the phytotoxic effects of volatile substances emitted from the tomato plant (Lycopersicon esculentum) on receptor plants such as Bidens bipinnata, Plantago asiatica, Lactuca sativa, Eragrositis ferruginea and Achyranthes japonica were investigated. Bolatile substances from the leaves inhibited seedling growth of receptor plants in the laboratory tests. The inhibition response varied with the concentration of compounds. To identify the phytotoxic compounds from tomato plant a GC/MS method was employed. Forth compounds, such as trans-2-hexeal, linalool, phenylacetaldehyde, methlsalicylic acid and tetradecanaic acid were identified from the essential oil of tomato plants. The findings suggest that the tomato plant may have allelopathic potential.
The liquids of Lycopersicon esculentum were extracted with 70% aqueous MeOH and the concentrates were partitioned into EtOAc, n-BuOH, and H2O fractions. The repeated silica gel and octadecyl silica gel column chromatographies for the EtOAc fraction, whose activity was confirmed, led to isolation of one flavonol compound. The chemical structures of the compound were determined as quercetin (1) based on spectroscopic analyses including nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy. Through this study, the antioxidant efficacy was confirmed by demonstrating that the L. esculentum fraction showing an increase in glutathione mean (GM) and a decrease in glutathione heterogeneity (GH) uniformly raises the intracellular glutathione (GSH) level.
Plants of Lycopersicon esculentum, containing various organic compounds, are known to develop idioblasts in their epidermis. Lycopersicon esculentum have long been investigated in many areas, but structural aspects of the epidermis of various organs have not been carried out in detail. Thus, the present study attempted to reveal the patterns of idioblast development, particularly those of the reproductive organs, in L. esculentum epidermis using scanning electron microscopy. The present study mainly focused on patterns of the stomata and trichome types. Two types of stomata were developed in the flowers and fruits: anomocytic stomata (stomata type I) were distributed normally throughout the epidermis, whereas actinocytic raised stomata (stomata type II) were found variously in different epidermal tissues. For the trichomes, both glandular and non-glandular types were developed in the epidermis. The former included peltate glandular trichomes having four head cells (trichome type I) and capitate multicellular glandular trichomes (trichome type II). The latter included non-glandular short trichomes (trichome type III) and considerably elongated trichomes with basal rosette cells (trichome type IV). In paticular, the raised stomata were well-developed in the peduncles and the peltate glandular trichomes were prominent in the sepal and ovary epidermis. Transmission electron microscopy on the ontogeny and ultrastructural differentiation of these idioblasts, associated with the current result, will aid us in better understanding of the structure and functional relationship in the epidermal differentriation of Lycopersicon esculentum.
The fruits of tomato (Lycopersicon esculentum) were extracted with 70% aqueous methanol (MeOH) and the concentrates were partitioned into ethyl acetate (EtOAc), n-butanol (n-BuOH), and water (H2O) fractions. The repeated silica gel (SiO2) and octadecyl silica gel column chromatographies for the EtOAc fraction, whose activity was confirmed, led to isolation of one flavone compound. Nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy (MS) revealed the chemical structure of the isolated compound, 5,7,3'-trihydroxy-6,4',5'-trimethoxyflavone (1). LC-MS/MS analysis determined the content level of compounds 1 in the MeOH extract to be 4.02±0.12 ㎍/mg and in the TME-10 fraction to be 0.96±0.03 ㎍/mg. Through this study, the antioxidantive capacity was confirmed by demonstrating that the L. esculentum extract and their fractions showing an increase in glutathione mean and a decrease in glutathione heterogeneity uniformly raises the intracellular glutathione level.
The study was carried out to establish an efficient protocol for shoot organogenesis and plant regeneration from stem explant cultures of Lycopersicon esculentum cv. Micro-Tom. The regenerated shoots obtained from stem explant cultures on solid MS medium containing the different concentrations of BAP. The highest number of shoots (5.3) per explant and shoot growth (0.7 cm) was obtained on MS medium containing 4.0 mg/L BA. The additions of AgNO$_3$ and putrescine substantially improved the shoot regeneration frequency, at the optimal concentration of 7 mg/L and 50 mg/L respectively. The regenerated shoots (about 1 cm) were normal and could be easily rooted with 0.1 mg/L IBA treatment. The rooted plants were hardened and transferred to vermiculite with a 92% survival rate where they grew normally.
Effects of methyl jasmonate (MeJA) on ethylene production in tomato(Lycopersicon esculentum Mill.) hypocotyl segments and fruits were studied. Ethylene production in tomato hypocotyl segments was inhibited by the increasing concentratons of MeJA, and 450 $\mu$M of MeJA showed 50% inhibitory effect. Time course data indicate that this inhibitory effect of MeJA appeared after 3 h of incubation period and continued until 24 h. Inhibition of ethylene producton by MeJA was due to the decrease in 1-aminocyclopropane-1-carboxylic acid(ACC) synthase activity. However, MeJA treatment had no effect on ACC oxidase activity and the accumulaton of ACC oxidase mRNAs. MeJA also inhibited auxin-induced ethylene production by decreasing in ACC synthase activity. In contrast, MeJA stimulated ethylene production in tomato fruits. When 30 $\mu$L/mL MeJA was treated in a gaseous state, ethylene production doubled and this stimulating effect continued until 4 days. To investigate the mechanisms of MeJA on ethylene production, ACC synthase and ACC oxidase activities were examined after MeJA treatment. MeJA increased the activities of both ACC synthase and ACC oxidase, and induced ACC oxidase mRNA accumulation. These data suggest that MeJA plays distinct roles in the ethylene production in different tomato tissues. It is possible that MeJA affects differently the mechanisms of signal transuction leading to the ethylene biosynthesis.
The external qualities of cherry tomato fruits(Lycopersicon esculentum) grown in circulating deep flow hydroponic culture were non - destructively estimated in 1992 under warm climate conditions and evaluated in 1993-1994 under cool season. The fruit size of ‘Minicarol’ was by far the smallest compared to ‘Chelseamini’ and ‘Popo’. There were high correlations between external qualities of fruits-lengthwise growth harmonized with widthwise growth. The volume and/or weight of fruit could be non -destructively estimated by length and width of fruit. Volume=0.071$\times$ (length+width)$^3$+0.451, Fresh weight=0.072$\times$ (length+width)$^3$+0.542. These models were accurately evaluated by fruits harvested in 1994.
Lycopersicon peruvianum has a gametophytic self-incompatibility (GSI) mechanism controlled by a single genetic locus (S locus) with multiple alleles. S RNases, an allelic series of abundant stylar proteins, are products of the S locus in L. peruvianum and other Solanaceous plants. The $S_{11}$ RNase gene from L. peruvianum was introduced into a self-compatible (SC) species (Lycopersicon esculentum) to examine whether the expression pattern in the heterologous host mimics that in L. peruvianum. The resultant transgenic L. esculentum plants expressed the introduced gene highly in their styles, which is similar manner to the expresion in L. peruvianum. The $S_{11}$ RNase gene was expressed in the syle at a similar stage of flower development in both transgenic plants of L. esculentum and L. peruvianum without any morphological changes.
