사백산(瀉白散)이 Compound 48/80에 의하여 유도(誘導)된 Anaphylatic shock와 피하반응(皮下反應)에 미치는 영향(影響) (Effects of Sabaiksan on the Compound 48/80 Induced Anaphylatic Shock and Cutaneous Reaction)

  • Sabaiksan has been prescribed to treat various allergic diseases in herbal medicine which were induced by various vasoactive amine released from the mast cells. The constituents of Sabaiksan are Mori Cortex Radices(MCR), Lycii Cortex Radicis(LCR) and Glycyrrhizae Radix(GR). Recently, simple models of compound 48/80 induced anaphylactic shock and cutaneous reaction in vivo were developed to test various agents employed in the field of allergy and toxicology research. The purpose of this study is to evaluate the effects of Sabaiksan on compound 48/80 induced anaphylactic stock, cutaneous reaction and mesenteric mast cell degranulation rate in ICR mice, and on compound 48/80 induced peritoneal mast cell degranulation and histamine release in vitro. Groups of ICR mice were intraperitoneally pretreated with $100{\mu}{\ell}$ of saline, $MCR(2g/m{\ell}),\;LCR(2g/m{\ell}),\;GR(g/m{\ell})$ or Sabaiksan itself(MCR+LCR+GR) at 24, 12 and 1 hour before compound 48/80 solution ($10{\mu}{\ell}/gm$ B. W) were peritoneally given into them, and then mortality within 72 hours after the compound 48/80 injection, and mesenteric mast cell degranulation rate at 15 minutes after compound 48/80 injection were calculated. In vitro experiment, $400{\mu}{\ell}$ of rat peritoneal mast cell suspension$(10^6cell/m{\ell})$ were pretreated with $50{\mu}{\ell}$ of saline, $MCR(2g/m{\ell}),\;LCR(2g/m{\ell}),\;GR(g/m{\ell})$ or Sabaiksan itself at room temperature for 30 minutes, and then $50{\mu}{\ell}$ of compound 48/80 solution $(100{\mu}g/m{\ell})$ were added into it. 30 minutes after the addition of compound 48/80 solution, histamine release assay in the supernatant of peritoneal mast cell suspension were performed employing radioisotope enzymatic assay and morphologic changes of mast cells in each regular time point were photographed. Compared with controls, compound 48/80 induced anaphylactic shock was significantly inhibited by MCR and GR pretreatment into the ICR mice. Significant inhibition of compound 48/80 induced cutaneous reaction, mesenteric mast cell degranulation rate in vivo and histamine release from the rat peritoneal mast cells in vitro was observed only in MCR pretreated group. From the above results, it is suggested that MCR component of Sabaiksan may playa key role to suppress mast cell function since it has been applied to various allergic diseases.

전복 복합물의 항산화능 및 피로개선 효과 (Antioxidant and anti-fatigue effects of abalone (Haliotis discus hannai) composites containing natural plants)

  • 전복 음료를 개발하기 위하여 전복에 식물류를 혼합한 전복 복합물의 in vitro 항산화능을 측정하였으며, 정상군, 대조군, 전복 복합물의 성인 1일 1회분(APM-1) 급이군(EAPM-1) 및 3회분(APM-2) 급이군(EAPM-2)으로 구분하여 4주간 실험사육하였다. 전복 복합물의in vitro 항산화능과 총 페놀 및 플라보노이드 함량은 APM-2가 유의적으로 높았다. 혈중 총 지질 함량은 대조군에 비해 운동과 전복복합물을 병행급이한 EAPM-2군이 유의적으로 감소되었다. LDH 활성 및 BUN 함량은 대조군에 비해 전복 복합물 급이군에서 유의적으로 감소되었다. AST 활성은 전복 복합물 급이 시 대조군과 유의차가 없었으며, ALT 및 ALP 활성은 대조군에 비해 유의적으로 감소되었다. 혈청 및 간 조직에서 지질과산화물 함량은 대조군에 비해 전복 복합물 급이 군에서 유의적으로 감소하였다. 간 조직의 라디칼 소거능은 대조군에 비해 전복 복합물의 급이 시 유의적으로 증가하였다. 따라서 전복 복합물은 항산화능 및 페놀 화합물에 의존하여 트레드밀 운동에 의한 산화적 스트레스 해소 및 피로 회복 에 도움이 될 것으로 예상된다.

흑임자 첨가량을 달리한 구기자흑임자죽의 품질특성 (Quality Characteristics of Gugija.Heukimja Jook Containing Different Levels of Black Sesame Powder)

  • This study was to investigated optimum preparation conditions for Gugija Heukimja jook by response surface methodology (RSM). The important materials used for the Gugija Heukimja product were Lycium chinese, Seasamum indicum L., Ziziphus jujuba Mill. and rice. Moisture content, crude protein, crude fat, crude ash and crude fiber content were 15.62, 16.60, 13.62, 4.93 and 11.17% for Lycium chinese, 3.62, 20.94, 43.56, 5.67 and 10.72% for Seasamum indicum L., 24.12, 5.38, 0.73, 2.30 and 1.25% for Ziziphus jujuba Mill., and 12.36, 6.98, 0.46, 0.43 and 0.41% for rice, respectively. Optimal extraction conditions of brix, color, pH and total acidity in the materials for Gugija Heukimja jook were temperature $80^{\circ}C$, water volume 850 mL and extraction time of 63 min. The spreadability of jook with Lycium chinese and Ziziphus jujuba Mill. extracts was the highest when black sesame was 12 g pH was the lowest when black sesame was 9 g. PH showed a significant difference when the added black sesame ratio changed. The 'value L' of chromaticity was lower with an increase of the black sesame ratio, 'value a' was higher with an increase of the black sesame ratio, 'value b' was lower with an increase of the black sesame ratio. Brix was lower when black sesame in jook was added. Total acidity was highest (7.27%) when black sesame was 12 g. The degree of gelatinization tended to be lower with additional black sesame. Higher scores of sensory evaluation were found for the product containing 9 g black sesame with Lycii furctus and Ziziphus jujuba Mill. extracts ompared to the other samples with 3 and 6g. No significant difference in taste occurred for Gugija Heukimja jook prepared with over 9 g black sesame, while a great difference showed in color, pH and total acid. Therefore, the Gugija Heukimja jook prepared under the optimal conditions of 9 g black sesame with Lycium chinese and Ziziphus jujuba Mill. extracts exhibited higher qualities. Gugija Heukimja jook can use the easier for Yacksun jook. It is expected to become functional in the product's development.

  • A Bibliographical study was done about lumbago in oriental internal Medicine. The results are as follows. 1. Shenxu,sexual active, senile, Feng, Han, Shi, shiji, Tanyin and Qi lumbago are applicable to internal medical lumbago in oriental medicine, and Shenxu is essential pathogenesis. 2. Shenxu lumbago is characterized by continous pain, improved after chiropsia, deep pulse in Chi pulse, and used QingeWan, LiuweiDihuangWan and AnshenWan in herb-medicine. and Cortex Eucommiae, Radix Rehmanniae Praeparata and Fructus Psoraleae in drugs for the purpose of ZishenYijing, and related to lumbago caused by chronic prostatitis, calculus or tumor in kidney and diabetes mellitus. 3. Sexual active lumbago is dim-aching and weak pain in or after sexual life at low back and knee joint, and classified to Shenyang Buzu, ShenjingKuisun. GanshenYinxu and XinshenBujiao ; Liuwei Dihuang Wan, ZuoguiWan In herb-medicine. Cornu Cervi Pantotrichum, Fructus Lycii and Fructus Corni in drug were used for treatment. 4. Senile Lumbago is a kind of consumptive disease due to exhaused essential energy and caused by lack of Shenjing, Yangqi ; and described as repeated mild dim pain and ErzhiWan, QingeWan and MoyaoGao were used frequently, supposed to connected with osteoporosis, osteomalacia and osteoarthropathy deformons caused by aging. 5. Feng lumbago is attacked by Liuyin, and characterized by chilling and fever. stabbing pain from thoracic and lumbar vertebrae to pedes, and used WujiXan, XiaoxumingTang for treatment. Han lumbago is caused by cold's attacking Shenjing, distinguished for chilling,icing sense improved by heat on low back, and used WujiXan, JiangfuTang. Shi lumbago is caused by damp's inflowing Shenjing, described as stone-like lumbago which was subsided low back pain growing worse by gloomy rain, and used ShenshiTang. ShenzhuTang for the purpose of ZaoshiXingqi. And it is supposed that lumbago occured in the initial of urinary track infection was belong to those of Feng,Han and Sill's. 6. Sillji lumbago is caused by pathogen being in the spleen and the stomach, and used ChenxiangJiangqiTang, PingweiSan for treatment. And it is supposed that it belong to lumbago caused by gastrointerstinal disease such as peptic ulcer. gastroduodenal tumor and colonic inflammatory disease. 7. Tanyin lumbago is caused by Tanyin's flowing meridian, characterized by thoracolumbar verterber's heaviness, covered sense with something on low back and painless massage. Kuaiqi-drug are added to ErchenTang, DaotanTang for treatment, and it supposed that Tanyin lumbago's belong to that accompanied with metabolic disease such as obesity and gout. 8. Qi lumbago is caused by excessive stress such as melancholy and fury, described as multiple stabbing pain, an unexpected on and improvement, ranging back pain until flank and abdomine. Tiaoqi-drugs were added to RenshenShunqiSan, WuyaoShunqiSan for treatment, and it supposed to connecting with emotional lumbago such as hysteria, feigned illness and anxiety psychosis.

젖산발효에 의한 혼합과채음료 제조의 최적화 (Optimization for the Lactic Acid Fermentation of Mixed Fruit and Vegetable Juices)

  • 당근, 사과, 샐러리, 돌미나리, 대추, 구기자의 착즙액을 3 : 3 : 1 : 1/2 : 1 : 1/2의 비율로 혼합한 혼합과채즙에 동치미로부터 분리한 Leuconostoc mesenteroides subsp. mesenteroides, Lactococcus lactis subsp. lactis. Lactobacillus cellobiosus을 접종하고 당과 식염의 첨가와 배양온도의 최적조건을 발효 및 관능특성에 중점을 두고 조사하였다. 올리고당을 포함한 당은 25% 이하에서, 식염은 0.8% 이하에서 젖산균의 성장률이나 산 생선능에 크게 영향을 미치지 않았다. 배양온도가 높을수록 젖산균수와 산도가 증가하고 대장균수와 당도가 빠르게 감소하였다. $15^{\circ}C$에서는 발효가 진행됨에 따라 점질물이 형성되어 발효쥬스의 선호도가 저하되었다. 따라서 혼합과채즙의 젖산발효는 관능특성면에서 fructooligosaccharide 15%, 식염 0.2%, 배양온도 $25^{\circ}C$가 최적이었고 최적 발효종점은 3일로서 이때의 pH는 3.62였다. 최적상태에서 발효쥬스는 1 mL당 젖산균수 8, 효모수 4의 지수를 보였으며 발효초기에 $5.6{\times}10^2\;cfu/mL$이던 대장균은 검출되지 않았다.

매실과 오미자 추출물의 항균성과 기능성 고추장의 제조 (Antimicrobial Activity of Prunus mume and Schizandra chinenis H-20 Extracts and Their Effects on Quality of Functional Kochujang)

  • 천연물로부터 추출한 8가지 sample에 대하여 항균성의 효능이 있는 것을 탐색하여 의약연구의 기초자료 및 현대인의 기호에 맞는 기능성 식품 개발을 목적으로 하였다. 본 연구에서 항균활성 실험에서 6mm paper disc에 대한 공시 균주에 대한 오미자와 매실은 모든 사용한 균주에 대해 6-10 mm였으며, 대추와 구기자는 1mm 정도였고 솔잎 등은 거의 보이지 않거나 0.5 mm 정도 clear zone이 생성되었다. 1차screening test에서 항균력을 보인 B. subtilis, E. coli KCCM 11591, P. aeruginosa KCTC 1750, C. albicans 균주의 성장억제도는 사용균주 농도를 희석하여 S. chinenis H-20에 대한 농도의 의존성을 10cm petri dish에서 control과 비교할 때 $50\;{\mu}L$는 5배 정도 저해하였으며, $75\;{\mu}L$에는 완전 저해효능이 있었다. 배양초기와 배양5시간에 S. chinenis H-20를 $75\;{\mu}L$ 첨가한 것에 대한 항균력은 변화가 없었다. 즉 S. chinenis H-20은 유전정보전달을 저해하는 항생물질이 작용하는 것으로 추측할 수 있다. S. chinenis H-20을 배양 5시간에 첨가하여 control과 비교하였을 때 B. subtilis, S. aureus, E. coli를 완전히 저해하였다. 매실과 오미자를 첨가한 고추장의 관능평가를 볼 때 전체평가에서 찹쌀고추장이 3.39로 '보통'으로 나타났으며, 매실고추상은 4.29로 약간 '좋다'로 오미자고추장은 2.43으로 '약간 나쁘다'로 나타났다. 찹쌀고추장을 대조군으로 할 때 매실 고추장의 견고성은 4.45배, 오미자고추장은 1.78배 높았다.

한약재 추출물의 항균효과 검색 (Screening of Antimicrobial Activity for Medicinal Herb Extracts)

  • 천연식품보존료 개발 목적의 일환으로 항균효력이 있을 것으로 추정되는 한약재 중에서 목단피, 황백, 연교, 오미자, 자초, 길경 등 20종을 대상으로 물과 95% 에탄올로 항균성물질을 추출하여 각종 미생물에 대한 항균력을 검토하였다. 물 추출물의 경우, 모든 세균에 대하여 증식억제 효과가 나타난 것은 치자, 구기자, 오미자 등의 3종이었으며, 그중에서 오미자가 증식 억제력이 제일 강하였다. 에탄올 추출물의 경우, 대부분의 세균에 대하여 증식억제 효과가 나타난 것은 치자, 오미자, 자초 등의 3종이었으며, 목단피, 황백, 금은화 등의 3종은 그람양성균에만 효과가 나타났는데, 특히 황백의 경우, S. aureus에 대하여는 0.01%의 첨가로 균의 증식이 억제되었다. 한편 이들 한약재들의 물 추출물은 진균류에 대해서는 증식억제 효과가 좋지 않았으나, 에탄올 추출물의 경우, 목단피, 황백, 치자, 자초 등의 4종이 증식억제 효과가 우수하였고, 그중 황백이 균증식 억제력이 제일 강하였다. 전 시료를 통하여 에탄올 추출물이 물 추출물 보다 $2{\sim}100$배나 항균력이 높았으며, 특히 자초의 에탄올 추출물은 맛과 향 등의 관능검사결과 이미 이취가 적을 뿐만 아니라 항균력도 우수하여 천연식품 보존료로서 이용 가능성이 높았다.

천연 생약 복합 추출물이 Streutozotocin 유발 당뇨성 흰쥐의 혈당 및 인슐린, 혈청 과산화지질 농도에 미치는 효과 (Effects of a Natural Medicinal Multi-plant Extract on Blood Glucose, Insulin Levels, and Serum Malondialdehyde Concentrations in Streptozotocin-induced Diabetic Rats)

  • This study was designed to investigate a natural medicinal multi-plants extract (BG515), which consisted of multi extracts of Mori folium, Rehmannia glutinosa Liboschitz, Dioscorea japonica, Lycii fructus, and Astragalus radix, on blood glucose, insulin levels, and serum malondialdehyde (MDA) concentrations in streptozotocin-induced diabetic rats. Streptozotocin (STZ) induces a type 1 diabetes mellitus in rats. Type 1 is usually characterized by the presence of islet cell autoantibodies (ICA), autoantibodies to insulin (IAA), and autoantiboides to glutamic acid decarboxylase (GAD), which identify the autoimmune process that leads to $\beta-cell$ destruction. Thirty-five male Sprague-Dawley (SD) rats weighing $150\sim170g$ each (6 weeks old) were randomly divided into one control (Group A) and 4 STZ-induced diabetic groups, and were subjected to one of the following treatment for 12 weeks. Groups A and B were fed basal diets and Group C, D, and E received the same diets as groups A and B, but with supplements of 150 mg/kg, 300 mg/kg, and 600 mg/kg of BG515 orally for 12 weeks, respectively. Diabetes was induced in Groups B, C, D, and E by intravenous injection of 45 mg/kg of STZ per body weight in sodium citrate buffer (pH 4.5) via the tail vein. In the BG515 groups, we found increases in serum insulin levels, compared to the STZ-control group, but these data were not significant. In contrast, blood glucose and serum MDA concentrations decreased in the BG515 groups compare to the STZ-control group. At the 5th week, in all the BG515 administered groups, there were decreases in serum blood glucose levels compared to the STZ- control group, and this activity was very strong in the BG515-1 group at the 12th week. These results suggest that natural bio-complex compounds (BG515) may slightly suppress STZ-induced changes in serum MDA concentration via the maintenance of serum insulin levels, due to the prevention of $\beta-cell$ and glucagon destruction by STZ.

전통주에 이용되는 한약재의 특성 연구 (A Study about Characteristic of the Medicinal Herbs Added in the Traditional Korean Liquor)

  • Objective : The aim of this study is to analyze medicinal herbs, the ingredients of traditional Korean liquor, focusing on efficacy in the traditional Korean medicine. Through this study, for vitalizing traditional Korean liquor market, the researchers attempt to explain the superiority of traditional Korean liquor and the advantages of using herbs to brew alcoholic beverages. Method : We conducted a research targeting traditional Korean liquors in domestic market from July through september 2012, and finally 70 kinds of liquor were selected and analyzed. Results : Among traditional liquors, fermented liquors are most common. When comparing regions of brewery, the central region of Korea, Gyeonggi-do and Chungcheongnam-do, is major brew area. 37 liquors contain just one medicinal herb. followed 10 liquors contain two medicinal herbs. The most frequently used medicinal herb is Ginseng Radix. And Lycii Fructus, Schisandrae Fructus, Angelicae Gigantis Radix and Chrysanthemi Indici Flos are well used medicinal herbs to brew traditional liquors. In the classification of efficacy of medicinal herbs, tonifying and replenishing medicinal herbs are most popular as ingredients in the traditional alcohols, astringent medicinal herbs, exterior releasing medicinal herbs and heat clearing medicinal herbs are also frequently used. Investigation of tastes and properties of the herbs, it reached a conclusion as below: sweet taste is most common, followed by sour taste and bitter taste. Warm property is account for 50% approximately, followed by cold property. Conclusion : Medicinal herbs have been widely used in the traditional Korean liquor. It came out into the open that the liquor with medicinal herbs is very good for our body in terms of longevity and invigoration. However, liquor consumers concern about not only the efficacy but also the taste, scent and color. Therefore, further study about these things would be required to demonstrate the superiority of the traditional Korean liquor.

원산지별 국내 유통 한약재의 이산화황 잔류실태 조사 (Monitoring of Sulfur Dioxide Residues in Commercial Herbal Medicines at Domestic by Geographical Origins)

  • 국내에 유통되고 있는 국산과 수입산 한약재 11종 116건을 대상으로 잔류이산화황에 대한 모니터링을 수행한 결과 108건(93.1%)은 불검출이었고 6건(5.2%)는 이산화황 허용기준치 30 mg/kg을 초과하였고 2건은 30 mg/kg 미만으로 검출되었다. 116건의 이산화황 평균함량은 $17.6{\pm}144.2mg/kg$이고 최대 검출량은 구기자(1,546.3 mg/kg)이었고 중국산 현호색(66.6%)의 이산화황 검출 빈도가 가장 높게 나타났다. 국내산 한약재는 71건 중 1건(1.4%), 수입산 한약재는 45건 중 7건(15.6%)에서 이산화황이 검출되었고 국내산 1건(36.7 mg/kg)과 수입산 5건(118.1 mg/kg)이 허용기준치를 초과하여 국내산보다 수입산 한약재에 이산화황 잔류량이 많았다. 본 연구를 통해 이산화황 허용기준을 초과하는 부적합율은 다소 낮아지고 있지만 이산화황 허용기준을 초과하는 한약재들 중 일부 한약재에서는 잔류이산화황 함량이 높게 나타났고, 특히 국내산 약재에 비해 수입산 약재는 부적합율이 높게 나왔다. 한약재는 일반적으로 수세 및 가열과정을 거치므로 실제 섭취하는 형태의 탕액에서는 원재료보다 이산화황의 잔류량이 크게 감소하지만 완전히 제거되지는 않으므로 유통되는 한약재의 안전성 확립이 절실히 요구되어진다. 강화된 기준이 정착하여 안전한 한약재가 유통될 수 있도록 수입통관시 특별관리가 요구되며, 보다 적극적인 검사 요주의 품목에 대한 지속적인 잔류이산화황 모니터링이 필요할 것으로 사료된다.
