Cellulose Structures of Primary and Secondary Tissues in Pinus densiflora S. et Z. (소나무재의 1차조직과 2차조직 세포벽 중의 셀룰로오스 구조)

  • The microscopic characteristics and cellulose structures of primary and secondary tissues in Pinus densiflora S. et Z. were examined. Cells of primary tissue in cross section showed an irregular arrangement and round shape. Fiber lengths were 200 to $250{\mu}m$ in primary tissue, and 1,500 to $1,600{\mu}m$ in secondary tissue. Cell diameters in primary tissue were larger than those in secondary tissue; 40 to $50{\mu}m$ in former and 10 to $20{\mu}m$ in latter. Crystallite width and d-spacing of (200) in both tissues did not show any significant differences. However, crystallinity indices by Segal's method showed significant differences as 23% in primary tissue and 35% in secondary tissue. In the orientation of cellulose microfibril, primary tissues had a random pattern, whereas, secondary tissues presented an oriented pattern with 20 to 30 degree. The cellulose crystalline of primary tissue was easily transformed into cellulose II by mercerization, but that of secondary tissue hardly transformed. It is considered that the difference of crystal transformation in both tissues could be caused by the difference of lignification.

Plant Tissue Sensor for Determination of Cytidine (시티딘 정량을 위한 식물조직 센서)

  • Rose tissue containing cytidine deaminase converts cytidine to uridine and ammonia gas. Rose tissue sensor was constructed by immobilizing 50mg of a rose petal tissue on an NH3 gas sensor and the optimum condition of the sensor for the determination of cytidine was investigated. The tissue sensor showed a linear range of$7.0 {\times} 10^{-4}$$1.0{\times} 10^{-2}$M cytidine with a slope of 53 mV/decade in 0.2 M phosphate buffer, pH 8.4 at 37$^{\circ}C$. The detection limits were $3.0{\times}10^{-4}$ M and relative standard deviation was 3.4%. This sensor showed an excellent selectivity among various nucleosides and amino acids.

  • A biosensor for the measurement of L-glutamine has been constructed by immobilizing the slice of Wistar rat kidney and it's organelle on $NH_3$ gas-sensing electrode. The effects of pH, buffer solution, temperature and thickness of slice were investigated in order to optimize electrode response. The tissue sensor had the linearity in the range of L-glutamine concentration $8.0{\times}10^{-5}{\sim}1.0{\times}10^{-2} M$ with a slope of 53.8 mV/decade in 0.05 M phosphate buffer solution, pH 7.8 at $30^{\circ}C$, and optimum thickness of slice and response time were 30 ${\mu}m$ and 3∼5 min, respectively. The organelle sensor showed the linearity within L-glutamine concentration range of $1.2{\times}10^{-4}{\sim}5.0{\times}10^{-3} M$ with a slope of 54.0 mV/decade in 0.05 M phosphate buffer solution, pH 7.8 at $30^{\circ}C$, and response time was 6∼7 min, respectively. Thus, it is clear that the tissue and organelle sensor will be useful for L-glutamine measurements.

Relationship between Monounsaturated Fatty Acid Composition and Stearoyl-CoA Desaturase mRNA Level in Hanwoo Liver and Loin Muscle (한우 간 및 등심 조직에서 불포화지방산의 조성비율과 Stearoyl-CoA Desaturase mRNA의 발현 양상)

  • The Stearoyl-CoA Desaturase(SCD) is a key enzyme, which converting palmitic acid(16:0) and stearic acid (18:0) to pahnitoleic acid(16:1) and oleic acid(l8:1), respectively. The concentration of oleic acid(18:1) in meat of beef cattle could influence both palatability and perception of meat. This experiment has conducted to determine relationship between the compositions of monounsaturated fatty acids and the SCD mRNA level in bovine liver and loin muscle tissue. The compositions of palmitoleic acid(16:1) and oleic acid(18:1) in loin muscle were 5% and 46% of total lipid and in liver were 2% and 20% of total lipid, respectively. On the other hand, the compositions of palmitic acid(16:0) and stearic acid(18:0) in loin muscle were 25% and 45% of total lipid and in liver were 14% and 43% of total lipid, respectively. The ratio of monounsaturated to saturated fatty acids(the desaturation index) was used as a measure of SCD activity in tissues. The average desaturation index in loin muscle was higher about 3.6-fold than that in liver. The desaturation index of oleate/stearate and palmitoleatelpalmitate in loin muscle were higher 8-fold and 1.8-fold than those in liver, respectively, showing that the substrate specificity of SCD enzyme was very different between liver and muscle tissues. To determine whether the composition of monounsaturated fatty acids in liver and muscle are dependent on SCD expression, SCD mRNA level was examined by RT-PCR analysis. The SCD mRNA level in loin muscle was higher about 3-fold than that in liver. Thus, the quantitative relationship between the desaturation index of fatty acid and SCD mRNA was observed in liver and muscle. The difference in the compositions of monounsaturated fatty acids between bovine liver and muscle tissues may be due to different level of Stearoyl-CoA Desaturase mRNA.

Determination of Urea using Rose Tissue Sensor (장미조직센서를 이용한 요소의 정량)

  • The rose petal tissue biosensor has been constructed by immobilizing New carina rose tissue. Optimum conditions for the determination of urea were investigated using this sensor. Selectivity and life time of this sensor were also obtained. As a result, the biosensor showed the optimum response characteristics in 0.20M phosphate buffer solution at pH 8.0, $37^{\circ}C$ and 50mg of tissue amounts. This sensor was linear from $9.0{\times}10^{-5}$ to $4.0{\times}10^{-3}M$ urea with a slope of 42mV/decade. The limit of detection and response time are $7.0{\times}10^{-5}M$ and 17~19 min.

Tissue Specific Expression of Wound-Inducible RCaM-2 Promoter in Transgenic Tobacco Plants (상처에 의해서 유도되는 벼 calmodulin promoter의 transgenic 담배에서조직 특이적 발현)

  • To study calmodulin (CaM) gene expression and its regulation, rice CaM promoter (RCaM-2) was isolated and fused to $\beta-glucuronidase$ (GUS), reporter gene. X-Glue staining patterns revealed that GUS localization is high in meristemic tissues such as the stem apex, stolen tip, and vascular regions. GUS staining in the transverse sections of stem and petiole was restricted to the inside of the vascular system, and cortex and epidermis located outside of the vascular system usually did not show GUS staining even a plant that expressed strong activity. GUS activity was found to be tissue specific expressed and exhibited a dramatic transient increase in response to wounding. These results suggest that the 5'-flanking region of RCaM gene regulates wound-inducible expression.

Lactate Dehydrogenase and Monocarboxylate Transporters 1, 2, and 4 in Tissues of Micropterus salmoides (큰입우럭(Micropterus salmoides) 조직의 젖산탈수소효소 및 Monocarboxylate 수송체(MCT) 1, 2, 4)

  • The properties of lactate dehydrogenase (EC 1.1.1.27, LDH) and expression of monocarboxylate transporters (MCTs) 1, 2, and 4 were studied in tissues from Micropterus salmoides. Native-PAGE revealed that the LDH $A_4$ isozyme was predominantly located in skeletal muscle. The LDH $A_4$, $A_2B_2$, and $B_4$ isozymes were detected in heart, liver, eye, and brain tissues, while eye-specific $C_4$ isozyme was detected in eye tissue. In September, strong LDH $B_4$ isozyme activity was detected in heart tissue. High $A_4$ isozyme activity was noted in all other tissues except heart tissue. However, in November, strong $A_4$ isozyme activity was detected in heart tissue. The LDH/CS (Citrate synthase, EC 4.1.3.7) ratio in skeletal muscle and heart tissues indicated that anaerobic metabolism was high in those tissues. Native-PAGE after immunoprecipitation showed that eye-specific $C_4$ isozyme was more similar to the $A_4$ than the $B_4$ isozyme. The LDH $A_4$ isozyme was purified by affinity chromatography. The molecular weight of subunit A was 37,200. The LDH activity in tissues was consistently 11.05~28.32% due to inhibition by 10 mM pyruvate. The $K_m^{PYR}$ of LDH in eye tissue was very low. The optimum pH for LDH in tissues was pH 7.5~8.0. The LDH $A_4$ isozyme was detected in mitochondria of skeletal muscle, whereas the $B_4$ and $A_2B_2$ isozymes were detected in heart tissue mitochondria. Western blot analysis indicated that MCTs 1, 2, and 4 were located in the plasma membrane and mitochondria of skeletal muscle and heart tissues. The sizes of MCTs 1, 2, and 4 in skeletal muscle were 60, 54~38, and 63 kDa, while those in heart tissue were 57, 54~38, and 55.5 kDa, respectively. In conclusion, M. salmoides appears to use anaerobic metabolism predominantly when adapted to a hypoxic environment. In highly activated skeletal muscle and heart tissue, energy production is controlled by inward and outward flows of pyruvate and lactate through MCTs 1, 2, and 4 in the plasma membrane and mitochondria, with effective adjustment by LDH isozymes.

Changes of LDH Subunit Combinations Induced by Tetrodotoxin (Tetrodotoxin에 의하여 유발되는 LDH 하부단위체 조합의 변화)

  • In an attempt to make a scrutiny into a mechanism for the formation of quaternary structure of LDH isozymes, male mice were intraperitoneally exposed to a wide range of tetrodotoxin concentration and the changes in relative percentage of the five isozymes were monitored by electrophoresis and subsequent densitometry. The observations that a conspicuous increase of the $H_4$ isozyme was demonstrated in nearly all brain tissues, that the $M_4$ isozyme of skeletal muscle tissue was slightly increased while the $M_3H$ and $M_2H_2$ isozymes were decreased, that the M/H ratio was strikingly reduced in heart tissue and that assembly of $H_4$ isozyme was revealed in liver tissue although its rate was extremely low suggest that new intracellular ionic environment established by compulsory change in Donnan equilibrium could alter the LDH isozyme distribution. The reduction of assembly of $M_3H$ isozyme found in mouse tissues exposed to tetrodotoxin also seems to suggest that the subunit combination of 3M+H is very unfavorable in an intracellular environment deviated from its accustomed one. It was reaffirmed that there might occur TTX-insensitive sodium channels in plasma membrane.

In Pediatric Leukemia, Dose Evaluation according to the Type of Compensators in Total Body Irradiation (소아백혈병 환자의 전신방사선조사 시 조직보상체의 재질변화에 따른 선량평가)

  • Total body irradiation(TBI) and chemotherapy are the pre-treatment method of a stem cell transplantations of the childhood leukemia. in this study, we evaluate the Quantitative human body dose prior to the treatment. The MCNPX simulation program evaluated by changing the material of the tissue compensators with imitation material of pediatric exposure in a virtual space. As a result, first, the average skin dose with the material of the tissue compensators of Plexiglass tissue compensators is 74.60 mGy/min, Al is 73.96 mGy/min, Cu is 72.26 mGy/min and Pb 67.90 mGy/min respectively. Second, regardless of the tissue compensators material that organ dose were thyroid, gentile, digestive system, brain, lungs, kidneys higher in order. Finally, the ideal distance between body compensator and the patient were 50 cm aparting each other. In conclusion, tissue compensators Al, Cu, Pb are able to replace of the currently used in Plexiglass materials.

Preparation and Characterization of Sponge Using Porcine Small Intestinal Submucosa (돼지의 소장 점막하 조직을 이용한 스폰지의 제조 및 특성 결정)

  • Porcine small intestine submucosa (SIS) has been widely used as a biomaterial without immunorejection responses. Crosslinked SIS sponges were characterized for the possibility of the bio-interactive wound dressings and tissue engineered scaffolds. SIS powders were dissolved in 3% acetic acid aqueous solution at 48hrs followed by pouring into mold and then fabricated by freeze-drying method. SIS sponge was prepared by crosslinked with 1-ethyl-(3-3-dimethyl aminopropyl) carbodiimide hydrochloride (EDC) solution (deionized water: ethanol=5:95) with 1-100mM concentration for 24 hrs and Iyophilized. SIS sponges were characterized by scanning electron microscopy, differential scanning calorimeter, and Fourier transform infrared spectrometer and were tested their porosity and water absorption ability. It was observed that the concentration of EDC might be exceeded 50 mM to get good physical characteristics. In conclusion, it seems that SIS sponge could be very useful for the applications of wound healing and tissue construction.
