In vitro Culture Conditions for the Mouse Preantral Follicles Isolated by Enzyme Treatment

  • In order to investigate the factors affecting the culture of mouse preantral follicles in vitro, we examined the effect of culture media, protein supplements, and culture period on their growth. The oocyte diameter (initial size: $55.6{\pm}2.5{\mu}m$) was progressively increased during culture, and the maximum size ($72.0{\pm}2.4{\mu}m$) was reached on day 10 of the in vitro culture. The chromatin configuration in the germinal vesicle (GV) oocyte progressively shifted from a non-surrounded nucleolus (NSN) to a surrounded nucleolus (SN). On day 10 of the culture, most of the oocytes progressed to the SN pattern. The survival and metaphase II rates of the oocytes in alpha-minimal essential medium (alpha-MEM) were significantly higher (p<0.05) than those in Waymouth and tissue culture medium (TCM)-199. As a protein source, fetal bovine serum (FBS) was more suitable for the culture of mouse preantral follicles as compared to human follicular fluid (hFF) and bovine serum albumin (BSA); the optimal concentration of FBS was 5%. These results suggest that in a culture of mouse preantral follicles, alpha-MEM and 5% FBS are an optimal medium and a protein source, respectively; further, the 10 days of culture is required for the complete growth of oocytes in this culture system.

  • Index is a systematic guide to items contained in, or concepts derived from, a collection, Thus, it is represented as a paired set of index terms (t) and documents (D) : I= {(t,D) vertical bar t .mem. V, D .mem. W), where V is index vocabulary and W is document collection. Indexing is the process of analysing the informational content of records of knowledge and expressing the informational content in the language of the indexing system. It involves: 1) Selecting indexable concepts in a document; and 2) expressing these concepts in the language of the indexing system (as index entries): and an ordered list. Indexing process involves technical, semantic and syntactic problems. Technical problems are related to the accuracy of indexing, which is primarily governed by the indexer's ability of analysing subject, identifying indexable concepts, and coding. The proper levels of indexing exhaustivity, and index language specificity are also significant factors affecting the quality of index. Semantic problems are related to the choice of index terms and the form in which they should be used. Equivalent, hierarchical and affinitive/associative relationships of index terms are involved. Syntactic problems are largely related to the coordination of index terms. This process of coordination arises from the need to be able to search for the intersection of two or more classes defined by terms denoting distinct concepts. Finally, most valuable aspects of alphabetical subject indexing theories and practices are derived from those of Cutter, Kaiser, Ranganathan, Coates, Lynch and Austin, and discussed in details.

  • Vibro-impacts in manual transmissions result due to several nonlinearities such as multi-staged clutch characteristics and gear backlashes. For the sake of understanding the torsional system, one specific manual transmission with front engine and front wheel drive configuration is investigated with a linear model under the several assumptions substituting the nonlinear factors. First, this system is examined with the mathematical approaches by expressing the governing equations to find out the torsional motions. Second, this system is analyzed using the linear model in order to understand its modal and frequency response characteristics using eigensolution method and the FRF(Frequency Responses Function) analysis. Third, with the given results from the eigensolutions, several mode shapes are investigated related to the torsional motion characteristics. Fourth, the system characteristics with the FRFs are studied with the basic approach, with which the several key parameters will be suggested based upon the results in the further studies.

  • Recently, it is gradually raised necessity that interface is measured accurately and managed in industrial circles and medical world, An Ultrasonic wave transmitted from a focused beam transducer is being expected as a powerful tool for NDE of micro-defect. The ultrasonic NDE of the defect is based on the form of the wave reflected form the interface In this study, regarding to the thickness of film which is in opaque object and thickness measurement was done by MEM-cepstrum analysis of received ultrasonic wave. In measument results, film thickness which is beyond distance resolution capacity was measured accurately. Also, automatically repeated discrimination analysis method can be decided in the category of all kinds of defects on semiconductor package.

THE EFFICIENCY OF UTILIZATION OF METABOLIZABLE ENERGY OF MILK-REPLACER-FED CALVES AT WEANING PERIOD

  • A total of 22 energy balance trials were done for calves fed a liquid milk replacer, calf starter and second cut mixed hay during weaning period. Milk replacer supplied 50% of the total dietary energy, calf starter 42% and hay 8% Live weight of calves averaged 64.6 ($S.D.{\pm}7.8$) kg and daily gain 0.54 (${\pm}0.22$) kg. The metabolizability of gross energy averaged 0.751. A regression was calculated relating energy retention (ER, $kJ/kg^{0.75}$) to the intake of metabolizable energy (IME, $kJ/kg^{0.75}$): $$ER=0.69({\pm}0.09)IME-395,\;r=0.888,\;P<0.01,\;S.E.{\pm}7.1$$. Metabolizable energy for maintenance (MEm) was calculated to be $572kJ/kg^{0.75}$ when ER = 0. The amount of IME over MEm for an individual animal (MEg, $kJ/kg^{0.75}$) was regressed on average daily gain (ADG, kg) by the method of regression through the origin: $$MEg=364({\pm}55)ADG,\;r=0.634,\;P<0.01,\;S.E.{\pm}12$$. The amount of ME required for maintenance and growth was estimated to be $936kJ/kg^{0.75}$.

A regression was calculated relating energy retention (ER, $kJ/kg^{0.75}$) to the intake of metabolizable energy (IME, $kJ/kg^{0.75}$): ER = 0.72 (${\pm}0.12$) IME - 330, r = 0.702, P < 0.01, $S.E.{\pm}18.0$. Metabolizable energy for maintenance (MEm) was calculated to be $458kJ/kg^{0.75}$ when ER = 0. The amount of IME over MEm for an individual animal (Meg, $kJ/kg^{0.75}$) was regressed on averaged daily gain (ADG, kg): Meg = 413 (${\pm}91$) ADG + 0.2, r = 0.650, P < 0.01, $S.E.{\pm}21$. The amount of ME requirement for suckling calves was estimated using values obtained above.

  • The aim of this note is to study some properties of Schrodinger operators, the magnetic case, $H_{0}$ (a)=1/2(-i.del.-a)$^{2}$; H(a)= $H_{0}$ (a)+V, where a=( $a_{1}$,.., $a_{n}$ ).mem. $L^{2}$$_{loc}$ and V is a potential energy. Also, we are interested in solutions, .psi., of H(a).psi.=E.psi. in the sense that (.psi., $e^{-tH}$(a).PSI.)= $e^{-tE}$(.psi.,.PSI.) for all .PSI..mem. $C_{0}$ $^{\infty}$( $R^{n}$ ) (see B. Simon [1]). In section 2, under some conditions, we find that a semibounded quadratic form of H9a) exists and that the Schrodinger operator H(a) with Re V.geq.0 is accretive on a form domain Q( $H_{0}$ (a)). But, it is well-known that the Schrodinger operator H=1/2.DELTA.+V with Re V.geq.0 is accretive on $C_{0}$ $^{\infty}$( $R^{n}$ ) in N Okazawa [4]. In section 3, we want to discuss $L^{p}$ estimates of Schrodinger semigroups.ups.

Estimation of Vibration Field of a Cylindrical Structure Derived by Optimal Sensor Placement Methods (센서최적배치 기법에 의한 원통형 구조물의 진동장 예측)

  • This study is concerned with the estimation of vibration-field of a cylindrical structure by modal expansion method(MEM). MEM is a technique that identifies modal participation factors using some of vibration signals and natural modes of the structure: The selection of sensor locations has a big influence on predicted vibration results. Therefore, this paper deals with four optimal sensor placement( OSP) methods, EFI, EFI-DPR, EVP, AutoMAC, for the estimation of vibration field. It also finds optimal sensor locations of the cylindrical structure by each OSP method and then performs MEMs. Predicted vibration results compared with reference ones obtained by forced response analysis. The standard deviations of errors between reference and predicted results were also calculated. It is utilized to select the most suitable OSP method for estimation of vibration field of the cylindrical structure.

  • A major problem of frequency analysis in the field of low-frequencies such as building or construction vibration is the way of signal processing which is appropriate to obtain included frequency content from the finite process to be measured. Therefore, it is the aim of the investigation reported herein to develop the signal processing algorithm which is analyzed without losing the reliability of the measurements in low-frequency domain. To accomplish the research objective, it was analyzed the problems on the way of signal processing in low-frequency domain, and compared the response characteristics of FFT with those of MEM (Maximum Entropy Method) about the low-frequency of vibration. This evaluation of the response characteristics is used in determining appropriate signal processing algorithm into the low-frequency domain.
