Photosensitized Lysis of Egg Lecithin Liposomes by L-Tryptophan and N-Acetylphenylalanyl-L-Tryptophan

  • The photosensitized lysis of egg lecithin lipid membranes (liposomes) have been performed to UV-B light (270-320 nm) by L-tryptophan(L-Trp) and its peptide such as N-acetylphenylalanyl-L-tryptophan(NAPT) incorporated in the liposomes(ca. 0.1% by weight) or in the external buffer (0.1-0.3 mM). Requirement of oxygenation suggests that the lysis of liposomes is caused by the photosensitized oxidation of lipids. There was significant protection against lysis photosensitized by Trp in the external buffer by low concentration of ferricyanide (0.8 mM), but there was no effect on the lytic efficiency by $N_3^-$ which is singlet oxygen($^1O_2$) quencher, indicative of an electron transfer mechanism involved in the photosensitization. The small change of the lytic efficiency with increasing pH from 4 to 9 was interpreted by large target theory and subsequently indicates that superoxide($O_2^-$) may be an active intermediate for the oxidation. The efficiency of photosensitization of Trp was higher than that of NAPT under the same experimental condition. The weak lytic efficiency of liposomes photosensitized by NAPT was enhanced by incorporating NAPT in liposomes, but it was again quenched by ${\beta}$-carotene incorporated in the bilayer of liposomes. These results indicate that a portion of liposome lysis may be due to $^1O_2$ formation from the excited NAPT.

Improvement of the Detection Technique of Listeria monocytogenes through Modification of the Enrichment Medium and DNA Extraction Buffer (증균배지 및 DNA 추출법 개량을 통한 Listeria monocytogenes의 검출기법 개선 연구)

  • In this study we developed an enrichment medium and lysis buffers to detect Listeria monocytogenes in meat and processed meat products under various lysis conditions. The enrichment efficiency of L. monocytogenes medium listed in the Food Standards was compared, and thus, Listeria Enrichment Broth (LEB) was modified by adding supplements such as carbon source and minerals. The lysis buffers were developed to extract L. monocytogenes DNA quickly and efficiently under various lysis conditions. L. monocytogenes was most rapidly grown in LEB containing 0.1% pyruvate and 0.1% ferric citrate. A lysis buffer mixed with 0.5% or 1% N-lauroylasrcosine sodium salt, 0.5 N NaOH and 0.5 M EDTA for 30 min at room temperature was found to be the best in terms of DNA purity and yield. These results indicate that developed enrichment medium and lysis buffer can be used to detect L. monocytogenes in meat and processed meat products rapidly and efficiently.

  • Total RNA was isolated from two months old wheat, rice, tobacco and sweet potato. The procedure used was simple and provided pure RNA preparation. Lysis of plant tissue in a buffer with guanidine thiocyanate and CsCl density gradient centrifugation separated RNA from the rest of the cellular components. Subsequent cholroform/1-butanol extraction and ethanol precipitation were necessary to ensure contaminant-free RNA preparation. Storage of the lysed plant tissue in the buffer with guanidine thiocyanate preserved the sample for two months without noticeable RNA degradation.

  • Background: The polymerase chain reaction (PCR) is a very sensitive method for the detecting of mycobacterial DNA. There are many reports revealing the efficacy of PCR for the diagnosis of M. tuberculosis, but there are many different methods for DNA extraction from Mycobacterium tuberculosis. Bead beater method is a very useful method for DNA extraction from clinical spectimens, but its procedures are relatively complicated and time-consuming. So we studied other methods for the DNA extraction from Mycobacterium tuberculosis $H_{37}Rv$ and some clinical specimens (5 smear positive sputa and 5 smear negative CSF). Method: We extracted the mycobacterial DNA with 6 different methods from H37Rv strain and clinical specimens. The methods included SDS-microwave oven method, NaOH lysis method, Triton X-100-Proteinase K method, Lysis buffer method, SDS-proteinase K method and bead beater method. The target DNA was 123bp of IS6110 and was detected by examination of ethidium bromide-stained agarose gels. Results: Among 6 methods, SDS-proteinase K method, bead beater method, lysis buffer method and triton X-100-proteinase K method were excellent, but SDS-proteinase K method was the best method in the aspect of simplicity and cost-effectiveness. Conclusion: We suggest that SDS-porteinase K method is a simple and convinient method and might be the best method for the extraction of mycobacterial DNA.

  • The purpose of this study was to evaluate bacteriocin activity against human flora. Lactobiocin, a bacteriocin produced by Lactococcus sp. HY 449, inhibited the growth of Starphylococcus epidermidis, Starphylococcus aureus, Streptoccoccus pyogenes and Propionibacterium acnes. When crude bacteriocin was added to indicator cells during logarithmic growth, the optical density(O.D 650nm) of cells without bacteriocin increase after 5h of incubation. Whereas in the presence of bacteriocin, the O.D of cell suspensions decreased. The similar patterns were observed for absorbance readings at 280 nm and 260 nm. The release of cellular components when cell were treated with Lactobiocin suggests some degree of membrane damage or cell lysis. Scanning electron microscopy of cells following treatments with Lactobiocin in PBS buffer revealed disruptures of cell morphology. These results indicate that bacteriocin appears to cause cell lysis of tested strains. In cytotoxicity on human fibroblast, LD$\_$50/ of Lactobiocin was ca. 50 mg/ml and no change was observed cell proliferation at the same concentration. Any irritation and allergic reaction did not observed when evaluated by human patch test for Lactobiocin.

New Protein Extraction/Solubilization Protocol for Gel-based Proteomics of Rat (Female) Whole Brain and Brain Regions

  • The rat is an accepted model for studying human psychiatric/neurological disorders. We provide a protocol for total soluble protein extraction using trichloroacetic acid/acetone (TCA/A) from rat (female) whole brain, 10 brain regions and the pituitary gland, and show that two-dimensional gel electrophoresis (2-DGE) using precast immobilized pH (4-7) gradient (IPG) strip gels (13 cm) in the first dimension yields clean silver nitrate stained protein profiles. Though TCA/A precipitation may not be "ideal", the important choice here is the selection of an appropriate lysis buffer (LB) for solubilizing precipitated proteins. Our results reveal enrichment of protein spots by use of individual brain regions rather than whole brain, as well as the presence of differentially expressed spots in their proteomes. Thus individual brain regions provide improved protein coverage and are better suited for differential protein detection. Moreover, using a phosphoprotein-specific dye, ingel detection of phosphoproteins was demonstrated. Representative high-resolution silver nitrate stained proteome profiles of rat whole brain total soluble protein are presented. Shortcomings apart (failure to separate membrane proteins), gel-based proteomics remains a viable option, and 2-DGE is the method of choice for generating high-resolution proteome maps of rat brain and brain regions.

Korean Paddy Soil Microbial Community Analysis Method Using Denaturing Gradient Gel Electrophoresis (Denaturing gradient gel electrophoresis를 이용한 한국의 논 토양 미생물 다양성 분석 방법)

  • Soil microbes are important integral components of soil ecosystem which have significant and diverse role in organic matter decomposition, nitrogen cycling, and nitrogen fixation. In this study an effective denaturing gradient gel electrophoresis (DGGE) method was employed for paddy soil microbial diversity survey. For optimum paddy soil microbial DNA extraction, different methods such as Lysis buffer, skim milk bead, sodium phosphate buffer, Epicentre Soil Master DNA extraction kit (Epicentre, USA) and Mo Bio Power Soil DNA kit (MO BIO, USA) methods were utilized. Among all the method, using Mo Bio Power Soil kit was most effective. DGGE analysis of Bacteria was carried out at 6% polyacylamide gel and 45-60% denaturing gradient in the optimal conditions. Whereas DGGE analysis of fungi was done at 6% polyacrylamide gel and 45-80% denaturing gradient in the optimal conditions. By applying the above assay, it was found that variation within the microbial community of paddy soil occurs by a factor of time. DGGE assay used in this study through for a variety of soil microbial analysis suggests the potential use of this method.

  • Polymerase chain reaction (PCR) is a powerful technique in molecular biology and is widely used in various fields. By amplifying DNA fragments, PCR has facilitated gene cloning procedures, as well as molecular genotyping. However, the extraction of DNA from samples often acts as a limiting step of these reactions. In particular, the extraction of PCR-compatible genomic DNA from higher plants requires complicated processes and tedious work because plant cells have rigid cell walls and contain various endogenous PCR inhibitors, including polyphenolic compounds. We recently developed a novel solution, referred to as AnyDirect, which can amplify target DNA fragments directly from whole blood without the need for DNA extraction. Here, we developed a simple lysis system that could produce an appropriate template for direct PCR with AnyDirect PCR buffer, making possible the direct amplification of DNA fragments from plant leaves. Thus, our experimental procedure provides a simple, convenient, non-hazardous, inexpensive, and rapid process for the amplification of DNA from plant tissue.

Control Effect of Dinoflagellate Bloom by Powder of Marine Rock and Fungus Culture Supernatant (해양암석 분말과 곰팡이 배양액에 의한 적조생물 편조류의 구제효과)

  • To see effect of marine rock powder and fungal culture supernatant, we analyzed the biodegradation rates of harmful marine dinoflagellate, Heterosigma akashiwo and Prorocentrum minimum for developing the effective control methodology of algal bloom. Relatively low removal rates were observed in the treatment of marine rock powder or buffer solution alone. However, the lysis of H. akashiwo and P. minimum was enhanced in the combined treatments of marine rock powder with fungal supernatant. The effective concentration and exposure time of fungal supernatant for the lysis of H. akashiwo and P. minimum were 5 ml/l and 30 minutes, respectively. These results suggest that the fungal supernatant may be a biocontrol agent for the control of algal blooms in seawater.

  • To detect proper lytic assay conditions of the crude zumolyase from Arthrobacter luteus, effets of the various factors involved in the lytic system of Sacchromyces sake cultured with shaking in the malt extracts medium were investigated. The results are summarized as follows : 1. The susceptibilities of viable cells of S. sake from logarithmic growth phase to the lytic enzmye were much greater than those of the cells in lag and stationary phases. The cells cultured for 18 hr were the most susceptible to the enzyme. 2. Lytic activity of the enzyme toward the viable cells of S. sake was very low. It was, however, enhanced 4 folds of more by the pretreatment of the cells with 0.05 M sodium sulfite. 3. Lytic activity of the enzyme toward commercial baker's yeast cells was negligible, and the effect of sodium sulfite on the lysis of the cells also was nothing but a little. 4. The lyophilized cells of the baker's yeast showed more susceptibility to the lytic enzyme than viable cells of the yeast. No definite effect of sodium sulfite on the lysis of the lyophilized cells, however, was observed either baker's yeast of S. sake. 5. It appeared that the relationship between the reaction rate and the enzyme concentration on the lysis of the yeast cell walls followed enzyme kinetic theory, but one between the reaction rate and concentration of the yeast cells as substrates showed different pattern from that in enzyme kinetic theory. 6. After the preparation of crude zymolyase was kept at $7^[\circ}C$ for 10 days in the 0.05 M phosphate buffer, pH 7.5, the remainning lytic activity was about 80 %.
