Differential Response of Etiolated Pea Seedlings to Inoculation with Rhizobacteria Capable of Utilizing 1-Aminocydopropane-1-Carboxylate or L-Methionine

  • The majority of soil microorganisms can derive ethylene from L-methionine (L-MET), while some rhizobacteria can hydrolyze 1-aminocyclopropane-1-carboxylate (ACC) due to their ACC-deaminase activity. In this study, three strains having either ACC-deaminase activity (Pseudomonas putida biotype A, $A_7$), or the ability to produce ethylene from L-MET (Acinetobacter calcoaceticus, $M_9$) or both (Pseudomonas fluorescens, $AM_3$) were used for inoculation. The highly ethylene specific bioassay of a classical 'triple' response in pea seedlings was used to investigate the effect of the inoculation with the rhizobacteria in the presence of 10 mM ACC or L-MET. The exogenous application of ACC had a concentration-dependent effect on the etiolated pea seedlings in creating the classical 'triple' response. The inoculation with P. putida diluted the effect of ACC, which was most likely due to its ACC-deaminase activity. Similarly, the application of $Co^{2+}$ reduced the ACC-imposed effect on etiolated pea seedlings. In contrast, the inoculation of A. calcoaceticus or P. fluorescens in the presence of L-MET caused a stronger classical 'triple' response in etiolated pea seedlings; most likely by producing ethylene from L-MET. This is the first study, to our knowledge, reporting on the comparative effect of rhizobacteria capable of utilizing ACC vs L-MET on etiolated pea seedlings.

남성근로자의 대사증후군 유병에 영향을 미치는 직업군 및 생활습관 위험인자: 후향적 코호트 조사연구 (Influence of Occupational Type and Lifestyle Risk Factors on Prevalence of Metabolic Syndrome among Male Workers: A Retrospective Cohort Study)

  • Purpose: This study examined the influence of occupational type and lifestyle habits on the prevalence of metabolic syndrome (MetS) among Korean male workers. Methods: Through secondary analysis of their four-year health examination data, 3,892 subjects were divided into four subgroups according to the presence of MetS now and four years ago. Results: Nineteen percent (n=739) suffered from MetS and these 739 subjects were classified into following occupations: 7.1% were office workers, 17.6% were non-office workers, and 42.2% were drivers. Multiple logistic regression analyses showed that when the data adjusted for age, the predicting factors on the prevalence of MetS were heavy drinking (OR 1.34, 95% CI 1.09~1.64) and the occupation of non-office workers (OR 2.99, 95% CI 2.13~4.18) and drivers (OR 7.97, 95% CI 4.89~10.83) among workers without MetS four years ago. Among workers already with a history of MetS, the predicting factors were less exercise (OR 1.55, 95% CI 1.02~2.35) and drivers (OR 2.21, 95% CI 1.03~2.94). Conclusion: Heavy drinking and less exercise and drivers were reported as influencing factors on the prevalence of MetS by this sample. The findings suggest that employers need to provide their employees with screening and management program for those at risk of MetS.

Met inactivation by S-allylcysteine suppresses the migration and invasion of nasopharyngeal cancer cells induced by hepatocyte growth factor

  • Purpose: Past studies have reported that S-allylcysteine (SAC) inhibits the migration and invasion of cancer cells through the restoration of E-cadherin, the reduction of matrix metalloproteinase (MMP) and Slug protein expression, and inhibition of the production of reactive oxygen species (ROS). Furthermore, evidence is emerging that shows that ROS induced by radiation could increase Met activation. Following on these reports of SAC and Met, we investigated whether SAC could suppress Met activation. Materials and Methods: Wound healing, invasion, 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium (MTT), soft agar colony forming, western blotting, and gelatin zymography assays were performed in the human nasopharyngeal cancer cell lines HNE1 and HONE1 treated with SAC (0, 10, 20, or 40 mM) and hepatocyte growth factor (HGF). Results: This study showed that SAC could suppress the migration and invasion of HNE1 and HONE1 cell lines by inhibiting p-Met. An increase of migration and invasion induced by HGF and its decrease in a dose dependent manner by SAC in wound healing and invasion assays was observed. The reduction of p-Met by SAC was positively correlated with p-focal adhesion kinase (p-FAK) and p-extracellular related kinase (p-ERK in both cell lines). SAC reduced Slug, MMP2, and MMP9 involved in migration and invasion with the inhibition of Met-FAK signaling. Conclusion: These results suggest that SAC inhibited not only Met activation but also the downstream FAK, Slug, and MMP expression. Finally, SAC may be a potent anticancer compound for nasopharyngeal cancer treated with radiotherapy.

생산직 근로자의 수면시간과 수면의 질이 대사증후군 및 대사증후군 구성요소 유병률에 미치는 영향 (Effects of Sleep Duration and Quality on Prevalence of Metabolic Syndrome and Metabolic Syndrome Components in Korean Blue-collar Workers)

  • Purpose: The prevalence of metabolic syndrome (MetS), a cluster of metabolic abnormalities has rapidly increased in Korea. Sleep may play a role in determining its prevalence. However, the relationships between MetS and the duration and quality of sleep are not yet clear. This study aimed to examine the associations between the duration and quality of sleep and the prevalence of MetS. Methods: Study participants included 348 Korean blue-collar workers employed by six small-sized companies in Korea. The data were collected using an interviewer-administered questionnaire, and logistic regression analysis was conducted to estimate the effects of the factors related to MetS. Results: The multiple logistic regression analysis revealed that the independent factors that contributed to the prevalence of MetS were being male (adjusted odds ratio [aOR]=4.87, 95% confidence interval [CI]=1.58~15.0) and lower sleep quality (aOR=5.12, 95% CI=1.90~14.30). Sleep duration was related to the prevalence of some MetS components, but it was not associated with MetS prevalence. Conclusion: Sleep quality was negatively associated with MetS prevalence when covariates, such as sleep duration, were controlled. When developing a MetS risk-reduction program, focus should be given to sleep quality as well as sleep duration in an intervention for Korean blue-collar workers.

Daily walnut intake improves metabolic syndrome status and increases circulating adiponectin levels: randomized controlled crossover trial

  • BACKGROUND/OBJECTIVES: Several previous studies have investigated whether regular walnut consumption positively changes heart-health-related parameters. The aim of this study was to investigate the effects of daily walnut intake on metabolic syndrome (MetS) status and other metabolic parameters among subjects with MetS. SUBJECTS/METHODS: This study was a two-arm, randomized, controlled crossover study with 16 weeks of each intervention (45 g of walnuts or iso-caloric white bread) with a 6 week washout period between interventions. Korean adults with MetS (n = 119) were randomly assigned to one of two sequences; 84 subjects completed the trial. At each clinic visit (at 0, 16, 22, and 38 weeks), MetS components, metabolic parameters including lipid profile, hemoglobin A1c (HbA1c), adiponectin, leptin, and apolipoprotein B, as well as anthropometric and bioimpedance data were obtained. RESULTS: Daily walnut consumption for 16 weeks improved MetS status, resulting in 28.6%-52.8% reversion rates for individual MetS components and 51.2% of participants with MetS at baseline reverted to a normal status after the walnut intervention. Significant improvements after walnut intake, compared to control intervention, in high-density lipoprotein cholesterol (HDL-C) (P = 0.028), fasting glucose (P = 0.013), HbA1c (P = 0.021), and adiponectin (P = 0.019) were observed after adjustment for gender, age, body mass index, and sequence using a linear mixed model. CONCLUSION: A dietary supplement of 45 g of walnuts for 16 weeks favorably changed MetS status by increasing the concentration of HDL-C and decreasing fasting glucose level. Furthermore, consuming walnuts on a daily basis changed HbA1c and circulating adiponectin levels among the subjects with MetS. This trial is registered at ClinicalTrials.gov as NCT03267901.

성장기의 쥐와 늙은 쥐 조직의 Glutathione Peroxidase 활성에 대한 Methionine 투여의 효과 (Effect of Methionine Supplementation on Glutathione Peroxidase Activity in Young and Old Murine Tissues)

  • 자라나는 과정의 젊은 흰쥐와 14개월 된 늙은 흰쥐, 생쥐들을 대상으로 Met 투여에 따른 간, 혈장, 신장, 비장 등의 glutathione peroxidase(GSHPx) 효소 변화를 측정 비교하였다. 성장기의 쥐에 있어서 Se을 Selenite 형태로 투여했을 때 0.5%의 식이 Met 공급은 같은 Se을 Se-Met 형태를 공급했을 때 보다 효소 활성이 증가하였다. 그러나, 14개월 된 늙은 쥐에서는 Met 투여에 대한 효소 활성은 큰 변화가 없었다. 일반적으로 식이 Se 증가는 효소 활성과 거의 정비례하나 1ppm Se 수준 이상에서는 완만하게 증가 또는 거의 정체하는 상태를 보이지만, 늙은 쥐에서는 Se의 식이 수준이 2ppm이 되어도 여전히 효소 활성이 증가하는 경향을 보였다. 성장기의 생쥐 실험에서도 2ppm Se 수준에서의0.3%와 0.8%의 식이 Met 투여는 효소 활성에 큰 영향이 없었다. 그러나 실험식이 투여 18일 경엔 4일째 보다 Met 투여군이 4.2배의 증가를 보인 반면 비투여군은 2.5배의 증가에 머물렀다. 18일경 이후 효소 활성이 감소하여 42일재는 반으로 감소함을 보였다. Se의 생체 활성은 특히 식이 Se 수준이 낮을 때 Met에 의해 증가되는 폭이 컸으며 Se 수준이 높을수록 상대적으로 감소함을 보였다. 위의 제반 결과에서 Met의 요구량이 Se-Met에 의해 대체되고 있음을 짐작할 수 있다. 그러므로 식이 Met 수준 상태에 따라 효소의 활성이 달라질 수 있다는 것을 시사한다.

메티오닌과 셀렌이 에탄올 중독된 흰쥐의 뇌지질과산화에 미치는 영향 (Effect of Methionine Levels on Brain Lipid Peroxidation in Ethanol-treated Rats of Selenium Deficiency)

  • 에탄을 투여로 인한 뇌조직의 지질과산화에 미치는 식이성 Met과 Se의 효과를 구명코자 Se을 결핍시킨 횐쥐를 대상으로 Met을 수준(0, 3, 9g/kg diet)별로 급여 하였으며, 실험기간은 5주와 10주로 사육하여 뇌조직의 알코을 대사효소와 유리기 제거 효소계의 활성을 관찰하였다. 알코을 대사효소인 ADH 활성은 5주와 10주군에서 에탄을 투여시 증가되었고 Met정상급여군이 결핍과 과량급여군에 비해 유의적인 증가를 나타내었다. AIDH 활성은 에탄을 투여로 유의하게 감소되었고 특히 Met와 Se 동시 결핍시 감소가 가장 현저하였다. 투여기간이 길수록 활성은 감소하는 경향이었다. MAO활성은 에탄올 투여군이 대조군에 비해 유의적으로 증가하였으며, Met급여에 의해 활성은 억제되는 것으로 나타났다. SOD 활성은 에탄올 투여로 유의적인 감소를 보였으며 5주 및 10주 모두 Met 과량급여군이 결핍과 정상급여군에 비해 유의적으로 증가하였다. GSH-Px 활성은 에탄을 투여로 유의적으로 감소되 었으며, Met과 Se 동시 결핍군에서 가장 감소정도가 큰 것으로 나타났다. Met 급여수준이 증가할수록 그 활성은 유의적으로 증가하였으며, 10주군이 5주군에 비해 활성이 증가하였다. 에탄올 투여로 증가된 GST 활성은 Met결핍에 의해 더욱 가중되었으며 10주군이 5주군에 비해 GST 활성이 유의적으로 증가된 것으로 보아 장기간의 Met 결핍은 에탄을 중독을 심화시킬 것으로 생각된다. Catalase 활성은 에탄을 투여로 유의적인 증가를 나타내었고 Met 결핍군이 정상급여군 및 과량 급여군에 비하여 유의적인 증가를 보였다. GSH함량은 에탄올에 의해 유의적으로 감소되었는데 Met 급여 수준이 증가할수록 에탄올로 인해 감소된 GSH함량이 유의적으로 증가되었으며, 10주군이 5주군에 비해 증가 정도가 현저하였다. 과산화의 지표인 LPO 함량은 에탄을 투여시 증가되었고 Met 결핍군은 정상급여군에 비해 유의적으로 증가되었으며, 특히 Met 과량급여시 5주군에 비해 10주군이 유의적으로 증가하였다. 이 상의 결과에서 Se 결핍시 Met 급여로 인해 에탄올 대사효소의 활성 증가 및 에탄올에 의해 증가된 뇌의 지질과산화를 줄이는 것으로 보아 Met이 생체의 노화를 방지할 수 있을 것으로 생각되며, Se 결핍으로 증가된 지질과산화 반응에 대한 보호와 적응 수단으로써 방어 효소의 활성도가 높아진 것으로 사료된다.

요통환자의 근에너지 기법 시술 전과 후의 척추기립근 변화-경근전도를 통해 (The Clinical Study of Muscle Energy Techniques(MET) in Elector Spinae Muscle on Low Back Pain Patients-Through Meridian Electromyography)

  • Objectives : To evaluate the clinical utility of MET in elector spinae muscle on low back pain patients. Methods : We compared electrical activity between a before MET and a after MET in elector spinae muscle on low back pain patients in same group(n=30) in dynamic flexion-reextension state during five seconds. We analyzed amplitudes and areas of electrical activity and asymmetry index(AI). Results : 1. After MET in elector spinae muscle on low back pain patients were significantly decreased in Visual Analogue Scale(VAS) than before MET in elector spinae muscle on low back pain patients(p<0.05). 2. After MET in elector spinae muscle on low back pain patients were lower electrical activity than before MET in elector spinae muscle on low back pain patients but it is not a pointless observation(p<0.05). 3. AI of the after MET in elector spinae muscle on low back pain patients significantly decreased compared with before MET in elector spinae muscle on low back pain patients(p<0.05). Conclusions : According to above results, there is clinical effect MET on low back pain patients.

Discovery of an Indirubin Derivative as a Novel c-Met Kinase Inhibitor with In Vitro Anti-Tumor Effects

  • The c-Met protein is a receptor tyrosine kinase involved in cell growth, proliferation, survival, and angiogenesis of several human tumors. Overexpression of c-Met has been found in gastric cancers and correlated with a poor prognosis. Indirubin is the active component of Danggui Longhui Wan, which is a traditional Chinese antileukemic recipe. In the present study, we tested the anti-cancer effects of an indirubin derivative, LDD-1937, on human gastric cancer cells SNU-638. When we performed the in vitro kinase assay against the c-Met activity, LDD-1937 inhibited the activity of c-Met. This result was confirmed by immunoblot and immunofluorescence of phosphorylated c-Met. Immunoblot analysis showed that LDD-1937 decreased the expression of the Erk1/2, STAT3, STAT5, and Akt, downstream proteins of c-Met. In addition, LDD-1937 reduced the cell viability and suppressed colony formation and migration of SNU-638 cells. Furthermore, LDD-1937 induced $G_2/M$ phase arrest in the SNU-638 cells by decreasing the expression levels of cyclin B1 and CDC2. Cleaved-PARP, an apoptosis-related protein, was up-regulated in cells treated with LDD-1937. Overall, this study suggests that LDD-1937 may be a novel small-molecule with therapeutic potential for selectively inhibiting c-Met and c-Met downstream pathways in human gastric cancers overexpressing c-Met.

Measurement and Decomposition of Socioeconomic Inequality in Metabolic Syndrome: A Cross-sectional Analysis of the RaNCD Cohort Study in the West of Iran

  • Objectives: Socioeconomic inequality in metabolic syndrome (MetS) remains poorly understood in Iran. The present study examined the extent of the socioeconomic inequalities in MetS and quantified the contribution of its determinants to explain the observed inequality, with a focus on middle-aged adults in Iran. Methods: This cross-sectional study used data from the Ravansar Non-Communicable Disease cohort study. A sample of 9975 middleaged adults aged 35-65 years was analyzed. MetS was assessed based on the International Diabetes Federation definition. Principal component analysis was used to construct socioeconomic status (SES). The Wagstaff normalized concentration index (CIn) was employed to measure the magnitude of socioeconomic inequalities in MetS. Decomposition analysis was performed to identify and calculate the contribution of the MetS inequality determinants. Results: The proportion of MetS in the sample was 41.1%. The CIn of having MetS was 0.043 (95% confidence interval, 0.020 to 0.066), indicating that MetS was more concentrated among individuals with high SES. The main contributors to the observed inequality in MetS were SES (72.0%), residence (rural or urban, 46.9%), and physical activity (31.5%). Conclusions: Our findings indicated a pro-poor inequality in MetS among Iranian middle-aged adults. These results highlight the importance of persuading middle-aged adults to be physically active, particularly those in an urban setting. In addition to targeting physically inactive individuals and those with low levels of education, policy interventions aimed at mitigating socioeconomic inequality in MetS should increase the focus on high-SES individuals and the urban population.
