Study on the Activity of GOT and GPT in the Hepatotoxic Rat Treated (구기자 투여 간손상 흰쥐에서 GOT 및 GPT의 활성화 연구)

  • The present study was undertaken in order to investigate betaine production by tissue culture and its medicinal effect in Lycium chinone Mill. In order to ulvestigate the protective effect of L. chinense on the hepatoxicity induced by $CCl_4$, 0.5 g/kg water extract of the compound mixture (leaves, roots and shoots) of L. chinense and its callus were fed to rat (SD line) once a day. As a result, the activity of GOT and GPT in the group fed compound mixture (GOT 760.4 and GPT 540 Karmen unit) and callus (G0T 772.1 and GPT 556.4 Karmen unit) was decreased in the blood serume relative to the controlled rat group (GOT 949 and GPT 640 Karmen unit) and the same result was obtained in the group fed with 0.1 g/kg sylimarin (the activity of GOT and GPT was shown 492.6 and 320.4 Karmen unit respectively. These results strongly indicate that water extracts of the mixture and callus from L. chinense do have the same decreasing effect of GOT and GPT in the hepatotoxic rat induced by $CCl_4$.

  • Transformation system of rolC gene, dwarf gene in Lycium chinenese Mill. established by using system. Pin-punctured leaves induced numerous adventious buds in abaxial side when cultured on 3/2 MS medium containing 2.0 mg/L zeatin. Survival rate and shoot regeneration frequency of leaf explants decreased as kanamycin sulfate level increased. Shoot buds were not regenerated on 3/2 MS medium containing 10 mg/L kanamycin sulfate and 2.0 mg/L zeaein. Of the level tested, 10 mg/L of kanamycin sulfate was optimum in selection of kanamycin sulfate resistant plant. Co-culture time of bacteria and leaf explants was affected at the frequency of shoot regeneration and survival of leaf explants. Leaf explants co-cultivated during above 48hr severely decreased survival rate and shooting rate. Best result on survival rate and shooting rate were obtained when exposed for 24 h. 80 explants of 105 leaf explants survived on 3/2 MS medium containing 2.0 mg/L zeatin and 10 mg/L kanamycin sulfate, and 15 shoots was regenerated on the same medium. To select kanamycin sulfate resistant plant, regenerate as cultured on 3/2 MS medium containing 10 mg/L kanamycin sulfate, and obtained 5 kanamycin resistant plants. Southern blot analysis conformed that the rolC gene was incorporated into the genomic DNA of kanamycin resistant plants.

Free Radicals Scavenging Activity of Bulro Kugi (Lycium chinense Mill) Fruit, Leaf and Root (불로 구기 부위별 자유라디칼 소거효과에 관한 연구)

  • Free radical-scavenging activities of Korean Bulro Kugi (Lycium chinense Mill) fruit, leaf, and root were evaluated. Total phenolic contents of fruit, leaf, and root were $1,078.4{\pm}61.0$, $939.9{\pm}19.8$, and $3,792.2{\pm}106.6mg/L$, and their flavonoids were $396.7{\pm}15.2$, $1,952.9{\pm}21.3$, and $425.3{\pm}13.5mg/L$, respectively (p<0.001). HPLC revealed main polyphenolic compounds in fruit were p-coumaric and syringic acids in fruit, p-coumaric, syringic, and procatechuic acids in leaf, and p-coumaric, syringic, caffeic, and procatechuic acids in root. Highest radical-scavenging activities of superoxide anion and hydroxyl were found in leaf and root, respectively (p<0.001).

  • Chemical and flavor components of Bulro Kugi (Lycium chinense Mill) fruit leaf, and root were compared. Fructose and glucose were detected in fruit and leaf, and sucrose in root, respectively. Citrate was the highest among organic acids in fruits, and malate in leaf and root Capsaicin was detected in leaf and root. Volatile flavor compounds were extracted by simultaneous distillation and extraction method using Likens and Nickerson's extraction apparatus. Concentrated flavor extract was analysed, and 128 compounds, including 22 acids 15 alcohols, 12 aldehydes, 8 esters, 10 furans, 26 hydrocarbons, 4 phenols, 2 pyrroles, 1 pyrazine, and 28 miscellaneous components. were identified by GC and GC-MS. Main volatile compounds were hexadecanoic acid and 2-furancarboxaldehyde in hot-air dried fruit, hexadecanoic acid and 1-hexadecene in fresh fruit, 3, 7, 11, 15-tetranethyl-2-hexadecan-1-ol and hexadecanoir acid in leaf, and hexadecanoic acid in root.

Studies on Outbreak of Diseases and Pests and Effect of Environmental Friendly Control Materials in Boxthorn (Lycium chinenseMill.) Organic Cultivation (구기자(Lycium chinense Mill.) 유기재배시 병해충 발생 및 친환경제제의 방제효과)

  • This study was carried out to develop environmental friendly control for major diseases and pests on Boxthorn (Lycium chinense Mill.). Outbreak of Eighteen diseases and pests were found at the Boxthorn organic yards in Chung-nam province. Among them Powdery mildew (Erysiphe polygoni de Cand.), Hypophyllous mold (Pseudocercospora chengtuensis (Tai)), Western flower trips (Frankliniella occidentalis (Pergande)), Green peach aphid (Myzus pericae (Sulzer)) and Corn earworm (Helicoverpa armigera) needed to be controled by environmental friendly methods for high fruit yield of organic Boxthorn. In summer(Jun) test Bacilus subtilis QST 713 wettable powder and Sulfur wettable powder were effective and in autumn (Sep.) test Sulfur, Copper hydroxide and Paraffinic oil were relatively effective in Powdery mildew. In Hypophyllous mold control test Paraffinic oil and Bacilus subtilis GB - 0365 were effective with above 70% control value. And it was possible to control Western flower trips by natural enemy (Orius laevigatus) by 80% control value. Corn earworm was possible to control by Bacillus thuringiensis subsp. aizawai GB413 flowable and Bacillus thuringiensis aizawa 0423 wettable powder application above 70%. And Green peach aphid was controllable with environmental friendly materials, such as, Bacillus subtilis (Seoncho), Bacillus subtilis (Jinsami) above 80% and Ginkgo nut extract above 70% control value.

Physicochemical and Sensory Properties of Baikseolgi Made with Kugija (Lycium chinense Mill.) Powder

  • Baikseolgi was made with Kugija powder with the intention to take advantage of its functional properties. Appropriate amount of Kugija powder (0$\sim$8%) was mixed with rice flour, sugar, salt, and water and then, steamed for predetermined time and their physicochemical and sensory properties were measured. pH decreased significantly while titratable acidity increased significantly with the addition of Kugija (p<0.05). The addition of Kugija powder did not significantly affect the moisture content (p>0.05) although it appeared to decrease with the higher amount of Kugija. Lightness (L-value) decreased significantly with the addition of Kugija (p<0.05), indicating that the color of Baikseolgi became dark as also indicated by the visual observation. Redness (a-value) and yellowness (b-value), on the other hand, increased significantly as the amount of Kugija increased in the sample (p<0.05). Both hardness and firmness decreased significantly with the addition of Kugija (p<0.05). Eight percentage Kugija Baikseolgi was significantly stronger in Kugija flavor (7.9), chewiness (6.1), sweetness (5.0), and yellowness (8.2) attributes than other samples, whereas control was significantly higher in mouthfeel (6.0) and hardness (6.0) attributes than others (p<0.05). Finally, consumer test indicated that 4% Kugija sample received the highest score in appearance (8.13) and overall acceptability (8.07) attributes.

  • In this report, an experiment has been conducted to test the quantitative changes of nucleic acids in the nuclei of the epidermal cell of the galls, caused by Eriophyes kuko Kishida on the leaf of Lycium chinense Mill by means o( microspectrophotometric techniques, the two-wave-length methods. And the sizes of the epidermal cells and nuclei have been measured. The experimental results were summarized as follows: 1) It has been found that as the gall grows, the diameter of the epidermal cells and their nuclei increased and they were larger than those of tile healthy ones. 2) Microspectrophotometric measurement of nuclei by the 'Two-wave-length method' after staining with Feulgen reagent showed no changes in DNA content in the early stage of the gall. As the gall matured, however, DNA content of the gall increased more than that of the healthy leaf. 3) RNA-measurement of nuclei stained with Azur-B in DNase treated epidermal cells of the gall revealed that temporary increase in RNA content occurred in early to middle stages after the gall formation. As the gall matured, however, RNA content of the gall decreased more as against that of the healthy leaf.

  • Light and electron microscopic studies were made to investigate the morphological changes in growing galls on the leaf of Lycium chinense MILL caused by Eriophyes kuko Kishda. The results are summarized as follows: 1. Light microscopy At the early stage of the invasion of E. kuko on the back side of the young leaf of L. chinense, the.epidermal cells become hypertrophic and develope a gall. As the gall grows, the cells of both palisade and spongy-layers become hypertrophic and these tissues are hard to be distinguished because of their irregular outgrowth. As the gall grows, the nuclei of the gall also become hypertrophic and larger than these of normal cells. 2. Electron microscopy Under electron microscopy the mitochondria, the golgi apparatus and the plastids of the advanced galls are degenerated and disintergrated and the cell walls become thicker than normal ones. The characteristic star bodies and the ring-form structures are found in the mature gall cells.

  • This Paper deals With the biology and the life Cycle of Eriophyes kuko KISHIDA. 1. The host of Eriophyes kuko KISHIDA is Lycium chinense MILL (Boxthorn), Solanum nigrum LINNE(Morel) and Capacium annum LINNE(Red pepper) etc. 2. Eriophyes kuko KISHIDA is migrated and propagated by physical carriers. The direction of the wind has a great influence upon migration and dispersal. 3. The number of e99s deposited by a adult female is $28.5\pm0.449$ egg period is $7.0\pm0.447$ and larval period is $4.33\pm0.667$ days respectively. 4. The size of gall diameter on the leaf of Lycium Chinense MILL after boring in each date by Eriophyes kuke KISHIDA is as follows, 1st day after boring is $290.5\pm0.3415\mu$ End 25st day after boring is $3260.5\pm0.5157\mu$ in average. 5. Adult periods are from 25 to 30 days and the egg laying period is 6 days. 6. Eriophyes kuko KISHIDA has 6 to 7 generations in a year, and hibernate they over winter in the gall as an adult. The first adult appears in late May.
