(γ-Aminobutyric Acid Transporter 2 Binds to the PDZ Domain of Mammalian Lin-7 ((γ-Aminobutyric acid transporter 2와 mammalian Lin-7의 PDZ결합)

  • Neurotransmitter transporters, which remove neurotransmittesr from the synaptic cleft, are regulated by second messenger such as protein kinases and binding proteins. Neuronal ${\gamma}-aminobutyric$ acid transporters (GATs) are responsible for removing the inhibitory neurotransmitter ${\gamma}-aminobutyric$ acid (GABA) from the synaptic cleft. ${\gamma}-aminobutyric$ acid transporters 2 (GAT2/BGT1) is involved in regulating neurotransmitter recycling, but the mechanism how they are stabilized and regulated by the specific binding protein has not yet been elucidated. Here, we used the yeast two-hybrid system to identify the specific binding protein(s) that interacts with the C-terminal region of GAT2 and found a specific interaction with the mammalian LIN-7b (MALS-2). MALS-2 protein bound to the tail region of GAT2 but not to other GAT members in the yeast two-hybrid assay. The "T-X-L" motif at the C-terminal end of GAT2 is essential for interaction with MALS-2. In addition, this protein showed specific interactions in the glutathione S-transferase (GST) pull-down assay. An antibody to GAT2 specifically co-immunoprecipitated MALS associated with GAT2 from mouse brain extracts. These results suggest that MALS may stabilize GAT2 in brain.

  • Sodium hyaluronate (NaHA), water soluble polymer having ultra-high molecular weight, is characterized by using on-line frit inlet asymmetrical flow field-flow fractionation (FI-AFlFFF) and multiangle light scattering (MALS). This study demonstrates the capability of power programming FI-AFlFFF for the separation of NaHA and the applicability of FI-AFlFFF with MALS for the characterization of molecular weight distribution and their structural information. Since sample injection and relaxation in FI-AFlFFF are achieved by using hydrodynamic relaxation, separation of high molecular weight polymers can be achieved smoothly without halting the separation flow. Experiments are carried out with the two different NaHA products (a raw NaHA sample and a thermally degraded NaHA product) and molecular weight distribution and conformations in solution are determined. Influence of sample filtration on the change of molecular weight distribution is also discussed.

  • This project aims to develop a partnership between France and Korea; the field of paper conservation with two mains objectives. The first object is to study the possibilities of implementing the use of traditional Korean paper in conservation workshops in France and Europe. The second one is to establish a long term collaborative research program to investigate the degradation mechanisms of ancient papers and particularly early waxed manuscripts founded in Korea and France which are now brittle and fallen into parts. This problem deserves an investigation using analytical tools such as SEC/MALS, GC/MS to understand the interactions between wax and cellulose during the ageing process and to recommend proper conservation treatment. A research plan could be establish for both institution in order to facilities an exchange of visiting scholars (scientist, post-doc) to implement and share knowledge and for carrying out the research.

  • A large number of accidents at an underground place have been happening, including the gas explosion under construction of subway, the fires of underground utility and underground shopping malls, and other explosion, at home and abroad recently. These accidents make the function of a city ineffective due to the paralyses of electricity and communications net as well as the loss of property, and cause people to feel unsecured with accompaniment of a heavy of toll of lives. This study suggests safety assessment items and fire protection assessment technique for underground shopping malls by extracting dangerous elements in the management stage through examination of related accidents, documents and present conditions.

Genotypes of Hepatitis C Virus in Relapsed and Non-respondent Patients and their Response to Anti-Viral Therapy in District Mardan, Khyber Pakhtunkhawa, Pakistan

  • Hepatitis C is a blood-borne infectious disease of liver, caused by a small enveloped, positive-single stranded RNA virus, called the hepatitis C virus (HCV). HCV belongs to the Flaviviridae family and has 6 genotypes and more than 100 subtypes. It is estimated that 185 million people are infected with HCV worldwide and 5% of these are in Pakistan. The study was designed to evaluate different genotypes of HCV circulating in District Mardan and to know about the behavior of these genotypes to different anti-viral regimes. In this study 3,800 patients were exposed to interferon alfa-2a plus Ribavirin treatment for 6-months and subjected to real-time PCR to check the viral response. Among these 3,677 (97%) patients showed no detectable HCV RNA while 123 (3%) patients (non-responders) remained positive for HCV RNA. Genotypes of their analyzed showed that most of them belonged to the 3a genotype. Non-responders (123) and relapsed (5) patients were subjected to PEG-interferon and Ribavirin therapy for next 6 months, which resulted into elimination of HCV RNA from 110 patients. The genotypes of the persisting resistant samples to anti-viral treatment were 3b, 2a, 1a and 1b. Furthermore, viral RNA from 6 patients remained un-typed while 4 patients showed mixed infections. HCV was found more resistant to antiviral therapy in females as compared to mals. The age group 36-45 in both females and males was found most affected by infection. In general 3a is the most prevalent genotype circulating in district Mardan and the best anti-viral therapy is PEG-interferon plus Ribavirin but it is common practice that due to the high cost patients receive interferon alfa-2a plus Ribavirin with consequent resistance in 3% patients given this treatment regime.

Fluorescence-labelling for analysis of protein in starch using asymmetrical flow field-flow fractionation (AF4)

  • Starch is a mixture of amylose (AMY) and amylopectin (AMP) which are different in physical properties such as molar mass (M), rms radius ($R_g$) and hydrodynamic diameter ($d_H$). The rheological and functional properties of starch are influenced by various factors including the molecular size, molar mass distribution (MD) and the concentration ratio of AMY and AMP. It is also important to analyze proteinaceous material in starch as they affect the flavor and texture of food to which starch is added. In this study, asymmetrical flow field-flow fractionation (AF4) was employed for separation and quantitation of AMY and AMP in starches (Amaranth, potato, taros and quinoa). AF4 was coupled with a multi-angle light scattering (MALS) and a refractive index (RI) detector for determination of the absolute M, MD and molecular structure. It was found that AMP has the M and $R_g$ ranging $3.7{\times}10^7{\sim}6.5{\times}10^8g/mol$ and 84 ~ 250 nm, respectively. Also the existence of branch was confirmed in higher M. In addition, proteinaceous material in starch was analyzed by AF4 coupled with a fluorescence detector (FS) after fluorescence-labeling. AF4-FS with fluorescence-labelling showed a potential for investigation on existence of proteinaceous material and the interaction between proteinaceous material and polysaccharide in starch.

Depolymerization of Alginates by Hydrogen Peroxide/Ultrasonic Irradiation (과산화수소/초음파를 이용한 알지네이트의 저분자화)

  • A high molecular weight natural sodium alginate (HMWSAs) was depolymerized by hydrogen peroxide ($H_2O_2$) with ultrasonic irradiation. The effects of the reaction conditions such as reaction temperature, reaction time, hydrogen peroxide concentration and ultrasonic irradiation time on the molecular weights and the end groups of the depolymerized alginates were investigated. It was revealed that depolymerization occurred through the breakage of 1,4-glycosidic bonds of sodium alginate and the formation of formate groups on the main chain under certain conditions. The changes in molecular weight were monitored by GPC-MALS. The molecular weight of 2 wt% alginate solution decreased from 450 to 15.9 kDa for 0.5 hrs at 50 $^{\circ}C$ under an appropriate ultrasonic irradiation. The PDI(polydispersity index)s of the alginate depolymerized in this study were considerably narrow in comparison with those obtained from the other chemical degradation method. The PDIs were in the range of 1.5~2.5 in any reaction conditions employed in this study.

The Effects of Kimchi Intake on Lipid Contents of Body and Mitogen Response of Spleen Lymphocytes in Rats (김치의 급여가 흰쥐의 체내 지질함량과 비장 면역세포 증식능력에 미치는 영향)

  • Effects of kimchi on lipid metabolism and immune function were studied in experiments using 63mals SD rats fed 6 inds of Baechu-kimchi containing diet during 4 weeks. Three kinds of freeze dried kimchi differ in fermentation period (not fermented, 3-, 6-week-fermented at 4$^{\circ}C$) were added at 5%, 10% of the diet containing 15% lard. The levels of serum total lipid and triglyceride and the content of liver total lipid and triglyceride of all kimchi groups were lower than those of a control group. But the levels of serum and liver cholesterol is not affected by kimchi intake. The triglyceride concentration of epididymal fat pad and feces of kimchi groups were higher than those of a control group. The food efficiency ratio, epididymal fat pad weight of 3-, 6-week-fermented kimchi 10% groups were significantly lower than control and not-fermented kimchi groups. Especially 6-week-fermented kimchi groups showed adipocytes, less in number and larger in size than those of other groups. The blastogenesis of spleen lymphocytes to LPS was higher in rats fed fermented kimchi diets than rats fed control and not-fermented kimchi diet. These results suggest that kimchi stimulates lipid mobilization to epididymal fat pad and lipid excretion via feces, so lower serum and liver triglyceride concentration. The fermented kimchi stimulate the proliferation of B cell and lower the lipid accumulation in epididymal fat pad, especially kimchi fermented for 6 weeks at 4$^{\circ}C$ lower the adipose cell number.

The Effects of mCIMT using PNF on the Upper Extremity Function and Activities of Daily Living in Patients with Subacute Stroke (고유수용성촉진법을 이용한 수정된 강제유도 운동치료가 아급성 뇌졸중 환자의 상지 기능과 일상생활수행능력에 미치는 영향)

  • Purpose: The purpose of this study was to investigate the effects of a modified constraint induced movement therapy (mCIMT) using proprioceptive neuromuscular facilitation (PNF) on the upper extremity function and activities of daily living (ADLs) in patients with subacute stroke. Methods: Fourteen participants with subacute stroke were randomly assigned to a group using both mCIMT and PNF or a group using mCIMT alone. Each group underwent twenty sessions (1 h/d, 5 d/wk) for 4 weeks. Patients were assessed with the action research arm test (ARAT), the Fugl-Meyer assessment for the upper extremities (FMA-UE), the Modified Barthel Index (MBI), and motor activity logs (MALs; amount of use [AOU] and quality of movement [QOM]). Results: Both the experimental group and the control group showed significant intragroup improvement in the ARAT, FMA-UE, MBI, and MAL-AOU (p<0.05). The group using both mCIMT and PNF exhibited greater improvement in the ARAT, FMA-UE, MBI, and MAL-AOU than did the group using mCIMT alone. Statistical analyses showed significant differences in the ARAT (p=0.01), FMA-UE (p=0.01), MBI (p=0.00), and MAL-AOU (p=0.01) between the groups. Conclusion: This study applied mCIMT combined with PNF for subacute stroke patients, and the results showed significant improvements in the patients' upper extremity function and ADLs. Therefore, mCIMT using PNF may be more effective than mCIMT alone in improving upper limb function and ADLs in patients with subacute stroke.

Excretroy Urography in the Minipig (미니돼지에서 배설성요로조영술의 적용)

  • Excretory urography was performed on eight healthy minipigs to evaluate radiographic anatomical status and excretive function of the urinary tracts and to get an optimal procedure. The right and left kidneys were located in $T13{\sim}L4$and $T12{\sim}L3.5$, respectively. The left kidneys were located more cra-nially than the right. The everage lengths and widths of kidneys were $8.50{\pm}0.58$ $(mean{\pm}SD)$and $4.30{\pm}0.39$ cm on the ventrodorsal projection and $8.70{\pm}0.76$, and $4.10{\pm}0.40$ cm on the lateral projection, respectively. The lengths of kidneys were twice the width. When the lengths of kidneys were com-pared with lengths of the second lumbar vertebras, the ratios of kidney lengths to L2 lengths were $3.62{\pm}0.30$ on the ventrodorsal projection and $3.63{\pm}1.10$ on the lateral projection. The lengths and 쟝손 of renal pelvis including major calices were $4.01{\pm}0.46$, and 2.20{\pm}0.41$ cm, respectively. The lengths and 쟝손 of minor calices were $0.49{\pm}0.06$, and $0.10{\pm}0.01$ cm. The lengths and widths of the ureters were $12.25{\pm}2.05$, and 2.94{\pm}0.86$ cm. The nephrogram stage was reached in 3{\sim}5$ minutes, and the pyelogram phase in $5{\sim}15$ minutes individually after injection of contrast medium. The result of this study shows similar information and procedure of excretory urography for other laboratory ani-mals and leads to the potential use of this method for minipigs.
