liver microsomes. These results suggested that the inhibitory effect of troleandomycin on $AFB_1$ metabolism is due to the formation of MI complex with P450 3A4. And the reaction mechanism of troleandomycin by human liver microsomes might be dlfferent from that one by rat liver microsomes.

Biological roles and an evolutionary sketch of the GRF-GIF transcriptional complex in plants

  • GROWTH-REGULATING FACTORs (GRFs) are sequence-specific DNA-binding transcription factors that regulate various aspects of plant growth and development. GRF proteins interact with a transcription cofactor, GRF-INTERACTING FACTOR (GIF), to form a functional transcriptional complex. For its activities, the GRF-GIF duo requires the SWITCH2/SUCROSE NONFERMENTING2 chromatin remodeling complex. One of the most conspicuous roles of the duo is conferring the meristematic potential on the proliferative and formative cells during organogenesis. GRF expression is post-transcriptionally down-regulated by microRNA396 (miR396), thus constructing the GRF-GIF-miR396 module and fine-tuning the duo's action. Since the last comprehensive review articles were published over three years ago, many studies have added further insight into its action and elucidated new biological roles. The current review highlights recent advances in our understanding of how the GRF-GIF-miR396 module regulates plant growth and development. In addition, I revise the previous view on the evolutionary origin of the GRF gene family.

  • The MiDF Cement Composite is a high-performance construction material with low defects that dehydrates surplus water through pressurization and minimizes air gap between particles. In other words, the performance expression of the MiDF cement complex is affected by pressurized conditions. Thus, this study analyzed the physical characteristics of MiDF cement complex according to the power and pressure of the ga-power and the time of application and intends to use it as a basic data for optimal mixing.

  • We study fibred Riemannian spaces with almost complex structures which are induced by the almost complex structure or the almost contact structure on the base and fibre. We show that if the total space is a complex space form, then the total space is locally Euclidean. Moreover, we deal with the fibred Riemannian space with various Kaehlerian structures.

  • Exploring microRNA (miRNA) and mRNA regulatory interactions may give new insights into diverse biological phenomena. Recently, miRNAs have been discovered as important regulators that play a major role in various cellular processes. Therefore, it is essential to identify functional interactions between miRNAs and mRNAs for understanding the context- dependent activities of miRNAs in complex biological systems. While elucidating complex miRNA-mRNA interactions has been studied with experimental and computational approaches, it is still difficult to infer miRNA-mRNA regulatory modules. Here we present a novel method, termed layered hypernetworks (LHNs), for identifying functional miRNA-mRNA interactions from heterogeneous expression data. In experiments, we apply the LHN model to miRNA and mRNA expression profiles on multiple cancers. The proposed method identifies cancer-specific miRNA-mRNA interactions. We show the biological significance of the discovered miRNA- mRNA interactions.

Rationally designed siRNAs without miRNA-like off-target repression

  • Small interfering RNAs (siRNAs) have been developed to intentionally repress a specific gene expression by directing RNA-induced silencing complex (RISC), mimicking the endogenous gene silencer, microRNAs (miRNAs). Although siRNA is designed to be perfectly complementary to an intended target mRNA, it also suppresses hundreds of off-targets by the way that miRNAs recognize targets. Until now, there is no efficient way to avoid such off-target repression, although the mode of miRNA-like interaction has been proposed. Rationally based on the model called "transitional nucleation" which pre-requires base-pairs from position 2 to the pivot (position 6) with targets, we developed a simple chemical modification which completely eliminates miRNA-like off-target repression (0%), achieved by substituting a nucleotide in pivot with abasic spacers (dSpacer or C3 spacer), which potentially destabilize the transitional nucleation. Furthermore, by alleviating steric hindrance in the complex with Argonaute (Ago), abasic pivot substitution also preserves near-perfect on-target activity (∼80-100%). Abasic pivot substitution offers a general means of harnessing target specificity of siRNAs to experimental and clinical applications where misleading and deleterious phenotypes from off-target repression must be considered.

A Study on the Complex type & Space Character of Japanese Community Mixed-support Facility (일본 공동체복합지원시설의 복합화 유형과 공간에 관한 연구)

  • After 2004, as the community mixed-support facility in Japan was admitted as a private business of public facilities based on the Special Law for Urban Regeneration, the community mixed-support facilities in Japan are developed into various types with the view of the urban regeneration and urban renewal, more different point of view, and a tendency of establishment of the community facility has been considerably changed for the past twenty years. The reasons of the change are the social demand for the reformation of facility and the modification of subsidy constitution according to the establishment of equal facility. Some examples of that are the construction of lifelong education community and the change into a equivalent society between male and female and the adjusting to the aging and the welfare society and the social situation of more consumption recently, and in order to emerge from former building system, new facilities related to the contents are being expanded. The complex of the community mixed-support facility has an advantage in that it supplies the services for the complex of the community mixed-support facility, constitutes the communal character and the sense of solidarity among residents and can lead the development of region through the exchange of regional information. Through the Japanese community mixed-support facility which has many cases of it, this study would recognize the suggestion, the feature of it and examine the complex types.

A Variable Ordering Method for OPKFDDs using Complex Terms (Complex term을 이용한 OPKFDD의 입력변수 순서 방법)

  • OPKFDD(Ordered Pseudo-Kronecker Functional Decision Diagram)는 각 노드에서 다양한 decomposition을 취할 수 있는 Ordered-DD(Decision Diagram)의 한 종류이다. OBDD(Ordered Binary Decision Diagram)에서 각 노드는 Shannon decomposition 만을 이용하는 반면, OPKFDD는 각 노드마다 Shannon, positive Davio, negative Davio decomposition 중의 하나를 사용하도록 하며 많은 경우 매우 적은 수의 노드로 함수를 표현할 수 있다. 그러나 각 노드마다 각기 다른 확장 방법을 선택할 수 있는 특징 때문에 입력 노드에 대한 확장 방밥과 입력 변수 순서의 결정에 의해서 OPKFDD의 크기가 좌우되며 이에 대한 최적의 해를 구하는 것은 매우 어려운 문제로 알려져 있다. 본 논문에서는 DD 크기를 기준을 노드 수로 하여 기존의 BDD(Binary Decision Diagram) 자료구조에서 OPKFDD를 효율적으로 유도해내는 방법을 제시하고 complex term을 이용하여 이를 최소화하는 알고리즘을 제시한다. 그리고 입력변수 순서 결정을 위하여 다출력함수의 경우 함수간의 포함관계를 고려한 그룹-sifting과 각 노드의 확장 방법을 제안하고 실험 결과를 제시한다.
