The monomer and dimer structures of the amyloid fragment Aβ(1-16) sequence formed in H2O were investigated using electrospray ionization mass spectrometry (MS) and tandem MS (MS/MS). Aβ16 monomers and dimers were indicated by signals representing multiple proton adduct forms, [monomer+zH]n+ (=Mz+, z = charge state) and [dimer+zH]z+ (=Dz+), in the MS spectrum. Fragment ions of monomers and dimers were observed using collision-induced dissociation MS/MS. Peptide bond dissociation was mostly observed in the D1-D7 and V11-K16 regions of the MS/MS spectra for the monomer (or dimer), regardless of the monomer (or dimer) charge state. Both covalent and non-covalent bond dissociation processes were indicated by the MS/MS results for the dimers. During the non-covalent bond dissociation process, the D3+ dimer complex was separated into two components: the M1+ and M2+ subunits. During the covalent bond dissociation of the D3+ dimer complex, the b and y fragment ions attached to the monomer, (M+b10-15)z+ and (M+y9-15)z+, were thought to originate from the dissociation of the M2+ monomer component of the (M1++M2+) complex. Two different D3+ complex geometries exist; two distinguished interaction geometries resulting from interactions between the M1+ monomer and two different regions of M2+ (the N-terminus and C-terminus) are proposed. Intricate fragmentation patterns were observed in the MS/MS spectrum of the D5+ complex. The complicated nature of the MS/MS spectrum is attributable to the coexistence of two D5+ configurations, (M1++M4+) and (M2+M3+), in the Aβ16 solution.
Poly-N-vinylcarbazole/polyaniline (PVK-PANI) composites are synthesized by varying target loading concentrations of aniline (0.025 - 0.1 M). The surface morphology of the composites is studied by scanning electron microscopy (SEM) and atomic force microscopy (AFM). The temperature-dependent DC conductivity of PVK-PANI composite films was studied at the temperature range of 300 - 500 K. The data suggest that the conductivity increase with an increase in aniline concentration in the composite with an increase in temperature. Further based on the conductivity behavior we can suggest that the PVK-PANI composites show a semiconducting behavior with a positive temperature coefficient of resistivity (TCR). The enhanced conductivity and the positive TCR of the PVK-PANI composite films may be due to the strong interaction between PANI and PVK in the composite films.
An alternative molecular porphyrin-phthalocyanine aggregate was prepared and characterized with UV-visible and X-ray absorption spectroscopies. UV-visible experiments evidence 1-dimensional porphyrin-phthalo-cyanine array formed by mixing $SnTPPCl_2 ({\lambda}_{max}=429,\;{\varepsilon}=2.4{\times10^ 5 /M{\cdot}cm)\;and\;NiPc(OBu)_8({\lambda}_{max}=744 nm,\;{\varepsilon}= 2.0{\times}10^ 5 /M{\cdot}cm)$ in solution. In the UV-visible spectrum of the porphyrin-phthalocyanine array, $(SnPNiPc)_n$, a new Q-band appeared at 844 nm with decrease of the Q-band peak of $NiPc(OBu)_8$ at 744 nm. The red-shift of Q-band evidences an alternative porphyrin-phthalocyanine array formed in solution through metal-halide interaction rather than ${\pi}-{\pi}$ facial interaction, in which nickel of $NiPc(OBu)_8$ coordinates with chloride of $SnTPPCl_2$ through self assembly. Ni K-edge XANES (X-ray absorption near edge structure) spectra also support the axial ligation of nickel to chloride. The square planar structure of $NiPc(OBu)_8$ turns to an octahedral structure in (SnPNiPcSnP) by axial ligation. A higher energy-shift (0.2 eV) of the preedge peak of (SnPNiPcSnP) indicaties partial oxidation of nickel by charge transfer from NiPc$(OBu)_8$ to SnTPPCl$_2$.

This experiment was to study the effects of application rates of mineral N and cattle slurry on the dry matter yield of Orchardgrass grown in different cutting frequency. Annual rates of mineral N of 0($N_0$), 90kg($N_1$), 180kg($N_2$) and 270kg/ha($N_3$) in 3 cuttings, and 0($N_0$), 120kg($N_1$), 240($N_2$) and 360kg/ha($N_3$) were applied as urea in 4 cuttings. Cattle slurry applied at rates of $30m^3(S_1)$, $60m^3(S_2)$ and $90m^3/ha(S_3)$, suppling 90kg, 180kg and 240kg N/ha in 3 cuttings, and at rates of $40m^3(S_1)$, $80m^3(S_2)$ and $120m^3/ha(S_3)$, suppling 120kg, 240kg and 360kg N/ha in 4 cuttings, respectively. The results were summarized as follows; 1. Mineral N and cattle slurry application rates(N), and cuts(C) were significant differences at the 0.1% level(p<0.001), and 5% level(p<0.05) for the interaction of $N{\times}C$ in both cuttings. 2. The annual dry matter yields obtained were 8.8ton-10.1ton/ha at rates of $N_1-N_3$, and 7.1ton-9.5ton/ha at rates of $S_1-S_3$ in 3 cuttings. The annual dry matter yields obtained were 10.9ton-13.9ton/ha at rates of $N_1-N_3$, and 6.9ton-11.2ton/ha at rates of $S_1-S_3$ in 4 cuttings, respectively. 3. Relative efficiency of cattle slurry N for the dry matter production of Orchardgrass pasture as compared to mineral N were 91.4%(ranged from 72.2% to 109.7%) in 3 cuttings, and 75.1%(ranged from 48.3% to 107.9%) in 4 cuttings, respectively. 4. The annual cattle slurry application rates required to maintain highest dry matter yields were estimated to be 90m3/ha(270kg N/ha) and 80m3/ha(240kg N/kg) in 3 and 4cuttings.

Insulin-like growth factor binding protein 5 (IGFBP-5) plays an important role in controlling cell survival, differentiation and apoptosis. Apoptosis can be induced by an extrinsic pathway involving the ligand-mediated activation of death receptors such as tumor necrosis factor receptor 1 (TNFR1). To determine whether IGFBP-5 and TNFR1 interact as members of the same apoptosis pathway, recombinant IGFBP-5 and TNFR1 were isolated. The expression and purification of the full-length TNFR1 and truncated IGFBP-5 proteins were successfully performed in E. coli. The binding of both IGFBP-5 and TNFR1 proteins was detected by surface plasmon resonance spectroscopy (BIAcore), fluorescence measurement, electron microscopy, and size-exclusion column (SEC) chromatography. IGFBP-5 indeed binds to TNFR1 with an apparent $K_D$ of 9 nM. After measuring the fluorescence emission spectra of purified IGFBP-5 and TNFR1, it was found that the tight interaction of these proteins is accompanied by significant conformational changes of one or both. These results indicate that IGFBP-5 acts potently as a novel ligand for TNFR1.

The hepatitis C virus (HCV) internal ribosome entry site (IRES) is an RNA structure located in the 5'-UTR of the HCV RNA genome. The HCV IRES consists of four domains I, II, III, and IV, where domains II - IV are recognized by 40S ribosomal subunit and the domain III is bound to eukaryotic initiation factor 3 (eIF3) for translation initiation. Here, we have characterized the tertiary interaction between an L-/K- rich peptide and the HCV IRES domain IV. To probe the peptide binding interface in RNA, we synthesized $^{13}C$- and $^{15}N$-double labeled RNA and the binding site was identified by using the chemical shift perturbation (CSP) NMR methods. Our results showed that the peptide binds to the upper stem of the IRES domain IV, indicating that the tertiary interaction between the IRES domain IV and the peptide would disrupt the initiation of translation of HCV mRNA by blocking the start codon exposure. This study will provide an insight into the new peptide-based anti-viral drug design targeting HCV IRES RNA.

There is relatively little numerical study on the behavior of eccentrically loaded high strength rectangular concrete-filled steel tubular (CFST) slender beam-columns with large depth-to-thickness ratios, which may undergo local and global buckling. This paper presents a multiscale numerical model for simulating the interaction local and global buckling behavior of high strength thin-walled rectangular CFST slender beam-columns under eccentric loading. The effects of progressive local buckling are taken into account in the mesoscale model based on fiber element formulations. Computational algorithms based on the M$\ddot{u}$ller's method are developed to obtain complete load-deflection responses of CFST slender beam-columns at the macroscale level. Performance indices are proposed to quantify the performance of CFST slender beam-columns. The accuracy of the multiscale numerical model is examined by comparisons of computer solutions with existing experimental results. The numerical model is utilized to investigate the effects of concrete compressive strength, depth-to-thickness ratio, loading eccentricity ratio and column slenderness ratio on the performance indices. The multiscale numerical model is shown to be accurate and efficient for predicting the interaction buckling behavior of high strength thin-walled CFST slender beam-columns.
Repairing damaged DNA and removing all physical connections between sister chromosomes is important to ensure proper chromosomal segregation by contributing to chromosomal stability. Here, we show that the depletion of non-SMC condensin I complex subunit H (NCAPH) exacerbates chromosome segregation errors and cytokinesis failure owing to sister-chromatid intertwinement, which is distinct from the ultra-fine DNA bridges induced by DNA inter-strand crosslinks (DNA-ICLs). Importantly, we identified an interaction between NCAPH and GEN1 in the chromatin involving binding at the N-terminus of NCAPH. DNA-ICL activation, using ICL-inducing agents, increased the expression and interaction between NCAPH and GEN1 in the soluble nuclear and chromatin, indicating that the NCAPH-GEN1 interaction participates in repairing DNA damage. Moreover, NCAPH stabilizes GEN1 within chromatin at the G2/M-phase and is associated with DNA-ICL-induced damage repair. Therefore, NCAPH resolves DNA-ICL-induced ultra-fine DNA bridges by stabilizing GEN1 and ensures proper chromosome separation and chromosome structural stability.
Complement fragment iC3b serves as a major opsonin for facilitating phagocytosis via its interaction with complement receptors CR3 and CR4, also known by their leukocyte integrin family names, αMβ2 and αXβ2, respectively. Although there is general agreement that iC3b binds to the αM and αX I-domains of the respective β2-integrins, much less is known regarding the regions of iC3b contributing to the αX I-domain binding. In this study, using recombinant αX I-domain, as well as recombinant fragments of iC3b as candidate binding partners, we have identified two distinct binding moieties of iC3b for the αX I-domain. They are the C3 convertase-generated N-terminal segment of the C3b α'-chain (α'NT) and the factor I cleavage-generated N-terminal segment in the CUBf region of α-chain. Additionally, we have found that the CUBf segment is a novel binding moiety of iC3b for the αM I-domain. The CUBf segment shows about a 2-fold higher binding activity than the α'NT for αX I-domain. We also have shown the involvement of crucial acidic residues on the iC3b side of the interface and basic residues on the I-domain side.
The goal of this study was to analyze the effects of genistein, a widely used tyrosine kinase inhibitor, on cloned Shaw-type $K^+$ currents, Kv3.1 which were stably expressed in Chinese hamster ovary (CHO) cells, using the whole-cell configuration of patch-clamp techniques. In whole-cell recordings, genistein at external concentrations from 10 to $100{\mu}M$ accelerated the rate of inactivation of Kv3.1 currents, thereby concentration-dependently reducing the current at the end of depolarizing pulse with an $IC_{50}$ value of $15.71{\pm}0.67{\mu}M$ and a Hill coefficient of $3.28{\pm}0.35$ (n=5). The time constant of activation at a 300 ms depolarizing test pulses from -80 mV to +40 mV was $1.01{\pm}0.04$ ms and $0.90{\pm}0.05$ ms (n=9) under control conditions and in the presence of $20{\mu}M$ genistein, respectively, indicating that the activation kinetics was not significantly modified by genistein. Genistein $(20{\mu}M)$ slowed the deactivation of the tail current elicited upon repolarization to -40 mV, thus inducing a crossover phenomenon. These results suggest that drug unbinding is required before Kv3.1 channels can close. Genistein-induced block was voltage-dependent, increasing in the voltage range $(-20\'mV{\sim}0\'mV)$ for channel opening, suggesting an open channel interaction. Genistein $(20{\mu}M)$ produced use-dependent block of Kv3.1 at a stimulation frequency of 1 Hz. The voltage dependence of steady-state inactivation of Kv3.1 was not changed by $20{\mu}M$ genistein. Our results indicate that genistein blocks directly Kv3.1 currents in concentration-, voltage-, time-dependent manners and the action of genistein on Kv3.1 is independent of tyrosine kinase inhibition.
