Characteristics of $G_{418}$-sensitive mitochondrial ATPase/ATP synthase from pleurotus florida (사철느타리버섯 중 $G_{418}$-sensitive 미토콘드리아성 ATPase/ATP synthase의 특성)

  • The mitochondrion was purified at 44% sucrose layer from pleurotus florida by using ultracentrifuge and sucrose density gradient method. Optimum pH and temperature of ATPase and ATP synthase were pH 7.4, $60^{\circ}C$ and pH 7.5, $57^{\circ}C$ respectively, also their Km values were determined as 11.6mM and 8.4mM. ATPase was activated at 5~6mM ATP substrate concentration, then ATP synthase was 5~10mM range ADP. ATPase/ATP synthase were $Mg^{2+}$-dependent enzyme, partially inhibited by their substrate, and then showed an none competitive inhibition pattern by $G_{418}$. Amino acid composition of ATPase/ATP synthase was as follows, hydrophobic amino acid residue was 50.5%, small residue, 56.1%, hydrogen bonding residue, 43.7% and helix breaking residue, 55.2%. Phosphatidyl choline, phosphatidyl ethanolamine and phosphatidyl glycerol were contained but not phosphatidyl inositol and phosphatidyl serine. Palmitate(51.31%), stearate(18.32%) and unsaturated fatty acids($C_{18:1}$, $C_{18:2}$ and $C_{16:1}$) were predominated.

A Study on the Characteristics of Flow Fields in a Coal Gasifier (석탄 가스화기내의 유동장 특성 연구)

  • In this study numerical computations were performed to predict reacting flow fields of gasification processes of pulverized subbituminous coal in a cylindrical coal gasifier. To check the size effects of particles on flow fields in the gasifier, simulations were performed for five cases with four sizes of particles such as 40 $\mu\textrm{m}$, 60 $\mu\textrm{m}$, 80 $\mu\textrm{m}$ and 100 $\mu\textrm{m}$. Each case has a unique size of particles with one more case that has evenly mixed four sizes of particles. Predictions showed that the gasification which uses coals of the mixed sizes reveals more preferable gas velocity and temperature distributions than that uses coals of a unique size. Predicted gas temperature at the exit of the gasifier ranged 1,400 to 1580$^{\circ}C$, 1,480 to 1,700$^{\circ}C$, 1,600 to 1740$^{\circ}C$, 1630 to 1790$^{\circ}C$ and 1500 to 1680$^{\circ}C$ for particle sizes of 40 $\mu\textrm{m}$, 60 $\mu\textrm{m}$, 80 $\mu\textrm{m}$, 100 $\mu\textrm{m}$ and the evenly mixed four sizes, respectively.

  • Background : Surfactant protein B(SP-B) and surfactant protein C(SP-C) are important in accelerating surface spreading of surfactant phospholipid. The glucocorticoids accelerate the morphologic differentiation of epithelial cells into type II cells and increase the rate of phosphatidylcholine synthesis. The hydrophobic surfactant protein has been shown to be upregulated by glucocorticoids in vitro, however, its regulation in vivo is not well established. Methods : The authors investigated the effects of glucocorticoid on the accumulation of mRNA encoding SP-B and SP-C protein content of the lung. Adult rats were given different doses of subcutaneous dexamethasone and sacrificed at 24 hours and 1 week. SP-B and SP-C mRNA were measured by a filter hybridization method. Results : 1) The accumulation of SP-B mRNA at 24 hours after 0.2 mg/kg dexamethasone treatment was increased by 23.7%. 2) The accumulation of SP-B mRNA at 1 week after 2 mg/kg dexamethasone treatment was significantly increased by 96.6%(P<0.001). 3) The accumulation of SP-C mRNA at 24 hours after 0.2 mg/kg dexamethasone treatment was significantly increased by 42.7%(P<0.01). 4) The accumulation of SP-C mRNA at 1 week after 2 mg/kg dexamethasone treatment was significantly increased by 60.0% (P<0.01). Conclusion : The authors concluded that dexamethasone treatment in vivo resulted in increased levels of SP-B mRNA and SP-C mRNA. These results suggested that dexamethasone stimulates the synthesis of hydrophobic proteins associated with surfactant.

Comparative Quantitative Study of Surfactant Protein C mRNA by Filter Hybridization and Solution Hybridization in Rats (Filter Hybridization과 Solution Hybridization 방법에 의한 백서 Surfactant Protein C mRNA 정량측정의 비교)

  • Background : Surfactant protein C(SP-C) is a hydrophobic 5,000 dalton molecule. SP-C has the primary roles in accelerating surface spreading of a surfactant phospholipid. The filter hybridization and solution hybridization assays are both rapid and sensitive and can be used to measure the RNAs complementary to any cloned DNA sequence. Methods : The authors measured the SP-C mRNA levels quantitatively using solution hybridization and filter hybridization assays to obtain a standard curve equation to quantify the mRNA of unknown samples comparatively. Results : 1. The minimum level of the specimens by solution hybridization was 3 pg for SP-C mRNA. 2. The standard curve equation of the solution hybridization assay between the counts per minute(Y) and the SP-C mRNA transcript input(X) was Y=6.46 X+244. The correlation coefficient was 0.99. 3. The minimum detection level of specimens by filter hybridization was 0.1 ng for SP-C mRNA. 4. The standard curve equation of the filter hybridization assay between the counts per minute(Y) and SP-C mRNA transcript input(X) is Y=2541.6 X+252.7. The correlation coefficient was 0.99. Conclusions : A comparison of CPM/filter in the linear range allowed an accurate and reproducible estimation of the SP-C mRNA copy number. Filter hybridization and solution hybridization assays are both rapid and sensitive and can be used to measure the RNAs complementary to any cloned DNA sequence. It is ideally suited to situations where accurate quantitation of multiple samples is required.

  • Peroxidase in the fruit of Malus sieboldii (Regel) Rehder was partially purified by DEAE-cellulose column chromatography and Ultro-AcA 54 gel filtration. The optimum pH of peroxidase was 4.5 and optimum temperature was $80^{\circ}C$. The enzyme was stable at pH 5.0 and below $30^{\circ}C$, and inactivated by heat treatment at $80^{\circ}C$ for 15min. In the presence of 30mM $H_{2}O_2$ Km value on o-phenylenediamine as substrate was 1.65mM, and in the presence of 10mM o-phenylenediamine Km value on $H_{2}O_2$ was 7.97mM. L-Ascorbic acid and sodium L-ascorbate greatly inhibited the enzyme activity and among several metal ions $Mn^{2+}$ only increased the activity at 5mM.

Dealumination of $NH_4Y$-Zeolite to Convert to the Hydrophobic Zeolite by High-Temperature Steam Treatment (소수성 제올라이트로의 변환을 위한 고온 수증기처리에 의한 $NH_4Y$-제올라이트의 탈알루미늄)

  • This study was performed to change the hydrophilic $NH_4Y$-zeolite to the hydrophobic one for removal of VOCs by removing the $Al^{3+}$ in the zeolite-structure to increase the Si/Al ratio, for which the three pelleted $NH_4Y$-zeolite samples were contacted separately with the steam of $400^{\circ}C$, $500^{\circ}C$ and $600^{\circ}C$, respectively, in a stainless steel column for 4 hours. Then extraction of the ex-structure aluminum of the hydrolyzed zeolites with the nitric acids of 0.25, 0.50, 0.75, and 0.10 M at $90^{\circ}C$ in 500 mL-flasks, respectively, according to steam temperature were followed. XRD analysises of the dealuminated zeolites showed that the peaks of the zeolites that had been hydrolyzed with the steams of both $500^{\circ}C$ and $600^{\circ}C$ are distorted more with the increase of the concentration of nitric acid used for extraction of the ex-structure aluminums, however, those hydrolyzed with steam of $400^{\circ}C$ became amorphous phase when treated with the all nitric acids of four concentrations. Also the EDX analysises showed that the BET surface areas and TPVS of the zeolites that had been hydrolyzed with the steam of $600^{\circ}C$ were increased with the concentration of the nitric acid when the nitric acids of 0.25 M and 0.5 M had been used but decreased when the nitric acids of 0.75 M and 1.0 M had been used. These results led to the conclusion that both the $600^{\circ}C$ and $500^{\circ}C$-steam and the 0.5 M-nitric acid are appropriate to change the hydrophilic $NH_4Y$-zeolites to the hydrophobic one, which were proven by the measurement of the benzene and tolune-adsorbing capacities showing the same trend as the BET surface area and TPV The Si/Al ratios and water-adsorbing capacities of the dealuminated zeolites were increased and decreased, respectively, with the concentration of the nitric acids so that it showed that the hydrophobicity is increased.

  • COMS DATS C&M software is an integrated management system providing control and monitoring functionalities for COMS IDACS (Image Data Acquisition and Control System). DATS C&M S/W consists of a system management module, a control and monitoring module, a data management module, and a trend analysis module. COMS SOC is supposed to operate IDACS as a backup of MSC. Especially, for the backup operation, the control and monitoring module of DATS C&M S/W is designed to support the synchronization of the two IDACS systems. This paper describes design, implementation, and result of development of DATS C&M S/W.

  • Coastal Zone is a integrated space including both coastal waters and inland. C.Z.M is a strategic practice system which can integrate and coordinate the coastal zone for preserving and making use of coastal resources. Through foreign and domestic C.Z.M Cases, this study analyzes the operating mechanism in the three aspects: role sharing among the administrative agencies, mediation and cooperation among the interested parties, and inducements of voluntary participation. And it achieves the common characteristics of C.Z.M: building the assessment system for establishing desirable policy directions, the stable support system for sustainable coastal management, the mediation system for cooperation of stakeholder, and the education system for reinforcing the management ability and inducing the voluntary participation. Finally it suggests the implications to build the coastal zone management system properly on the basis of common values in this mechanism.

Numerical study of strength reduction-induced capillary rise effect for unsaturated soil

  • Previous studies postulated insignificant capillary rise (hc) effect above the water table (Hw) for unsaturated soils. In addition, these studies utilised dry unit weight above Hw. This paper, therefore, addresses the effect of these postulations on strength where the influence of hc using a modified upper bound approach, Discontinuity Layout Optimization (UNSAT-DLO) for a simulated soil was predicted. Two different parametric studies to model passive earth pressure and bearing capacity problems are carried out to provide an insight into the effect of capillary rise on strength. Significant increase in strength, owing to unsaturated conditions, was obtained where the maximum increase was when suction slightly less or greater than the air entry suction. On the other hand, the results showed a negative effect of hc. For example, up to 8.24% decrease in passive thrust (Pp) was obtained at Hw=0 m when hc rose 1 m from 0 m. To put this into perspective, this was equivalent to a decrease of about 2° in 𝜙 at Hw=0 m and hc =0 m in order to obtain the same result at hc =1 m. For the bearing capacity problem, the effect was seen to be higher, up to 18.4% decrease in N𝛾 was obtained when hc rose from 0 m to 2.5 m at Hw =0 m. In addition, the results revealed a negative influence of assigning dry unit weight above Hw or hc.. However, considerable increase in strength was obtained when unsaturated unit weight above hc was assigned.

Effect of M.A.P. and C.A. storage on quality of Mushrooms(Agaricus bisporus) during storage (양송이 버섯의 MAP및 CA저장 효과)

  • In order to study the effect of modified atmosphere packaging(M.A.P.) and controlled atmosphere(C. A.) storage on keeping freshness of mushrooms (Agricus-bisporus). Mushrooms was packaged with polyethylene(P.E.) film (40${\mu}{\textrm}{m}$, 60${\mu}{\textrm}{m}$, 80${\mu}{\textrm}{m}$) and C.A. conditions(CO2 concentration of 2%, O2 concentration of 2%) and storage at 0$\pm$1$^{\circ}C$, RH(relative hummidity) 92$\pm$1%. Gas composition in film was changed rapidly at early of storage, but it kept a constant level after 14 days of storage, and then kept at the level of 6~9% CO2 and 2~5% O2. Weight loss was 5% in non-packed mushrooms after 3 days of storage, but P.E. film, CO2 treated, C.A. conditions were 5% after 21 days of storage. Flesh firmness of the mushrooms was continuously decreased throughout storage period and the lowest of flesh firmness changed was 80${\mu}{\textrm}{m}$-P.E. film packing. Discoloration of the piteous of mushrooms appears to be the most Important factors to determine its marketability, L value of it appears to be of the high values at P.E. film packing and C.A. conditions during period. Large amount of ethanol and acetaldehyde were produced from the 7 days during storage, large contents of mannitol and trehalose were at the 14 days and 7 days during storage.
