Expression, Purification and NMR Studies on MC4R-TM2 Mutant

  • Melanocortin-4 receptor (MC4R) subtype is associated with obese humans. Especially, in a patient with severe early-onset obesity, novel heterozygous mutation in the MC4R gene was detected, resulting in an exchange of aspartic acid to asparagine in $90^{th}$ amino acid residue located in the predicted second trans-membrane domain (TM2). Mutations in the melanocortin-4 receptor (MC4R) gene are the most frequent monogenic causes of severe obesity which have been described as heterozygous with loss of function. In order to compare structure difference between MC4R wild type (MC4R-TM2-wt) and mutant (MC4R-TM2-D90N), we designed both MC4R-TM2-wt and MC4R-TM2-D90N construct in pET 21b vector. In this study, we optimized high-yield purification procedure for recombinant TM2-D90N. Eluted recombinant protein was resolubilized under urea condition for thrombin cleavage reaction and we conducted the high-performance liquid chromatography (HPLC) with reverse phase column under 1% acetonitrile, 0.01% TFA buffer solution. The molecular size of purified target peptide was confirmed by Tricine-SDS page analysis. To characterize MC4R-TM2-D90N, we have performed $^{15}N$-isotope labeling of peptide using M9 media and purified labeled target peptide for hetero-nuclear NMR spectroscopy.

Comparsion of Dst forecast models during intense geomagnetic storms (Dst $\leq$ -100 nT)

  • We have investigated 63 intense geomagnetic storms (Dst $\leq$ -100 nT) that occurred from 1998 to 2006. Using these events, we compared Dst forecast models: Burton et al. (1975), Fenrich and Luhmann (1998), O'Brien and McPherron (2000a), Wang et al. (2003), and Temerin and Li (2002, 2006) models. For comparison, we examined a linear correlation coefficient, RMS error, the difference of Dst minimum value (${\Delta}$peak), and the difference of Dst minimum time (${\Delta}$peak_time) between the observed and the predicted during geomagnetic storm period. As a result, we found that Temerin and Li model is mostly much better than other models. The model produces a linear correlation coefficient of 0.94, a RMS (Root Mean Square) error of 14.89 nT, a MAD (Mean Absolute Deviation) of ${\Delta}$peak of 12.54 nT, and a MAD of ${\Delta}$peak_time of 1.44 hour. Also, we classified storm events as five groups according to their interplanetary origin structures: 17 sMC events (IP shock and MC), 18 SH events (sheath field), 10 SH+MC events (Sheath field and MC), 8 CIR events, and 10 nonMC events (non-MC type ICME). We found that Temerin and Li model is also best for all structures. The RMS error and MAD of ${\Delta}$peak of their model depend on their associated interplanetary structures like; 19.1 nT and 16.7 nT for sMC, 12.5 nT and 7.8 nT for SH, 17.6 nT and 15.8 nT for SH+MC, 11.8 nT and 8.6 nT for CIR, and 11.9 nT and 10.5 nT for nonMC. One interesting thing is that MC-associated storms produce larger errors than the other-associated ones. Especially, the values of RMS error and MAD of ${\Delta}$peak of SH structure of Temerin and Li model are very lower than those of other models.

Performance analysis of an MC-CDMA system by using an adaptive beamforming technique (적응 빔 형성 기법을 사용한 MC-CDMA 시스템의 성능분석)

  • This paper presents an adaptive beamforming algorithm for an MC-CDMA system with an adaptive array antenna. By employing an antenna array at the receiver of an MC-CDMA system, the performance of an MC-CDMA system, which is known to be effective for high data rate transmission due to its robustness to multipath fading and its simplicity for using a simple one-tap equalizer, is shown to be significantly improved. The proposed algorithm for adaptive beanforming in an MC-CDMA system is derived by (1) calculating the error signals between the pilot symbols of desired user and the received pilot signals in frequency domain, (2) transforming the frequency-domain error signals into time-domain error signals, (3) updating the filter coefficients of the adaptive beamformer in the direction of minimizing the MSE. Convergence behavior and performance improvement of the proposed approach are demonstrated through computer simulation by applying it to the conventional MC-CDMA system.

  • In this paper, it was estimated the BER performance of the individual system type by applying the SFH type and the FFH type to the FH MC-DS CDMA system that combined the strong points of MC DS/CDMA and that of Frequency Hopping under the Nakagami fading channel with Multi-tone Jamming. An increase in the number of hoppings means that it can improve the BER performance by reducing the receiving burden of the FH MC DS/CDMA by achieving a frequency diversity effect. Therefore, I verified that the performances of the FH MC DS/CDMA system could be improved by increasing the number of hoppings.

  • We propose 2-channel filter bank method instead of FFT method to decrease sub-channel interference for using of efficient frequency resource in MC-CDMA method. Since a prototype filter of filter bank having wavelet characteristic is designed having more less side-lobe, the nearest co-channel interference and inter symbol interference are decreased efficiently. Since the spreading signal of suggesting MC-CDMA system is being demanded for less chip rate and is not being considering for autocorrelation characteristic, the Walsh code can be used as a optimal orthogonal signal set. We consider bit error rate and signal to noise ratio to estimate the performance of suggested system on condition that white noise channel and arbitrary sinusoidal jammer are existing. As a result of comparing to traditional FFT-based MC-CDMA simulation result, our suggested system has shown better performance than traditional MC-CDMA method on the side of minimizing interference effect.

Influence of Methylcellulose on Properties of Wheat Gliadin Film Cast from Aqueous Ethanol

  • Present work was focused on the influence of methylcellulose (MC) on steady rheology of wheat gliadin solution and the properties of glycerol plasticized gliadin films. The presence of MC below 0.99 wt% improved viscosity and flow activation energy of the 10 wt% gliadin solution significantly. In the casting films containing 0.2 g glycerol/g dry protein, the MC component aggregated in the gliadin matrix. The blend films containing less than 7.7 wt% MC exhibited higher Young's modulus (E) and tensile strength (${\sigma}_b$) and lower elongation at break (${\epsilon}_b$) in comparison with the pure gliadin film, which was related to the intermolecular interaction between MC and gliadins, the brittle fracture of the aggregated MC component, and the increase in glass transition temperature ($T_g$) of the gliadin phase. Increasing MC content led to a slight increase in water vapor permeability (WVP) without significant influence on the moisture absorption (MA).

  • A modular multilevel-clamped composited multilevel converter ($M-MC^2$) is proposed. $M-MC^2$ enables topology reconfiguration, power device reuse, and composited clamping. An advanced five-level converter ($5L-M-MC^2$) is derived from the concept of $M-MC^2$. $5L-M-MC^2$ integrates dual three-level T-type modules and one three-level neutral point clamped module. This converter can also integrate dual three-level T-type modules and one passive diode module by utilizing the device reuse scheme. The operation principle and SPWM modulation are discussed to highlight converter performance. The proposed $M-MC^2$ is comprehensively compared with state-of-the-art five-level converters. Finally, simulations and experimental results are presented to validate the effectiveness of the main contributions of this study.

  • MC-DS-CDMA is technique where a single data symbol is transmitted at multiple subcarriers which are orthogonal to each other. With this technique, frequency diversity can be achieved. The performance of PSAM MC-DS-CDMA system with the application of STBC antenna diversity is analyzed. The BER performance of this system over multipath fading environment is evaluated, considering the channel estimation ernr, carrier frequency offset, and the nonlinear high power amplifier.

  • In MC-CDMA systems effects of delay spread of the channel are reduced with increased symbol duration by simultaneously transmitting data symbols on the parallel subcarriers. However, the increased symbol duration causes the system to be more vulnerable to time selective fading. In this paper, we investigate the effects of time selective fading characteristics of the mobile channel from the viewpoint of desired signal power to inter-carrier interference power ratio at the combiner output of the MC-CDMA receiver.

  • In this paper, we propose an adaptive Beamforming Algorithm, called MC-LMS, for adaptive antenna applied to an MC-CDMA system. New method for updating the weight vector based on the MSE criterion is derived. Computer simulations show that MC-LMS algorithm is capable of rejecting co-channel interference that affects the MC-CDMA system. Thus, the BER performance of the MC-CDMA system is much higher than that of the MC-CDMA system without using adaptive antenna and that of the DS-CDMA system with adaptive antenna in multi-path Rayleigh fading channel.
