The Anti-melanogenic Effect of Geranium krameri Extract (선이질풀 추출물의 멜라닌 생성 억제 효과)

  • We investigated the application of functional materials by examining a variety of physiological activities of Geranium krameri extract obtained from the National Institute of Horticultural and Herbal Science. Geranium krameri extract had a low cytotoxicity against murine melanoma B16F10 cells. At concentrations with little or no cytotoxicity, Geranium krameri extract showed high DPPH radical scavenging activity ($IC_{50}$, $8.72{\mu}g/mL$) and anti-microbial activities against Bacillus subtilis, Escherichia coli, and Candida albicans. Additionally, Geranium krameri extract inhibited tyrosinase activity ($IC_{50}$, $456.86{\mu}g/mL$) and decreased melanin content ($IC_{50}$, $50.35{\mu}g/mL$). The treatment of B16F10 cells with Geranium krameri extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggest that Geranium krameri extract inhibits melanin synthesis in murine melanoma B16F10 cells by suppressing intracellular tyrosinase expression, as well as directly inhibits tyrosinase activity simultaneously.

Effect of Abelmoschus esculentus extract on anti-melanogenesis and skin barrier function (오크라 추출물의 멜라닌 생성저해 및 피부장벽개선 효과)

  • We investigated the application of functional materials by examining a variety of physiological activities of Abelmoschus esculentus extract. A. esculentus extract had a low cytotoxicity against murine melanoma B16F10 cells. At concentrations showing little or no cytotoxicity, A. esculentus extract showed high a DPPH radical scavenging activity ($ID_{50}$, $5.24{\mu}g/mL$), inhibited tyrosinase activity ($ID_{50}$, $102.12{\mu}g/mL$), and decreased melanin content ($ID_{50}$, $17.85{\mu}g/mL$). The treatment of B16F10 cells with A. esculentus extract suppressed the protein expression of tyrosinase in a dose-dependent manner. These findings suggest that A. esculentus extract inhibits melanin synthesis in murine melanoma B16F10 cells by suppressing intracellular tyrosinase expression, as well as directly inhibits tyrosinase activity simultaneously. Additionally, A. esculentus extract promotes the expression of involucrin, which is related to skin barrier protection. These results indicate that A. esculentus extract is an appropriate material for improving skin barrier function.

Anti-inflammatory Effect of Extracts from Folk Plants in Ulleung Island (울릉도 민속식물 추출물의 항염증 효과)

  • We investigated the effect of extracts($50{\mu}g/mL$) from 31 folk plants in Ulleung Island on nitric oxide(NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Actinidia arguta(leaf, twig), Hovenia dulcis(leaf), Camellia japonica(leaf, twig), Sambucus sieboldiana var. pendula(leaf, twig) and Fallopia sachalinensis(root) showed significant NO inhibition activity(42%~88% NO inhibition rate). Of them, we selected 3 samples(A. arguta, H. dulcis and C. japonica) showing more than 50% NO inhibition activity and little effect on cell viability. We found that the extracts suppressed the NO and prostaglandin $E_2(PGE_2)$ produced by LPS-activated RAW264.7 cells in dose-dependent manners.

  • The mature spores of present Myxobolus sp. was ovoid in front view with no distinct redges of folds, lemon-shaped in side view with a straight sutural ridge. Spore valves showed symmetrical and smooth. Spores were 9 to $12{\mu}m$ (Mean=$10.4{\pm}0.7$, n=50)in length, 6 to $9{\mu}m$ (Mean=$7.7{\pm}0.6$, n=50) in width and 5.0 to $7.5{\mu}m$ (Mean=6.2, n=7) in thickness. Two polar capsules of spore were pyriform in shape, equal or mearly equal in size, 3 to $6{\mu}m$(Mean=$4.6{\pm}0.6$, n=50) in length, 2 to $3{\mu}m$(Mean=$2.2{\pm}0.3$, n=50) in width, Polar filaments of spore were composed with six to seven coils within capsules. Extended polar filaments were 55 to $135{\mu}m$ (Mean=78.7, n=50)in length. The shape and measurements of the present Mysobolus sp. spores were very similar with the spore of M. cyprinicola Reuss, 1906.

Somatic embryogenesis induction in four cassava landraces in East Java, Indonesia

  • Manihot esculenta Crantz, commonly known as cassava, is a staple aliment that is a significant source of revenue for farmers. The embryogenic callus is crucial in the genetic engineering of various crop species, including cassava. Four cultivar cassava landraces from East Java were assessed for their ability to produce friable embryogenic callus (FEC) for protoplast isolation. In this study, four cassava cultivars; (Kaspro, Kuning, Gajah, and Gendruwo); were used to obtain FEC, which involved the culture of immature leaf lobes (ILLs) and apical buds (ABs) media containing MS supplemented with 33 μM picloram and 2 μM of CuSO4 (M1) or MS supplemented with 50 μM 2,4-D and 2 μM CuSO4 (M2). The highest FEC induction efficiency ranged from 72% to 57%, and the highest FEC number ranged from 4.7 to 3.7 with AB explants in media containing MS + 33 μM pilocram and 2 μM CuSO4 (M1). On the other hand, the efficiency of somatic embryogenesis induction ranged from 67% to 53%, and the number ranged from 4.4 to 3.4. The efficiencies of FEC induction ranged from 48% to 42%, and the number ranged from 3.1 to 2.6 with AB explants in media containing MS + 50 μM 2,4-D and 2 μM CuSO4 (M2); the efficiency of FEC induction ranged from 56% to 50%, and the value ranged from 3.6 to 2.4 with ILL explants. The FEC induction of the Gendruwo cultivar, which was examined using AB and ILL explants, demonstrated the lowest efficiency. Nevertheless, all four cultivars showed the ability to generate FEC, even though their effectiveness differed depending on the explant genotype and the applied media.

Establishment of Baseline Sensitivity of Phytophthora capsici Causing Pepper Phytophthora Blight to Carboxylic Acid Amide Fungicides (Carboxylic acid amide계 살균제에 대한 고추 역병균 Phytophthora capsici의 감수성 기준 설정)

  • Baseline sensitivity to benthiavalicarb, iprovalicarb and dimethomorph included into carboxylic acid amide (CAA) group was evaluated in 180 isolates of Phytophthora capsici over 4 years from 2005 to 2008. $EC_{50}$ (effective concentration inhibiting mycelial growth by 50%) value of benthiavalicarb ranged from $0.015{\mu}g\;mL^{-1}$ to $0.049{\mu}g\;mL^{-1}$ with a mean of $0.033{\mu}g\;mL^{-1}$. The mean values of $EC_{50}$ of iprovalicarb and dimethomorph were 0.411 (0.197 - 0.556) ${\mu}g\;mL^{-1}$ and 0.271 (0.101 - 0.798) ${\mu}g\;mL^{-1}$, respectively. Although there was no increasing tendency in $EC_{50}$of benthiavalicarb and iprovalicarb during 4 years, $EC_{50}$ of dimethomorph was increased gradually by laps of time. There was no cross-resistance between each fungicide used in this study and metalaxyl. Among fungicides included into CAA group, there was a positive correlation between benthiavalicarb and iprovalicarb, and between dimethomorp and mandipropamid.

Antioxidant and Tyrosinase Inhibitory Activities of Dicaffeoylquinic Acid Derivatives Isolated from Gnaphalium Affine D. DON (떡쑥 추출물로부터 분리된 Dicaffeoylquinic Acid 유도체들의 항산화 및 타이로시네이즈 저해 활성)

  • In this study, three dicaffeoylquinic acids (DCQAs) isolated from Gnaphalium affine D. DON. extracts were structurally identified and evaluated for their antioxidant activities, cellular protective effects, and tyrosinase inhibitory activities. The ethyl acetate fraction of G. affine was chromatographed, which yielded 3 DCQA derivatives of 1-3 : 3,5-dicaffoylquinic acid (3,5-DCQA, 1), 4,5-dicaffeoylquinic acid (4,5-DCQA, 2), 1,5-dicaffoylquinic acid (1,5-DCQA, 3). The structure of each compounds was determined using $^1H$ NMR and MS analyses. Compounds of 1-3 showed strong free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}=3.70$, 5.80, and $5.50{\mu}M$, respectively) compared to those of a commonly used lipophilic antioxidant, (+)-${\alpha}$-tocopherol ($21.90{\mu}M$). Cellular protective effects of 1-3 compounds on the $^1O_2$ sensitized photohemolysis of human erythrocytes were similar to (+)-${\alpha}$-tocopherol. 1-3 compounds also exhibited higher tyrosinase inhibitory effects ($IC_{50}=0.15$, 0.16, and 0.13 mM) compared to arbutin (0.33 mM), known as a skin-whitening agent. These results indicate that three DCQA derivatives may be applied as an antioxidant and a skin whitening agent in food or cosmetic industries.

Inhibitory Effects of Carex humilis Extract on Elastase Activity and Matrix Metalloproteinase-1 Expression (산거울 추출물의 Elastase 활성 저해 및 Matrix Metalloproteinase-1 발현 억제 효과)

  • In order to evaluate anti-wrinkle activity of Carex humilis extract, free radical scavenging activity, elastase inhibitory activity and reduction of expression Matrix metalloproteinase-1 (MMP-1) mRNA and MMP-1 protein were investigated. The roots of Carex humilis were extracted with 95 % ethanol and successively partitioned with organic solvents with increasing polarity of the solvents. Each fraction of organic solvent were investigated by using free radical scavenging activity and elastase inhibitory activity test. Among them, EtOAc fraction showed antioxidant activity ($SC_{50}$=4.89 ${\mu}g/mL$) and elastase inhibitory activity ($IC_{50}$=23.5 ${\mu}g/mL$). EtOAc fraction was developed on silica gel by open-column chromatography and consecutively re-developed on C18 resin by prep-HPLC to give ${\alpha}$-viniferin as a major component, which was confirmed by spectrometric analysis. In the assay on expression of MMP-1 mRNA by RT-PCR and protein by western-blot, EtOAc layer (10 ~ 100 ${\mu}g/mL$) was reduced about 50 ~ 60 %, 50 ~ 65 % respectively and ${\alpha}$-viniferin (0.5 ~ 2 ${\mu}g/mL$) was inhibited about 60 ~ 75 %, 55 ~ 65 % respectively in human fibroblast. Therefore, our findings suggest that EtOAc layer of Carex humilis containing ${\alpha}$-viniferin can be useful as an active ingredient for cosmeceuticals of anti-wrinkle effects.

Evaluation of Physicochemical Properties and Biological Activities of Steamed and Fermented Deodeok (Codonopsis lanceolata) (증숙 및 발효 더덕의 이화학적 특성 및 생리활성 변화)

  • This study was designed to evaluate the physicochemical properties and biological activities of steamed and fermented Codonopsis lanceolata. The treatments included NS-NF (non-steamed and non-fermented), NS-LF (non-steamed and L. rhamnosus fermented), S-NF (steamed and non-fermented), and S-LF (steamed and L. rhamnosus fermented). Total polyphenol amounts of S-NF and S-LF were significantly increased to more than 26 mg GAE/g. The highest DPPH scavenging activities were observed for S-NF and S-LF, showing $EC_{50}$ values of 0.8 and 0.6 mg/mL, respectively. The growths of Staphylococcus aureus, Listeria monocytogenes, Salmonella Typhimurium, and Shigella boydii were effectively inhibited by S-LF (MIC < 9 mg/mL). The NS-LF and S-LF ($EC_{50}$ <6 mg/mL) effectively inhibited ${\alpha}$-Glucosidase and tyrosinase activities compared to NS-NF ($EC_{50}$ <17 mg/mL). The S-LF exhibited the highest acetylcholinesterase (AChE) inhibitory activity ($IC_{50}$ <32 mg/mL). Therefore, the results suggest that the application of the steaming process combined with probiotic fermentation can effectively enhance the biological and pharmacological activities in C. lanceolata.

Evaluation of Separation Distance from the Temporary Storage Facility for Decontamination Waste to Ensure Public Radiological Safety after Fukushima Nuclear Power Plant Accident (후쿠시마 원전 사고 이후 일반인의 방사선학적 안전성 확보를 위한 제염폐기물 임시저장시설 이격거리 평가)

  • The object of this study was to evaluate the separation distance from a temporary storage facility satisfying the dose criteria. The calculation of ambient dose rates took into account cover soil thickness, facility size, and facility type by using MCNPX code. Shielding effects of cover soil were 68.9%, 96.9% and 99.7% at 10 cm, 30 cm and 50 cm respectively. The on-ground type of storage facility had the highest ambient dose rate, followed by the semi-ground type and the underground type. The ambient dose rate did not vary with facility size (except $5{\times}5{\times}2m\;size$) due to the self-shielding of decontamination waste in temporary storage. The separation distances without cover soil for a $50{\times}50{\times}2m\;size$ facility were evaluated as 14 m (minimum radioactivity concentration), 33 m (most probably radioactivity concentration), and 57 m (maximum radioactivity concentration) for on-ground storage type, 9 m, 24 m, and 45 m for semi-underground storage type, and 6 m, 16 m, and 31 m for underground storage type.
