USEFULNESS OF CERVICAL VERTEBRAE MATURATION STAGE AS A MANDIBULAR MATURATION INDICATOR (하악골 성장 지표로서 경추골 성숙도의 유용성)

  • This study was to investigate if cervical vertebrae maturation stages are as useful as hand-wrist maturation stages in evaluating the mandibular growth. The subject consisted of 292 girls aged from 8 to 16 years with normal occlusion. They were classified according to diagnosis by using studycast, lateral cephalogram, and handwrist X-ray film. The results were as follow: 1. Cervical vertebrae and hand-wrist maturation stages increased with age. 2. All mandibular measurements (Ar-Go, Go-Me, N-Go, S-Gn, N-Me) increased linearly with cervical vertebrae maturation stages. 3. Ar-Go, Go-Me, N-Go, S-Gn increased linearly with hand-wrist maturation stages. 4. Ar-Go, Go-Me, N-Go, S-Gn increased relatively rapidly between cervical vertebrae maturation stages 3 and 4. Go-Me and S-Gn increased relatively rapidly between hand-wrist maturation stages 6 and 7. 5. Ar-Go, Go-Me, N-Go, S-Gn, N-Me had high correlations with cervical vertebrae maturation stages as well as hand-wrist maturation stages. These results suggest that cervical vertebrae maturation stages are reliable on evaluating the mandibular growth.

Earlier Metabolizable Energy Intake Level Influences Heat Production during a Following 3-Day Fast in Laying Hens

  • The present study was conducted to estimate energy requirements for maintenance in laying hens by using indirect calorimetry and energy balance. A total of 576 28-wk-old Nongda-3 laying hens with dwarf gene were randomly allocated into four ME intake levels (86.57, 124.45, 166.63 and 197.20 kcal/kg body weight $(BW)^{0.75}$ per d) with four replicates each. After a 4 d adaptation period, 36 hens from one replicate were maintained in one of the two respiration chambers to measure the heat production (HP) for 3 d during the feeding period and subsequent 3 d fast. Metabolizable energy (ME) intake was partitioned between heat increment (HI), HP associated with activity, fasting HP (FHP) and retained energy (RE). The equilibrium FHP may provide an estimate of NE requirements for maintenance (NEm). Results showed that HP, HI and RE in the fed state increased with ME intake level (p<0.05). Based on the regression of HP on ME intake, the estimated ME requirements for maintenance (MEm) was 113.09 kcal/kg $BW^{0.75}$ per d when ME intake equals HP. The FHP was decreased day by day with the lowest value on the third day of starvation. Except for lowest ME intake level, the FHP increased with ME intake level on the first day of starvation (p<0.05). The FHP at the two higher ME intake levels were greater than that at the two lower ME intake levels (p<0.05) but no difference was found between the two lower ME intake levels. Linear regression of HP from the fed state to zero ME intake yielded a value of 71.02 kcal/kg $BW^{0.75}$ per d, which is higher than the extrapolated FHP at zero ME intake (60.78, 65.23 and 62.14 kcal/kg $BW^{0.75}$ per d for the first, second and third day of fasting, respectively). Fasting time, lighting schedules, calculation methods and duration of adaptation of hens to changes in ME intake level should be properly established when using indirect calorimetry technique to estimate dietary NE content, MEm and NEm for laying hens.

Effect of Butylated Hydroxytoluene on Experimental Hepatocarcinogenesis Induced by 3'-Methyl-4-Dimethylaminoazobenzene I. Ultrastructural study (실험적인 간암 유발과정에서 Butylated Hydroxytoluene이, 미치는 영향 I : 미세구조적인 연구)

  • A morphologic study on the effect of butylated hydroxytoluene (BHT) on experimental hepatocarcinogenesis induced by 3'-methyl-4-dimethyl aminoazobenzene (3'-MeDAB) was investigated. A total of 110 Sprague-Dowley male rats weighting about 200 g each were used for the experiment, and divided into 4 groups; the 3'-MeDAB, BHT, 3'-MeDAB/BHT treated group, and the control group. Four to eight rats of each group were sacrified on the 4th, 8th, 14th and 16th experimental weeks, with continuous pelletized feeding containing 0.09% 3'-MeDAB and 0.5% BHT. The liver was examined by transmission and scanning electron microscopy. The results were as follows; Electron microscopically, the fine structure of the hepatocytes remained consistently abnormal up to 16 weeks after the 3'-MeDAB treatment. There was no significant difference in the groups observed earlier than in the ones observed later. Many subcellular changes were observed : nuclear change, decreased glycogen, mitochondrial abnormalities, disaggregated rough endoplasmic reticulum, marked proliferation of smooth endoplasmic reticulum, dilatation and distortion of bile canaliculi, increased lysosomes, apoptotic bodies, migration of bile ductule cell. In the BHT treated group, the ultrastructural changes of hepatocytes were not significant, except for the lipid droplets and proliferated smooth endoplasmic reticulum among hepatocytes depending on the experimental duration. The various subcellular changes of 3'-MeDAB/BHT treated groups were simillar to those of the 3'-MeDAB treated group, but the degree of changes in the 3'-MeDAB/ BHT treated group decreased compared with those of the 3'-MeDAB treated group. These results suggest that dietary butylated hydroxytoluene has a protective/inhibitory effect on the hepatocarcinogenesis induced by 3'-methyl-4-dimethyl -aminoazobenzene.

Imyosan induces caspases-mediated apoptosis in human colorectal cancer HCT116 cells (이묘산(二妙散)에 의한 대장암 세포주 HCT116의 Caspases 활성화를 매개로 한 세포사멸)

  • The purpose of this study was to investigate the effect of Imyosan on apoptosis in human colorectal cancer HCT116 cells. Phellodendron amurense Rupr. and Atratylodes lancea D.C. compose Imyosan. First of all, to study the cytotoxic effect of methanol extract of Imyosan (IMS-MeOH) on HCT116 cells, the cells were treated with various concentrations of IMS-MeOH and then cell viability was determined by XTT reduction method. IMS-MeOH reduced viability of HCT116 cells in a dose and time-dependent manner. To confirm the induction of apoptosis, the c1eavage of poly ADP-ribose polymerase (PARP), a substrate for caspase-3 and a typical sign of apoptosis, and the activation of caspase-3, procaspase-8 and procaspase-9 were examined by western blot analysis. IMS-MeOH decreased procaspase-3, procaspase-8 and procaspase-9 levels in a dose-dependent manner and induced the clevage of PARP. IMS-MeOH triggered the mitochondrial apoptotic signaling by increasing the release of cytochrome c from mitochondria to cytosol. Furthermore, IMS-MeOH also downregulated the anti-apoptotic Bcl-2 and upregulated the pro-apoptotic-Bax. Therefore, these results suggest that IMS-MeOH induced HCT1l6 cell death through the mitochondrial pathway. To explore whether the activities of caspases was required for induction of apoptosis by IMS-MeOH, caspase-3, -8, -9 activity measured by using substrates, respectively. IMS-MeOH increased caspase-3, -8, -9 activity. Co-treatment with inhibitors of caspase-3, -8, -9 and IMS-MeOH significantly blocked IMS-MeOH-triggered apoptosis in HCT1l6 cells. These results suggest that IMS-MeOH induces caspases-mediated apoptosis.

Effect of methyl jasmonate on the glucosinolate contents and whole genome expression in Brassica oleracea (유묘기 양배추류에서 메틸자스모네이트에 의한 글루코시놀레이트 함량 변화 및 전사체 발현 분석)

  • In this study, we analyzed the changes in glucosinolate content and gene expression in TO1000DH3 and Early big seedling upon methyl jasmonate (MeJA) treatment. Analysis of glucosinolate contents after MeJA treatment at $200{\mu}M$ concentration showed that the total glucosinolate content increased by 1.3-1.5 fold in TO1000DH3 and 1.3-3.8 fold in Early big compared to those before treatment. Aliphatic glucosinolates, progoitrin and gluconapin, were detected only in TO1000DH3, and the changes in the content of neoglucobrassicin were the greatest at 48 hours after MeJA treatment in TO1000DH3 and Early big. The transcriptomic analysis showed that transcripts involved in stress or defense reactions, or those related to growth were specifically expressed in TO1000DH3, while transcripts related to nucleosides or ATP biosynthesis were specifically expressed in Early big. GO analysis on transcripts with more than two-fold change in expression upon MeJA treatment, corresponding to 12,020 transcripts in TO1000DH3 and 13,510 transcripts in Early big, showed that the expression of transcripts that react to stimulus and chemical increased in TO1000DH3 and Early big, while those related to single-organism and ribosome synthesis decreased. In particular, the expression increased for all transcripts related to indole glucosinolate biosynthesis, which is associated with increase in glucobrassicin and neoglucobrassicin contents. Upon MeJA treatment, the expression of AOP3 (Bo9g006220, Bo9g006240), TGG1 (Bo14804s010) increased only in TO1000DH3, while the expression of Dof1.1 (Bo5g008360), UGT74C1 (Bo4g177540), and GSL-OH (Bo4g173560, Bo4g173550, Bo4g173530) increased specifically in Early big.

The effects of Somok on apoptosis of human liver cancer HepG2 cell. (소목(蘇木)이 사람 간암 세포주인 HepG2의 세포사멸에 미치는 영향과 그 경로)

  • The purpose of this study was to investigate the anticancer effects of Caesalpiniae Lignum (Somok) on HepG2 cells, a human liver cancer cell line. To study the cytotoxic effect of Caesalpiniae Lignum methanol extract (CL-MeOH) on HepG2 cells, the cells were treated with various concentrations of CL-MeOH and then cell viability was determined by XTT reduction method and trypan blue exclusion assay. CL-MeOH reduced proliferation of HepG2 cells in a dose-dependent manner. To confirm the induction of apoptosis, HepG2 cells were treated with various concentrations of CL-MeOH. The activation of caspase 3 and the cleavage of poly ADP-ribose polymerase (PARP), a substrate for caspase-3 and a typical sign of apoptosis, was examined by western blot analysis. CL-MeOH decreased procaspase 3 level in a dose-dependent manner and induced the clevage of PARP at concentration> $200{\mu}/ml$. Mitogen-activated protein (MAP) kinase signaling cascades are multi-functional signaling networks that influence cell growth, differentiation, apoptosis, and cellular responses to stress. CL-MeOH-induced MAPK activation was examined by Western blot for phosphorylated ERK, p38 and JNK. CL-MeOH significantly increased p38 phosphorylation and JNK phosphorylation in a dose-dependent manner. Inhibition of p38 function using the selective inhibitor SB20358O results in inhibition of apoptosis by CL-MeOH. These results suggest that CL-MeOH-induced apoptosis is MAP kinase-dependent apoptoric pathway. These results suggest that CL-MeOH is potentially useful as a chemotherapeutic agent in human liver cancer.

A Study on Work Analysis and Productivity of Food Service in Elementary Schools using Work Sampling (Work Sampling 을 이용한 국민학교급식의 작업분석과 생산성에 관한 연구)

  • This study was carried out to investigate work analysis by use of work sampling and productivity of school feeding at three elementary schools in Seoul. The results were as knows: 1. In the mean work percent of dietitians The direct work functions showed the highest, as the mean work percent was 40.4% for me direct work functions, 29.9% for me indirect work functions and 29.8% for me delays. Among the direct work functions, routine clerical was 30.9%, and receiving was 9.5%. 2. In the mean work percent of bakers The direct work functions (73.5%) marked the highest. Among those, cooking was 41.8%, transportation was 15.9%, and cleaning was 8.5%. 3. In the mean work percent of workers The direct work functions (781%) were me highest. Among those, the decreasing order was cooking (32.3%), cleaning (27.8%), and transportation (13.8%). 4. Work percent per labor hour The work percent per labor hour of dietitians was higher in the morning than in me afternoon, while they continued to work even during the lunch time. Bakers usually worked in me morning, but rarely worked in me afternoon. Workers had lunch and break time after their food service. In me school aided by parents volunteers, the total work time of workers was reduced by an hour and their work percent in me afternoon was high. 5. The number of mea1s served per labor hour was 32.6 meals, and me labor time spent per served mea1 was 1.8 minutes.

Mechanism of Action of Anticancer Drug Aziridinylbenzoquinones: Involvement of DT-diaphorase (DNA에 결합하는 항암제의 작용기전)

  • Aziridinylbenzoquinones such as 3, 6-diaziridinyl-1, 4-benzoquinone (DZQ) and its 2, 5-methyl analog (MeDZQ) require bioreductive activation in order to elicit their anticancer activities. To determine the involvement of DTD in the activation of these drugs, we have used a ligation-mediated polymerase chain reaction to map the intracellular alkylation sites in a sing1e copy gene at the nucleotide level. We have performed this analysis in two human colon carcinoma cells, one proficient (HT-29) and one deficient (BE) in DT-diaphorase (DTD) activity. In the DTD proficient HT-29 cell line, DZQ and MeDZQ were found to alkylate both 5'-(A/T)G(C)-3' and 5'-(A/T)A-3' sequences. This is consistent with the nucleotide preferences observed when DZQ and MeDZQ are activated by purified DTD to reactive metabolites capable of alkylating DNA in vitro [Lee, C. -S., Hartley, J. A., Berardini, M. D., Butler, J., Siegel., D., Ross, D., & Gibson, N. W. (1992) Biochemistry, 31: 3019-3025]. Surprisingly in the DTD-deficient BE cell line a pattern of alkylation induced by DZQ and MeDZQ similar to that observed in the DTD-proficient HT-29 cells was observed. This suggests that reductive enzymes other than DTD can be involved in activating DZQ and MeDZQ to DNA reactive species in vivo.

Effect of Addition of Potato peel , Guar gum , Polydextrose on Quality of Backsulgies (감자 껍질 , Guar gum 및 Polydextrose 첨가에 의한 백설기의 품질특성 변화)

  • The physicochemical, rheological and sensory characteristics of 'BACKSULGIES', which was added with potato peel, guar gum or polydextrose, were investigated. The maximum acceptable addition ratio of dietary fiber to 'BACKSULGI' was 10%. And optimal addition ratio was 3% for all samples. The water binding capacity was affected by dietary fiber sources and incubation conditions (temperature and time). The Guar gum had me highest value of water binding capacity. The solubility was highly related with water binding capacity and me swelling power was increased with temperature increment. The degree of gelatinization was not significantly different with dietary fiber sources. But me values of gelatinization of 'BACKSULGIES' added with dietary fibers were significantly higher than mose of 'BACKSULGI' with no dietary fiber. Generally hardness and brittleness incresed along with storage time. But me hardness of 'BACKSULGIES' added with dietary fibers was significantly lower man those of 'BACKSULGI' with no dietary fiber. The retardation effect of dietary fibers for retrogradation of 'BACKSULGIES' was also proved by time constant determination of Avrami equation. Sernsory evaluation revealed that me addition of dietary fibers did not reduce the organoreptic quality. Therefore potato peel 3%, guar gum 3%, polydextrose 3% were optimum addition ratio which could be accepted as conventional 'BACKSULGI'. As me results of this study, it was proved mat the additions of dietary fibers to 'BACKSULGI' had the retardation effect of retrogradation.

  • Pd(II) p-tolylamide Pd(2,6-$(Ph_2PCH_2)_2C_6H_3$)(NH($C_6H_4Me$-p)) (1) was metathetically prepared by the reaction of Pd(2,6-$(Ph_2PCH_2)_2C_6H_3$)Cl with NaNH($C_6H_4Me$-p). Treatment of 1 with carbon dioxide affords the palladium(II) carbamate Pd(2,6-$(Ph_2PCH_2)_2C_6H_3$)(OC(O)NH($C_6H_4Me$-p)) (2), quantitatively. Complex 2 reacts with HX (X = Cl, OTf) to give Pd(2,6-$(Ph_2PCH_2)_2C_6H_3$)X, $NH_2$(p-Tol) and $CO_2$. Reaction of the palladium(II) carbamate with MeI produced Pd(2,6-$(Ph_2PCH_2)_2C_6H_3$)I along with generation of methyl N-tolylcarbamate MeOC(O)NH($C_6H_4Me$-p), exclusively.
