A Study on the Optimum Stocking Density of Abalone, Haliotis discus hannai, reared in Net Cage Culture (북방전복, Haliotis discus hannai의 해상가두리 양성 시 적정 수용밀도에 관한 연구)

  • The effects of different stocking densities on the growth and survival rate of the abalone, Haliotis discus hannai, were investigated in marine net cage for two years. Stocking density was set 15, 30, 45 and 60 percentage $(=per.)/m^2$ with share to cross-sectional area per shelter. The primary rearing period (PRP) and the secondary rearing period (SRP) were conducted by a year. One year mean water temperature of PRP and SRS showed the difference about $2^{\circ}C$. In the growth (initial mean shell length of abalone : $36.14{\pm}2.28mm$) of PRP, the absolute growth rate (ARG), daily growth rate (DGR) and specific growth rate (SGR) of the $15per./m^2$ were higher than those of density groups (P < 0.05). Survival rates of all density groups were showed no significant difference. In the growth (mean shell length of abalone : $55.26{\pm}6.93mm$) of SRP, ARG, DGR and SGR of stocking density groups showed no significant difference except for $45per./m^2$ density group. Survival rate in the low-density (15, $30per./m^2$) was more than 70%, and those of the high-density (45, $60per./m^2$) were less than 31% and 9%, respectively. These results showed that the appropriate stocking density for $15per./m^2$ was seven hundred fifty number per one net cage ($2.4{\times}2.4m$), during PRP using 3-4 cm abalone in length. Also for the secondary rearing period, the optimal stocking density (shell length 5-6 cm of abalone) consider with the economical efficiency was determined to be $30per./m^2$, resulting the productivity improved.

Extraction & Purification of ${\beta}$-carotene from Recombinant Escherichia coli (재조합 대장균으로부터 고순도 베타-카로틴의 추출 및 정제)

  • This paper aimed to develop a solvent extraction and purification process to recover high-purified ${\beta}$-carotene from recombinant Escherichia coli. Cells harvested from the culture broth were treated through numerous steps: dehydration, solvent extraction, crystal formation and separation. To optimize the extracting condition, experiments were carried out to investigate the effect of cell disruption, temperature, organic solvents, solvent-biomass ratio on the yield of ${\beta}$-carotene extracted from cells. The result indicated that no significant differences of extraction yield were observed from cells with or without step of cell disruption. Among different extracting solvents, the highest extraction yield of ${\beta}$-carotene, 30.3 mg-${\beta}$-carotene/g-dry cells, was obtained with isobutyl acetate at solvent-biomass ratio 25 mL/g-dry cells at $50^{\circ}C$. Notably, in case of acetone, the extraction yield was quite low when using acetone itself, but increased almost up to the highest value when combining this solvent and olive oil. The purity of ${\beta}$-carotene crystals obtained from crystallization and separation was 89%. The purity degree was further improved up to 98.5% by treating crude crystals with additional ethanol washing.

Study on the Environmental Factors and Symptoms of VDT Syndrome (VDT 증후군의 환경적 요인과 증상에 대한 연구)

  • Purpose: Recently incidence of VDT syndrome has gradually increased as extensive use of computers. VDT syndrome reported by VDT workers include musculoskeletal disorder, neuropsychiatric disoders and eye symptoms such as eye strain, tired eyes, irritation and blurred vision. The environmental factors of VDT syndrome include electromagnetic waves, size, brightness and lighting of computer screen, height of a monitor and a worktable, working hours, kind of task, distance between screen and workers, indoor humidity and temperature, indoor air contamination and ventilation. In this study, we investigated the environmental factors related to body symptoms and health effects included in VDT syndrome. Methods: Study subjects were total 120 persons (54 male, 66 female) with age from 19 to 28. We surveyed the body symptoms and physical discomfort when doing an activity in a short distance such as reading book or paper, computer work. The questionnaire included main body symptoms, self-consciousness symptoms of eye, satisfaction of working environment, pain of the wrist when using keyboard and mouse. Results: Most of people (70%) felt physical pain from long time work of computer, paper, electrical apparatus. They mainly complained pain of neck and low back (57.1%), eye (45.2%) and head (31%). With the environmental factors, 78.3% of the subjects complaint pain of eye from inappropriate illumination. Most of the symptoms included 'eye fatigue'(38.3%), 'dryness of eye'(31.9%) and 'blurred vision'(23.7%). Subjects in this study complained discomfort of their chairs and most of them experienced pain in the wrist when using keyboard or mouse. Conclusions: When people use electrical apparatus or work with paper, people would get their eye fatigue and feeling of physical fatigue because of not harmonizing various environmental factors such as light, space, posture, worktable with theirselves. Therefore, workers should develop preventive method such as self-control of adequate break time to avoid fatigue while VDT work. Work environment should be changed to ergonomic design for optimal visual environment to prevent musculoskeletal disorder through constant research.

Proficiency Testing for the Gas-chromatographic Analysis of Procymidone, Chlorpyrifos and Metolachlor Residues in Soil (가스크로마토그래피를 이용한 토양 중 프로사이미돈과 클로르피리포스, 메톨라클로르의 잔류분석 숙련도시험)

  • The proficiency testing for the residue laboratories of pesticide registration was conducted in order to improve the reliability and the ability for pesticide residue analysis. On October 2011 the testing was carried out using the soil collected and kept as the moistened state for five years, which is expected to very low residue levels of pesticides. The soil was fortified with chlorpyrifos, metolachlor and procymidone in a manner similar to prepare soil sample for indoor soil degradation test, and then sub-samples were prepared for the distribution to participants. Some of them were randomly selected for confirm of homogeneity and to ensure the stability of samples at room temperature. Samples were consisted of two soil treated as different levels, one of which was used to the assesment and another used to confirm. In addition, provide three standard solutions, respectively concentration of 10 mg/L, and untreated soil. Forty seven institutions submitted results. The medians of results were used as the assigned values for pesticide residues. Fitness for purpose standard deviation of proficiency test was calculated by applying 20% RSD as the coefficient of variation allowed in the soil residue test. Z-score was applied for evaluation of individual pesticides, and the average of the absolute value of the Z-score for the overall assessment of pesticides. Laboratories evaluated the absolute value of the Z-score less than 2 to fit the case of chlorpyrifos and procymidone were 44, metolachlor 40.

Spatial Distribution of Pigment Concentration Around the East Korean Warm Current Region Derived from Satellite Data - Satellite Observation in May 1980 - (위성원격탐사에 의한 동한난류 주변 해역의 색소농도 공간적 분포 -1980년 5월 관측을 중심으로 -)

  • Spatial distribution of Phytoplankton Pigment Concentration (PPC) and Sea Surface Temperature (SST) around the East Korean Warm Current (EKWC) was described, using both Coastal Zone Color Scanner (CZCS) images and Advanced Very High Resolution Radiometer (AVHRR) images in May, 1980. Water mass in this region can be classified into five categories in the horizontal profile of PPC and SST, nLw (normalized water-leaving radiance) images: (1) coastal cold water region associated with concentrations of dissolved organic material or yellow colored substances and suspended sediments, (2) cold water region of thermal frontal occurred by a combination of phytoplankton absorption and suspended materials, (3) warm water overlay region by the phytoplankton absorption than the suspended materials; (4) warm water region occurred by the low phytoplankton absorption, and (5) offshore region occurred by the high phytoplankton absorption. In particular, the highest PPC (>2.0 mg/m^3) area appeared in the CZCS and AVHRR images with a band shaped distribution of the thermal front and ocean color front region, which is located the coastal cold waters alonB western thermal front of the warm streamer of the EKWC. In this region, the highest PPC occurred by a combination of the high absorption of the phytoplankton (443 nm) and highest reflectance of suspended materials (550 nm). Another high PPC ($\simeq$$6\;mg/m^3$) appeared in the warm water overlay region inside warm streamer. High phytoplankton pigment concentration of this region was corresponding to the short wavelength of 443 nm, which represented phytoplankton absorption of the CZCS image.

Seasonal Distribution of Water Masses and Spatio-temporal Characteristics of Nutrients in the Coastal Areas of Gangwon Province of the Korean East Sea in 2009 (2009년 강원 연안의 계절별 수괴 분포 및 영양염의 시공간적 특성)

  • In order to investigate the distribution of water masses and spatio-temporal variation of nutrients in the coastal areas of Gangwon province of the Korean East Sea, a survey of the physico-chemical parameters (temperature and salinity) and nutrients ($NO_2$-N, $NO_3$-N, $NH_4$-N, $PO_4$-P, and $SiO_2$-Si) was carried out at 5 locations (Goseong, Sokcho, Yangyang, Gangneung, and Donghae) in February, May, August, and November 2009. The water masses included in the study area were divided into 4 groups; 1) Tsushima Surface Water (TSW), 2) Tsushima Middle Water (TMW), 3) North Korean Cold Water (NKCW), and 4) East Sea Proper Water (ESPW). The distribution of water masses was affected by the change of season. In February, surface water was derived from the TMW. The TSW was not observed in May, but only observed in August. In November, as the influence of the TSW weakened, that of the NKCW strengthened. Considering the vertical profiles of nutrients, the concentrations in all the seasons were very low within the surface water, but increased rapidly near the thermocline. Most of nutrient concentrations, except for dissolved silicate, remained constant below the depth of 200 m. However, the dissolved silicate concentration increased with depth, suggesting that silicate has a delayed regenerative pattern. The ESPW had the highest nutrient concentration, followed by the NKCW, TMW, and TSW. In February, May, and November, the N/P ratio in most of the water masses was similar to or larger than the Redfield ratio, indicating that nitrogenous nutrients did not act as a limiting factor for phytoplankton growth. However, in August, the N/P ratio in the TSW was less than the Redfield ratio, and the concentration of $NO_2$-N+$NO_3$-N was 0.86 ${\mu}m$, indicating that nitrogenous nutrients did act as a limiting factor for phytoplankton growth in the study area.

Assessing Future Climate Change Impact on Hydrologic Components of Gyeongancheon Watershed (기후변화가 경안천 유역의 수문요소에 미치는 영향 평가)

  • The impact on hydrologic components considering future potential climate, land use change and vegetation cover information was assessed using SLURP (Semi-distributed Land-Use Runoff Process) continuous hydrologic model. The model was calibrated (1999 - 2000) and validated (2001 - 2002) for the upstream watershed ($260.4\;km^2$) of Gyeongancheon water level gauging station with the coefficient of determination and Nash-Sutcliffe efficiency ranging from 0.77 to 0.60 and 0.79 to 0.60, respectively. Two GCMs (MIROC3.2hires, ECHAM5-OM) future weather data of high (A2), middle (A1B) and low (B1) emission scenarios of the IPCC (Intergovernmental Panel on Climate Change) were adopted and the data was corrected by 20C3M (20th Century Climate Coupled Model) and downscaled by Change Factor (CF) method using 30 years (1977 - 2006, baseline period) weather data. Three periods data of 2010 - 2039 (2020s), 2040 - 2069 (2050s), 2070 - 2099 (2080s) were prepared. To reduce the uncertainty of land surface conditions, future land use and vegetation canopy prediction were tried by CA-Markov technique and NOAA NDVI-Temperature relationship respectively. MIROC3.2 hires and ECHAM5-OM showed increase tendency in annual streamflow up to 21.4 % for 2080 A1B and 8.9 % for 2050 A1B scenario respectively. The portion of future predicted ET about precipitation increased up to 3 % in MIROC3.2 hires and 16 % in ECHAM5-OM respectively. The future soil moisture content slightly increased compared to 2002 soil moisture.

Fertilizer demand estimated in aspect of crop nutrition (작물영양면(作物營養面)에서 본 비료(肥料)의 수요전망(需要展望))

  • Fertilizer(N,P,K) demand for crop production in 1980, 1990 and 2000 was estimated according to the two proposed models, one of which is fertilizer use efficiency model expressed in $Fn=(Y/E){\cdot}(1-Cs)Eu$, where Fn:fertilizer demand, Y:Crop production estimated, E:nutrient efficiency, Cs:fraction of natural resource nutrient in plant, Eu:fertilizer use efficiency and the other fertilization efficiency model expressed in Fn=Y(1-Cys)/Fe, where Cys:fraction of yield without fertilizer, Fe:fertilization efficiency. Total crop uptake of nutrient and its noncycling portion were estimated as criteria for fertilizer demand and nutrient maintenance. Total crop uptake of N,P,K was 600,000 M/T in 1965 700,000 M/T in 1974 and estimated to 880,000 M/T in 1980, 1,170,000 M/T in 1990 and 1,410,000 M/T in 2000. Fertilizer demand appeares to be about 90% of total crop uptake according to fertilizer use efficiency model and about 87% according to fertilization efficiency model. The noncycling nutrient was about 29% of total crop uptake. Fertilizer demand was almost same to the uptake amount in nitrogen, 1.5 times of uptake in phosphorus and half of uptake in potassium. Varietal development, improvement of soil fertility and cultivation method and development of fertilizer forms appears to decrease fertilizer demand by increasing efficiency term in two models while environmental stress such as low temperature appears to give reverse effect resulting in higher fertilizer demand. Fertilizer consumption in 1974 seemed to be unreasonably high especially in nitrogen and phosphorus and thus the effective use of fertilizer appeared as an urgent problem considering that large fields are still remained in lower fertility.

Comparison of Growth of the Pacific Oysters, Crassostrea gigas, Cultured with Korean and Japanese Spats (한국산 및 일본산 굴, Crassostrea gigas 종패의 성장비교)

  • To study the growth of transplanted Pacific oysters, Crassostrea gigas, we sampled Korean and Japanese oysters attached in Chinhae Bay near Gaduk Island and in Seto inland sea in Japan, respectively, suspended in Pukman Bay. Water Temperature ranged from 11.2 to 27.8$^{\circ}C$ (mean 19.84 ${\pm}$ 5.47$^{\circ}C$) on the surface, and 11.1 to 23.6$^{\circ}C$ (mean 18.31 ${\pm}$ 4.18$^{\circ}C$) on the bottom. Salinity ranged from 31.45 to 34.57 (mean 33.10 ${\pm}$ 1.16) on the surface, and from 31.69 to 34.35 (mean 33.24 ${\pm}$ 1.06) on the bottom. salinity was the lowest in September and October, and the highest in December. Growth of oysters in shell height showed a significant difference after being suspended at the farm, reaching 70.3 ${\pm}$ 12.5 mm in the Korean oysters and 96.2 ${\pm}$ 14.6 mm in the Japanese oysters in December. While the Korean oysters showed relatively low growth rate and cessation of growth after sudden growth between June and July, the Japanese oysters showed continuous growth during the whole farming period, although stepwise growth was observed. It was not until September that meat weight showed a significant difference between the two. After September, there was a sudden increase in the Japanese oysters, reaching 7.5 ${\pm}$ 2.9 g in December, but growth of the Korean oysters showed slow growth rate during whole farming period, reaching 4.6 ${\pm}$ 1.9 g in December. here was an obvious decrease in the meat weight of Japanese oysters in December, which might be attributed to restriction of food. Condition factors rebounded in October in the Korean oysters and in September in the Japanese oysters, respectively, attaining 12.8 in the Korean oysters and 15.3 in the Japanese oysters at the end of investigation on December. Shell length-height regression equations were as follows: Korean oysters: S$\sub$h/=2.922S$\sub$t/,-4.8024 (r$^2$= 0.8541) Japanese oysters: S$\sub$t/=3.623S$\sub$h/,-5.1239 (r$^2$=0.7782) This showed the possibility of morphological transformation in the shell of the Korean oysters since shell height was longer than those reported by Bae et al. (1976) and Lee et al. (1992).

Reproductive Cycle of Ribbed Gunnel Dictyosoma burgeri (그물베도라치 Dictyosoma burgeri의 생식주기)

  • The morphology of gonad and reproductive cycle of ribbed gunnel (Dictyosoma burgeri) were investigated on the basis of histological observation. The specimens were monthly sampled in the coastal waters of Jeju from November 2001 to February 2003. The ovaries and testis of this species are categorized as cystovarian and lobule type, respectively. The gonadosomatic index (GSI) of female increased in November and maintained high values from December to February. The GSI of male was similar to that of female although it was decreased in February. The reproductive cycle can be grouped into the following successive stage in the ovary: growth (October to November), mature (November to February), spawning (January to February), and degenerating and recovery (March to September). And in the testis, the stage observed were: multiplication (August to November), growth (November to January), mature and spawning (November to February), and degenerating and recovery (January to September). The minimum maturation size of D. burgeri was over 15.0 cm and fecundity ranged from 2,194 to 6,581 eggs. The relationship between the fecundity and fish body was calculated in the fecundity (F) equation as: $F=0.4057TL^{3.1425}$ ($R^2=0.7621$) for total length (TL); $F=149.88BW^{0.9579}$ ($R^2=0.7982$) for body weight (BW), respectively. The fecundity was correlated positively with TL and BW. The histological observations of the gonads suggested that major spawning of this species probably occurs between January to February, when low water temperature ($13{\pm}0.3^{\circ}C$) period.
