Temporal and Spatial Characteristics in the Pollen Flow of Living Modified Rice (유전자변형 벼로부터 일어나는 화분비산의 시공간적 특성)

  • Pollen flow is one of the essential components in the ecological risk assessment of transgenic crops, because pollen can act as a vehicle to disseminate transferred alien genes. Pollen flow pattern of a cultivated rice variety and Living modified (LM) rice was studied at diurnal and distance changes under field. We measured airborne pollen density at the distances of -1, 0.5, 0, 1, 2, 3, 4, 5, 7, 9, 11 and 13 m from rice cultivation and recorded the direction and speed of wind using weather station in the conventional rice paddy field during the flowering period of rice. Diurnal changes in pollen density were observed as a peak between 10:00 to 13:00 hr. The density of airborne rice pollen geometrically decreased with the increase of distance from pollen sources. It is therefore necessary to carry out a detailed investigation of pollen flow of a particular species, where ecological risk assessment requires an accurate estimation of pollen flow including both distance and intensity of pollen dispersal. The rice pollen flow was significantly influenced by weather conditions, particularly by wind direction and speed. The precise determination of the local wind conditions at flowering time therefore appears to be of primary importance for setting up suitable isolation distance from transgenic rice in the field.

Functional characterization of a CCCH type zinc-finger protein gene OsZF2 by ectopic overexpression of the gene in rice (과발현 형질전환벼에서 CCCH type zinc-finger protein 유전자 OsZF2 기능 분석)

  • We have previously isolated a CCCH type zinc-finger protein gene, OsZF2 (Oryza sativa Zinc Finger 2), from the cold-treated rice cDNA library. To investigate the potential role of OsZF2, transgenic rice lines over-expressing OsZF2 under the control of CaMV 35S promoter have been developed through Agrobacterium-mediated transformation. Elevated level of OsZF2 transcripts was confirmed by RNA gel blot analysis in transgenic rice. Under the 100 mM NaCl condition, the transgenic rice showed significantly enhanced growth rate in terms of shoot length and fresh weight, implicating that OsZF2 is likely to be involved in salt response of rice. In the field condition, however, the transgenic rice showed a dwarf phenotype and flowering time was delayed. Genome expression profiling analysis of transgenic plants using the 20K NSF rice oligonucleotide array revealed many up-regulated genes related to stress responses and signaling pathways such as chaperone protein dnaJ 72, salt stress-induced protein, PR protein, disease resistance proteins RPM1 and Cf2/Cf5 disease resistance protein, carbohydrate/ sugar transporter, OsWAK kinase, brassinosteroid LRR receptor kinase, and jasmonate O-methyltransferase. These data suggest that the CCCH type zinc-finger protein OsZF2 is a upstream transcriptional factor regulating growth and stress responsiveness of rice.

Analysis of the Climate inside Multi-span Plastic Greenhouses under Different Shade Strategies and Wind Regimes

  • In this work, the effects of shade combination, shade height and wind regime on greenhouse climate were quantified. A two-dimensional (2-D) computational fluid dynamics (CFD) model was developed based on an 11-span plastic greenhouse in eastern China for wind almost normal to the greenhouse orientation. The model was first validated with air temperature profiles measured in a compartmentalized greenhouse cultivated with mature lettuce (Lactuca sativa L., 'Yang Shan'). Next, the model was employed to investigate the effect of shade combinations on greenhouse microclimate patterns. Simulations showed similar airflow patterns in the greenhouse under different shade combinations. The temperature pattern was a consequence of convection and radiation transfer and was not significantly influenced by shade combination. The use of shade screens reduced air velocity by $0.02-0.20m{\cdot}s^{-1}$, lowered air temperature by $0.2-0.8^{\circ}C$ and raised the humidity level by 0.9-2.0% in the greenhouse. Moreover, it improved the interior climate homogeneity. The assessment of shade performance revealed that the external shade had good cooling and homogeneity performance and thus can be recommended. Furthermore, the effects of external shade height and wind regime on greenhouse climate parameters showed that external shade screens are suitable for installation within 1 m above roof level. They also demonstrated that, under external shade conditions, greenhouse temperature was reduced relative to unshaded conditions by $1.3^{\circ}C$ under a wind speed of $0.5m{\cdot}s^{-1}$, whereas it was reduced by merely $0.5^{\circ}C$ under a wind speed of $2.0m{\cdot}s^{-1}$. Therefore, external shading is more useful during periods of low wind speed.

Comparison of Herbicidal Action between Pyrazosulfuron - ethyl and Imazaquin (Pyrazosulfuron - ethyl과 Imazaquin의 살초작용 비교)

  • To know whether pyrazosulfuron-ethyl(PYR) and imazaquin(IMA), known as a acetolactate synthase(ALS) inhibitors, have a same herbicidal action pattern in rice(Oryza sativa) or barnyardgrass (Echinochloa crus-galli), an inhibition pattern and a response characteristics in combination with dymron or butachlor were investigated. In contrast to the phytotoxicity of rice treated with IMA, the one treated with PYR was completely tended to be recovered after 25 days after treatment. Safening effect of dymron against PYR was effectively developed to transplanted-rice, while such an effect was not shown in combination with IMA. In combination with PYR and butachlor, antagonistic effect was observed in both simultaneous or sequential treatment on bamyardgrass, however, additive effect was rather shown in combination with IMA and its activity was dominantly dependent on the first applied compound. $I_{50}$ of PYR and IMA on the ALS extracted from barnyardgrass was $4{\times}10^{-7}$M and $2.8{\times}10^{-6})$M, respectively. Butachlor did not affect their activities on ALS in vitro. These results suggest that PYR and IMA might have a different action each other in the pathway to a final herbicidal activity even though their primary action site is ALS.

Response of Ascorbate Peroxidase and Dehydroascorbate Reductase in Lettuce (Lactuca sativa L.) Leaves Exposed to Cold Stress (저온 처리한 상추의 잎 내에서 ascorbate peroxidase와 dehydroascorbate reductase의 반응)

  • To investigate the relationship between cold stress and the activity of ascorbate peroxidase(APX), dehydroascorbate reductase (DHAR), mRNA expression level of two enzymes, hydrogen peroxide content was studied in lettuce leaves under stress condition imposed by cold stress at $4^{\circ}C$ for 24 hr in the dark and following recovery at $20^{\circ}C$ from cold stress. Hydrogen peroxide content increased gradually in lettuce leaves during cold stress, but decreased slightly following recovery from cold stress. Soluble protein content, however, decreased gradually during cold stress, and then rapidly returned to normal levels following recovery. Total chlorophyll content decreased gradually during cold stress, and then keep constant following recovery. The patterns of chlorophyll a and b content similar to that of total chlorophyll content, and carotenoid content didn't change. The ratio of chlorophyll a and total chlorophyll was increased during cold stress, but decreased with rapid during cold stress, and then the ratio returned to normal levels following recovery. During cold stress, the activity of APX and DHAR in the lettuce leaves increased dramatically, and also transcript levels of mRNA of APX and DHAR, as determined by probing 32P-labeled single stranded RNA of APX and DHAR, highly increased and returned to normal levels following recovery, respectively. Relationship between APX and DHAR activity and hydrogen peroxide highly related ($R^2$=0.8715 and 0.8643), whereas between hydrogen peroxide and total chlorophyll content and soluble content related reversely ($R^2$=0.5021 and 0.8915).

Thermal Stability of the Major Color Component, Cyanidin 3-glucoside, from a Korean Pigmented Rice Variety in Aqueous Solution (한국산 유색미의 주요성분인 Cyanidin 3-glucoside의 수용액에서의 열안정성)

  • Thermal stability of the major color component, cyanidin 3-glucoside, isolated from Korean pigmented rice (Oryza sativa var. Suwon 415) were investigated to explore possible application of value-added natural colors as food additives. The anthocyanin showed red and blue color with maximum absorption peaks at 511 nm and 572 nm in acidic (pH 2.0) and alkaline (pH 9.0) buffer solutions, respectively, and the thermal degradation reactions were carried out with different temperature ranges at $50{\sim}95^{\circ}C$. Degree of degradation was determined with UV/Vis spectra which indicate characteristic absorption patterns with sharp isosbestic points at 350 nm (pH 2.0), and 275, 310, and 405 nm (pH 9.0). Thus the reaction follows simple first-order kinetics. The anthocyanin was very stable against heat at acidic pH and relatively stable at alkaline pH with half-life values of 50.3 hr and 0.6 hr at $70^{\circ}C$, respectively. The activation energies and Arrhenius frequency factors of the pigment were 26.9 kcal $mol^{-1}\;and\;6.0{\times}10^{11}\;s^{-1}$, at pH 2.0, and 15.2 kcal $mol^{-1}\;and\;1.4{\times}10^{6}\;s^{-1}$, pH 9.0, and respectively.

Expression Analysis of OsCPK11 by ND0001 oscpk11 Mutants of Oryza sativa L. under Salt, Cold and Drought Stress Conditions (염분, 저온 및 가뭄 스트레스 조건에서 벼 ND0001 oscpk11 돌연변이체의 OsCPK11 발현 분석)

  • Calcium-dependent protein kinases (CDPKs) are known to be involved in regulating plant responses to abiotic stresses such as salinity, cold temperature and dehydration,. Although CDPKs constitute a large multigene family consisting of 31 genes in rice, only a few rice CDPKs' functions have been identified. Therefore, in order to elucidate the functions of OsCPK11 in rice, this study was intended to focus on the expression pattern analysis of OsCPK11 in wild type and ND0001 oscpk11 mutant plants under these abiotic stresses. For the salt, cold and drought stress treatment, seedlings were exposed to 200 mM NaCl, 4℃ and 20% PEG 6,000, respectively. RT-PCR and quantitative real-time PCR were performed to determine the expression patterns of OsCPK11 in wild type and ND0001 mutant plants. RT-PCR results showed that OsCPK11 transcripts in the wild type and heterozygous mutant were detected, but not in the homozygous mutant. Real-time PCR results showed that relative expression of OsCPK11 of wild type plants was increased and reached to the highest level at 24 hr, at 6 hr and at 24 hr under salt, cold and drought stress conditions, respectively. Relative expression of OsCPK11 of ND0001 homozygous plant was significantly reduced compared to that of wild type. These results suggested that oscpk11 homozygous mutant knocks out OsCPK11 and OsCPK11 might be involved in salt, cold and drought stress signaling by regulating its gene expression.

Vascular Plant Diversity of Sambong and Jaung Mountains in Paju City, Border Area of the Korean DMZ (DMZ접경지역에 위치한 파주시 삼봉산·자웅산 일대의 관속식물 다양성)

  • This study conducted a flora survey in the areas of Mt. Sambong (282m) and Mt. Jaung (263m), located in Paju-si, Gyeonggi-do. A total of 434 taxa, including 6 forms, 34 varieties, 11 subspecies, 383 species, 271 genera, and 94 families, were surveyed in the two mountain areas. The ferns were identified as 24 taxa from 10 families, the gymnosperms as 8 taxa from 2 families, the dicotyledonous plants as 322 taxa from 73 families, and the monocotyledonous plants as 80 taxa from 9 families. Five taxa, including Salix koriyanagi, Cimicifuga heracleifolia, Forsythia koreana, Paulownia coreana, and Hemerocallis hakuunensi, were identified as the Korean endemic plants. Eight taxa, including Cimicifuga heracleifolia, Tylophora floribunda, and Iris minutiaurea as Vulnerable species, andAristolochia contorta, Viola diamantiaca, Lithospermum erythrorhizon, Lilium distichum, andArisaema heterophyllumas Least Concern species, were designated as rare and endangered plants designated by the Korea Forest Service. The invasive alien plants were 40 taxa, including Rumex crispus, Phytolacca americana, Cerastium glomeratum, Silene armeria, Chenopodium album, Chenopodium ficifolium, Magnolia obovata, Potentilla supina, Amorpha fruticosa, Medicago sativa, Robinia pseudoacacia, Trifolium repens, Euphorbia supina, Hibiscus trionum, and Oenothera biennisaccounting for 9.2 percent of the 434 taxa of the surveyed plants. As this area is a mountainous area close to settlement facilities, it is believed that ecological preservation measures and measures of specialized plants should be implemented.

Effects of Seed Soaking of Kinetin on Alleviating Copper Toxicity during Germination in Rice (Kinetin 침종처리가 벼 발아중 구리 독성 경감에 미치는 영향)

  • The study was carried out to determine the proper concentration of plant growth regulator, kinetin on alleviating copper toxicity for two rice cultivars of seed germination. The results were as followings : Soaking treatment of kinetin 10$^{-3}$ M increased the germination rate of both cultivars, Ilpumbyeo and Hyangmibyeo 1 by 92% and 88% as compared with copper treatment (60ppm). But the soaking treatment effect of plant growth regulator, kinetin was not recognized at the kinetin 10$^{-4}$ M and 10$^{-5}$ M. Chlorophyll content of both rices was higher than that of Hyangmibyeo 1. Copper content of Ilpumbyeo was higher in leaf than in seed part. At the 3 days after treatment of copper 60ppm, both cultivars of treatment of kinetin 1O-3M showed the somewhat thin bands at the 35 and 40kDa compared with others. A new protein band pattern was only appeared to kinetin 1O-3M at approximately 54.4kDa(M. W) at the 7 days after treatment of copper 60ppm in llpumbyeo cultivar, SOD activity of copper 60ppm treatment increased in 3DAT, but there were no significant differences in 5 and 7DAT of two cultivars. Free proline contents of copper 60ppm treatment in llpumbyeo were remarkably increased about 4.996$\mu$M. In particular, free proline content of kinetin l0$^{-3}$ M in Ilpumbyeo was 5.008$\mu$M in 3DAT. In case of Hwangmibyeo 1, free proline content of copper 60ppm was 5.825$\mu$M compared with an untreated control showing 2.34l$\mu$M. The effects of kinetin treatment were recognized to promote the root growth and germination rate under copper toxicity(60ppm) condition in both cultivars.

Lucerne transient streak virus; a Recently Detected Virus Infecting Alfafa (Medicago sativa) in Central Saudi Arabia

  • A survey was conducted to determine the status of Lucerne transient streak virus (LTSV) in three high-yielding alfalfa regions in central Saudi Arabia (Riyadh, Qassim, and Hail) during 2014. Three hundred and eight symptomatic alfalfa, and seven Sonchus oleraceus samples were collected. DAS-ELISA indicated that 59 of these samples were positive to LTSV. Two isolates of LTSV from each region were selected for molecular studies. RT-PCR confirmed the presence of LTSV in the selected samples using a specific primer pair. Percentage identity and homology tree comparisons revealed that all Saudi isolates were more closely related to each other but also closely related to the Canadian isolate-JQ782213 (97.1-97.6%) and the New Zealand isolate-U31286 (95.8-97.1%). Comparing Saudi isolates of LTSV with ten other sobemoviruses based on the coat protein gene sequences confirmed the distant relationship between them. Eleven out of fourteen plant species used in host range study were positive to LTSV. This is the first time to document that Trifolium alexandrinum, Nicotiana occidentalis, Chenopodium glaucum, and Lathyrus sativus are new host plant species for LTSV and that N. occidentalis being a good propagative host for it.
