Clinicopathological Significance and Diagnostic Accuracy of c-MET Expression by Immunohistochemistry in Gastric Cancer: A Meta-Analysis

  • Purpose: The aim of the present study was to elucidate the clinicopathological significance and diagnostic accuracy of immunohistochemistry (IHC) for determining the mesenchymal epidermal transition (c-MET) expression in patients with gastric cancer (GC). Materials and Methods: The present meta-analysis investigated the correlation between c-MET expression as determined by IHC and the clinicopathological parameters in 8,395 GC patients from 37 studies that satisfied the eligibility criteria. In addition, a concordance analysis was performed between c-MET expression as determined by IHC and c-MET amplification, and the diagnostic test accuracy was reviewed. Results: The estimated rate of c-MET overexpression was 0.403 (95% confidence interval [CI], 0.327~0.484) and it was significantly correlated with male patients, poor differentiation, lymph node metastasis, higher TNM stage, and human epidermal growth factor receptor 2 (HER2) positivity in IHC analysis. There was a significant correlation between c-MET expression and worse overall survival rate (hazard ratio, 1.588; 95% CI, 1.266~1.992). The concordance rates between c-MET expression and c-MET amplification were 0.967 (95% CI, 0.916~0.987) and 0.270 (95% CI, 0.173~0.395) for cases with non-overexpressed and overexpressed c-MET, respectively. In the diagnostic test accuracy review, the pooled sensitivity and specificity were 0.56 (95% CI, 0.50~0.63) and 0.79 (95% CI, 0.77~0.81), respectively. Conclusions: The c-MET overexpression as determined by IHC was significantly correlated with aggressive tumor behavior and positive IHC status for HER2 in patients with GC. In addition, the c-MET expression status could be useful in the screening of c-MET amplification in patients with GC.

Association between metabolic syndrome components and cardiac autonomic modulation in southern Indian adults with pre-metabolic syndrome: hyperglycemia is the major contributing factor

  • Metabolic syndrome (MetS) involves multi-factorial conditions linked to an elevated risk of type 2 diabetes mellitus and cardiovascular disease. Pre-metabolic syndrome (pre-MetS) possesses two MetS components but does not meet the MetS diagnostic criteria. Although cardiac autonomic derangements are evident in MetS, there is little information on their status in pre-MetS subjects. In this study, we sought to examine cardiac autonomic functions in pre-MetS and to determine which MetS component is more responsible for impaired cardiac autonomic functions. A total of 182 subjects were recruited and divided into healthy controls (n=89) and pre-MetS subjects (n=93) based on inclusion and exclusion criteria. We performed biochemical profiles on fasting blood samples to detect pre-MetS. Using standardized protocols, we evaluated anthropometric data, body composition, baroreflex sensitivity (BRS), heart rate variability (HRV), and autonomic function tests (AFTs). We further examined these parameters in pre-MetS subjects for each MetS component. Compared to healthy controls, we observed a significant cardiac autonomic dysfunction (CAD) through reduced BRS, lower overall HRV, and altered AFT parameters in pre-MetS subjects, accompanied by markedly varied anthropometric, clinical and biochemical parameters. Furthermore, all examined BRS, HRV, and AFT parameters exhibited an abnormal trend and significant correlation toward hyperglycemia. This study demonstrates CAD in pre-MetS subjects with reduced BRS, lower overall HRV, and altered AFT parameters. Hyperglycemia was considered an independent determinant of alterations in all the examined BRS, HRV, and AFT parameters. Thus, hyperglycemia may contribute to CAD in pre-MetS subjects before progressing to MetS.

  • Purpose: The purpose of this study was to analyze the changes of the relative impulse of the foot when carrying 5 types of schoolbag during level walking. Methods: Forty-four normal subjects were divided into 5 groups according to the method of carrying a bag: carrying no bag (condition 1), a backpack (condition 2), a shoulder bag (condition 3), a cross bag (condition 4) and a one-hand bag (condition 5). To evaluate the partial relative impulse, 7 areas were measured on the sole of a foot: the hallux, toe, 1st metatarsal head (Met1) and the 2nd & 3rd metatarsal heads (Met2/3), the 4th & 5th metatarsal heads (Met4/5), mid foot and heel. Repeated one-way analysis of variance (ANOVA) and independent t-tests were used to analyze the statistical data. Results: In the right foot, the relative impulses in every areas revealed no significant difference (p>0.05), however, the relative impulses at Met1, Met2/3, Met4/5 and heel were significantly different between the different conditions (p<0.05). There were significant differences on comparing the relative impulse between left and right foot within one condition, the mid foot in condition 2, as well as the hallux and Met1 in condition 3 (p<0.05). In condition 4, the toe, Met1, Met4/5 and mid foot were significantly different (p<0.05). The values at the hallux, toe and Met1 in condition 5 also were significantly different (p<0.05). Conclusion: These findings showed that the symmetrical relative impulse of the foot was present in the case of carrying a backpack, but there was an extra load on specific regions and asymmetrical relative impulses in the case of carrying a shoulder bag, a across bag and a one-hand bag.   • This study was performed to investigate the characteristics of nutrient intake according to metabolic syndrome in Korean elderly. Data on 1,200 elderly (men 545, women 655) over 65 years was obtained from the Korea National Health and Nutrition Examination Survey 2010. The subjects were divided into metabolic syndrome (MetS) and normal groups based on NCEP-ATP III guidelines and by applying the Asia-Pacific definition with waist circumference. The prevalence of MetS was 48.9% in all subjects, 35.6% in men and 58.9% in women, respectively. All nutrient intake was significantly lower in MetS than normal subjects, except iron, vitamin A, carotene, and retinol. In the carbohydrate: protein: fat (C:P:F) ratio, MetS showed a higher rate for carbohydrate, but a lower rate for protein and fat than normal. MetS was lower in calcium, vitamin B1, and vitamin C by the nutrient adequacy ratio than normal. Mean adequacy ratio of MetS (0.69) was lower than that of the normal group (0.72). These results demonstrated that undernutrition, not excess intake of specific nutrients - energy, fat, or cholesterol was associated with the risk for MetS in Korean elderly with MetS. Therefore, it is recommended to have a healthy dietary pattern with diverse nutrients for the prevention of MetS.

사상체질에 따른 대사증후군과 수족냉증 분포 차이와 역상관관계 (Inverse Association and Differences in the Distribution of Metabolic Syndrome and Cold Hypersensitivity in the Hands and Feet According to Sasang Constitution)

  • Objectives This study aimed to examine the differences in the distribution of metabolic syndrome(MetS) and cold hypersensitivity in the hands and feet(CHHF) according to Sasang constitution, and to determine whether CHHF and MetS have an inverse association. Methods MetS and its components, CHHF, Sasang constitution data from 1,998 participants in the Korean medicine Daejeon Citizen Cohort study(KDCC) were obtained. The participants were divided into a non-CHHF(n = 1,270, 63.6%), intermediate(n = 220, 11.0%) and CHHF(n = 508, 25.4%) group according to the thermal sensitivity questionnaire. Sasang constitution was diagnosed by Korea Sasang Constitutional Diagnostic Questionnaire(KS-15). One-way ANOVA and the chi-square test were used for participants' general characteristics and thermal sensitivity and MetS related factors. ANCOVA and logistic regression were used to compare the differences and the odds ratios(ORs) for MetS and its components. Results The MetS and CHHF prevalence rates of the Taeeumin, Soeumin, and Soyangin were 27.6%, 3.8%, 7.7%, and 18.3%, 42.3%, 26.4% respectively. The ANCOVA for MetS components showed that the waist circumference was significantly lower in the CHHF group as compared to the non-CHHF group in total and Soyangin. The logistic regression for MetS prevalence showed that CHHF had a significant inverse association in total(OR = 0.611) and Taeeumin(OR = 0.521). Conclusions The MetS prevalence had the highest in Taeeumin, followed by Soyangin and Soeumin, while the prevalence of CHHF was highest in Soeumin, followed by Soyangin and Taeeumin. In addition, it was confirmed that CHHF and MetS had an inverse association independently.

The Wine Yeast Strain-Dependent Expression of Genes Implicated in Sulfide Production in Response to Nitrogen Availability

  • Sulfur metabolism in S. cerevisiae is well established, but the mechanisms underlying the formation of sulfide remain obscure. Here, we investigated by real-time RT-PCR the dependence of expression levels of MET3, MET5/ECM17, MET10, MET16, and MET17 along with SSU1 on nitrogen availability in two wine yeast strains that produce divergent sulfide profiles. MET3 was the most highly expressed of the genes studied in strain PYCC4072, and SSU1 in strain UCD522. The strains behaved differently according to the sampling times, with UCD522 and PYCC4072 showing the highest expression levels at 120 h and 72 h, respectively. In the presence of 267 mg assimilable N/l, the genes were more highly expressed in strain UCD522 than in PYCC4072. MET5/ECM17 and MET17 were only weakly expressed in both strains under any condition tested. MET10 and SSU1 in both strains, but MET16 only in PYCC4072, were consistently upregulated when sulfide production was inhibited. This study illustrates that strain genotype could be important in determining enzyme activities and therefore the rate of sulfide liberation. This linkage, for some yeast strains, of sulfide production to expression levels of genes associated with sulfate assimilation and sulfur amino acid biosynthesis could be relevant for defining new strategies for the genetic improvement of wine yeasts.

The Effects of Supplementing Methionine plus Cystine to a Low-protein Diet on the Growth Performance and Fat Accumulation of Growing Broiler Chicks

  • This experiment was conducted to determine the effects of a low-protein diet supplemented with DL-methionine plus L-cystine (Met + Cys) on the growth performance and fat accumulation of female broiler chicks during the growing period (3-6 wks old). A low-protein diet (17% CP; 3,200 ME kcal/kg) was supplemented with Met + Cys (1.1 : 1.0) at levels 0.75, 0.94, 1.25, 1.31 or 1.50% of diet, respectively. Another diet with 21% CP and 3,200 ME kcal/kg served as the control group. All essential amino acids were adjusted to meet the National Research Council (1984) requirement for chicks. Feed and water were given ad libitum. Body weight of the chicks fed the low-CP diets supplemented with Met + Cys were heavier than those of the control birds. Feed conversion ratio and feed intakes were not significantly different between and among the treatment groups. Similary, abdominal fat content was not significantly different among the various treatments except that of the chicks fed the low CP diet with 1.25% Met + Cys which was higher than that of the control group. Fatty acid synthetase (FAS), acetyl-CoA carboxylase (ACC) activities and carcass protein content were not influenced by dietary treatments. Carcass fat content was lowest in chicks fed low CP diet with 0.75% Met + Cys and highest in the group that received 1.50% Met + Cys supplementation. Liver triglyceride increased as Met + Cys supplementation level increased. Various lipid fraction concentrations (cholesterol ester, free cholesterol, and phospholipid) in the serum went up as Met + Cys increased up to 1.25% after which it levelled off. Results of this experiment suggest that it is possible to reduce dietary protein level from 21% to 17% for growing broiler chicks by the supplementation of Met + Cys when other EAA were sufficient.

Adenovirus-mediated Expression of Both Antisense Ornithine Decarboxylase and S-adenosylmethionine Decarboxylase Induces G1 Arrest in HT-29 Cells

  • To evaluated the effect of recombinant adenovirus Ad-ODC-AdoMetDCas which can simultaneously express both antisense ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (AdoMetDC) on cell cycle distribution in colorectal cancer cell and investigated underlying regulatory responses, human colorectal cancer cells HT-29 were cultured in RPMI 1640 medium and infected with Ad-ODC-AdoMetDCas. Cell cycle progression was detected by flow cytometry analysis. The expression levels of cell cycle regulated proteins were measured by Western blot analysis. The mRNA level of cyclin D1 was measured by RT-PCR. And a luciferase reporter plasmid of cyclin D1 promoter was constructed to observe the effect of Ad-ODC-AdoMetDCas on cyclin D1 promoter activity. The results showed that recombinant adenovirus Ad-ODC-AdoMetDCas significantly induced $G_1$ arrest, decreased levels of cyclin D1 protein and mRNA and suppressed the promoter activity. Ad-ODC-AdoMetDCas also inhibited nuclear translocation of $\beta$-catenin. In conclusion, downregulation of ODC and AdoMetDC mediated by Ad-ODC-AdoMetDCas transfection induces $G_1$ arrest in HT-29 cells and the arrest was associated with suppression of cyclin D1 expression and inhibition of $\beta$-catenin nuclear translocation. As a new anticancer reagent, the recombinant adenovirus Ad-ODC-AdoMetDCas holds promising hope for the therapy of colorectal cancers.

The Association Between Circulating Inflammatory Markers and Metabolic Syndrome in Korean Rural Adults

  • Objectives: This study was performed to investigate the associations between the metabolic syndrome (MetS) and inflammatory markers. Methods: This cross-sectional analysis was performed using data from 1578 Koreans aged 40-69 years residing in a rural area. We investigated associations between MetS and circulating high sensitivity C-reactive protein (hs-CRP), white blood cells (WBC) and adiponectin. MetS was defined using the criteria proposed by the National Cholesterol Education Program Adult Treatment Panel III (ATP-III). Results: Increased WBC counts and hs-CRP levels and decreased adiponectin levels were observed in subjects with MetS. WBC, hs-CRP and adiponectin levels linearly deteriorated with an increase in the number of MetS components (all ptrend <0.005). Finally, adjusted odds ratios (ORs) for the risk of MetS by increase/decrease in 3 inflammatory markers were calculated by multivariate logistic regression analyses. In terms of changes in inflammation markers, in men, the adjusted ORs (95% confidence interval) were 1.15 (1.01-1.31) for WBC, 1.64 (1.02-2.64) for hs-CRP, and 0.19(0.08-0.45) for adiponectin, whereas corresponding adjusted ORs (95% Cls) in women were 1.27 (1.15-1.40), 0.98 (0.67-1.42), 0.09 (0.04-0.18), respectively. Conclusions: Serum adiponectin levels and WBC counts were found to be strongly associated with MetS in both sexes. However, hs-CRP lost its significance after adjusting for BMI and other inflammatory markers in women. This study shows that inflammatory response is associated with MetS in the Korean population. Further prospective studies are necessary to confirm the contribution made by inflammatory markers to the development of MetS.

재래종 옥수수 수집종에 대한 특성조사 제5보 다수다벽 재래종 옥수수계통의 특성변이 (Investigation on Korean Local Maize Lines V. Variabilities of Plant Characters of Multi-eared and Tillered Lines(MET))

  • 재래종의 수집종 가운데서 선접된 다종다벽계통 (약칭 : MET)의 일반적 대성을 분석하여 청예용 옥수수의 육종을 위한 기초자료를 얻고져 수행한 시험결과를 요약하면 다음과 같다. 1. 주요 특성들의 환경변이와 유전변이를 추정하였던 바 MET계통의 분벽성, 개체당 이삭수(수수)는 환경변이보다도 유전변이가 큰 것으로 추정되었다. 개체당 건물중 및 건엽중 등도 유전변이가 커 MET 계통의 유전적 특성으로 확인되었다. 그러나 건기중, 입중 등은 환경변이가 컸었다. 2. 몇가지 특성의 평균치를 두 품종간에 비교하면 다음과 같다. 가. 분벽기수 및 이삭수 : 수원 19호는 환경변이 (파종기, 재식밀도)에 관계없이 분벽기이 없고 이삭수도 변하지 않았으나 MET계통은 분벽기수 및 이삭수가 크게 달라졌다. 나. 개체당 건물중, 건엽중 및 건기중 : 파종기나 재식밀도에 따라 다르기는 하지만 MET계통의 개체당 건물이 수원 19호보다 비교적 높았으며 건엽중은 월등히 높았고(1.5-2.5)배 개체당 건기중도 비교적 높았다. 다. 개체당 입중 및 입중 대 건기엽중비 : MET계통의 개체당 입중은 수원 19호보다 5~40% 낮았다. 따라서 MET계통의 입중 대 건기엽중비가 평균 35%였고 수원 19호는 41%로서 높았다. 3. MET계통은 파종 2주 후부터 분벽하기 시작하여 3 ~ 5일간격으로 다음 분벽이 되었으며 생육초기에 왕성하여 최고 6~7개에 이르나 2~3개만이 유효기으로 신장되였다. 유효기비율은 소식일수록 높았다. 분벽의 발생은 주기의 최하절읍부터 시작되는 데 하부절위가 짧아 한절위에서 분벽이 여러개 발생하는 것처럼 보였다. 제 1, 2분벽기의 기장은 주기의 것에 비하여 10cm 정도씩 짧았으며 재식밀도에 따라 주기 및 제일, 이분벽기의 기장은 소식(60$\times$60cm)한 경우 밀식(60$\times$20cm)한 경우와 같이 분벽들에 의한 밀식의 효과가 있었고 중간재식밀도(60$\times$40cm)인 경우에는 소ㆍ밀도의 중간되는 기장을 보였다. 4. MET계통은 개체당 이삭수가 많은 것에 반하여 이삭의 크기는 수원 19호의 \frac{1}{3}도 못되었다. 각분벽기에 달리는 이삭수는 소식의 경우 주기에 4개, 제일, 이분벽기에 2~3개씩 달렸다. MET계통의 입은 경입종이며 황색입이 주가되고 백색입이 약간 섞여 있었으며 100입중은 14~15gr. 의 소입종이었다. MET계통은 착수고가 수원 19호보다 훨씬 높았다. 5. 주요 특성간의 상간 : 수원 19호에 비하여 MET 계통은 주요 특성간에 높은 상관계수를 보여 유전상관도 높을 것이라는 것을 간접적으로 나타내주었다. 6. MET계통은 생장속도가 수원 19호보다 훨씬 늦었으며 개화기도 2~3주나 늦었다. 7. 흑조위축병 : 수원 19호와 같이 MET계통도 흑조위축병에 대해 이병성이었다. 이병성은 늦게 파종하거나 밀식할수록 높아지는 경향이 있었다.
