초음파가 잉어 Cyprinus carpio의 성장 및 단기적 행동에 미치는 영향 (Effect of Ultrasound on the Growth and Short-term Behaviour of the Carp, Cyprinus carpio)

  • 본 연구에서는 잉어의 생태 반응에 미치는 초음파(강도 31 dB re 1 ${\mu}Pa$, 주파수 $14{\sim}15$ kHz)의 영향을 구명하기 위하여 치사율, 체중변화, 건강성 평가 및 행동패턴을 관찰하였다. 실험기간 동안 초음파에 노출된 잉어 Cyprinus carpio의 치사율은 대조군을 포함하여 처리군 모두 5% 이하로 실험구 사이의 치사율은 뚜렷하게 구별되지 않았다(P>0.05). 각각의 실험구에서 측정한 실험어류의 평균 체중은 실험종료 시 대조군에 비해 처리군에서 약간 증가하는 경향을 보였으나 실험구간의 뚜렷한 차이는 보이지 않았다. 대조군과 고처리군의 일부 개체들은 가벼운 표피손상과 꼬리지느러미의 출혈 흔적이 관찰되었으나 눈, 지느러미, 기생충 감염, 흉선의 출혈 및 아가미에서는 이상증상이 전혀 관찰되지 않았다(P>0.05). 근거리에서 일시적으로 초음파를 발생시켰을 때 어류는 음원으로부터 회피하는 방향성 움직임을 보였으나 원거리에서 초음파에 노출된 잉어는 뚜렷한 행동변화를 보이지 않았다. 결론적으로 낮은 강도의 초음파에 노출된 어류는 초음파 발생에 따른 심각한 스트레스는 받지 않을 것으로 판단된다.

초음파에 의한 carrageenan 저분자화의 최적 조건 (Optimal Processing Parameters of Low Molecular Weight Carrageenan by Ultrasound)

  • [ $\lambda$ ]-carrageenan 고유의 특성을 유지하면서 용해성을 높인 저분자 $\lambda$-carrageenan을 제조하여 보다 효율적으로 식품산업에 이용하기 위하여 초음파를 이용하여 $\lambda$-carrageenan을 저분자화하는 최적조건을 조사한 결과 $\lambda$-carrageenan의 초음파 처리시 각 처리조건들과 저분자화 효과와의 상관관계를 나타내는 반응표면식을 구한 결과, carrageenan 농도와 초음파 강도가 저분자화에 미치는 주 영향인자인 것으로 나타났다. 그리고 저분자화를 위한 초음파 처리의 최적 조건은 초음파 처리온도 $10^{\circ}C$, 초음파 강도 121.64 $W/cm^2$, 시료 carrageenan 농도 $2\%$, 초음파 처리시간 40분이었다. $\lambda$-carrageenan 및 저분자화된 $\lambda$-carrageenan의 분자량을 조사한 결과 대조구 $\lambda$-carrageenan의 분자량은 약 250,000이었으며 초음파처리에 따른 저분자화 된 $\lambda$-carrageenan의 분자량은 각각 p-1의 경우 약 200,000정도였으며, p-2의 경우 약 60,000정도이었다.

세포 배양 가능한 커버슬립형 초음파 변환자 (Cell-cultivable ultrasonic transducer integrated on glass-coverslip)

  • 초음파 뇌 자극술을 통하여 뇌 심부의 국소 지역에 있는 뇌 세포의 활성화를 유도할 수 있으며, 이를 통하여 저하된 뇌 기능을 치료하는데 효과가 있음이 보고되어 왔다. 반면, 초음파 자극의 종류에 따라 신경 변조의 효율과 방향이 달라질 수 있음이 알려져 있어, 적절한 초음파 자극의 종류를 확립하는 연구가 중요하다. 따라서, 본 논문에서는 이를 효과적으로 최적화 하기 위해 세포 배양시 사용되는 커버슬립 기반의 초음파 변환자를 제안하고자 한다. 균일한 초음파 자극을 전도하기 위해서 폴리머 압전소자(Poly-vinylidene fluoride-trifluorethylene, PVDF-TrFE)를 스핀 코팅하고 패를린 절연층을 상단에 적층시켜 음압 출력을 극대화 시켰다. 개발된 초음파 변환자 융합 커버슬립은 초음파자극기 표면에 배양된 수십개의 신경세포에 균일하고 정확한 초음파 자극을 전달 할 수 있고, 자극에 따른 세포의 반응을 형광 현미경으로 실시간 관찰 가능하다. 따라서, 동일한 초음파 자극에 대한 세포의 반응 신호를 최대 수십개 세포로부터 동시에 획득 가능하므로, 반응 신호를 평균 한다면 낮은 강도의 초음파 자극에 따른 뇌 세포의 미세한 반응을 검출할 수 있을 뿐만 아니라, 초음파 변환자와 물의 표면 등에서 발생하는 정현파에 의한 자극의 왜곡 현상을 줄일 수 있어서 사용자가 원하는 초음파 자극을 정확하게 세포로 전달 가능하다. 이렇게 개발된 초음파 변환자를 통해 변환자 표면에 배양된 별세포에서 6 MHz, 0.2 MPa의 저강도 초음파 자극에 의해 유도된 칼슘 반응을 성공적으로 관찰할 수 있었다.

  • Purpose: Osteoporosis, is a major health problem for the elderly and post-menopausal women and shown to alter the properties of bone as well as impair bone healing around titanium implants in both human and animals. The objective of this study was to examine the effect of LIPUS with adipose-derived stem cells on the healing process around a titanium implant in rats with osteoporosis. Methods: Sixteen osteoporosis-induced rats were divided into two groups: an adipose-derived stem cell injected with Low Intensity Pulsed Ultrasound (LIPUS) application group and a control group. Titanium screw implants (diameter, 2.0 mm: length, 3.5 mm, Cowell Medi, Korea) were placed into both tibia of 16 rats, on 8 rats as the control group and the other 8 rats as the experimental group. Rats were sacrificed at different intervals from 1, 2, 4 and 8 weeks after implantation for histopathologic and immunohistochemical analyses. Results: Histopathological analysis revealed newly formed bone in experimental group earlier than that in control group. Especially at 1 week after implantation, more amounts of new bone matrix and collagen around the implant in the experimental group were seen compared with the control group. Immunohistochemical analysis showed that the levels of osteoprotegerin (OPG) expression in the experimental group were increased at early stages compared with that of control group until 2 weeks after implantation. But after 2 weeks, the expression level of OPG similar in both groups. The expression levels of receptor activator of nuclear factor kB ligand (RANKL) were stronger in the experimental group than the control group until 2 weeks after implantation. After 4 weeks, expression of RANKL in experimental group was similar to the control group. Conclusion: The results of this study suggest that LIPUS with Adipose-Derived Stem Cells in implantation could promote bone healing around titanium implants in rats with osteoporosis.

역학적 강도 분석을 이용한 저강도 초음파의 조사 부위의 골다공증 골절 방지 효과 평가 (In vivo Evaluation of Osteoporotic Fracture Prevention of the site to which low Intensity Ultrasound is Irradiated using Mechanical Strength Simulations)

  • Purpose: The aim of present study is to evaluate a possibility of clinical application for the effect of low intensity ultrasound stimulation (LIUS) in mechanical characteristics of bone on osteoporotic fractures prevention. Materials and Methods: Eight virgin ICR mice (14 weeks old, approximate weight 25g) were ovariectomized (OVX) to induce osteoporosis. The right hind limbs were then stimulated with LIDS (US Group), whereas left hind limbs were not stimulated (CON Group). Both hind limbs of all mice were scanned by in-vivo micro-CT to acquire two-dimensional (2D) images at 0 week before stimulation and 3 weeks and 6 weeks after stimulation. Three-dimensional (3D) finite element (FE) models generated by scanned 2D images were used to determine quantitatively the effect of LIUS on strength related to bone structure. Additionally, distributions of Hounsfield units and elastic moduli, which are related to the bone quality, for the bones in the US and CON groups were determined to analyze quantitatively a degree of improvement of bone qualities achieved by LIUS. Results: The result of FE analysis showed that the structural strength in US Group was significantly increased over time (p<0.05), while that in CON Group was statistically constant over time (p>0.05). High values of Hounsfield units obtained from voxels on micro-CT images and high values of elastic moduli converted from the Hounsfield units were dominantly appeared in US Group compared with those in CON Group. Conclusion: These finding indicated that LIUS would improve the mechanical characteristics of osteoporotic bone via the effects of bone structure (bone strength) and quality (Hounsfield unit and elastic modulus). Therefore, the LIUS may decrease effectively the risk of osteoporotic fracture in clinics.

  • Introduction: Osteoporosis is a major health problem in the elderly that involves changes in the properties of bone as well as impaired bone healing around a titanium implant in both humans and animals. This study examined effect of low intensity pulsed ultrasound (LIPUS) on the bone healing process around a titanium implant in osteoporosis-induced rats. Materials and Methods: Sixteen rats were divided into two groups. A control group with osteoporosis induced by removing both ovaries and an experimental group of rats that were applied with LIPUS after osteoporosis had been induced. A screw type titanium implant (diameter, 2.0 mm: length, 3.5 mm, Cowell-Medi, KOREA) was placed into the tibias of 16 rats. The control and experimental group contained 8 rats each. The rats were sacrificed at 1, 2, 4, and 8 weeks after implantation to examine the histopathology and immunochemistry. Results: The histopathology examination revealed earlier new bone formation in the experimental group than the control group. In particular, at 1 week after implantation, more new bone matrix and collagen were observed around the implant of the experimental group compared to the control group. Immunochemistry analysis showed that the level of OPG expression of the experimental group was higher in the early stages than in the control group. After 8 weeks, the levels of OPG expression were similar in both groups. The expression level of receptor activator of nuclear factor kB ligand (RANKL) was stronger in the experimental group than the control group. After 4 weeks, the level of RANKL expression was similar in both groups. Conclusion: These results suggest that the application of LIPUS to implantation can promote bone healing around titanium in osteoporosis animals.

고강도 집속 초음파에 의한 온도민감성 리포솜으로부터 약물 방출 (Drug Release from Thermosensitive Liposomes by High-Intensity Focused Ultrasound)

  • 종양 조직으로 약물을 효과적으로 전달하기 위하여 리포솜의 개발이 활발이 연구되고 있다. 그러나 리포솜이 종양조직에 효과적으로 축적됨에도 불구하고, 낮은 약물 방출 때문에 리포솜의 치료 효과가 제한적이다. 따라서 우리는 외부 자극에 의하여 약물방출을 최대화 시킬 수 있는 온도민감성 리포솜을 개발하였다. 모델약물인 독소루비신은 pH 전위차 방법에 의하여 리포솜 내부에 봉입하였다. 리포솜의 입자 크기는 $142.0{\pm}6.24nm$ 이었고, 표면전하는 $-10.55{\pm}1.12mV$ 이었다. 온도민감성 리포솜으로부터 약물의 방출은 형광광도계로 측정하였으며 $42^{\circ}C$ 이상에서 5분 이내에 80% 이상의 방출률을 나타냈다. 초음파에 의해 온도민감성 리포솜으로부터 방출된 독소루비신의 세포독성은 초음파를 조사하지 않은 온도민감성 리포솜보다 월등히 우세하였다. 이번 연구에서 우리는 초음파에 의하여 온도민감성 리포솜으로부터 온도에 민감한 약물방출을 증명하였고, 이것은 외부 자극에 의한 종양조직의 약물 농도를 증가시킬 수 있는 암치료에 효과적일 것이다.

맥동성 초음파가 흰쥐 창상치유에 미치는 조직학적 변화 (The Histological Observation of the Effects of Pulsed Ultrasound on Wound Healing of Rats)

  • The purpose of this study was to evaluate the effects of pulsed ultrasound on wound healing and observe during the wound healing process the distribution of mast cells according to histopathologic findings. Eighty Sprague Dawley rats which were divided into 4 groups received full thickness skin wounds on the back. Each of the 5 animals was sacrificed immediately and then sacrificed again 1, 3, 6, and 12 days after injury. Specimens from the wounds were removed during healing and routinely processed with a hematoxylin-eosin stain and a toluidine blue stain. The authors then observed the distribution of mast cells under a light microscope. The results of this study were as follows: The rate of wound healing and the length of the wounds of the pulsed ultrasound group II was significantly faster than group I on day 6 and day 12 (p<.001). Group III showed the most significant effect after12 days (p<.001). Group IV also showed a significant effect at 12 days (p<.01). A low-intensity ultrasound .5 $W/cm^2$ resulted in a fast healing rate. During the wound healing process mast cells had a tendency to decrease in the acute inflammatory phase. During the wound healing process mast cells were thought to contribute to the healing of the wound.

  • The purpose of study was to evaluate effects of low-intensity ultrasound and laser on healing of bone fracture.   • Introduction: This study examined the effect of the application of low intensity pulsed ultrasound on bone healing after an injection of adipose tissue-derived stem cells (ADSCs) during the implantation of a titanium implant in the tibia of diabetes-induced rats. Materials and Methods: Twelve Sprague-Dawely rats were used. After inducing diabetes, the ADSCs were injected into the hole for the implant. Customized screw type implants, 2.0 mm in diameter and 3.5 mm in length, were implanted in both the tibia of the diabetes-induced rats. After implantation, LIPUS was applied with parameters of 3 MHz, 40 mW/$cm^2$, and 10 minutes for 7 days to the left tibiae (experimental group) of the diabetesinduced rats. The right tibiae in each rat were used in the control group. At 1, 2 and 4 week rats were sacrificed, and the bone tissues of both tibia were harvested. The bone tissues of the three rats in each week were used for bone-to-implant contact (BIC) and bone area (BA) analyses and the bone tissues of one rat were used to make sagittal serial sections. Results: In histomorphometric analyses, the BIC in the experimental and control group were respectively, $39.00{\pm}18.17%$ and $42.87{\pm}9.27%$ at 1 week, $43.74{\pm}6.83%$ and $32.27{\pm}6.00%$ at 2 weeks, and $32.62{\pm}11.02%$ and $47.10{\pm}9.77%$ at 4 weeks. The BA in experimental and control group were respectively, $37.28{\pm}3.68%$ and $31.90{\pm}2.84%$ at 1 week, $20.62{\pm}2.47%$ and $15.64{\pm}2.69%$ at 2 weeks, and $11.37{\pm}4.54%$ and $17.69{\pm}8.77%$ at 4 weeks. In immunohistochemistry analyses, Osteoprotegerin expression was strong at 1 and 2 weeks in the experimental group than the control group. Receptor activator of nuclear factor kB ligand expression showed similar staining at each week in the experimental and control group. Conclusion: These results suggest that the application of low intensity pulsed ultrasound after an injection of adipose tissue-derived stem cells during the implantation of titanium implants in the tibia of diabetes-induced rats provided some positive effect on bone regeneration at the early stage after implantation. On the other hand, this method is unable to increase the level of osseointegration and bone regeneration of the implant in an uncontrolled diabetic patient.
