카드뮴투여가 Balb/c 마우스의 면역반응에 미치는 영향 (Effect of Cadmium Chloride on the Immune Responses in Balb/c Mouse)

  • This study was designed to investigate the antibody production to sheep red blood cells(SRBC) and proliferation of mitogen-stimulated spleen cells in Balb/c mice which received cadmium chloride. The mice were divided into three independent groups which were one control and two experimental groups by the cadmium treatment or not. No specific treatment was done for the control group. One of two experimental groups, which is called 'pre-treatment group' in this paper, was subcutaneously injected with low dose of cadmium chloride(0.5 mg/kg/day) for 5 consecutive days before the primary SRBC immunization. The other called 'non-pretreatment group' was only pretreated with normal saline. Both experimental groups were intraperitoneally injected with high dose of cadmium chloride(5 mg/kg) 8 hours before the primary immunization. Mice were intraperitoneally immunized twice with 2% SRBC suspension containing $10^8$ cells. The results obtained were as follows, 1. The PFG responses to SRBC were significantly increased in two experimental groups, cadmium pretreatment and non-pretreatment compared with that of control group(p<0.05). 2. The total antibody titers to SRBC in cadmium treated groups were similar to that of control group, but titers of IgG antibody were significantly elevated(p<0.01). 3. The proliferation response of spleen lymphocytes to various mitogens was suppressed in proportion to the concentration of cadmium and the degree of cadmium accumulation in liver was increased in the cadmium treated groups. These results suggest that cadmium chloride could affect on mouse immune response, especially its cell mediated immune response could be decreased while its humoral immune response could be increased, which may not be influenced by the administration methods or pretreatment of cadmium to mouse.

시험관내에서 홍화의 물 추출물이 T 및 B 림프구의 활성에 미치는 영향 (Effect of Water Extract of Carthamus tinctorious L. on In Vitro Activity of T and B Lymphocytes)

  • Based on the traditional application of Carthamus tinctorious L. (CF) as a component of Korean medicinal decoctions, in the present study, we investigated in vitro an immunomodulatory activity of water extract of CF(WECF). Water extract of CF significantly increased the in vitro proliferative responses of spleen cells (SPC). However, addition of WECF during anti-CD3 activation resulted in a significant decrease in SPC proliferation. Flow cytometric analysis showed that WECF addition chanced T and B cell frequencies in anti-CD3-activated spleen cell populations. Using purified cells, it was revealed that WECF is mitogenic to B cells but rather inhibitory to T cell Proliferation. Upon anti-CD3 stimulation, high concentration (1 mg/ml) of WECF significantly inhibited T cell proliferation until day 2 of stimulation. At day 3, anti-CD3-activated cells exposed to WECF recovered their proliferation to the level comparable to control. Although B cell proliferation was also inhibited in proliferation at day 1, it recovered sooner and then was rather augmented by WECF at day 3. These data indicate that WECF down-regulates lymphocyte proliferation at early phase of activation but T cells are more vulnerable than B cells to WECF, However, CD4+ and CD8+ T cells did not differ in WECF-mediated immunotoxicity. Data of propidium iodide (PI) staining showed that WECF accelerates activated T cell, but not B cell, apoptosis and WECF concurrently inhibited cytokine production of activated T cells. Taken together, WECF exhibits B cell mitogenic activity and differential toxicity more pronounced to T cells, suggesting a possible in vivo application of WECF for specific control of T cells without alteration of B cell activity.

Adenine attenuates lipopolysaccharide-induced inflammatory reactions

  • A nucleobase adenine is a fundamental component of nucleic acids and adenine nucleotides. Various biological roles of adenine have been discovered. It is not produced from degradation of adenine nucleotides in mammals but produced mainly during polyamine synthesis by dividing cells. Anti-inflammatory roles of adenine have been supported in IgE-mediated allergic reactions, immunological functions of lymphocytes and dextran sodium sulfate-induced colitis. However adenine effects on Toll-like receptor 4 (TLR4)-mediated inflammation by lipopolysaccharide (LPS), a cell wall component of Gram negative bacteria, is not examined. Here we investigated anti-inflammatory roles of adenine in LPS-stimulated immune cells, including a macrophage cell line RAW264.7 and bone marrow derived mast cells (BMMCs) and peritoneal cells in mice. In RAW264.7 cells stimulated with LPS, adenine inhibited production of pro-inflammatory cytokines $TNF-{\alpha}$ and IL-6 and inflammatory lipid mediators, prostaglandin $E_2$ and leukotriene $B_4$. Adenine impeded signaling pathways eliciting production of these inflammatory mediators. It suppressed $I{\kappa}B$ phosphorylation, nuclear translocation of nuclear factor ${\kappa}B$ ($NF-{\kappa}B$), phosphorylation of Akt and mitogen activated protein kinases (MAPKs) JNK and ERK. Although adenine raised cellular AMP which could activate AMP-dependent protein kinase (AMPK), the enzyme activity was not enhanced. In BMMCs, adenine inhibited the LPS-induced production of $TNF-{\alpha}$, IL-6 and IL-13 and also hindered phosphorylation of $NF-{\kappa}B$ and Akt. In peritoneal cavity, adenine suppressed the LPS-induced production of $TNF-{\alpha}$ and IL-6 by peritoneal cells in mice. These results show that adenine attenuates the LPS-induced inflammatory reactions.

Immunosuppressive Effect of Prodigiosin on Murine Splenocyte and Macrophages

  • Prodigiosin was isolated from marine bacteria Hahella chejuensis which has been recently discovered from Marado, Cheju Island, Republic of Korea. Immunosuppressive properties have been reported for prodigiosin members such as undecylprodigiosin, metacycloprodigiosin, prodigiosin, and its synthetic analogue PNU156804 (PNU). However, the effect of this agent on the function of macrophage and splenocyte has not been characterized in detail. In the present study, we examined the effects of prodigiosin for its ability to alter the function of murine macrophage and NK cell, and the proliferation of splenocytes. When thioglycollate-elicited macrophages pre-exposed to prodigiosin (1-50 ng/ml) were stimulated with LPS/IFN-$\gamma$, pretreatment with prodigiosin resulted in the inhibition of tumoricidal activity of macrophage in a concentration-dependent manner. Tumoricidal activity of NK cell was also inhibited by prodigiosin. Moreover, we found that prodigiosin was able to cause a dose-dependent inhibition of murine lymphocyte responsiveness to Con A and LPS although T-mitogenic response was the more sensitive one. Taken together, the present results point out that prodigiosin has a suppressive effect on the mitogen-induced proliferation of murine lymphocytes and the function of macrophage and NK cell.

Synergistic Effect of Interleukin-18 on the Expression of Lipopolysaccharide-Induced IP-10 (CXCL-10) mRNA in Mouse Peritoneal Macrophages

  • Interleukin (IL)-18, a member of the family of IL-l cytokine, is one of the principal inducers of $interferon-{\gamma}(IFN-{\gamma})$ in T lymphocytes and natural killer cells. The objective of the present study was to evaluate the effect of IL-18 on the expression of chemokine IP-10 (CXCL-10) mRNA in mouse peritoneal macrophages. IL-18 had very weak direct effect or synergistic effect with IL-12 on the expression of IP-10 mRNA in C57BL/6 mouse peritoneal macrophages. However, IL-18 pretreatment was found to playa cooperative role in the expression of lipopolysaccharide (LPS)-induced IP-10 mRNA. For the expression of LPS-induced IP-10 mRNA, the synergistic effect was detected after 16 h of IL-18 pretreatment prior to LPS stimulation. The expression level of CD14 in cells stimulated with LPS was not changed by IL-18 pretreatment, and the level of $IFN-{\gamma}$ production during IL-18 pretreatment plus LPS stimulation was barely discernible ($0.36{\pm}0.31pg/ml$). Namely, the synergistic effect of IL-18 pretreatment was not related to a change of LPS receptor, CD14 expression, and the production of $IFN-{\gamma}$ by the interaction between IL-18 and LPS. The synergistic effect of IL-18 pretreatment on the expression of LPS-induced IP-10 was related to not NF-kB but AP-1 activation, and associated with the extracellular signal-regulated kinase (ERK) pathway, one of the mitogen-activated protein kinase signaling pathways. These results provide useful information that may elucidate the mechanisms underlying the effect of IL-18 on the expression of IP-10 mRNA.

Immune-Enhancing Effects of Crude Polysaccharides from Korean Ginseng Berries on Spleens of Mice with Cyclophosphamide-Induced Immunosuppression

  • Panax ginseng C. A. Meyer is well known as traditional herbal medicine, and ginseng berries are known to exhibit potential immune-enhancing functions. However, little is known about the in vivo immunomodulatory activity of Korean ginseng berries. In this study, crude Korean ginseng berries polysaccharides (GBP) were isolated and their immunomodulatory activities were investigated using cyclophosphamide (CY)-induced immunosuppressive BALB/c mice. In CY-treated mice, oral administration of GBP (50-500 mg/kg BW) remarkably increased their spleen sizes and spleen indices and activated NK cell activities. GBP also resulted in the proliferation of splenic lymphocytes (coordinating with ConA: plant mitogen which is known to stimulate T-cell or LPS: endotoxin which binds receptor complex in B cells to promote the secretion of pro-inflammatory cytokines) in a dose-dependent manner. In addition, GBP significantly stimulated mRNA expression levels of immune-associated genes including interleukin-1β (IL-1β), IL-2, IL-4, IL-6, tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), toll-like receptor 4 (TLR-4), and cyclooxygenase-2 (COX-2) in CY-treated mice. These results indicate that GBP is involved in immune effects against CY-induced immunosuppression. Thus, GBP could be developed as an immunomodulation agent for medicinal or functional food application.

고사리 단백다당(Pteridium aquilinum Glycoprotein, PAG)이 마우스 면역활성에 미치는 영향 (Effects of the Glycoprotein Isolated from Pteridium aquilinum on the Immune Function of Mice)

  • 고사리 수추출물에서 분리된 단백다당획분(Pteridium aquilinum glycoprotein, PAG)이 마우스 면역계에 미치는 영향을 알아보기 위하여 BALB/C 마우스의 복강내에 14일간 반복 투여한 후 마우스의 체중 및 면역장기의 무게변화를 관찰하면서 마우스 항체생성능과 비장세포 증식능에 대하여 조사하였다. PAG를 용량별$(0{\sim}500{\;}{\mu}g/kg)$로 투여한 결과 마우스 체중변화와 면역장기에는 커다란 효과를 관찰할 수 없었으나, HEL에 대한 항체생성능에 있어서는 $500{\;}{\mu}g/kg$ 투여시 뚜렷한 증가를 보였다. 시험물질을 투여한 마우스로부터 비장세포를 분리하여 phytohemagglutinin (PHA) 혹은 lipopolysaccharide (LPS)에 대한 비장세포 증식능을 측정한 결과, PAG 투여군에 있어서는 PHA와 LPS유도 비장세포 증식능이 용량 의존적으로 증가되었음이 관찰되었다. 비장세포 증식능에 대한 in vitro 시험결과 LPS에 대한 반응증가는 단백다당획분(PAG)의 세포에 대한 직접적인 작용에 의하며, PHA에 대한 반응증가는 세포에 대한 직접적인 작용은 아님을 알 수 있었다. PAG의 항체생성능 증가에 기여하는 활성부위를 규명하기 위하여 periodate산화 및 pronase소화를 실시하여 얻어진 각각의 조단백, 조다당분들과 고사리 수추출물에서 정제된 정제다당획분이 항체생성능에 미치는 영향에 대하여 시험하였다. 시험결과 조다당투여군과 정제다당 투여군에서는 항체생성능 증가가 관찰되지 않았으나 periodate 산화물인 조단백 투여군에서 뚜렷한 항체생성능을 확인할 수 있었으며, 이와 같은 실험결과 PAG의 단백성분이 항체생성능 증가에 있어서 주요 활성 본체임을 알 수 있었다., 660 mmHg 및 560 mmHg 압력 중 560 mmHg의 감압 하에서 5회 반복 수세하는 것이었다. 분자구조 변형에 의한 것이라 사료된다./TEX>일 경에 점질물과 산패취가 발생하여 상품으로서의 가치를 상실하였고, $-20^{\circ}C$ 저장구는 저장 100일 까지 상품으로서 가치를 유지하였다.등의 네 가지 요인으로 나누어서 각각의 효과를 요인간으로 비교하면 한일 양국이 함께 가격의 효과가 가장 작다. 그러나 그 이외 요인별 효과의 상대적 중요성은 양국간에 다른데 한국은 소비지출의 효과가 출생연도나 연령 효과보다 크지만 일본은 경제적 요인인 소비지출보다 세대주의 출생연도나 연령 등의 비경제적 요인의 효과가 크다.가능한 전분으로, 완전한 결정성구조를 가진 전분이 아닌 것으로 사료된다.147.33 peak area%인 것과 유사(類似)하게 높았다.tarrow}6$보다 $4{\beta}{\rightarrow}8$의 결합방식이, 그리고 procyanidin 류에서는 gallate를 갖는 물질이 이를 함유하지 많은 물질보다 더욱 높은 ACE의 저해효과를 나타내었으며, 또한 hydroxyl 기가 많을수록 효소 저해효과도 증가하는 것으로 나타났다. 이와 같은 결과는 chocolate의 주원료인 cacao bean의 polyphenol 성분 또한 녹차 등에서 볼 수 있는 생리활성효과에 손색없는 것으로 판단되고, 이러한 기능성에 기초하여 chocolate, 음료 등의 식품이나 의약품의 기능성 소재로서의 산업적 응용 가능성이 높은 것으로 사료된다.대장균군이 역시 난각에서 가장 높은 빈도로 분리되었고, 난황(Yolk)에서는 극히 낮은 수준의 세균오염도를 보였다. 다양한 동물종유래 S. aureus 균주들의 유전학적 분석목적에 가장 신뢰도 높고 감별능력이

수은이 마우스의 체액성 및 세포성 면역반응에 미치는 영향 (Effect of Mercury Chloride on Humoral and Cell-mediated Immune Responses in Mice)

  • 수은이 마우스의 세포성 및 체액성 면역반응에 미치는 영향을 밝히고자 실험을 통해 얻은 결과를 다음과 같이 요약하는 바이다. $10^{-2}mM$이상의 농도에서 수은은 시험관내의 마우스 비장세포에 대한 명백한 독성 작용(toxic effect)을 나타냈다. 수은에 의한 마우스 생존을(survival rate)은 0.8mg(per 100g body weight) 이하의 농도에서는 100%생존율을 나타냈으며, 0.9mg 이상의 농도에서는 5일 이내에 100% 치사율을 나타냈다. Subtoxic 농도의 수은 투여에 따른 Arthus 및 DTH반응의 결과는 $Arthus(T_3)$반응에 있어서 수은 투여군과 대조군 사이에 유의한 차이가 나타나지 않았던 반면, $T_{24},\;T_{48}$의 DTH반응은 수은 투여군이 대조군에 비하여 억제되었다. 수은 투여 마우스의 면양적혈구(SRBC)에 대한 항체반응을 측정한 결과 면양적혈구에 대한 총응집소가는 수은 투여 농도에 의존적으로 대조군의 값에 비해 억제되었다. 시험관 내에서 마이토젠에 대한 비장세포의 증식 반응은 수은 농도에 의존적으로 억제되었다. 수은 투여군의 IgE 생산은 농도 증가에 따라 $1.7\sim2.0$배의 수준으로 증가하였으며, $IgG_1$의 경우도 증가의 경향을 보였으나 대조군에 비해 큰 차는 아니었다. 이상의 결과를 살펴볼 때 수은 중독시 출현할 수 있는 면역반응의 변화로서, 초기에는 세포성 면역반응의 감소가 현저할 것으로 사료되나 중독이 진행됨에 따라 Ig농도의 상승을 제외한 체액성 면역반응의 전반적인 감소가 수반될 것으로 예측된다.

오리나무유래 디아릴헵타노이드 허수테논의 T 세포활성억제 및 항아토피 효능연구 (T-lymphocyte Inactivation and Anti-atopic Effects of Diarylheptanoid Hirsutenone Isolated from Alnus japonica)

  • 본 연구에서는 오리나무유래 HST의 T 세포 비활성화 효능을 확인하고 그 기전을 탐색하여 새로운 항아토피 천연소재로의 개발 가능성을 제시하고자 하였다. HST는 T 세포 mitogen으로서 anti-CD3 mAb가 처리 된 마우스 비장에서 Th 사이토카인(IL-2, IFN-${\gamma}$, IL-4, IL-5, IL-10)의 발현을 효과적으로 억제하였으며, T 세포 early activation marker인 CD25 유전자 발현이 INCA-6에서 매우 효과적으로 억제되는 것으로 보아, NFAT inactivator-유사 NFAT 탈인산화 억제 기전을 가지는 것으로 예측되었다. 또한 세포주기조절 단백질인 p21과 p27의 유전자도 HST에 의해 upregulation이 되어 효과적인 T 세포 증식 및 분화를 억제할 것으로 생각되었다. 이러한 세포주기조절 효과는 AD를 악화시키는 세균인 S. aureus 성장억제 실험에서 확인되어 향후 항박테리아 효능을 갖는 우수한 항아토피피부염 천연소재로서 HST의 활용 가치가 높을 것으로 판단된다.

Effect of Zedoariae rhizoma on Bronchial Inflammation and Allergic Asthma in Mice

  • There are detailed descriptions of the clinical experiences and prescriptions of asthma in traditional Korean medicine. Zedoariae rhizoma is one of the Korean herbal medicines used to treat bronchial asthma and allergic rhinitis for centuries. However, the therapeutic mechanisms of this medication are still far from clear, In this study, a house-dust-mite (Dermatophagoides pteronyssinus [Der p])-sensitized murine model of asthma was used to evaluate the immunomodulatory effect of Zedoariae rhizoma on the allergen-induced airway inflammation in asthma. Three different protocols were designed to evaluate the treatment and/or long-term prophylacitic effect of Zedoariae rhizoma in Der p-sensitized mice. Cellular infiltration and T-cell subsets in the bronchoalveolar lavage fluid (BALF)of allergen-challenged mice were analyzed. Intrapulmonary lymphocytes were also isolated to evaluate their response to allergen stimulation. When Zedoariae rhizoma was administered to the sensitized mice before AC (groups A and C), it suppressed airway inflammation by decreasing the number of total cells and eosinophil infiltration in the BALF, and downregulated the allergen- or mitogen-induced intrapulmonary lymphocyte response of sensitized mice as compared to those of controls. This immunomodulatory effect of Zedoariae rhizoma may be exerted through the regulation of T-cell subsets by elevation or activation of the CD8+ and double-negative T-cell population in the lung. However, the administration of Zedoariae rhizoma to sensitized mice 24 h after AC (group B) did not have the same inhibitory effect on the airway inflammation as Zedoariae rhizoma given before AC. Thus, the administration of Zedoariae rhizoma before AC has the immunomodulatory effect of reducing bronchial inflammation in the allergen-sensitized mice. On the other hand, to determine the potentiality of prophylactic and/or therapeutic approaches using a traditional herbal medicine, Zedoariae rhizoma, for the control of allergic disease, we examined the effects of oral administration of Zedoariae rhizoma on a murine model of asthma allergic responses. When oral administration of Zedoariae rhizoma was begun at the induction phase immediately after OVA sensitization, eosinophilia and Th2-type cytokine production in the airway were reduced in OVA-sensitized mice following OVA inhalation. These results suggest that the oral administration of Zedoariae rhizoma dichotomously modulates allergic inflammation in murine model for asthma, thus offering a different approach for the treatment of allergic disorders.
