The Role of Lipid Peroxidation and Glutathione on the Glycochenodeoxycholic Acid-Induced Cell Death in Primary Cultured Rat Hepatocytes

  • Intracellular accumulation of bile acids in the hepatocytes during cholestasis is thought to be pathogenic in cholestatic liver diseases. The objective of this study was to determine the role of lipid peroxidation and glutathione on the bile acid-induced hepatic cell death mechanism in primary cultured rat hepatocytes. To induce hepatic cell death, we incubated primary cultured rat hepatocytes with glycochenodeoxycholic acid $(GCDC;\;0{\sim}400\;{\mu}M)$ for 3 hours. In electron microscopic examination and agarose gel electrophoresis, low concentration of GCDC treatment mainly induced apoptotic feature. Whereas $400\;{\mu}M$ GCDC treated cells demonstrated both apoptosis and necrosis. Lipid peroxidation was increased dose-dependently in GCDC treated hepatocyte. And this was also accompanied by decreased glutathione. Therefore, oxygen free radical damage may play a partial role in GCDC-induced hepatic cell death.

Pathogenicity of viral hemorrhagic septicemia virus (VHSV) isolated from olive flounder Paralichthys olivaceus to masu salmon Oncorhynchus masou

  • The pathogenicity of viral hemorrhagic septicemia virus (VHSV) from olive flounder Paralichthys olivaceus was investigated with masu salmon Oncorhynchus masou fry. The cumulative mortality of fish challenged with FYeosu05 isolate at $10^{6.5}$ $TCID_{50}$/fish was 60%. No mortality was observed in fish challenged with the isolates at $10^{5.5}$ $TCID_{50}$/fish and in mock-challenged fish. The affected fish showed darkening of the body, expanded abdomen, pale gills and enlarged spleen. VHSV from $10^{6.3}$ to $10^{7.8}$ $TCID_{50}$/g-tissue was re-isolated from the dead fish. These results suggest that the VHSV from olive flounder is pathogenic to masu salmon fry, although with low virulence.

Occurrence of Bacterial Soft Rot of Lily Bulb Caused by Pectobacterium carotovorum subsp. carotovorum and Pseudomonas marginalis in Korea

  • Soft rot symptom was observed on lily bulb in the fields and at a low temperature storage house from 1999 to 2000 in Korea. The small dark-brown lesion appeared on the bulb, and enlarged and developed into the inner scales of the bulb. The bulb became water soaked and gave out unpleasant odor. Two different pathogenic bacteria were isolated from infected tissues. The causal bacteria were identified as Pectobacterium carotovorum subsp. carotovorum (Erwinia carotovora subsp. carotovora) and Pseudomonas marginalis based on bacteriological characteristics. Pathogenicity of the bacteria was proven by Koch's postulations. This is the first report of bacterial soft rot of lily bulb in Korea caused by the two bacteria.

  • Alternaria longissima Deighton & Macgarvie was recorded for the first time on seeds of sesame in low percentages. Detailed descriptions of the habit characters, conidial morphology and cultural characters are described. This fungus, which is generally regarded as a saprophyte, was shown to be a pathogen to sesame plants producing zonate leaf spots, foliage blight, stem necrosis and spots on capsules, but does not show any pathogenicity to rice and sorghum plants.

  • The tetrahydropahnatine alkaloid was assayed against spore germination of some saprophytic and pathogenic fungi e.g., Alternaria solani, A. brassicicola, A. brassicae, A. alternata, Erysiphe pisi, Curvularia lunata, C. pallescens, C. maculans, Curvularia species, Colletotrichum species, C. musae, Helminthosporium echinoclova, H. pennisetti, H. spiciferum, and Heterosporium sp. It inhibite spore germination of all the fungi tested. Colletotrichum spp. Curvularia lunata, Helminthosporium spiciferum and Heterosporium sp. were most sensitive as complete inhibition of spore germination was observed at very low concentration(200 ppm).

  • Erythema nodosum is a nodular erythematous eruption usually limited to the low extremities. Erythema nodosum seems to be Gwadungjun(瓜藤纏) and Damp-heat flow(濕毒流注) in oriental medicine. The symptom of erythema nodosum is similar to that of Gwadungjun, so we investigated the literatures of oriental medicine and recet medicine. The results were summarized as follows; 1. In Oriental medicine, erythema nodosum regarded as Damp-heat flow and Gwadungjun. 2. Inflammatory nodus of lower extremity, ie, similar to Gwadungjun, Damp-heat flow, Bichonbal(비천發), Samnibal(三里發), Ududok(魚두毒), etc. 3. Erythema nodosum results from inner damp-heat(內有濕熱), affection due to pathogenic wind(外感風邪). 4. Treatment is to clear away heat(淸熱), remove dampness(利濕), heat from blood(凉血), cold-damp(祛寒), and promote blood flood(活血通絡). 5. We can use Bangpungtongsungsan(防風通聖散), Sopunghwa1hyultang(疏風活血湯), and Danguijumtongtang(當歸拈通湯), etc.

Preliminary Study on the Recycled Composting System for Reducing Bulking Agent Cost (부자재 비용 저감을 위한 순환 퇴비화 시스템의 예비연구)

  • This study was performed to find the recycling performance and to provide design data during recycle solid composting system to reduce bulking agent cost, Dairy manure amended with rice hulls and recycled compost was composted in a laboratory -scale vessel by continuous aeration for 10 days. The temperature and ammonia emission variations according to the ratios of bulking agents during the primary aeration stage were surveyed. Also, the influence of fresh compost quality on the recycling performance were anlayzed. While recycled composting system were operatured. The temperature in compost was maintained in the range of 40∼60$^{\circ}C$ needed for biodegradation and death of pathogenic organisms, but the ammonia emission was different by the ratios of recycled compost. The ammonia emission increased due to the low C/N ratio(17.6), high pH value(8.1) of the recycled compost.

Genome Architecture and Its Roles in Human Copy Number Variation

  • Besides single-nucleotide variants in the human genome, large-scale genomic variants, such as copy number variations (CNVs), are being increasingly discovered as a genetic source of human diversity and the pathogenic factors of diseases. Recent experimental findings have shed light on the links between different genome architectures and CNV mutagenesis. In this review, we summarize various genomic features and discuss their contributions to CNV formation. Genomic repeats, including both low-copy and high-copy repeats, play important roles in CNV instability, which was initially known as DNA recombination events. Furthermore, it has been found that human genomic repeats can also induce DNA replication errors and consequently result in CNV mutations. Some recent studies showed that DNA replication timing, which reflects the high-order information of genomic organization, is involved in human CNV mutations. Our review highlights that genome architecture, from DNA sequence to high-order genomic organization, is an important molecular factor in CNV mutagenesis and human genomic instability.

Antimicrobial Activity of Biosurfactants

  • Biosurfactant has antibiological activities. Due to its low toxicity, biosurfactant can be applied to human health care products. Posseing the antibiological activities as well as the surfactant properties implies that biosurfactant can be widely used in the cosmetic industry. In this presentation, applications of biosurfactant have been reviewed including the antimicrobial activities of sophorolipid. Sophorolipid, a biosurfactant produced from Candida bombicola ATCC 22214, showed antimicrobial activity against Bacillus subtilis, Staphylococcus xylosus, Streptococcus mutans, and Propionibacterium acne at 4, 1, 1, 0.5 ppm as MIC (minimum inhibitory concentration). Also 100 ppm of sophorolipid inhibited 50% of cell growth of plant pathogenic fungus, B. cinera. However, sophorolipid showed no effect on the Escherichia coli., indicating its selective antimicrobial activity depending on the cell wall structure. Treatment of B. subtilis with sophorolipid increased the leakage of intracellular enzyme, malate dehydrogenase, indicating the possible interaction of sophorolipid with cellular membrane. Between lactone-type and acid-type sophorolipid, the former showed higher antimicrobial activity.
