Zinc and Selenium Requirements for Glutathione Peroxidase Activity and Cell Survival in Chinese Hamster Ovary Cells Overexpressing Metallothionein

  • Many defined cell culture media were formulated over 3() years ago and may be deficient in certain micronutrients whose essentiality has only subsequently been recognised. The objective of this study was to evaluate whether alpha-minimal essential medium (MEM) supplemented with 10% foetal bovine serum contained sufficient selenium for optimal activity of the selenium containing enzymes cytosolic glutathione peroxidase (cGPx) and phospholipid hydroperoxide glutathione peroxidase (PHGPx) in cultured Chinese hamster ovary (CHO) cells. Additionally, the effect of zinc deficiency and metallothionein (MT) overexpression on cGPx and PHGPx activity was studied. The addition of 100 nM of selenous acid to the culture medium increased cGPx expression by 10-fold and PHGPx by about 2-fold in both wild-type CHO-K1 cells and CHO-K1 cells overexpressing mouse MT-1. Zinc deficiency had no significant effect on enzyme activity, but cells overexpressing mouse MT-1 had higher levels of cGPx activity. Zinc deficiency decreased cell survival but overexpression of MT-1 was partially protective, probably because its presence in quantity favoured the uptake, sequestration and cellular retention of any remaining zinc. This study demonstrates that selenium in complete alpha-MEM is insufficient for optimal cGPx and PHGPx activity and may compromise the cellular response to oxidative stress.

Binary Sequence Generator with a Large Number of Output Sequences (다수열 출력 이진 수열 발생기)

  • The number of output sequence was proposed as a characteristic of binary sequence generators for cryptographic application, but in general most of binary sequence generators have single number of output sequence. In this paper, we propose two types of binary sequence generators with a large number of output sequences. The first one is a Switched-Tap LFSR (STLFSR) and it applies to the generalized nonlinear function and the Geffe's generator as example. The other is a generalized memory sequence generator(GMEM-BSG) which is an improved version of the Golic's memory sequence generator (MEM-BSG) with a large number of output sequences, and its period, linear complexity, and the number of output sequence are derived.

A COMPARATIVE STUDY OF PRESERVING ABILITY OF HUMAN PERIODONTAL LIGAMENT CELLS STORED IN DIFFERENT STORAGE MEDIA (수종의 저장용액에서 치주인대세포의 생존율 비교)

  • Preservation of the remaining periodontal ligament cells on an avulsed tooth is very important to the successful outcome of replantation. HBSS is recommended as the most suitable storage medium for the avulsed tooth that cannot be replanted immediately. But their availability near the site of an accident is doubtful. The purpose of this in vitro study was to compare periodontal ligament cells stored in different storage media obtained easily on the spot. Human periodontal ligament cells were collected from the premolar teeth extracted for orthodontic treatment. Cells were cultured in ${\alpha}-MEM$ culture medium containing 20% FBS, at $37^{\circ}C$ 100% humidity, in a 5% $CO_2$ incubator. Cells were cultured in 96 well culture plate, $5{\times}10^4$ cells per well with ${\alpha}-MEM$ and incubated for 24 hours. After discarding the medium, those cells were cultured in ${\alpha}-MEM$ contained with 10% FBS, pasteurized milk, sterilized saline, unstimulated saliva and bench-dried state at $25^{\circ}C$ room temperature for 30, 60, 90, 120, 180 minutes respectively. And then each group was measured using MTT assay. The results were as follows. 1. Between the group of each time, there was statistically significant difference. Periodontal ligament cells viability was highest in pasteurized milk and was reduced stepwisely in sterilized saline, unstimulated saliva and bench-dried state(p<0.05). 2. between the time of each group, there was statistically significant difference(p<0.05) but was no statistically significant difference at 90-120 minutes in pasteurized milk and at 60-90 minutes and 120-180 minutes in sterilized saline(p>0.05). In conclusion, HBSS as storage medium of an avulsed tooth is not practical on the spot. Insteadily pasteurized milk can be recommended to maintain the periodontal ligament cells viability.

  • On the development of hirame(Paratichtys olivaceus) culture, outbreak of scuticociliata infection was reported to cause severe damage in Japan. To establish effective measures for isolation and cultivation of this ciliate, we tried to culture this pathogenic ciliate using medium for bacteria and fish cell lines in vitro. Scuticociliata from the brain tissues of infected fish was aseptically inoculated to CHSE-214 cells cultured in MEM-10 without antibiotic. Scuticociliata grew well and the number of ciliate reached $10^6\;cells/ml$ at temperatures of $15^{\circ}C$ to $20^{\circ}C$ for 10d. The number of ciliate cultured in the cell lines is 10 times higher than the numbers cultured in the liquid medium alone. This ciliata could be cloned by dilution method. Scuticociliata isolated could grow well on 42 different cell lines that were established from marine fish, warm freshwater fish, and salmonids. This ciliate could be preserved in liquid nitrogen for more than 6 months. Subsequently, we observed the optimal temperature and salinity for growth, and tested the sensitivities of this organism to formaldehyde, flagyl(Metronidazole), Ekuteshin(Combination compound of sulfamonometoxin and ormethoprim), and ozonixation. Optimal temperature for growth was $25^{\circ}C$ and salinity was 1.0 to 1.5%. Washed scuticociliata was killed by formaldehyde at the concentration of 50ppm for 10min, but was not completely killed even at a high concentration of 400ppm for 20min in MEM-5. Flagyl and Ekuteshin can inhibit the growth of scuticociliata at the concentration of 1,000 and $100{\mu}g/ml$ in MEM-10, respectively. More than 99% of this scuticociliata could be killed by ozonization at a dose equivalent to $1.0mg/\ell$ oxidant for 30sec in sea water. Isolated scuticociliata showed the pathogenicity to the cultured hirame by artificial infection(I. P. injection, $10^5\;cells$/fish). The number of scuticociliata in the water could be counted by most probable number(MPN) method using tissue culture, and the minimum detectable number was $1.8\;cells/\ell$. The number in the reservoir tank for water supply to the culture tank was 110 cells/l. After cleaning by elimination of the sediments from of the reservoir tank and disinfected with formaldehyde, number of scuticociliata decreased and was counted less than $1.8\;cells/\ell$ and infection rate of cultured hirame was decreased.

  • If R is ring and M is a right (or left) R-module, then M is called a faithful R-module if, for some a in R, x.a=0 for all x.mem.M then a=0. In [4], R.E. Johnson defines that M is a prime module if every non-zero submodule of M is faithful. Let us define that M is of prime type provided that M is faithful if and only if every non-zero submodule is faithful. We call a right (left) ideal I of R is of prime type if R/I is of prime type as a R-module. This is equivalent to the condition that if xRy.subeq.I then either x.mem.I ro y.mem.I (see [5:3:1]). It is easy to see that in case R is a commutative ring then a right or left ideal of a prime type is just a prime ideal. We have defined in [5], that a chain of right ideals of prime type in a ring R is a finite strictly increasing sequence I$_{0}$.contnd.I$_{1}$.contnd....contnd.I$_{n}$; the length of the chain is n. By the right dimension of a ring R, which is denoted by dim, R, we mean the supremum of the length of all chains of right ideals of prime type in R. It is an integer .geq.0 or .inf.. (b) If R is prime ring and dim$_{r}$R=n then dim$_{l}$ R=n if dim$_{l}$ R=n if dim$_{l}$ R<.inf. (c) Let R be a principal right and left ideal domain. The effects of calcium sulfate on periodontal ligament cells (Calcium sulfate제재가 치주인대세포에 미치는 영향)

  • Calcium sulfate has a long history of medical use as an implant material. The biocompatibiliry of the material has been clearly established. Bone ingrowth concomitant with resorption occurs rapidly with efficient conduction of bone from particle to particle. Calcium sulfate also has a potential for functioning as a good bamer membrane. The purpose of this study was to compare the biocompatibility of different types of calcium sulfate grafting materials including an expelimental calcium sulfate compound on periodontal ligament cells in vitro as a preliminary test towards the development of a more convenient and useful form of grafting material which could promote regeneration of periodontal tissue. Human periodontal ligament cells were collected from the premolar teeth extracted for orthodontic treatment. cells were cultured in a.MEM culture medium containing 20% FBS, at $37^{\circ}C$ and 100% humidity, in a 5% CO2 incubator. Cells were cultured into 96 well culture plate $1{\times}104$ cells per well with $\alpha$-MEM and incubated for 24 hours. After discarding the medium, those cells were cultured in $\alpha$-MEM contained with 10% FBS alone (control group), in medcal-grade calcium sulfate(MGCS group), in plaster(plaster group), experimental calcium sulfate paste(CS paste group) for 1, 2, 3 day respectively. And then each group was characterized by examining of the cell counting, MTI assay, collagen synthesis. The results \vere as follows. 1. In the analysis of cell proliferation by cell counting, both medical-grdde calcium sulfate group and plaster group showed no stastically significant difference at day 1, 2, 3 accept for plaster group at day 1 compared to control group, but there was stastically significant difference between CS paste group and all other groups at day 1, 2, 3(P<0.05). 2. In the analysis of cytotoxicity by MIT assay, both medical-grade calcium sJlfate group and plaster group showed no stastically significant difference compared to control group at day 1, 2, 3 but there was stastically significant difference between CS paste group and all other groups at day 1, 2, 3(P<0.OS). 3. In the analysis of collagen synthesis by immunoblotting assay, high level was detected for medical-grade calcium sulfate group and plaster group at day 1, 2, 3 compared to CS paste group. On the basis of these results, medical-grade calcium sulfate and plaster was shown to possess biocompatibility whereas the CS paste had unfavourable outcome. This observation shows a need for modification of the materials contained in calcium sulfate paste.

Sound quality metrics to express the discomfort of overload excavator noise during operation (과부하 굴삭기 소음의 불쾌감 표현인자)

  • In this paper, we tried to find out sound quality metrics to express discomfort of overload excavator noise and to develop sound quality indexes through multiple regression analysis by using them. For this purpose, the interior noise of cabin under overload condition was recorded for six excavator models with different noise properties and Jury test was carried out by PCM (Paired Comparison Method) and MEM (Magnitude Estimation Method). Jury test result with low consistency was classified into two groups with different preference tendencies by cluster analysis and multiple regression analysis was conducted in order to find out which sound quality metrics have significant effects on discomfort(low preference). As a result, we figured out that the sound quality metrics to express the discomfort were the partial loudness (= $PN_{10Bark}$) between 0 and 10 Bark in case of group1 and the difference between engine noise(= $dB_{EG}$) and hydraulic system noise ($dB_1$) in case of group2. Using the results of preference ranking and tendency analysis of PCM followed by the correlation analysis between PCM and MEM, the more reliable results were adopted by excluding the data with low consistency obtained from Jury test via MEM.

Preparation and Characterization of Silicone Hydrogel Lens Containing Poly(ethylene glycol) (PEG를 포함한 실리콘 수화젤 렌즈의 제조 및 특성)

  • Silicone hydrogels incorporated with poly(ethylene glycol)(PEG) were prepared and characterized to evaluate the effects of PEG on contact lenses. The silicone hydrogels were copolymerized with methacryloxypropyl tris(trimethylsiloxy) silane (TRIS), methyl methacrylate (MMA), N,N-dimethyl acrylamide (DMA) and PEG-containing monomers such as poly(ethylene glycol) methyl ether methacrylate (PEG- MEM). The silicone hydrogels were characterized using Fourier transform infrared spectroscopy (FT-IR), electron spectroscopy of chemical analysis (ESCA), and scanning electron microscopy (SEM). Water absorbance, water contact angle and light transmittance of the silicone hydrogels were evaluated. The experiments of protein adsorption were also carried out to evaluate the protein adsorption in tears. The peak intensity of C-O bond was increased by the incorporation of PEG-containing monomers and thus PEG incorporation into silicone hydrogels could be confirmed. Phase separation was not shown by the SEM observation of the cross-section of silicone hydrogels. Water absorbancy was increased, while water contact angle and light transmittance were decreased with increasing incorporation of the PEG-containing monomers. The absorption of proteins in tears, albumin, lysozyme and $\gamma$-globulin, on the surface of silicone hydrogels was decreased with increasing incorporation of the PEG-containing monomers.

  • 초소형의 전자 부품, 의료기기 및 광부품 등의 핵심부품들에 관한 산업계와 소비자의 욕구에 발맞추어 과학 기술의 진보가 요구되고 있다. 전통적인 금속 가공 방법과 MEMS와 같은 미세 부품 성형 방법의 중간적인 형태의 Milli-Structure 생산 기술은 근간의 추이에 발맞추어 차세대 국가경쟁력 강화를 위한 핵심기술로 주목받고 있다. 본 논문에서는 미세 부품 성형에 관련하여 국외 개발 동향을 검토하고 국내의 연구 상황을 본 연구팀의 연구를 위주로 하여 미세 소성 가공과 미세 기계 가공으로 분류하여 검토해 보기로 한다.
