Batch and Continuous Culture Kinetics for Production of Carotenoids by ${\beta}$-Ionone-Resistant Mutant of Xanthophyllomyces dendrorhous

  • A ${\beta}$-ionone-resistant mutant strain isolated from the red yeast Xanthophyllomyces dendrorhous KCTC 7704 was used for batch and continuous fermentation kinetic studies with glucose media in a 2.5-1 jar fermentor at $22^{\circ}C$ and pH 4.5. The kinetic pattern of growth and carotenoid concentration in the batch fermentations exhibited a so-called mixed-growth-associated product formation, possibly due to the fact that the content of intracellular carotenoids depends on the degree of physical maturation toward adulthood. To determine the maximum specific growth rate constant (${\mu}_m$) and Monod constant ($K_s$) for the mutant, glucose-limited continuous culture studies were performed at different dilution rates within a range of $0.02-0.10\;h^{-1}$. A reciprocal plot of the steady-state data (viz., reciprocal of glucose concentration versus residence time) obtained from continuous culture experiments was used to estimate a ${\mu}_m$ of $0.15\;h^{-1}$ and $k_s$ of 1.19 g/l. The carotenoid content related to the residence time appeared to assume a typical form of saturation kinetics. The maximum carotenoid content ($X_m$) for the mutant was estimated to be $1.04\;{\mu}g/mg$ dry cell weight, and the Lee constant ($k_m$), which was tentatively defined in this work, was found to be 3.0 h.

연속결정화 방법에 의한 13X 제올라이트 결정성장 (Crystal growing of sodium type 13X zeolite by continuous crystallization method)

  • NaX seed결정(10~20 $\mu$m)을 $3.5Na_2$O : $Al_2O $: $2.1SiO_2$ : $1000H_2$O 용액에 0.5~2.0g 첨가한 후 연속결정화법으로 50 $\mu$m의 균일한 결정을 성장시켰다. 연속결정화법에 의한 결정성장을 시험하기 위하여, 모액을 7일, 5일, 3일, 2일과 1일 간격으로 공급하였다. Seed 첨가의 결과는 첨가하지 않은 용액과 비교하여 보다 균일하고 큰 결정들을 얻었다. 합성용액에 seed의 첨가는 반응물과 물리적인 접촉 면적을 초래하여 합성 겔의 핵성장 없이 seed 결정의 결정성장을 확인할 수 있다.

Studies on Solvent Sublation of Trace Heavy Metals by Continuous Flow System as Ternary Complexes of 1,10-Phenanthroline and Thiocyanate Ion

  • A continuous flow system has been developed to determine trace Cu(II), Mn(II), Ni(II) and Zn(II) in a large volume of water samples by a solvent sublation technique. The mixed solution of 1,10-phenanthroline(phen) and thiocyanate ion was used as ligands for the formation of their ternary complexes. The continuous system was constructed in this laboratory with a peristaltic pump, a mini shaker, three mixing bottles and a flotation cell by connecting each part with a polyethylene tube. The flotation conditions such as the flow rate of sample solution and the injection rates of ligand, buffer and surfactant solutions have been investigated to obtain the best sublation efficiencies. Each solution flowed into the flotation cell through each polyethylene tube by the peristaltic pumps. The ternary complexes were floated and extracted into MIBK in a flotation cell of 2 L by bubbling a nitrogen gas. The absorbances of extracted analytes in MIBK were directly measured by graphite furnace-AAS. The concentrations of 1,10-phenanthroline and thiocyanate ion were $2.6\;{\times}\;10^{-3}$ M and $2.3\;{\times}\;10^{-2}$ M in the mixed solution, respectively. The pH of sample solution was adjusted to 5.0 with a buffer solution and 1%(m/v) sodium lauryl sulphate solution was added as a surfactant to support the effective flotation of the complexes. The $N_2$ gas was bubbled at 30 mL/min for 90 minutes for 20 L of sample. Reproducible results of less than 10% RSD and recoveries of 80-120% could be obtained in real samples.

고정화 Aminopeptidase M 컬럼 반응기를 이용한 메치오닐 인간성장호르몬으로부터 천연형 인간성장호르몬의 연속생산 (Continuous Production of Authentic Human Growth Hormone from Methionyl Human Growth Hormone Using the Column Reactor of Immobilized Aminopeptidase M)

  • Aminopeptidase M(ApM)을 Cellufine Formyl 에 고정화시켜 고정화 효소의 반응특성을 고찰하고, 고정화 ApM을 충진한 column reactor를 이용하여 메치오닐 인간 성장 호르몬(met-hGH)으로부터 천연형 인간 성장 호르몬(hGH)의 연속 생산을 검토 하였다. Cellufine Formyl 19 gel당 2.3mg의 ApM 이 결합되었을 때 met-hGH의 hGH로의 전환능력이 가장 우수하였다. Soluble enzyme과 고정화 효 소의 반응 최적 pH는 7.0, 반응 최적 온도는 $55^{\circ}C$로 통일하였으나 고정화에 의해 pH 벙위가 보다 넓 어졌으며, column reactor에서 연속 운전시 최적온 도 역시 $55^{\circ}C$로 나타났다. Column reactor를 이용 한 천연형 hGH의 연속 생산시 met-hGH가 100% 전환되는 조건에서 hGH 수율과 생산성은 각각 약 77%와 약 0.8mg hGH/ml.h이었다. 반응기 크기 를 5배 증가시켰을 때 두 반융기에서 유속 SV 값이 통일하면 met-hGH 전환율과 hGH 수율이 통일하 였으며, 90일간의 연속 운전 결과로 예측한 column reactor의 반감기는 45t에서 228일, $55^{\circ}C$에서 81 일로 비교척 안정하였다.

  • Automatic monitoring of cutting process is one of the most important technology in machining. AE sensing technology has been applied to monitoring process and proved to be effective in detecting tool abnor- malities such as tool wear and fracture. In this experimental study. AE signals were detected from the tool holder for continuous and interrupted cutting, which obtained from changing workpice material configuration, under control of constant cutting speed from CNC lathe. From statistical and frequency analysis, the AE signals were analyzed to obtaining the characteristics of continuous and interrupted cutting conditions and tool failure. The Kurtosis values decreased but RMS voltages increased as the cutting speed increased, in both continuous and interrupted cutting. RMS voltage is suddenly increased but Kurtosis value is suddenly decreased when tool failure condition. Power spectrum density of AE signals when tool failure reaches extreme value around 0.065 cycles/ .mu. m.

Behaviour of continuous prestressed concrete beams with external tendons

  • External prestressing has been applied to both new construction and retrofitting of existing reinforced and prestressed concrete structures. Continuous beams are preferred to simply supported beams because of economy, fewer movement joints and possible benefits from moment redistribution. However, this paper argues that continuous prestressed concrete beams with external unbonded tendons demonstrate different full-range behaviour compared to reinforced concrete (RC) beams. Applying the same design approach for RC to external prestressing may lead to design with a lower safety margin. To better understand the behaviour of continuous prestressed concrete beams with unbonded tendons, an experimental investigation is performed in which nine such specimens are tested to failure. The full-range behaviour is investigated with reference to moment-curvature relationship and moment redistribution. The amounts of moment redistribution measured in the experiments are compared with those allowed by BS 8110, EC2 and ACI 318. Design equations are also proposed to estimate the curvature ductility index of unbonded prestressed concrete beams.

  • In this paper, the study focus on continuous query in EPC Information Services(EPCIS) middleware which is a component of RFID system. We can consider EPCIS as a data stream system with a repository. In our work continuous query is implemented in two query execution model. One is standing query model another is traditional query execution model in which continuous query run over database periodically. Furthermore a balance strategy is presented. It is used to determine which continuous query implementation model is suitable for the query. Finally we conclude our work and issue some research topic for future work.

공기주입방식에 의한 매립지가스 안정화에 관한 현장연구 (Field Study on Stabilization of Landfill Gas by Air Injection Mode)

  • 본 연구에서는 공기주입 방식과 공기주입 시간이 매립지가스 안정화에 미치는 영향을 평가하고 매립지굴착사업 진행시 굴착작업 시간을 판단하고자 실시하였다. 간헐주입의 경우 혐기성에서 호기성으로의 전환되는데 연속주입보다 8배 정도 긴 시간이 소요되어 매립지굴착사업과 같이 혐기성 가스를 빠른 시간 내에 호기성 가스로 치환하여 굴착작업을 수행해야 하는 경우에는 연속주입이 더 적합할 것으로 판단되었다. $0.2m^3/min$ 유량으로 공기를 연속주입한 후 중단하였을 때 공기주입 시간과 호기성 조건($CH_4$ < 5%)이 유지되는 시간과는 비교적 높은 상관도($r^2$ = 0.95)를 나타내고 있었다. 공기주입 시간을 1이라고 볼 때 공기주입 중단 후 호기성은 약 1.5배 정도의 긴 기간 동안 유지되고 있었다.

  • LCD-BLU is one of kernel parts of LCD and it consists of several optical sheets: LGP, light source and mold frame. The LGP of LCD-BLU is usually manufactured by etching process and forming numerous dots with $50\sim300{\mu}m$ diameter on the surface. But the surface of the etched dots of LGP is very rough due to the characteristics of the etching process during the mold fabrication, so that its light loss is high along with the dispersion of light into the surface. Accordingly, there is a limit in raising the luminance of LCD-BLU. In order to overcome the limit of current etched dot patterned LGP, optical pattern with continuous microlens was designed using optical simulation CAE. Also, a mold with continuous micro-lens was fabricated by UV-LiGA reflow process and applied to 7 inch size of navigator LCD-BLU in the present study.

Lactobacillus brevis의 고정화 균체에 의한 γ-aminobutyric acid의 연속 생산 (Continuous Production of γ-aminobutyric Acid by Immobilization of Lactobacillus brevis)

  • Lactobacillus brevis BH-21의 균체를 고정화하여 column형 reactor를 이용하여 $\gamma$-aminobutyric acid의 연속적 생산을 검토하였다. Lactobacillus brevis BH-21에 의한 $\gamma$-aminobutyric acid 생산조건은 고정화 담체로는 3.0% sodium alginate가 좋았고 직경 2.2 mm의 bead를 사용하여 기질의 주입속도는 10 mL/h이 최적 조건이었다. Lactobacillus brevis BH-21의 고정화 균체와 유리 균체를 사용하여 $\gamma$-aminobutyric acid의 생산을 최적 조건하에서 1, 2, 3, 및 4차 발효 동안 48시간마다 신선한 기질을 주입하면서 비교ㆍ조사한 결과 고정화 균체를 사용 시에 생산량이 더 많았다. Column형 reactor에 의한 연속 발효 시 발효 25일까지 생산이 우수하였고 발효 30일까지는 $\gamma$-aminobutyric acid를 생산할 수 있었다.
