음곡(KI10) 구기자 약침이 난소적출 생쥐의 골다공증에 미치는 영향 (Effects of the Herbal-Acupuncture with Lycii Fructus Extract at KI10 (Eumgok) on Osteoporosis in Ovariectomized ddy mice)

  • Objective & Methods : The purpose of this study is to observe the effects of herbal-acupuncture with Lycii Fructus (LF-HA) at KI10 (Eumgok) on osteoporosis in ovariectomized (OVX) ddy mice. We carried out several experimental items which were known to be related with osteoporosis. Results : 1. Tibial BMD was significantly increased in the LF-HA group. 2. Tibial Ca level was significantly increased in the LF-HA group. 3. Osteoclast like cell in tibia was significantly decreased in the LF-HA group. 4. Histological change of tibia was improved, the trabecular bone thickness was significantly increased and the growth plate length was significantly decreased in the LF-HA group. Conclusion : These results suggest that LF-HA at KI10 has therapeutic effects on osteoporosis especially by way of suppressing osteoclast cells in ovariectomized ddy mice.

한약재복합 추출물의 인간피부섬유아세포 HS68에 대한 항노화 효과 (Antiaging Activity of Mixed Extracts from Korean Medicinal Herbs on HS68 Skin Fibroblast)

  • Objectives : The purpose of this study was to investigate antiaging and antioxidant effects on cultured human skin fibroblast with 80% ethanol extracts of plants including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Methods : An ethanol extract of three medicinal plants including stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Extracts were assessed to determine the mechanism of antioxidant and antiaging activities. Antioxidant activity of extract was evaluated by two different assays as 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and super oxide dismutase (SOD) like activities. These extracts were tested for cell viability on HS68 skin fibroblast by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. We investigated the effects of Ultraviolet-B irradiation on cytotoxicity, type 1 collagen, elastin level and oxidative damage in cultured human skin fibroblast (HS68). Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. Results : The extracts obtained dose-dependently increased the scavenging activity on DPPH radical scavenging activity and SOD like activity. The extracts of complex herbal medicine showed low cytotoxicity as more than 100% cell viability in 100ppm/ml concentration. HS68 fibroblasts were survived 70% at 120 $mJ/cm^2$ UVB irradiation and treated tumor necrosis factor (TNF)-alpha. The levels of aging factors and cytotoxicity were decreased by ethanol extract of complex herbal medicine. Conclusions : These results suggest that ethanol extracts of complex medicinal plants of including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus may have value as the potential antioxidant and antiaging medicinal plant.

  • Objective : This study was conducted to observe the anti-hyperlipidemic and anti-inflammatory effects, as well as the metabolic benefits, of pear extract and herbal drug mixture (Lycii Fructus, Coicis Semen, Alimatis Rhizoma, and Astragali Radix) on rats fed with a high fat diet. Methods : The animals used were male rats and the control group was fed a high fat diet only. The experimental groups were divided into four. Exp I group was fed a high fat diet with a mixture of pear extract and 3% Lycii Fructus; Exp II group was fed a high fat diet with a mixture of pear extract and 3% Coicis Semen; Exp III group was fed a high fat diet with a mixture of pear extract and 3% Alimatis Rhizoma; and Exp IV group was fed a high fat diet with a mixture of pear extract and 3% Astragali Radix for 4 weeks. Results : The body weight gain increased in all groups, but attenuated gradually in the experimental groups compared to the control group. The food intakes were significantly lower in Exp I and Exp III groups than the control group. The concentrations of serum total cholesterol (TC), HDL-cholesterol, LDL-cholesterol, and triglyceride (TG) were significantly higher in Exp II than in the control group, and lower in Exp III group than in the control group. Also the concentration of serum free fatty acid was significantly lower in the Exp III group than in the control group. In inflammatory activities, the Exp II group was significantly lower than the control group. Conclusion : The results indicated that Exp III group (administered a mixture of pear extract and Alimatis Rhizoma) most efficiently reduced fat accumulation and body weight, while the Exp II group (administered the mixture of pear extract and Coicis Semen) had the highest elevated lipid metabolism and immune activity.

Mild stressed rat을 이용한 수종한약의 항우울효과에 대한 실험적 연구 (Anti-depressant Effect of Herbal Extracts in the Chronic Mild Stressed Rat)

  • This study was performed to investigate the anti-depression effect of herbal extracts (Rehmanniae Radix Preparat, Corni Fructus, Lycii Fructus, Pinelliae Rhizoma, Nelumbinis Semen) in the chronic mild stressed rat. After adaptation period for 2 weeks, each herbal extract was administered to the rats divided 5 groups. For comparison, St. John's wart was administered to another group in the same environment. The chronic mild stress was induced to rats by overnight illumination for 2 days before the forced swimming test. Struggling time, first latency and rest duration were calculated during the forced swimming test to evaluate the anti-depressant effect. The results were as follows: 1. In the result of evaluation of the struggling time, there was no significance in Pinelliae Rhizoma (-1.1%), Corni Fructus (-3.9%), Rehmanniae Radix Preparat (15.2%), Lycii Fructus (4.9%), St. John's wart (25.2%). There was a significant increase only in Nelumbinis Semen (43.9%) (P=0.0037). 2. In the result of evaluation of the first latency time, there was no significance in Lycii Fructus (-21.37%), Rehmanniae Radix Preparat (38.4%), Pinelliae Rhizoma (65.5%), Corni Fructus (29.2%), St. John's wart (75.8%). There was a significant increase only in Nelumbinis Semen (90.2%) (P=0.0116). 3. In the result of evaluation of the first rest duration, there was no change in St. John's wart, and Decrease was shown in Pinelliae Rhizoma (62.4%), Corni Fructus (31.6%) Rehmanniae Radix Preparat (63.1 %), Lycii Fructus (12.4%), Nelumbinis Semen (59.0%), but there was no significance in all. 4. Nelumbinis Semen is considered to have a superior anti-depression effect than the other herbal extracts, and to have a superior and-depression effect than St. John's wart being used commonly.

구기자와 구기엽 추출물이 난소적출 흰쥐의 골다공증에 미치는 영향 (Effects of Lycii Fructus and Lycii Folium Extracts on Osteoporosis in Ovariectomized Rats)

  • 본 연구는 구기자와 구기엽 추출물이 난소를 절제한 흰쥐의 체중 증가, 골밀도, 에스트로겐 수용체의 발현에 미치는 영향을 밝히고자 시행하였다. 실험군은 흰쥐 24마리를 가장수술군(Sham군), 난소를 절제한 대조군(OVX군), 난소절제 후 구기자 추출액을 투여한 군(LCF군), 난소절제 후 구기엽 추출액을 투여한 군(LCL군)으로 구분하였다. 8주 후 난소를 절제한 OVX군($330{\pm}5.39g$), LCF군($315{\pm}2.99g$) 및 LCL군($318{\pm}2.06g$)의 체중은 Sham군($281{\pm}1.71g$)에 비하여 증가하였다. 혈청 osteocalcin 활성은 OVX군($107{\pm}3.52ng/mL$)에 비하여 LCF군($444.6{\pm}26.9ng/mL$)과 LCL군($407{\pm}18.9ng/mL$)에서 증가하였다. Alkaline phosphatase의 활성은 OVX군($95{\pm}2.9U/L$)에 비하여 LCF군($108{\pm}2.7U/L$)과 LCL군($407{\pm}18.9ng/mL$)에서 증가하였다. 8주 후 넙다리뼈를 실체현미경으로 관찰한 결과 뼈 기질밀도는 난소절제군에서 감소하였으나, 구기자와 구기엽을 추출액을 투여한 군에서는 뚜렷이 회복되었다. 에스트로겐 수용체의 mRNA 발현은 OVX군에서는 거의 발현되지 않았으나, LCF군과 LCL군에서 뚜렷이 증가하였다. 이상의 결과로 보아 구기자와 구기엽 물추출물이 난소절제에 의하여 유발된 흰쥐의 뼈 기질 회복 또는 손실 지연에 효과가 있다고 할 수 있을 것이다.

Determination of Betaine in Fructus Lycii Using Hydrophilic Interaction Liquid Chromatography with Evaporative Light Scattering Detection

  • A simple new method was developed for the determination of betaine in Fructus Lycii using hydrophilic interaction liquid chromatography with evaporative light scattering detection (HILIC-ELSD). Good chromatographic separation and reasonable betaine retention was achieved on a Kinetex HILIC column ($2.1{\times}100mm$, $2.6{\mu}m$) packed with fused-core particle. The mobile phase consisted of (A) acetonitrile and (B) 10 mM ammonium formate (pH 3.0)/acetonitrile (90/10, v/v). It was used with gradient elution at a flow rate of 0.7 mL/min. The column temperature was set at $27.5^{\circ}C$ and the injection volume was $10{\mu}L$. The ELSD drift tube temperature was $50^{\circ}C$ and the nebulizing gas (nitrogen) pressure was 3.0 bar. Stachydrine, a zwitterionic compound, was used as an internal standard. Calibration curve over $10-250{\mu}g/mL$ showed good linearity ($R^2$ > 0.9992) and betaine in the 70% methanol extract of Fructus Lycii was well separated from other peaks. Intraand inter-day precision ranged from 1.1 to 3.0% and from 2.4 to 5.3%, respectively, while intra- and inter-day accuracy ranged from 100.0 to 107.0% and from 94.3 to 103.9%, respectively. The limit of quantification (LOQ) was $10{\mu}g/mL$ and the recoveries were in the range of 98.2-102.7%. The developed HILIC-ELSD method was successfully applied to quantitatively determine the amount of betaine in fourteen Fructus Lycii samples from different locations, demonstrating that this method is simple, rapid, and suitable for the quality control of Fructus Lycii.

수확시기에 따른 구기자 품종의 성분변화 (Changes in Composition of Gugija (Lycii Fructus) Species According to Harvest Time)

  • 시험장에서 육성한 품종인 명안, 불로, 청대, 장명, 청운과 육성중인 청양6호, 청양7호 및 청양재래의 가용성 무질소물, 조단백질, 조지질, 회분, 총 폴리페놀, 엑스, 베타인의 함량을 8, 9, 10 및 11월의 수확시기별로 조사한 결과, 가용성 무질소물 및 엑스는 육성중인 청양6호가 가장 높은 함량을 보였고, 조단백질, 총폴리페놀 및 베타인의 함량에서는 육성중인 청양 7호가 가장 높은 함량을 보였으며, 조지질은 청대에서, 조회분은 청운에서 가장 높았다. 건과비율은 육성중인 청양6호에서 가장 높았고, 건과비율이 높을수록 가용성 무질소물의 함량은 증가 (r = 0.70)하는 반면 조단백질 (r = -0.83) 및 조회분 (r = -0.71)등은 감소하였다. 일반성분 중에는 서로 일정한 상관을 보였는데, 가용성 무질소물의 함량이 증가하면 엑스의 함량 (r = 0.81)은 높아지는 반면, 단백질 (r = -0.88), 지질 (r = -0.82) 및 회분 함량 (r = -0.81)은 감소하고, 총폴리페놀 함량이 증가하면 베타인 함량이 증가 (r = 0.90)하는 경향이었다. 품종특성과 성분을 분석한 결과 육성중인 청양 6호는 가용성 무질소물 및 엑스분이 높게 관찰되었을 뿐만 아니라, 건과비율이 높아 타 품종에 비해 쉽게 건조될 수 있어 건조비용을 줄일 수 있는 것으로 예상되었고, 청양 7호는 조단백질, 총폴리페놀 및 베타인 함량이 높아 기능성 품종으로서의 가치를 보여주었다.

구기자 추출액을 첨가한 다식의 품질특성 및 항산화 효과 (Quality Characteristics and Antioxidative Effects of Dasik added with Lycii Fructus Extract)

  • The purpose of this study was to investigate the quality characteristics and antioxidative effect of Dasik prepared with Lycii fructus extract(LD). LD were added in ratios (w/w) of 0(C), 5(LD1), 10(LD2), and 15%(LD3), and then proximate compositions, physicochemical properties, sensory evaluations and antioxidative effect of the Dasik were measured. LD1~LD3 samples showed higher contents of moisture, crude lipid, crude protein and crude ash as well as pH and sugar content compared to control (p<0.001). The total phenol content of LD samples (LD1, 31.5% ; LD2, 66.1% ; and LD3, 69.7%) was higher than that of control (4.71%). DPPH radical scavenging activity of LD3 showed the highest activity of 65.3%, whereas that of control (27.0%) was the lowest among samples. Additionally, ABTS radical scavenging activity of LD samples was higher than that of control (C, 38.9% ; LD1, 53.6%; LD2, 80.5% ; and LD3, 83.3%). The results of texture analysis on all samples showed that control had the highest but LD3 was the lowest (p<0.001). In sensory evaluation, the scores of appearance, flavor, taste and overall preference for LD2 was significantly higher than the samples(C, LD1, and LD2). Form the findings, this study suggests that 10% addition of LD was effective for preparation of Dasik in the aspects of the consumer acceptability and functionality.

LPS로 유도된 급성 간독성에 대한 구기자 추출물의 보호 효과 (The Preventive Effects of Lycii fructus Extract Against LPS-induced Acute Hepatotoxicity)

  • The purpose of this study was to investigate the preventive effects of Lycii fructus Extract (LFE) against the acute hepatotoxicity-inducing lipopolysaccharide (LPS) in the liver. LFE of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. On the day 21, 1.5 ml/kg of LPS dissolved in saline was injected 4 hours before anesthetization. We examined the levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), lactate dehydrogenase (LDH) in serum of rats, superoxide dismutase (SOD) in mitochondrial fraction, and malondialdehyde (MDA), catalase (CAT), glutathione peroxidase (GPx) in liver homogenate. LPS-treatment markedly increased the levels of GOT, GPT, LDH and MDA, and significantly decreased those of SOD, CAT and GPx. But LFE-pretreatment decreased the levels of GOT, GPT, LDH and MDA, by 17.7%, 27.5%, 40.7% and 56.9%, respectively and increased those of SOD, CAT and GPx, by 90.5%, 78.9% and 83.8%, respectively. These results showed that the LFE had the preventive effects against the acute hepatotoxicity-inducing LPS in the liver.

볶음 조건에 따른 구기자 열수 추출물의 이화학적 특성 (Physico-chemical Properties of Boxthorn(Lycii fructus) Hot Water Extracts by Roasting Conditions)

  • 건 구기자와 볶음조건을 달리한 볶은 구기자의 열수 추출물에 대하여 여러 가지 이화학적 특성을 조사하였다. 건 구기자의 일반성분은 수분 17.5%, 회분 5.4%, 지방 14.7%, 단백질 18.9%, 섬유질 12.8%, 당질 31.7%였다. 열수 추출시 추출수율은 볶음온도와 시간이 증가할수록 가용성 고형분이 더 빨리 추출되었으며, 색도는 명도가 감소하고 적색도가 증가하여 추출물이 더 진한 갈색을 나타내었고 탁도 역시 증가하였다. pH는 볶음온도와 시간이 증가할수록 5.08에서 4.38로 감소하였고 산도는 0.09%에서 0.16%로 증가하였다. 유리당 함량은 볶음온도와 시간이 증가할수록 감소하였고 특히 glucose함량이 0.45%에서 0.12%로 크게 감소하였다. 지방산 조성은 건 구기자에 비해 볶음온도와 시간이 증가할수록 linoleic acid의 비율이 감소하고 palmitic acid의 비율이 증가하였다. 관능검사 결과는 볶음온도와 시간이 증가할수록 단 냄새, 탄 냄새, 신 맛, 탄 맛, 떫은 맛 등은 증가하였고 단 맛과 구수한 맛은 약간 감소하였다.
