It has been suggested that oxygen-derived free radicals play an important role in the pathophysiology of acute gastric ulceration induced by NSAIDs and ischemia-reperfusion. Taurine is hypothetized to exert its protective effect on NSAIDs-induced gastric injury by its antioxidant properties. Protective effect of taurine on indomethacin-induced gastric mucosal lesion and its protection mechanism were investigated. Intragastric administration of 25 mg/kg of indomethacin induced hemorrhagic lesions on the glandular stomach in rats. Pretreatment with 0.25 or 0.5 g/kg of taurine one day before or for 3 days significantly reduced the gastric lesion formation and inhibited the elevation of lipid peroxide level in gastric mucosa. The luminol-dependent chemiluminescence of rat peritoneal neutrophils increased immediately after treatment of FMLP or indomethacin. Taurine (5-20 mM) inhibited chemiluminescence of neutrophils activated by FMLP. Human neutrophils (polymorphonuclear leukocytes) significantly adhered to the confluent monolayer of human umbilical vein endothelial cells (HUVEC) after coincubation with indomethacin. This neutrophil adhesion induced by indomethacin to HUVEC was prevented by taurine in a dose-dependent manner. These results indicate that the protective effect of taurine against NSAIDs-induced gastric mucosal injury is due to its antioxidant effect, which inhibits lipid peroxidation and neutrophil activation.
The objective of this study was to investigate the sonophysical and sonochemical effects induced by acoustic cavitation in heterogeneous systemin a 28 kHz double-bath reactor using calorimetry, the aluminiumfoil erosion test, and the luminol test. With no glass beads, calorimetric power in the inner vessel increased as much as the outer sonoreactor lost and total calorimetric power was maintained for various liquid height conditions (0.5 ~ 7 cm) in the vessel. Higher calorimetric energy was obtained at higher liquid height conditions. Similar results were obtained when glass beads were placed with various beads heights (0.5 ~ 2.0 cm) and relatively high calorimetric energy was obtained in spite of large attenuation in the glass beads layer. An aluminium foil placed between the bottom of the inner vessel and the glass beads layer was damaged, indicating significant sonophysical effects. Much less damage was detected when the foil was placed above the beads layer due to large attenuation of ultrasound. Sonochemical effects, visualized by sonochemiluminescence (SCL), also decreased significantly when the beads were placed in the vessel. It was established that the optimization of the liquid height above the solid-material layer could enhance the sonophysical and sonochemical effects in the double-bath sonoreactors.

In this study, we used a chemiluminescence reaction to investigate the removal effect of pet soil, such as dog blood, urine and feces. The soiled fabrics were washed with a standard laundry course of 30℃ and a washing time of 30 min and a pet care laundry course of 40-60℃ and a washing time of 100 min. The detergency was evaluated by the surface reflectance and chemiluminescence reaction (bloodstain detection by luminol test and urine-stain and feces-stain detection by UV blacklight test) before and after washing. The surface reflectance results did not show any difference in detergency for both courses, whereas the chemiluminescence reaction did. The detergency of the pet care course compared to the standard course was 101% according to the surface reflectance and 120% according to the chemiluminescence reaction. Therefore, residual stains not detected by surface reflectance can be evaluated through chemiluminescence reaction, and it was confirmed that pet stains can be managed more hygienically by washing for a long time at a high temperature.
Since the typical horn-type ultrasonic equipment induces a reaction at the probe tip, the sonochemical reaction has a limitation that it occurs only in a specific area. As one of the ways to overcome this limitation, an ultrasonic device with multi-stepped horn equipped with several oscillators has been developed. The objective of this study was to investigate the sonochemical effects induced by acoustic cavitation system in 20 kHz multi-stepped ultrasonic horn using calorimetry, KI dosimetry and the luminol test. The sonochemical effects of multi-stepped ultrasonic horn were compared with that of the typical horn-type 20 kHz ultrasonic device. The effect of immersion depth and power on the sonochemical reaction was investigated in the ultrasonic system with multi-stepped ultrasonic horn. Higher calorimetric energy was obtained at higher immersion depth and power conditions. Sonochemical effects increased significantly when using the high immersion depth and input power. However, as the input power increased, the cavitation reaction zone concentrated around the ultrasonic horn. Additionally, the experiments to examine the effect of liquid temperature was conducted. The smaller sonochemical reaction was obtained for the higher liquid temperature. The effect on temperature seems to be closely related to liquid conditions such as viscosity and vapor pressure of water.
A molecularly imprinted polymer (MIP) was synthesized by radiation-induced polymerization (RIP), where the ferulic acid was used as a template molecule, 4-vinylpyridine as a monomer and ethylene glycoldimethacrylate (EGDMA) as a cross-linking monomer. The MIP was packed in a glass column using a slurry method for use in medium pressure liquid chromatography (MPLC). The MPLC column was tested for separation and purification of ferulic acid from the rice oil. When repeated three times, the MPLC separation/purification yielded the ferulic acid with the purity higher than ~99%. The chemiluminescence of the luminal (5-amino-2,3-dihydro-1,4-phtalazinedione) measured on a potato disc slide (5.0 mm thick) was enhanced in the presence of ferulic acid, while, without the ferulic acid, the chemiluminescence of luminol on the potato slice disc was not observed, which suggests the ferulic acid obtained from the rice oil can be useful for immunoassay.
Many herbs have been used as therapeutics in Korean traditional medicine. In view of their clinical indications, anti-oxidant activity may contribute to their pharmacological effects. However, anti-oxidant information on these plants has not been available. In this study, seventy herbs which have been used in Korean traditional medicine were selected and screened for anti-oxidant activity using their water extracts. The anti-oxidant activity was assessed by their ability to inhibit three oxidation reactions; luminol/Fenton reagent, 2, 7-dichlorodihydrofluorescein(DCHF)/Fenton reagent and DCHF/peroxynitrite. In each assay, 70 herbs were divided into two groups; anti-oxidant group which inhibited the respective oxidation reaction and was majority(about 60 herbs), and pro-oxidant group which enhanced the oxidation reaction but was minority(more or less 10 herbs). When the herbs were listed in the order of their anti-oxidant strength, the orders obtained from each assay were found to be quite similar. The upper top rankers(more or less 10 herbs) in each assay showed strong activity compared to the others. The uppermost rankers in each assay were Rubus coreanus Miquel/Rubus schizostylus(覆盆子), Schisandra chinensis Baillon/Schizandra chinensis(五味子) and Terminalia chebula Retziusl Terminalia chebula(訶子). Of the pro-oxidant herbs, about 4-5 herbs were strongly pro-oxidant, which enhanced the control oxidation reactions to 150-300%. But the meaning of this observation is not known since few of them in one assay were also anti-oxidant in other assays. The results obtained in the present study may serve as information for understanding pharmacological effects of these herbs and developing new drugs from them.

Although neutrophils function in both defense and tissue destruction, their defensive roles have rarely been studied in association with periodontitis. We hypothesized that peripheral neutrophils are pre-activated in vivo in periodontitis and that hyperactive neutrophils would show enhanced phagocytic ability as well as an increased production of reactive oxygen species (ROS). Peripheral blood neutrophils from patients with aggressive periodontitis and age/gender-matched healthy subjects (10 pairs) were isolated. The levels of CD11b and CD64 expression on the neutrophils and the level of plasma endotoxin were determined by flow cytometry and a limulus amebocyte lysate test, respectively. In addition, neutrophils were subjected to a flow cytometric phagocytosis assay and luminol-enhanced chemiluminescence for non-opsonized Fusobacterium nucleatum in parallel. The neutrophilsfrom most patients expressed increased levels of both CD11b and CD64. In addition, the plasma from these patients tended to contain a higher level of endotoxin than the healthy controls. In contrast, no differences were found between the two groups with regard to phagocytosis or ROS generation by F. nucleatum. The ability to phagocytose F. nucleatum was found to positively correlate with the ability to produce ROS. In conclusion, peripheral neutrophils from patients with aggressive periodontitis are hyperactive but not hyperreactive to F. nucleatum.
The rapid and spontaneous interaction between superoxide anion radical and nitric oxide to yield the potent oxidants. peroxynitrite artion and peroxynitrous acid, was investigated in phorbol myristate acetate(PMA)-stimulated RAW264.7 macrophases by means of lucigenin- or luminol-enhanced chemiluminescence method. When RAW264.7 macrophages were stimulated by PMA. peroxynitrite-induced chemiluminescence was clearly observed. To prove observed chemiluminescencc due to the reaction between superoxide anion radical and nitric oxide produced by RAW264.7 macrophases, N-nitrosoglutathione (GSNO), a nitric oxide-releasing compound. superoxide dismutase(SOD), an enzyme removing superoxide anion radical by dismutating superoxide artion radical to hydrogen peroxide, and N-acethyl cysteine(NAC), a scarvenging reagent both superoxide artion radical and nitric oxide, were added in the cell system. Peroxynitrite- induced chemilumincscence was increased by exogenous addition of GSNO. whereas observed chemiluminescence was decreased by SOD and NAC. These results suggest that PMA-stimulated RAW264.7 macrophages produce both superoxide anion radical and nitric oxide to form peroxynitrite.
The most common sources of biological trace material which are found in crime scene are the human bloodstains. Reliable identification in the forensic casework is important as it provides crucial insights into crime scene reconstruction and can thus contribute towards solving crimes. Blood-stains are routinely tested in forensic practise using various methods including the leucomalachite green (LMG) test, Kastle-Meyer phenolphthalein test, tetramethylbenzidine test, orthotolidine test, or the luminol chemoluminescence test with the latter cleaning attempts. All these presumptive thus indicative but not identifying tests take advantage of the peroxidase-like activity of the heme unit of the hemoglobin molecule in human blood. Therefore, false-positive results can be caused by the presence of strong oxidants, such as chlorine-containing detergents or by true peroxidases (e.g., from plants). In this study, composition for Gum guaiac was evaluated for the forensic identification of bloodstain and compared with the LMG. The sensitivity and specificity of the composition for Gum guaiac were examined more stable in bloodstain. The positive of Composition for Gum guaiac shown even with the 100,000-fold diluted bloodstain, which was no difference in comparison with LMG test. It was shown that composition for Gum guaiac was very stable to resist boiling for 20 minutes and the effect of bacteria did not affect the genetic analysis as well. The above result of the crime scene investigation, composition for Gum guaiac is easily expected to help identifying bloodstain in the evidences.
In this study, the antibacterial activity and the antioxidative effects, inhibitory effects on tyrosinase of Lespedeza cuneata G. Don extracts were investigated. MIC value of ethyl acetate fraction from L. cuneata G. Don on P. ovale (0.125%) showed that the antibacterial activity of the ethyl acetate fraction was higher than methyl paraben. The aglycone fraction of L. cuneata G. Don (14.63 ${\mu}g$/mL) showed the most prominent the free radical (1,1-diphenyl-2-picryl-hydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of L. cuneata G. Don fraction on $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The aglycone fraction of L. cuneata G. Don (0.07 ${\mu}g$/mL) showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of L. cuneata G. Don on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The L. cuneata G. Don extracts suppressed photohemolysis in a concentration dependent manner (1 ~ 50 ${\mu}g$/mL). The inhibitory effects ($IC_{50}$) of L. cuneata G. Don extracts on tyrosinase were determined with ethyl acetate fraction (104.83 ${\mu}g$/mL) and aglycone fraction (27.55 ${\mu}g$/mL) of L. cuneata G. Don extract. These results indicate that L. cuneata G. Don extract/fractions can function as high potential as bactericide against the pathogenic bacteria and antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Extract/fractions of L. cuneata G. Don could be applicable to new functional cosmetics for antiaging, antioxidant, and antibacterial activity.
