Role of MAPK Signaling Pathways in Regulating the Hydrophobin Cryparin in the Chestnut Blight Fungus Cryphonectria parasitica

  • We assessed the regulation of cryparin, a class II hydrophobin, using three representative mitogen-activated protein kinase (MAPK) pathways in Cryphonectria parasitica. Mutation of the CpSlt2 gene, an ortholog of yeast SLT2 in the cell wall integrity (CWI) pathway, resulted in a dramatic decrease in cryparin production. Similarly, a mutant of the CpBck1 gene, a MAP kinase kinase kinase gene in the CWI pathway, showed decreased cryparin production. Additionally, mutation of the cpmk1 gene, an ortholog of yeast HOG1, showed decreased cryparin production. However, mutation of the cpmk2 gene, an ortholog of yeast Kss1/Fus3, showed increased cryparin production. The easy-wet phenotype and accumulation of the cryparin transcript in corresponding mutants were consistent with the cryparin production results. In silico analysis of the promoter region of the cryparin gene revealed the presence of binding motifs related to downstream transcription factors of CWI, HOG1, and pheromone responsive pathways including MADS-box- and Ste12-binding domains. Real-time reverse transcriptase PCR analyses indicated that both CpRlm1, an ortholog of yeast RLM1 in the CWI pathway, and cpst12, an ortholog of yeast STE12 in the mating pathway, showed significantly reduced transcription levels in the mutant strains showing lower cryparin production in C. prasitica. However, the transcription of CpMcm1, an ortholog of yeast MCM1, did not correlate with that of the mutant strains showing downregulation of cryparin. These results indicate that three representative MAPK pathways played a role in regulating cryparin production. However, regulation varied depending on the MAPK pathways: the CWI and HOG1 pathways were stimulatory, whereas the pheromone-responsive MAPK was repressive.

  • Components of genetic variation were estimated for five metric traits using 24 biparental progenies (N. C. Design III) generated from F$_2$ generation of a commercial bivoltine silkworm hybrid, SH$_{6}$${\times}$NB$_4$D$_2$. Variance due to additive ($\sigma$$^2$A) and dominance ($\sigma$$^2$D) gene effects was significant for single cocoon weight and shell weight. However, magnitude of former was greater than latter indicating preponderance of additive gene action in the inheritance of these two traits. Average degree of dominance was in the range of partial dominance for all the traits. High estimates of heritability (ns) indicated operation of genes with large additive effects, hence, scope exists for improvement of present populations through a few cycles of selection.n.

  • A defensin is an inducible antibacterial peptide from a fleshfly and contains 40 residues basic peptide with six cysteines. For the consiruction of recombinant S cerevisiae expressing defensin, the structural gene coding for active defensin was chemically synthesized and fused in fiam to GAP promoter, MFul preprosequence and the GAL7 transcription terminator, generating a recombinant plasnlid pGMD18. S. ce~evisine 2805 Gells were transror~ned to uracil prototroph by the pGMDl8 arid the transformed cells showing antibacterial activity against 111. luteus TAM1056 were selected by growth inhibition zone assay. The optimal culture conditions for the unprovement of the defensin production of a selected tmdonnant were investigated. The optirmzed medium containing 0.4% yeast extract, 2% corn steep liquor, 2.5% glucose and 0.05% $C_2CO_3$, could be determined and the optimum lemperature. and initial pH could be detennnied as $28^{\circ}C$ and pH 3, ~mpectively. The optimized conditioiis revealed the trvofold Increase in the cell growth and the fourfold in the antibaclerial activity. coinpar-ed with tllc Yl'D medium.

  • The WILDHABER-NOVIKOV gear, one of the circular arc gears, has the large contact area between the convex and concave profiled mating teeth, moves from one end of the tooth to the other axially making a face contact. Hence it provides a large load capacity than the Involute gear and still satisfying the law of gearing. In order to analyze the gear stress, a photoelastic investigation was carried out. Photo elastic model of the WILDHABER-NOVIKOV gears were made of Araldite CT200 in this investigation. For both the many teeth gear and the few teeth gear segments, External gears of all addendum type WILDHABER-NOVIKOV gear and the involute gear were tested. Included were the models with various profile raddi at the same pressure angle 20 .deg. and module 13.5. The flank stresses and fillet stresses of these gears were observed in each case and compared with those of gears. From this investigation, the following results were obtained. A. In the case of having many teeth gear: As the profile radius is increased, the fillet stresses of the WILDHABER-NOVIKOV gear become the same or less than that of the INVOLUTE gea, and the flank stress becomes smaller than that of the INVOLUTE gear. Therefore the better design condition is satisfied with a large profile radius. B. IN the case of having a few teeth gear: As the profile radius is increased the flank stress of WILDHABER-NOVIKOV gear becomes smaller than that of the INVOLUTE gear, but the fillet stresses become larger than that of the INVOLUTE gear. Therefore the larger design condition is satisfied with small profile radius.

  • This study was performed to investigate effects of dietary folic acid supplementation on plasma homocysteine levels, thiobarbituric acid reactive substance s (TBARS) level s and liver SAM/SAH ratio in hyperhomocysteinaemia-induced pregnant rats. Forty-two female Sprague-Dawley rats were divided three groups (C: control diet, HFD: $0.3\%$ homocystine and 0 mg folic acid diet, HFS: $0.3\%$ homocystine and 8 mg/kg folic acid diet) according to homocystine and folic acid levels in the diet. They were fed experimental diets for 5 weeks prior to the mating and also during the entire period of pregnancy till gestational day 20. Dietary folic acid supplementation caused a significant decrease in plasma homocysteine levels which had been increased by a homocystine-diet, with a concomitant increase in plasma and liver folate levels. Liver TBARS levels in homocysteine-folic acid-deficient group (HFD) were higher than those in control group. Dietary folic acid supplementation increased hepatic SAM/SAM ratio in homocysteine-folic acid- sopplemetantion group (HFS) when compared to the HFD (p < 0.05). These data suggest that folate depletion and elevated plasma homocysteine may promote oxidative stress in rat livers and influence the remethylation cycle of the homocysteine metabolism detrimentally. In conclusion, dietary folic acid supplementation was found to be effective for lowering plasma homocysteine levels, relieving oxidative stress, and improving the methylation status in the body.

Effects of parental folate deficiency on the folate content, global DNA methylation, and expressions of FR${\alpha}$, IGF-2 and IGF-1R in the postnatal rat liver

  • We examined the effect of parental folate deficiency on the folate content, global DNA methylation, folate receptor-alpha (FR${\alpha}$), insulin-like-growth factor-2 (IGF-2) and -1 receptor (IGF-1R) in the liver and plasma homocysteine in the postnatal rat. Male and female rats were randomly fed a folic acid-deficient (paternal folate-deficient, PD and maternal folate-deficient, MD), or folic acid-supplemented diet (paternal folate-supplemented, PS and maternal-folate-supplemented, MS) for four weeks. They were mated and grouped accordingly: $PS{\times}MS$, $PS{\times}MD$, $PD{\times}MS$, and $PD{\times}MD$. Pups were killed on day 21 of lactation. The hepatic folate content was markedly reduced in the $PD{\times}MD$ and $PS{\times}MD$ and $PD{\times}MS$ as compared with the $PS{\times}MS$ group. The hepatic global DNA methylation was decreased in the $PD{\times}MS$ and $PS{\times}MD$ groups as much as in the $PD{\times}MD$ group, and all the three groups were significantly lower as compared to the $PS{\times}MS$ group. There were no significant differences in the hepatic FR${\alpha}$, IGF-2 and IGF-1R expressions among the groups. Positive correlations were found between the hepatic folate content and global DNA methylation and protein expressions of FR${\alpha}$, IGF-2 and IGF-1R, whereas an inverse correlation was found between hepatic folate content and plasma homocysteine level in the 3-week-old rat pup. The results of this study show that both paternal and maternal folate deficiency at mating can influence the folate content and global DNA methylation in the postnatal rat liver.

Adverse Effect of Nonylphenol on the Reproductive System in F1 Male Mice: A Subchronic Low-Dose Exposure Model

  • Nonylphenols (NPs) are widely used industrial materials, and are considered as potent endocrine disrupting chemical. Present study was undertaken to clarify the effect of subchronic low-dose NP exposure to F1 generation male mice. Mice were divided into 2 groups; (1) CON, control animals and (2) NP-50 ($50{\mu}g/L$), animals were treated with NP via drinking water. NP exposures were continuously conducted from parental pre-mating period until the postnatal day (PND) 55 of F1 offsprings. Mice were sacrificed on PND 55 and the tissue weights were measured. The initial body weights (at PND 21) and terminal body weights (PND 55) of the NP-50 animals were significantly lower than those of control animals (p<0.05). NP exposure induced a significant increase in the absolute weight of the testes (p<0.05). Conversely, the NP exposure caused significant decrease in the absolute weights of the epididymis (p<0.01), prostate (p<0.05) and seminal vesicle (p<0.05). Histopathological studies revealed that NP-treated animals exerted decreased seminiferous tubule diameters, reduced luminal area, and lower number of germ cells. Also some sloughing morphologies in the tubules were observed. In the caudal epididymis, fewer mature sperms and swollen epithelial cells were found in the NP-treated group. Our results confirmed that the subchronic low-dose NP exposure altered some male parameters and induced histopathological abnormalities in testis and epididymis of F1 mice. Since the NP dose used in this study is close to the average human daily NP exposure, our results could provide practically meaningful understanding of adverse effect of EDC in human.

Cultural characteristics of a new spore-less cultivar 'chungheung 1ho' for Lentinula edodes sawdust cultivation (톱밥 재배용 무포자 신품종 '청흥1호'의 생육특성)

  • The competitiveness of the mushroom industry can be increased by diversifying the domestic Lentinula edodes cultivar. Therefore, 'Chungheung 1ho' was cultivated by hybrid breeding method using strain 'E140025,' which was collected by the Chungheung Mushroom Farming Association. Regarding the morphological characteristics of the fruit, the cap of 'Chungheung 1ho' was smaller than that of the control cultivar, whereas, its stem was longer and thicker than that of the control cultivar. In addition, the weight of the individual fruits of the new cultivar was heavier than that of the control cultivar. The period for primordia formation was 24 days for the new cultivar, which was 16-20 days later than that of the control cultivar. Furthermore, it was found that the patterns of the PCR amplification band using six URP primers were different for 'Chungheung 1ho' than those of the mating strains.

  • Considerable progress in reproduction of dairy goats has been made, with advances in reproductive technology accelerating dairy goat production since the 1980s. Reproduction in goats is described as seasonal. The onset and length of the breeding season is dependent on various factors such as breed, climate, physiological stage, male effect, breeding system, and photoperiod. The reproductive physiology of goats was investigated extensively, including hypothalamic and pituitary control of the ovary related to estrus behavior and cyclicity etc. Photoperiodic treatments coupled with the male effect allow hormone-free synchronization of ovulation, but the kidding rate is still less than for hormonal treatments. Different protocols have been developed to meet the needs and expectations of producers; dairy industries are subject to growing demands for year round production. Hormonal treatments for synchronization of estrus and ovulation in combination with artificial insemination (AI) or natural mating facilitate out-of-season breeding and the grouping of the kidding period. The AI with fresh or frozen semen has been increasingly adopted in the intensive production system, this is perhaps the most powerful tool that reproductive physiologists and geneticists have provided the dairy goat industry with for improving reproductive efficiency, genetic progress and genetic materials transportation. One of the most exciting developments in the reproduction of dairy animals is embryo transfer (ET), the so-called second generation reproductive biotechnology following AI. Multiple ovulation and ET (MOET) program in dairy goats combining with estrus synchronization (ES) and AI significantly increase annual genetic improvement by decreasing the generation interval. Based on the advances in reproduction technologies that have been utilized through experiments and investigation, this review will focus on the application of these technologies and how they can be used to promote the dairy goat research and industry development in the future.

Effects of Larval Grafted Age for Artificial Queen-rearing on Queen Reproductive Potential and Growth of Apis cerana Colony

  • Queen honey bees (Apis cerana) can be reared artificially on demand by the use of grafting technique. The technique consists of grafting young worker larvae into queen cell cups and raising in a queenless strong colony. As the age of grafted larvae for queen rearing exhibited several aspects related to quality and reproductive potential of queen, this study were conducted to investigate the influence of age of grafted larva on morphological characteristics and lifespan of queen, and the growth of colony she headed. Our results demonstrated that queens reared from young worker larvae (i.e., less than 1-day old larvae) were significantly larger in size (i.e., body weight and thorax width) than that of queens reared from 2-day old worker larvae. Moreover, queens reared from younger worker larvae initiated egg-laying earlier, stored more spermatozoa in spermatheca and had a longer lifespan compared to queens raised from older worker larvae. We also found a significant positive effect of queen grafting age on the production of worker and drone brood, adult worker population in colonies headed by queens reared from younger larvae. These findings suggested that rearing queens from brood grafted at the earliest possible age could increase the reproductive potential of queen as well as fitness of colony she head.
